Determination of Isoflavonoid Glucosides in Rhizomes of Belamcanda chinensis by High Performance Liquid Chromatography

  • A new method for separation and quantitative determination of isoflavone glucosides in rhizomes of Belamcanda chinensis (Iridaceae) by high performance liquid chromatography was elaborated. A reverse-phase system with a Spheri-5 RP-18 column using MeOH:HOAc:$H_2O$(24 : 5 : 71) as a mobile phase was developed. The isoflavonoids were detected at 268 nm and the analysis was successfully carried out within 15 min.

  • Heterocyclic compounds with oxygen atoms are known to have potent biological effect. The flavonoids. isoflavonoids. and coumarins which form the bulk of these compounds are very polar and have limited use as drugs which have to pass through membranes. The non-polar property is increased by exchange oxygen to selenium as a part of heterocyclic compound. Our group is focused on synthesizing selenoheterocyclic compound with the above property.   • Flavonoids with oxygen atoms are known to have potent biological effect.They have been studied long as major antioxidants which protect cell membranes. Recent medical surveys show that increased intake of selenium decreases the risk of breast, colon, lung and prostrate cancer by preventing free radical generation. The flavonoids, isoflavonoids, and coumarins which form the bulk of these compounds are very polar and have limited use as drugs which have to pass through BBB(Brain Blood Barrier)The non-polar property is increased by exchange oxygen to selenium as a part of heterocyclic compound. Anti-Inflammatory Mode of Isoflavone Glycoside Sophoricoside by Inhibition of Interleukin-6 and Cyclooxygenase-2 in Inflammatory Response

  • Soy, high dietary intake for the oriental population, is a main source of isoflavonoids. Sophoricoside (SOP) an isoflavone glycoside was isolated from immature fruits of Sophora japonica (Leguminosae family) and its inhibitory effect on chemical mediators involved in inflammatory response was investigated in this study. SOP inhibited the interleukin (IL)-6 bioactivity with an $IC_{50}$ value of 6.1 $\mu$M whereas it had no effects on IL-1$\beta$ and TNF-a bioactivities. SOP was identified as a selective inhibitor of cyclooxygenase (COX)-2 activity with an $IC_{50}$ value of 4.4 $\mu$ M, but did not show inhibitory effect on the synthesis of COX-2. However, SOP had no effect on the production of reactive oxygen species including superoxide anions and nitric oxide. These results revealed that in vitro anti-inflammatory action of SOP is significantly different from that of genistein known as a phytoestrogen of soy products. This experimental study has documented an importance of dietary soy isoflavonoids as multifunctional agents beneficial to human health, and will help to clarify protective mechanisms of SOP against inflammatory conditions.

수목추출물의 생리활성에 관한 연구(VII) - 회화나무 Isoflavonoid의 항균활성 - (Studies on Biological Activity of Wood Extractives(VII) - Antifungal activity of Isoflavonoids from Sophora japonica -)

  • 항균활성이 우수한 회화나무 목부로부터 4종의 isoflavonoid 화합물을 단리하였으며, 기기분석 결과 3종의 isoflavone인 5,7-dihydroxy-4',6-dimethoxyisoflavone(irisolidone)을 비롯하여, 5,7-dihydroxy-4'-methoxyisoflavone(Biochanin A), 7-hydroxy-4'-methoxyisoflavone(formononetin) 그리고 1종의 isoflavanone인 7-hydroxy-4'-methoxyisoflavanone으로 각각 동정하였다. 이들 단리 물질의 항균활성을 배지점적법으로 조사한 결과 4종의 isoflavonoid 공히 낮은 항균활성을 나타내어, 회화나무 목부의 높은 항균활성은 본 실험에서 단리한 물질 이외의 물질이 관여하는 것으로 추정되며 금후 관련물질의 탐색이 필요할 것으로 사료되었다.

Isoflavonoids and Alkaloids from Spartidium saharae

  • A new isoflavone, $(+)-4'-O-methyl-8-C-{\beta}-D-glucopyranosylgenistein$, was isolated from the aerial parts of Spartidium saharae together with the known isoflavone, $(+)-8-C-{\beta}-D-glucopyranosylgenistein$ as well as dipiperidine alkaloids, (+)-ammodendrine and (+)-N-acetyl hystrine. Details of their structure elucidation are based on chemical and spectroscopic methods. N-formyl ammodendrine was detected by GC-MS. The potential chemotaxonomic value of the alkaloid content is explored. Cytotoxic activity has been determined for both alcoholic extract and isolated compounds.

Manipulating Isoflavone Levels in Plants

  • Metabolic engineering for production of isoflavones in nonlegume plants could distribute the health benefits of these phytoestrogens in more widely-consumed grains. Series of investigation to check the ability of the heterologous isoflavone synthase enzyme to interact with the endogenous phenylpropanoid pathway have been conducted. Overall, results provide possibility of production of isoflavonoids in several plant tissue systems including soybean and nonlegumes. In tissue that undergoes naturally enhanced synthesis of anthocyanins, genistein production was enhanced. In a monocot cell system, introduced expression of a transcription factor regulating genes of the anthocyanin pathway was effective in conferring the ability to produce genistein in the presence of the isoflavone synthase gene. However, in this case the intermediate accumulated to high levels indicating an inefficiency in its conversion. Introduction of a third gene, chalcone reductase, provided the ability to synthesize an additional substrate of isoflavone synthase resulting in production of the isoflavone daidzein. These research efforts provide insight into requirements for metabolic engineering for isoflavone production in nonlegume dicot and monocot tissues.

  • Two isoflavonoids were isolated from the flowers of Pueraria lobata (Flos puerariae) guided initially fractionation based on brine shrimp lethality assay. The structures were identified as 4',5,7-trihydroxy-6-methoxyisoflavone (tectorigenin) and 4',7-dihydroxy-6-methoxyisoflavone (glycitein), respectively, on the basis of their spectroscopic and physicochemical evidences.
