Iridoviridae과에 속하는 lymphocystis disease virus(LDV)는 비교적 큰 20면체 DNA 바이러스이며(Murphy et al., 1995), 림포시스티스병(lymphocystis disease, LD)을 유발하는 원인체로서 140여종 이상의 해수 및 담수 어종에서 보고된 바 있다(Wolf et al., 1966). 우리 나라의 경우, 주요 양식 어종인 넙치에 주로 감염되어 2차 감염과 빈혈 및 합병증 등에 의해 폐사를 유발할 뿐만 아니라 상품성을 저하시킴으로서 경제적 손실을 야기한다. (중략)

참돔 이리도바이러스 질병(red seabream iridovirus disease ; RSIVD)은 1990년 일본 시코쿠지역의 참돔 양식장에서 처음 발병된 후, 매년 발병지역이 확산되고 발병 어종이 다양해지고 있다. 참돔에서 분리된 RSIV는 icosahedral cytoplasmic deoxyribovirus로서 크기가 200∼240nm이며 형태학적 특징에 의해 iridoviridae로. 분류하고 있지만, 어류를 숙주로 하는 iridovirus과 중에서 lymphocystis virus속인 flounder virus(LCDV-1) 및 lymphocystis disease virus(LCDV-2)와 goldfish virus 1-lke virus속인 goldfish virus 1(GFV-1) 및 goldfish virus 2(GFV-2)와는 전혀 다른 바이러스로 알려져 있다. Ranaviruses are a primary cause of amphibian extinctions. More consistent ranavirus-infection reports and genetic characterizations of identified viruses are urgently needed, particularly from Asian countries. The objectives of this study were to obtain the partial major capsid protein (MCP) gene sequences (506 bp) of the ranavirus responsible for infecting frogs in South Korea, as our previous research had confirmed using qPCR, and to evaluate their genetic relationships with other previously reported ranavirus sequences. Three different ranavirus MCP sequences were obtained from Pelophylax nigromaculatus and Lithobates catesbeianus. All six different types of MCP sequence from the ranavirus identified in South Korea to date belonged to the Frog virus 3 (FV3)-like virus group in the genus Ranavirus. To better understand the origin and spread of ranaviruses in South Korea, further infection reports and full genome analyses of the identified ranaviruses are needed.
Background: Several species of amphibians in agricultural areas are often infected with ranaviruses; however, the biological or ecological factors that cause this infection are not well understood. In this study, we investigated whether local tadpole density, Gosner developmental stage, and weather conditions affected ranavirus infection in Dryophytes japonicus tadpoles in rice paddies over three months. Results: During the study, eight samplings were undertaken between June 6 and August 21, 2022. No die-off of tadpoles occurred, but 20 of 110 tadpoles (18.8%) were found to be infected with ranavirus. The tadpole density at the sampling site and Gosner stage of the sampled tadpoles were not related to the daily ranavirus infection rate. The mean daily highest temperature during the two weeks prior to the sampling date and the mean daily lowest and highest temperatures during the week prior to the sampling date were negatively related to the daily infection rate. Conclusions: Our results suggest that low and extreme temperatures caused by flooding and draining of paddy fields or climate change in summer could be a significant risk factor for ranavirus infection in summer-breeding frogs in agricultural areas.

The high mortality of cultured juvenile turbot, Scophthalmus maximus occurred in Gochang on June, 2003. The diseased fish was lethargic with reduced feed intake. Grossly, these fish showed pale body, abdominal extension and exophthalmia. The dominant internal gross features of diseased fish were severely enlarged spleen, pale gills and/ or liver. Diseased fish histologically showed basophilic enlarged cells in the kidney, spleen, gill, heart, stomach, intestine, liver, pancreas and adipose tissue. Transmission electron microscopy (TEM) reveled hexagonal virions in the cytoplasm of necrotic cells. The viral particles lead a central electron-dense core and an electron translucent zone, and were 136-159 nm in diameter. These results suggest that the virus belonging to the iridoviridae was responsible for the mortality of cultured juvenile turbot.

The red seabream iridovirus (RSIV), which belongs to the iridoviridae, causes infectious fish diseases in many Asian countries, leading to considerable economic losses to the aquaculture industry. Using the yeast surface display (YSD) technique, a new experimental system was recently developed for the detection and identification of a variety of marine viruses. In this study, a coat protein gene of RSIV was synthesized based on the nucleotide sequence database and subcloned into the yeast expression vector, pCTCON2. The expression of viral coat proteins in the yeast strain, EBY100, was detected by flow cytometry and Western blot analysis. Finally, they were isolated from the yeast surface through a treatment with ${\beta}$-mercaptoethanol. The data suggests that the YSD system can be a useful method for acquiring coating proteins of marine viruses.

Monitoring for megalocytivirus infection was conducted for ten months from March to December in 2013 in 15 aquatic farms culturing, red sea bream, rock bream, rock fish and black sea bream around Tongyoung coastal area in Korea, to assess spatial and temporal variability of detection prevalence, and to explore possible links with seawater temperature. In nested-PCR targeted major capsid protein (MCP) gene, asymptomatic megalocytivirus infection was detected in the externally healthy farmed fish with a significant prevalence in range from 0 to 58.3% for ten months. Higher prevalence of megalocytivirus (46.7% - 57.1%) was observed in high water temperature season from September to November than that in other months with lower prevalence of 0.0% to 20.0%. Even though an acute infection of megalocytivirus was occurred in rock bream (positive in the first PCR) with high mortality in one of fifteen farms, there was no expansion or transmission of the disease to the rock fish and red sea bream culturing in net cage just proximal to the rock bream cage in which disease outbreaked. Nucleotide sequence analysis of the cloned MCP gene isolated asymptomatically infected rock fish revealed that the megalocytivirus in this study was clustered together with the rock bream iridovirus (RBIV) under the subgroup II of the genus megalocytivirus (Iridoviridae), which is known to be the major megalocytivirus strain in Korea. The typical histopathological signs were not found in the spleen of rock fish asymptomatically infected by megalocytivirus. Experimental infection of rock bream with the spleen homogenate of the rock fish infected asymptomatically did not induce any mortality unlike the homogenate of infected rock bream with hih mortlity. However, these results may suggest that the asymptomatic infection of megalocytivirus in other fish species can be a potential risk threatening aquaculture industries as a transmission factor of megalocytivirus to susceptible fish species, especially rock bream.
