A Comprehensive review of raisins and raisin components and their relationship to human health

  • Purpose: This literature review was performed to assess the effect of raisins on human health. Methods: A review of Medline was conducted using the keywords: 'raisins, raisins and health, raisins and cardiovascular disease (CVD), raisins and cancer, raisins and diabetes, raisins and fiber, raisins and colon health, raisins and antioxidants, raisins and inflammation, raisins and dental caries'. The reference lists from previous review articles on raisins and human health and the California Raisin Marketing Board files were reviewed for additional studies. Results: Raisins have one of the highest polyphenolic content and antioxidant ORAC levels compared to other traditional dried fruits. Many of the polyphenols in raisins are well assimilated and bioavailable. Raisin consumption reduces low density lipoprotein (LDL) cholesterol, blood pressure and blood sugar, when compared to equal caloric carbohydrate snacks and is associated with a reduced risk of CVD. The anti-inflammatory and cancer chemopreventive effects of raisins are mixed. Raisin consumption reduces intestinal transit time and positively affects gut microbiota. Raisins produce sustained energy during long term athletic competitions equal to traditional sports energy gels, shots and jelly beans. Raisins produce a non-cariogenic oral environment and do not fit the American Academy of Pediatrics criteria to be considered a choking hazard. Conclusions: Based on the review of literature, consumption of raisins provide numerous health benefits for promoting general wellness and in the prevention of many chronic diseases including: CVD, type 2 diabetes mellitus (T2DM) gastrointestinal diseases, and dental caries.

김치 및 김치 유래 유산균의 건강 기능성에 대한 연구 동향 조사 (A survey of research papers on the health benefits of kimchi and kimchi lactic acid bacteria)

  • Purpose: This review article provides an overview of the trends of research papers on the health benefits of kimchi and kimchi lactic acid bacteria published from 1995 to 2017. Methods: All publications from 1995 to 2017 regarding kimchi and kimchi lactic acid bacteria were collected, reviewed, and classified. This review article covers the publications of the health benefits of kimchi and kimchi lactic acid bacteria on experimental, clinical trials, and epidemiology studies. Results: The number of publications on kimchi over the period were 590: 385 publications in Korean and 205 publications in English. The number of publications on the health benefits of kimchi and kimchi lactic acid bacteria were 95 in Korean and 54 in English. The number of publications on kimchi and kimchi lactic acid bacteria were 84 and 38, respectively, in the experimental models. Ten research papers on kimchi in clinical trials and 7 publications in epidemiology were found. Kimchi or kimchi lactic acid bacteria had protective effects against oxidative stress, mutagenicity, toxicity, cancer, dyslipidemia, hypertension, immunity, and inflammation in in vitro, cellular, and in vivo animal models. Moreover, kimchi had effects on the serum lipids, intestinal microbiota, iron status, obesity, and metabolic parameters in human clinical trials. In epidemiology, kimchi had effects on hypertension, asthma, atopic dermatitis, rhinitis, cholesterol levels, and free radicals. Conclusion: This review focused on the publications regarding the health benefits of kimchi and kimchi lactic acid bacteria, suggesting the future directions of studies about kimchi and kimchi lactic acid bacteria by producing a database for an evaluation of the health benefits of kimchi.

Effects of Saccharomyces cerevisiae and phytase co-fermentation of wheat bran on growth, antioxidation, immunity and intestinal morphology in broilers

  • Objective: The aim of this study was to investigate the effects of different amounts of wheat bran (WB) inclusion and postbiotics form by Saccharomyces cerevisiae and phytase co-fermented wheat bran (FWB) on the growth performance and health status of broilers. Methods: Study randomly allocated a total of 300 male broilers to a control and 4 treatment groups (5% WB, 5% FWB, 10% WB, and 10% FWB inclusion, respectively) with each pen having 20 broilers and 3 pens per treatment. Results: The WB does not contain enzymes, but there are 152.8, 549.2, 289.5, and 147.1 U/g dry matter xylanase, protease, cellulase and β-glucanase in FWB, respectively. Furthermore, FWB can decrease nitric oxide release of lipopolysaccharide stimulated chicken peripheral blood mononuclear cells by about two times. Results show that 10% FWB inclusion had significantly the highest weight gain (WG) at 1 to 21 d; 5% FWB had the lowest feed conversion rate at 22 to 35 d; 10% WB and 10% FWB inclusion have the highest villus height and Lactobacillus spp. number in caecum; and both 5% and 10% FWB can increase ash content in femurs. Compared to control group, all treatments increase mucin 2, and tight junction (TJ), such as occludin, claudin-1, zonula occludens-1, and mRNA expression in ileum by at least 5 folds. In chicken peripheral blood mononuclear cells, nicotinamide adenine dinucleotide phosphate-oxidase-1 mRNA expression decreases from 2 to 5 times, and glutamate-cysteine ligase catalytic subunit mRNA expression also increases in all treatment groups compared to control group. The mRNA expression of pro-inflammatory cytokines, including interleukin-6 (IL-6), nuclear factor-κB, and IL-1β, decreases in 5% and 10% FWB groups compared to control group. Conclusion: To summarize, both WB and FWB inclusion in broilers diets increase TJ mRNA expression and anti-oxidation and anti-inflammation, but up to 10% FWB groups have better WG in different stages of broiler development.

