탄소나노튜브를 이용하여 변형한 콜레스테롤 측정을 위한 바이오 센서를 개발하였다. 개발된 콜레스테롤 바이오 센서는 일회용으로 콜레스테롤의 효소반응에서 전자전달을 증진시키기 위하여 탄소잉크로 스크린 프린트된 전극을 탄소나노튜브를 이용하여 변형하였으며, 변형한 탄소나노튜브 전극위에 콜레스테롤 산화효소와 과산화효소, 전자매개 물질로 페로시안 칼륨을 도포하여 제작하였다. 탄소나노튜브를 이용하여 변형한 콜레스테롤 바이오 센서는 전기화학적으로 콜레스테롤 측정시 매우 신속하고 안정된 신호를 나타내었다. 개발된 콜레스테롤 센서는 소량의 (0.5 μL) 시료로 총 콜레스테롤 100~400 mg/dL 영역에서 5초 이내에 직선적인 감응을 나타내었으며 좋은 재현성을 나타내었다(CV 4.0% 이하).
A biosensor with PZT piezoelectric ceramic crystal was developed for the detection of formaldehyde gas. Poled PZT piezoelectric ceramic disk was made from ZrO2, TiO2 and Nb2O5, together with the addition of PbO and polyvinyl alcohol, through various processes of mixing, calcination drying, crushing, forming, sintering, polishing, ion coating and poling. Oscillator circuit of sensor was made of operational amplifier(AD811AN). Formaldehyde dehydrogenase was immobilized onto a piezoelectic ceramic crystal, together with the cofactors, reduced glutathione and nicotinamide adenine dinucleotide. The effect of flow rate on the sensitivity was determined by varing the flow rate of carrier gas from 24.7mL/min to 111.7mL/min through detector cell. The results indicated that as the flow rate was increased, the recovery rate was increased. And a significant increase in the sensitivity was observed in enhanced flow rate of carrier gas. Frequency difference(ΔF) of immobilized PZT piezoelectic disk increased proportionally to the concentration gas and reproduced to repeated exposures of formaldehyde gas(28ppm, Δ68Hz).
This study was carried out to develop enzyme biosensor for lactic acid bacteria. Lactic acids produced by lactic acid bacteria (LAB) was measured and good correlation $R^2=0.98$ between LAB count and lactic acids concentration was found. Hydrogen ion produced by L-lactate dehydrogenase (L-LDH) was measured by a potentiometer. Glutamic-pyruvic transminase (GPT) was used for eliminating inhibitor in the reaction. Polyacrylamide gel was used for immobilizing matrix of the sensor. The biosensor was tested and showed good feasibility with $R^2=0.99$ on validation.
We fabricated glucose oxidase (GOx)-modified biosensor for detection of glucose by physical immobilization of GOx after electrochemical polymerization of the conductive mixture monomers of the 3-thiophenecarboxylic acid (TCA) and thiophene (Th) onto ITO electrode in this study. We confirmed the successfully fabrication of GOx-modified biosensor via FT-IR spectroscopy, SEM, contact angle, and cyclic voltammetry. The fabricated biosensor has the detection limit of $0.1{\mu}M$, the linearity of 0.001-27 mM, and sensitivity of $38.75mAM^{-1}cm^{-2}$, respectively. The fabricated biosensor exhibits high interference effects to dopamine, ascorbic acid, and L-cysteine, respectively. From these results, the fabricated GOx-modified biosensor with long linearity and high sensitivity could be used as glucose sensor in human blood sample.
This study was performed to fabricate a batch-type SPR biosensing system using a thiolated E. coli antibody coupling, and to explore the feasibility of real-time detection of E. coii in a stagnant sample solution. In advance. “O” and “K” antigenic serotype E. coli antibodies were thiolated with sulfo-LC-SPDP and dithiothreitol, and immobilized by chemisorption in the gold surface of compact SPR sensors. When the SPR biosensor immobilized with E. coli antibody monitored a E. coli solution, it took 3 to 5 min to stabilize. The SPR biosensing system developed in this study was able to detect E. coli in the range above 10$^4$ CFU/mL at the 0.05 significant level. Also, the SPR biosensor had possibility to significantly detect E. coli in the range of 10$^2$ to 10$^4$ CFU/mL in E. coli solutions. Meanwhile, when the SPR biosensor immobilized with 5. coli antibody was cleaned with NaOH solutions, its ability to detect E. coli largely decreased due to wash-out of the immobilized antibody. In order to reuse the SPR sensor, it should be antibody-immobilized newly.
