회분식과 유가식 배양에 의한 Motierella alpina로부터의 Arachidonic acid의 생산

  • In the batch culture experiments, the addition of $MnSO_4$, was examined in flask culture, and then the optimal amounts of $MnSO_4$ was investigated in 2.5- L jar-fermenter. As a results, 0.005% $MnSO_4$ was found to enhance the ARA yield of 1.14- fold. Also the addition of $KH_2PO_4_4$ was investigated in 2.5-L jar-fermenter and the ARA yield was enhanced 1.20-fold. Fed batch culture study shown a relatively high productivity of cell mass (62.1 g/L) and ARA content (12.0 g/L) when 14% ammonia solution were alternatively used to control the pH and nitrogen source in the cultivation period.

Optimization of Culture Condition for the Hydrocinnamic Acid Production from Bacillus subtilis IJ-31 (Bacillus subtilis IJ-31에서 Hydrocinnamic Acid 생산을 위한 최적배양조건)

  • The metabolites released from cultures of rhizosphere bacteria can inhibit plant growth. Bacillus subtilis IJ-31 inhibited plant growth by the production of hydrocinnamic acid (HCA). The production of HCA by plant-growth inhibiting rhizobacterium B. subtilis IJ-31 was optimized. $90.5\;{\mu}g/ml$ of HCA was obtained under the condition of 1% rice bran as carbon source, 0.5% tryptone as nitrogen source, 0.1% $ZnCl_2$ as metal source at $37^{\circ}C$ for 60 h (pH 7.0). The optimal condition for the HCA production by B. subtilis IJ-31 in the jar fermenter was established using response surface methodology (RSM) of statistical analysis system(SAS) program. The production of HCA by B. subtilis IJ-31 in the jar fermenter culture reached $102.99\;{\mu}g/ml$ when 2.24% soil extracts was added and agitation speed was 290 rpm under the same condition. And the experimental value of HCA production is $102.5\;{\mu}g/ml$ in the same culture condition. The production of HCA by B. subtilis IJ-31 is higher as 12% than that from the flask culture.

  • The Study was carried out to investigate in the optimal mycelial growth and Exo-Polysaccharides of Fomitopsis pinicola. Jar fermentations were carried out to optimize the culture conditions for mycelial growth and exo- polysaccharide production. The optimal agitation speed and aeration rate were 200 rpm and 1.5 v.v.m., respectively. Under optimal culture conditions, the maximum mycelial growth and exo-polysaccharide production after 11 days with a 5 L jar fermenter containing the optimized medium were 10.21 g/L and 3.56 g/L, respectively. However, the fundamental information obtained this study is insufficient in the development of a efficient process for mycelial growth and exe-polysaccharide production from Fomitopsis pinicola.

  • The influence of agitation speed on the yeast growth was investigated in the production of probiotic feed from pulverized liquified food wastes by aerobic fermentation. A yeast Kluyvermyces marxianus was selected through a preliminary screening. The yeast was cultured by 2liter jar fermenter. in 10% solid(w/v) substrate of liquified food waste at $35^{\circ}C$ with each different agitation speed of 500, 900 and 1200 rpm. For the acceleration of enzyme excretion mixed culture with Aspergillus oryzae was also attempted and the results were compared to those of single culture. As results the viable cell number was increased by increasing agitation speed. But it showed highest value in 900rpm and then decreased in 1200rpm. The mixed culture increased amylase activity and growth rate, but did not seem to enhance the highest viable cell count in the final fermentation stage.

  • In order to produce phenylalanine without tyrosine co-production, we constructed various temperature-controllable expression vectors by insertion of lower expression of the tyrA gene into the plasmid pSY130-14. And tyrosine revertant to cultivate without addition of tyrosine, was selected from Escherichia coli strain AT2471[tyrA , thi ] by spontaneous mutation. The strain AT2471 harbouring plasmid pSY146A and the tyrosine revertant 5 harbouring plasmid pSY111-14 produced 12 g/l and 15 g/l of phenylalanine respectively in a 2.5 l jar fermenter at a constant temperature of $39^{\circ}C$ after 55 hours cultivation.