  • Objectives : Ulcerative colitis (UC) is an inflammatory bowel disease, which is one of chronic gastrointestinal disorders. Orostachys japonicus (OJ) has been used as a traditional medicine for various diseases including gastric cancer, gastric ulcers and intoxication. However, the regulatory effect of OJ on intestinal inflammation has not been fully understood, yet. The aim of this study was to investigate the effect of OJ on dextran sulfate sodium (DSS)-induced colitis in mice. Methods : To ascertain the pharmacological effects of OJ, the colitis mice were induced by drinking water containing 5% DSS for 7 days. Mice were randomized into groups receiving OJ (100 mg/kg), sulfasalazine (150 mg/kg) as a positive control, or water as a negative control. We evaluated the effects of OJ on DSS-induced the clinical signs, measuring weight loss and colon length. In addition, the inhibitory effect of OJ on the tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) level was determined by enzyme-linked immunosorbent assay in colitis tissue. Results : The results indicated that mice treated with DSS showed remarkable clinical signs, including weight loss, and reduced colon length. However, treatment with OJ significantly improved the weight loss and DAI as clinical symptoms. Moreover, OJ reduced the TNF-${\alpha}$ levels in DSS-treated colon tissues. Conclusions : Collectively, the findings of this study provide us with novel insights into the pharmacological actions of OJ as a potential medicine for use in the treatment of ulcerative colitis.

Effects of natural mono- and di-saccharide as alternative sweeteners on inflammatory bowel disease: a narrative review

  • Objectives: The incidence of inflammatory bowel disease (IBD) is increasing globally, and excessive added sugar consumption has been identified as one of the contributing factors. In the context of IBD, it is essential to explore functional sweeteners that can improve metabolic health and minimize the risk of IBD-related symptoms. This review article aims to shed light on the effects of natural mono- and di-saccharides as alternative sweeteners, specifically focusing on potential benefits for IBD. Methods: A comprehensive literature review was performed using PubMed and Google Scholar databases with articles published after the year 2000. The search terms 'IBD', 'added sugar', 'sweeteners', 'mono-saccharide', and 'di-saccharide' were combined to retrieve relevant articles. A total of 21 manuscripts, aligning with the objectives of the study, were selected. Papers focusing on artificial or high-intensity sweeteners were excluded to ensure relevant literature selection. Results: Multiple studies have emphasized the association between the high consumption of added sugars such as simple sugars and the increased risk of developing IBD. This is suggested to be attributed to the induction of pro-inflammatory cytokine productions and dysbiosis of the gut microbiota. Consequently, there is a growing demand for safe and functional sweeteners, in particular mono- and di-saccharides, that can serve as alternatives for IBD patients. Those functional sweeteners regulate inflammation, oxidative stress, and Intestinal barrier protection, and restore microbiome profiles in various IBD models including cells, animals, and humans. Conclusions: Understanding these mechanisms resolves the link between how sugar consumption and IBD, and highlights the beneficial effects of natural alternative sweeteners on IBD when they were administered by itself or as a replacement for simple sugar. Further, exploration of this relationship leads us to recognize the necessity of natural alternative sweeteners in dietary planning. This knowledge could potentially lead to more effective dietary strategies for individuals with IBD.

AOM/DDS로 대장암 유도 후 식용버섯균사체 유래 다당류의 대장암 억제효과 및 대장 미생물균총의 변화 (Anti-cancer Effects and Changes in Colonic Microflora of Polysaccharide Derived from Edible Mushroom Mycelium on AOM/DDS-induced Colon Cancer Model)

  • The objective of this study was to investigate the anticancer effects of EMPS (edible mushroom mycelium polysaccharide: Tremella fuciformis) in animal models with colorectal cancer induced by AOM/DSS. The experimental groups consisted of Nor (normal), NC (AOM/DSS), EMPS (EMPS 50, EMPS 100), and PC (Fluorouracil). The NC group had the highest number of colon tumors, whereas it was observed that tumor occurrence was significantly reduced in the EMPS consumption group. The expression of Bcl-2, an apoptosis inhibitor, was significantly lower in the EMPS 50 & 100 and PC groups. On the other hand, the mRNA gene expression of Bax, a factor that induces apoptosis, was significantly higher in the EMPS 50 & 100 and PC groups compared to the NC group. The mRNA expression levels of TNF-α and COX-2 significantly increased in the NC group, but showed a significant decrease in the EMPS and PC groups, indicating inhibition of the cancer-promoting response of cells. At the phylum level of the mice's intestinal microbial composition, the proportion of Bacteroidetes tended to decrease, while the proportion of Firmicutes tended to increase with EMPS administration. This suggests that changes in the gut microbiota caused by inflammation can be influenced by dietary intake.

