As an attempt to preserve resources in marine biological organisms, we first constructed a cDNA library from the red alga Porphyra yezoensis. The library construction method from P. yezoensis consists of three steps; those include protoplast presparation, RNA isolation, and phage library construction. Protoplast was prepared in order to remove much of the carbohydrate compounds which are characteristics of algal cell walls. Carbohydrate contamination in the purified RNA may inhibit further enzyme reactions, those carbohydrates should be removed. RNA samples prepared from protoplast still seemed to contain residual amount of carbohydrate because mRNA isolation with conventional method failed. We therefore developed a method with Poly ATtract mRNA isolation system. The constructed phage library was tested by analyzing cDNA insert in phage vector from randomly picked ten independent white plagues. All of the phages contained cDNA inserts with sizes ranging 0.5kb and 2.0kb.
The Coptis Rhizome is known for containing a number of isoquinoline type alkaloids. Berberine, coptisine and palmatine are the major constituents of alkaloids. The alkaloids were isolated and determined by forming complex from Coptis japonica (Ranunculaceae). For the determination of these alkaloids, a new spectrophotometric method was developed with a simple and selective sample clean-up using thiocyanatocobaltate[II] complex ion. The absorbance of alkaloidal complex in l,2-dichloroethane solution was measured at 625 nm. A calibration curve for the alkaloids isolated from Coptis Rhizome was linear over the concentration range of 0.2-0.3 mg/mι. The method proved to be rapid, simple and reliable for the isolation and the determination of the alkaloids in Coptis Rhizome.
Major equipment of combat vessels normally require the shock and vibration isolation to maintain its performance against environments of severe vibration and shock. In this paper, an optimal layout design method of resilient mounts for shock and vibration isolation is presented using simulated annealing optimization method. The reference levels of vibration and shock to isolate shipboard equipment are determined from the MIL-STD-167-1 and MIL-S-901 specifications. Through the numerical application, the validity of the presented method is investigated.
In this paper we propose a new fault detection and isolation (FDI) method for those faults of parameter change type. First, we design a residual generator based on the ${\delta}$-operator model of the plant by using the stable pseudo inverse system. Second, the parameter change is estimated by using the property of the block Hankel operator. Third, reliability with respect to stability is quantified. Fourth, the limitations for the meaningful diagnosis in our method are given. The numerical examples demonstrate the effectiveness of the proposed method.
In this paper, we propose a FDI method, which comes from singular value decomposition of measurement matrix fur redundant sensors. We analyze the performance of the proposed FDI method by comparing with the GLT method in two ways such as FDI performance and GN&C performance. Also, we propose a GN&C performance index by combining FDI and GN&C performance.

Mitochondria play a crucial role in many essential biological events by way of the electron transport chains and intermembrane proteins that they contain. Abnormalities in the mitochondria are strongly correlated with the development of diseases such as atherosclerosis, cancer, and diabetes. However, the study of mitochondria has been referred to as 'labor-intensive' because of the difficulty in isolating the organelles from their various sources, which can include cultured cells and tissues. Multiple companies provide mitochondria isolation kits, and it is possible for investigators to use different kits and apply different protocols depending on the source of the mitochondria. Therefore, we focused on producing an isolation buffer that could be applied to both cultured cells and tissues, and optimized an isolation protocol that could be used with both. Specifically, we adjusted the buffer condition that can be applied to human cervical cancer cells, fibroblasts, and tissues such as mouse liver and spleen. We also optimized the protocol to improve the efficacy and efficiency of the steps involved in the isolation of mitochondria. These methodological improvements may contribute to advanced research by allowing investigators to overcome the difficulties involved in isolation of mitochondria from biological samples.
In this paper, the method of isolation enhancement for the mobile phone MIMO antenna of LTE class 40(2300~2400MHz) was studied. Design of MIMO antenna was based on the hybrid antenna that operates both a monopole and an IFA(Inverted F Antenna). A structure for the isolation enhancement which controls induced electric field on the ground plane is located between MIMO antenna, and was not connected with the ground but apart 0.3mm. A MIMO antenna that operates on class 3~40(1710~2400MHz) of LTE service bands and a structure for the isolation enhancement at the class 10 band were designed. VSWR measurement of implemented antenna on the FR4 board showed within 3:1 at entire design bands, and isolation between antennas at the class 40 band was less than -30dB. Isolation was enhanced more than 20dB by the studied structure. ECC(Envelope Correlation Coefficient) for MIMO performance was under 0.1, and antenna average gain and efficiency measured in the anechoic chamber were -4.28~-1.40dBi and 37.32~72.36% respectively.
Because of the design and construction requirements, the nuclear structures need to maintain the structural integrity under both design state and extreme earthquake shaking. The base-isolation technology can significantly reduce the damages of structures under extreme earthquake events, and effectively protect the safeties of structures and internal equipment. This study proposes a base-isolation design for the AP1000 nuclear shield building on considering the performance requirements of the seismic isolation systems and devices of shield building. The seismic responses of isolated and nonisolated shield buildings subjected to design basis earthquake (DBE) shaking and beyond-design basis earthquake (BDBE) shaking are analyzed, and three different strategies for controlling the displacements subjected to BDBE shaking are performed. By comparing with nonisolated shield buildings, the floor acceleration spectra of isolated shield buildings, relative displacement, and base shear force are significantly reduced in high-frequency region. The results demonstrate that the base-isolation technology is an effective approach to maintain the structural integrity which subjected to both DBE and BDBE shaking. A displacement control design for isolation layers subjected to BDBE shaking, which adopts fluid dampers for controlling the horizontal displacement of isolation layer is developed. The effectiveness of this simple method is verified through numerical analysis.
This study focused on the effectiveness of seismic isolation technique in case of a reinforced concrete structure with soft story defined as the stiffness irregularity between adjacent stories. In this context, a seismically isolated 3-story reinforced concrete structure was analyzed by gradually increasing the first story height (3.0, 4.5, and 6.0 m). The seismic isolation system of the structure is assumed to be composed of lead rubber bearings (LRB). In the analyses, isolators were modeled by both deteriorating (temperature-dependent analyses) and non-deteriorating (bounding analyses) hysteretic representations. The deterioration in strength of isolator is due to temperature rise in the lead core during cyclic motion. The ground motion pairs used in bi-directional nonlinear dynamic analyses were selected and scaled according to codified procedures. In the analyses, different isolation periods (Tiso) and characteristic strength to weight ratios (Q/W) were considered in order to determine the sensitivity of structural response to the isolator properties. Response quantities under consideration are floor accelerations, and interstory drift ratios. Analyses results are compared for both hysteretic representations of LRBs. Results are also used to assess the significance of the ratio between the horizontal stiffnesses of soft story and isolation system. It is revealed that seismic isolation is a viable method to reduce structural damage in structures with soft story.
We consider some problems of the Modified SPRT(Sequential Probability Ratio Test) method for fault detection and isolation of inertial redundant sensor systems and propose an Advanced SPRT method to solve the problems of the Modified SPRT method. One problem of the Modified SPRT method to apply to inertial sensor system comes from the effect of inertial sensor errors such as misalignment, scale factor error and sensor bias in the parity vector, which make the Modified SPRT method hard to be applicable. The other problem is due to the correlation of parity vector components which may induce false alarm. We use a two-stage Kalman filter to remove effects of the inertial sensor errors and propose the modified parity vector and the controlled parity vector which removes the effect of correlation of parity vector components. The Advanced SPRT method is derived form the modified parity vector and the controlled parity vector. Some simulation results are presented to show the usefulness of the Advanced SPRT method to redundant inertial sensor systems.