  • 1. Penicillium sp. C 13-13 strain was obtained with the treatment of mutagenic agents(N.T.G.) and by single spore isolation method from the Penicillium sp. C8-14 strain, which was reported in the previous paper. 2. The above strain had a few spores and to obtain seed culture, it was cultured at $30^{\circ}C$ and initial pH $4.5{\sim}5.0$, with air rate 6l/min., and agitation 600 rpm for 48 hours in 10% wheat bran medium in 20l- Jar fermenter. When the broth that had above 70ml of mycelium was inoculated into wheat bran medium and incubated at $29{\sim}33^{\circ}C$ for 72 hrs, the cellulase activity of the koji was higher. 3. Adding calcium chloride and magnecium sulfate to the wheat bran medium to 1.5% and 0.015% respectively, the cellulase activity of the koji was higher than that of the control.

  • For the probiotic feed production from residual food waste, aerobic liquid fermentation was conducted by thermophilic bacteria. 11 Strains of bacteria were isolated from several soil sources and residual food waste. Screening was carried by shaking incubator for the separation of thermophilic strain at $55^{\circ}C$. The isolated strains were tested for enzyme activities such as ${\alpha}$-amylase and protease. 6 Bacterial strains were chosen and were adapted by repeated fermentation processes in food waste substrate. The viable cell count of them at final fermentation stages were shown as $3-7{\times}10^9/ml$ in 2L-jar fermenter. Among them B3, B6 showed higher enzyme activity. By the mixed fermentation of B3, B6 and Bacillus stearothermophilus, the highest viable cell count reached to $1.4{\times}10^{10}/ml$ in 8 hours.

  • For the probiotic feed production from residual food waste by using the yeast Kluyveromyces marxianus, aerobic liquid fermentation was attempted at $35^{\circ}C$. After grinding finely, optimal fermentation conditions of the substrate was investigated in shaking incubator. By controlling water content yeast growth was studied at each different solid content of 5, 10 and 15% respectively. The most active growth of the yeast was shown at 10%. For the stimulation of the cell growth, mixed culture with Aspersillus oryzae was conducted in a 2 litre-jar fermenter. As the results, the yeast growth rate was increased, but the maximum viable cell count amounted was slightly higher as $3.5{\times}10^9/ml$ than single culture.

Isolation and Characteristics of an Amylase-producing Fungus for Saccharifying Food Wastes (음식물쓰레기 당화를 위한 Amylase 생산균의 분리 및 특성조사)

  • In this study, an amylase-producing fungus, strain 15 was isolated from soil in order to saccharify food wastes with cellulolytic and amylolytic enzymes. The amylase production cultures were performed in Mandel's medium with 1% rice straw and 1% paper wastes as carbon sources. The strain produced various cellulolytic (FPase 0.25, xylanase 20.09, CMCase 3.15 U/mL-supernatant) and amylolytic ($\alpha$-amylase 1.20, gluco-amylase 0.70, $\beta$-amylase 2.40 U/mL-supernatant) enzymes in Mandel's medium. In 10 L jar fermenter, maximum amylase and FPase activities, 3.25 and 0.23 U/mL, were obtained when the culture was grown at 30$^{\circ}C$, 200 rpm and 0.6 vvm for 3 days. In 100 mL flask level and 10 L jar fermenter, amylase produced by the strain 15 showed similar cellulolytic and amylolytic enzyme activities with Trichoderma inhamatum KSJ1 isolated from rotten woods by previous researcher. The ability of saccharification to food wastes also showed similar degree. However, the isolate 15 appeared to be yellowish in YMEA plate comparing to Trichoderma inhamatum KSJ1 in greenish.