염증성 장질환 모델 및 크론병 환자에서의 점막상피 HuR 단백질의 변화 분석 (Tissue Distribution of HuR Protein in Crohn's Disease and IBD Experimental Model)

  • 염증성 장질환은 점막의 만성적 궤양과 염증을 동반하는 면역질환으로 알려져 있으며, 특히 TNF${\alpha}$와 같은 염증성 사이토카인은 주요한 생물학적 치료의 표적으로 이용되고 있다. 염증성 사이토카인의 유전자발현에서 전사물의 안정화는 매우 중요한 조절과정이며, 특히 본 연구에서는 이 안정화에 핵심적인 단백질인 HuR의 발현과 조직 분포에 대하여 동물모델과 환자의 조직에서 분석하였다. DSS를 처리함으로 유도되는 장염증 동물 모델에서 HuR 단백질의 발현량이 높았음을 확인했고, 점막의 상피조직 및 선조직 상피세포에서 상대적인 발현이 증대되었다. 또한 단백질의 활성측면에서 세포질로 이동된 HuR 단백질의 양도 상대적으로 증가하였다. 공간분포적으로 보면 DSS에 의한 화학적 점막자극에 의하여 초기에는 villi 하부에서의 발현정도가 상대적으로 villus 말단에 비하여 높게 유지되었다. 크론병 환자의 생검을 통하여 정상부위와 병변부위에서 HuR 단백질을 비교분석 하였다. 크론병 환자들의 병변에서는 지속적으로 HuR의 발현이 증대되어 있음을 확인했으며, 동물조직과 유사하게 병변부위의 장관상피세포 및 선 상피에서 주로 발현양이 높았다. 이러한 결과는 염증성 장질환에서의 HuR 단백질이 초기 염증성 인자의 발현에 중요한 역할이 예상되며, 구체적인 분자기전의 규명도 향후 기대된다. 이를 근간으로 하여 염증성 장질환의 진단과 치료의 표적개발에서 유용하게 응용하고자 한다.

소아 Helicobacter pylori 감염에서 위 상피세포의 증식과 세포사 (Gastric Epithelial Cell Proliferation and Apoptosis in Children with Helicobacter pylori Infection)

  • 목적: 소아기는 알코올이나 약물 등에 의한 자극이 거의 없는 시기로 H. pylori 감염의 자연 경과와 단기간의 영향을 연구하기에 적합한 시기이다. 최근 H. pylori 감염의 기전으로 중요시되고 있는 위상피세포 증식과 세포사에 대해 소아에서 알아보고자 하였다. 방법: 1996년 5월부터 2001년 6월까지 이화여자대학교 목동병원 소아과에서 소화기 증상으로 내시경을 시행하여 H. pylori 감염으로 진단된 58예와 감염 음성 40예를 대상으로 하였다. H. pylori 감염 양성은 조직학적으로 H. pylori 균이 관찰되고, CLO 검사와 ureC PCR이 전부 양성인 경우로 하였다. 위생검 조직에서 개정된 시드니 체계를 이용하여 조직 소견을 분석하고, proliferating cell nuclear antigen (PCNA) 발현으로 위 상피세포 증식의 정도를, in situ terminal deoxynucleotidyl transferasemediated dUTP nick-end labeling (TUNEL) 방법으로 세포사의 정도를 조사하였다. 결과: 1) H. pylori 감염 양성에서 다핵형 중성구의 활동성(P=0.000), 만성 염증(P=0.000), 상피손상(P=0.000), 림프여포(P=0.000)의 정도가 감염 음성에 비하여 유의하게 높았다. H. pylori 감염 양성에서 장형화생은 관찰되지 않았다. 2) H. pylori 감염 양성에서 세포 증식 지표는 $67.8{\pm}18.13$으로, 음성 $54.8{\pm}14.46$에 비하여 유의하게 높았다(P=0.000). 세포 증식 지표는 H. pylori 밀도가 증가할수록(r=0.277, P=0.007), 다핵형 중성구의 활동성이 증가할수 (r=0.280, P=0.007), 만성염증이 증가할수록(r=0.284, P=0.006) 증가하였다. 3) 세포사 지표는 H. pylori 감염 양성에서 $0.44{\pm}0.447$, 음성에서 $0.14{\pm}0.196$으로 감염 양성에서 음성보다 유의하게 높았다(P=0.000). 세포사 지표는 H. pylori 밀도가 증가할수록(r=0.472, P=0.000), 다핵형 중성구의 활동성이 증가할수록(r=0.370, P=0.001), 만성 염증이 증가할수록(r=0.483, P=0.000) 증가하였다. 4) 세포 증식 지표가 증가할수록 세포사 지표는 유의하게 증가하였다(r=0.353, P=0.003). 결론: H. pylori 감염 소아에서 세포 증식 지표와 세포사 지표가 유의하게 증가하였으며 상관성도 유의하였다. 이는 소아에서 위 상피세포 증식과 세포사가 H. pylori의 병인에 중요함을 시사하며, 앞으로 세포 증식과 세포사의 기전, 유발 요인 외에 다른 병독 인자와의 관련성에 대한 연구가 필요하리라 생각된다.