Rapid detection of foodborne pathogens has been a major challenge for the food industry. Salmonella contamination is well known in all foods including pasteurised milk. The possibility of specific detection of Salmonella Enteritidis by surface plasmon resonance (SPR) biosensor was explored using a commercially available portable SPR sensor. Self assembly technique was adopted to immobilize anti-Salmonella antibodies on the gold sensing surface of the SPR sensor. The concentration of polyclonal antibody for use in the SPR biosensor was chosen to 1.0 mg/mL. Experiments were conducted at near real-time with results obtained for one SPR biosensor assay within 1 hour. The limit of detection for Salmonella Enteritidis was determined to be $10^6$ CFU/mL in both PBS buffer and milk samples. The assay sensitivity was not significantly affected by milk matrix. Our results showed that it would be possible for employing the SPR biosensor to detect Salmonella Enteritidis in near real-time.
Trace uranium detection measurement was performed using DNA immobilized on a graphite pencil electrode(DGE). The developed probe was connected to the portable handheld voltammetric systems used for seawater analysis. The sensitive voltammogram was obtained within only 30 s accumulation time, and the anodic stripping working range was attained at 100~800 ${\mu}g/l$ U and 10~50 ${\mu}g/l$. The statistic relative standard deviation of 30.0 mg/l with the $15^{th}$ stripping was 0.2115. Here, toxicological and analytical application was performed in the seawater survey in a contaminated power plant controlling water. The results were found to be applicable for real-time toxicological assay for trace control.
본 연구에서는 탄소 유기물과 CoPh를 혼합하여 농약 잔류량을 측정 할 수 있는 바이오센서를 구현 하였다. 작동 전극은 탄소유기물과 CoPh를 섞어 사용하였고 비율은 CoPh를 7%로 제작하였다. CoPh가 7%인 경우 저농도 thiocholine 농도에서도 민감하게 반응하였다. 대표적인 농약인 카보후란에 대하여 센서의 감도 한계는 약$0.5{\mu}g/L$ 수준이며, 농약 농도에 따른 전극의 출력이 선형적인 결과를 얻었다. 또한 EPN 및 클로로피리포스 농약에 대하여 실험 한 결과 카보후란과 같이 농도에 따라 효과적으로 반응하는 것을 확인하였다. 이러한 센서는 현재 농약 잔류량 측정에 사용되는 비색법과 비교하여도 감도 및 선형성 면에서 뒤떨어지지 않으며, 간이 농약 잔류량 측정기 센서로 사용가능성이 있다.
Catechol and caffeine were simultaneously analyzed with a bismuth-immobilized carbon nanotube paste electrode (BPE) using square wave (SW) stripping voltammetry. Optimum analytical conditions were determined. Simultaneous working ranges of 100-1,500 $mgL^{-1}$ for caffeine and 5-75 $mgL^{-1}$ for catechol were obtained. In the separated cell systems, a working range of 0.1-2.1 $mgL^{-1}$ catechol with a correlation coefficient of 0.9935, and a working range of 10-210 $mgL^{-1}$ caffeine with a correlation coefficient of 0.9921 were obtained. A detection limit (S/N) of 0.15 $mgL^{-1}$ (7.7 ${\times}$$10^{-7}$ M) and a detection limit of 0.02 $mgL^{-1}$ (1.82 ${\times}$$10^{-7}$ M), respectively, manifested for catechol and caffeine. It was found that three macro-type electrode systems could be implanted in fish and rat neuro cells. For both ions, the ion currents were observed. The physiological impulse conditions and the neuronal thinking current were also obtained.
Conventional methods for pathogen detection and identification are labor-intensive and take several days to complete. Recently developed biosensors have shown potential for the rapid detection of foodborne pathogens. In this study, an impedimetric biosensor was developed for rapid detection of Salmonella typhimurium. To develop the biosensor, an interdigitated microelectrode (IME) was fabricated by using semiconductor fabrication process. Anti-Salmonella antibodies were immobilized based on either avidin-biotin binding or self assembled monolayer (SAM) on the surface of the IME to form an active sensing layer. To evaluate effect of antibody immobilization methods on sensitivity of the sensor, detection limit of the biosensor was analyzed with Salmonella samples innoculated in phosphate buffered saline (PBS) or food extract. The impedimetric biosensor based on SAM immobilization method produced better detection limit. The biosensor could detect 107 CFU/mL of Salmonella in pork meat extract. This method may provide a simple, rapid, and sensitive method to detect foodborne pathogens.