코팅된 젖산세균 분말의 저장 안정성에 미치는 효과 (Effect of Coating Materials on the Stability of Spray-Dried Lactobacillus Powder during Storage)

  • 본 연구에서는 분무건조법을 이용하여 젖산세균을 녹말로 캡슐화한 분말을 제조하고, 제조한 젖산세균 코팅 분말을 이용하여 다양한 수분활성도에 따른 평형수분함량을 측정하여 등온흡습곡선을 확립하고, pH 및 상대습도, 온도에 따른 젖산세균의 보호 효과를 알아보고자 하였다. 젖산세균 코팅 분말의 등온흡습곡선은 수분활성도에 따라 평형수분함량이 빠르게 증가하는 sigmoid형을 나타내었으나 시료간의 유의적인 차이를 나타내지 않았다. 상대습도에 따라 20, 30, $40^{\circ}C$에서 저장하였을 때, 상대습도가 낮을수록, 저장 온도가 낮을수록 젖산세균의 생존율이 높았으며, 시료들 중 생강을 첨가하여 코팅한 젖산세균 분말의 생존율이 가장 높았다. 75%의 상대습도에서 $20^{\circ}C$$35^{\circ}C$에서 각 4주간 저장 실험한 결과, 코팅하지 않은 젖산세균은 1주 후 사멸한 반면, 코팅한 젖산세균은 유지되었으며 특히 생강을 첨가하였을 경우 젖산세균 보호 효과가 가장 높았다. 소화 모방 실험을 통해 위액의 낮은 pH에서 녹말 코팅의 보호막 효과를 확인하였다. 이를 통해 녹말로 코팅한 젖산세균 분말은 식품 조성물, 식품 첨가용 조성물로 이용 가능하다. 한편, 프로바이오틱스 제품 제조 시 젖산세균이 8-10 log CFU/g 이상 존재해야 하는데, 본 연구에서는 분무건조 후 약 7-8 log CFU/g로 다소 감소하였지만 이는 사료 조성물 또는 사료 첨가용 조성물로 응용이 가능하다고 생각된다.

A Bacterial Metabolite, Compound K, Induces Programmed Necrosis in MCF-7 Cells via GSK3β

  • Ginsenosides, the major active component of ginseng, are traditionally used to treat various diseases, including cancer, inflammation, and obesity. Among these, compound K (CK), an intestinal bacterial metabolite of the ginsenosides Rb1, Rb2, and Rc from Bacteroides JY-6, is reported to inhibit cancer cell growth by inducing cell-cycle arrest or cell death, including apoptosis and necrosis. However, the precise effect of CK on breast cancer cells remains unclear. MCF-7 cells were treated with CK ($0-70{\mu}M$) for 24 or 48 h. Cell proliferation and death were evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and flow cytometry assays, respectively. Changes in downstream signaling molecules involved in cell death, including glycogen synthase kinase $3\beta$ ($GSK3\beta$), $GSK3\beta$, $\beta$-catenin, and cyclin D1, were analyzed by western blot assay. To block $GSK3\beta$ signaling, MCF-7 cells were pretreated with $GSK3\beta$ inhibitors 1 h prior to CK treatment. Cell death and the expression of $\beta$-catenin and cyclin D1 were then examined. CK dose- and time-dependently inhibited MCF-7 cell proliferation. Interestingly, CK induced programmed necrosis, but not apoptosis, via the $GSK3\beta$ signaling pathway in MCF-7 cells. CK inhibited $GSK3\beta$ phosphorylation, thereby suppressing the expression of $\beta$-catenin and cyclin D1. Our results suggest that CK induces programmed necrosis in MCF-7 breast cancer cells via the $GSK3\beta$ signaling pathway.
