  • IL-1RAcP는 일명 interleukin-1 receptor accessory protein이라 칭하며 interleukin-1 염증성 사이토카인과 interleukin-1 receptor I (IL-1RI) 결합체와 복합체를 형성하여 작용한다. IL-1RAcP는 면역반응, 스트레스 및 세포사멸과 관련이 있다. 본 연구의 목적은 참돔(Pagrus major)을 저수온(8℃, 33 psu) 및 저염분(20℃, 10 psu) 상태에 노출시킨 후, IL-1RAcP 유전자의 발현을 관찰하는 것이다. 연구결과, IL-1RAcP 유전자의 발현은 저수온(8℃, 33 psu) 및 저염분(20℃, 10 psu) 상태에서 유의적으로 증가하는 것으로 나타났다. 이 연구결과로서 IL-1RAcP 유전자는 저수온 및 저염분 등의 환경 스트레스에 대한 생체지표유전자로서 역할을 한다고 제의한다.

  • Inflammatory bowel diseases(IBD), including Crohn's disease and ulcerative colitis, are chronic inflammatory states of the intestinal tract. While the exact mechanisms inducing chronic inflammation are still unclear, it is hypothesized that the inflammation is caused in part by an inappropriate immune response to the intestinal microflora. Although inflammatory diseases are not directly linked to patient survival, symptoms of these diseases significantly decrease quality of life. The incidence rate is higher in western people than eastern people, but the incidence rate of IBD in eastern people, including Korean, is increasing. Recently, it has been reported that IL-17 is an important factor that appears to be involved in IBD induction and progression. This report reviews many recent papers reporting the relationship between IBD and IL-17, which may provide an understanding leading to new means of prevention and treatment for IBD.

Helicobacter pylori에 의한 세포독성 및 Interleukin-8 생성에 미치는 생약혼합물의 영향 (Inhibitory Effect of Medicinal Plant Extract on Cell Toxicity and Interleukin-8 Production Induced by Helicobacter pylori)

  • The effects of Helicobacter pylori and medicinal plants extract (Leweifang) on the viability and interleukin(IL)-8 production of gastric epithelial cell were investigated. Cells viability was significantly decreased when they incubated with H. pylori or H. pylori toxin. Co-incubation with Leweifang increased H. pylori or H. pylori toxin-inhibited cell growth in a concentration-dependent manner. The production of IL-8 was greatly increased in H. pylori-infected KATO III gastric epithelial cells in a concentration- and time-dependent manner. The increased production of IL-8 was significantly inhibited by Leweifang $(1,000{\sim}5,000{\mu}g/ml)$. These results indicate that Leweifang has protective effect on H. pylori-inhibited cell growth and H. pylori-induced gastric mucosal cell inflammation by suppressing the production of inflammatory cytokine (IL-8) from gastric epithelial cells.

나노 에멀젼이 잇몸 염증의 예방 및 치료에 미치는 효과에 대한 연구 (THE STUDY ON THE EFFECT OF NANOEMULSION FOR THE PREVENTION AND TREATMENT OF GINGIVAL INFLAMMATION)

  • We have found out the relationship of nanoemulsion containing nano vitamin C, E and propolis and gingival disease. We've confirmed effect of nanoemulsion through the experiment of in vivo and in vitro. We tested cell viability of gingival fibroblast cells by MTT assay and mRNA appearance of interleukin-$1{\beta}$, using mouse that was guided inflammation. Anti-microbacterial activity for Antibacterial effect's experiment was carried out by using S.aureus and E.coli. In addition, inflammation tissue has been observed with scanning electrical microscopy. In this study, expression of interleukin-$1{\beta}$ was decreased after adding nanoemulsion containing nanovitamin C, E and propolis. We've also obtained good results from the test of Antibacterial effect against S.aureus and E.coli. Also, swelling of inflammation tissues observed by scanning electrical microscopy has gone down. In conclusion, we have gained confidence that nanoemulsion containing nano vitamin C, E and propolis has very high Antibacterial effect against bacteria in oral. And it made us guess that inflammation of gingival reduces after decreasing interleukin-$1{\beta}$. Thus, we expect that nanoemulsion containing nano vitamin C, E and propolis gives good effects to patient having gingival disease.

Potential of Lilium lancifolium in Alleviating Pain and Inflammation in a Rat Model of Monosodium Iodoacetate-Induced Osteoarthritis

  • Lilium lancifolium (LL) is widely cultivated in East Asia and used to attenuate airway diseases. Our current study aimed to investigate the therapeutic effect of LL on pain level and inflammatory response in a rat model of monosodium iodoacetate (MIA)-induced osteoarthritis (OA). We first examined the effect of LL on inflammatory cytokines and inflammatory mediators in IL-1β-treated HTB-94 cells. The LL extract was found to significantly inhibit the levels of Interleukin-6 (IL-6), Interleukin-8 (IL-8), and prostaglandin-E2 (PGE-2) in Interleukin-1 β (IL-1β)-stimulated HTB-94 cells in a dose-dependent manner. Moreover, chronic oral administration of LL effectively restored the weight-bearing distribution in the rat model of MIA-induced OA. In addition, administration of LL inhibited inflammatory cytokines and inflammatory mediators, such as C-reactive protein (CRP), IL-6, leukotriene B4 (LTB-4), PGE-2, and matrix metalloproteinase-9 (MMP-9). Our findings collectively suggested LL as one of the potential therapeutic agents for OA, owing to its properties of reducing pain and inflammatory responses.

우심이 조직내의 Tumor necrosis Factor-${\alpha}$와 Interleukin-6의 함량 (The Contents of Tumor Necrosis Factor-${\alpha}$ and Interleukin-6 in Right Auricular Tissue)

  • Background: TNF-$\alpha$ plays a major role in producing left ventricular dysfunction cardio-myopathy pulmonary edema and inhibits the compensatory mechanism of congestive heart failure. IL-6 is an acute reactant of immune reaction and also known to control immune reaction but its function in the myocyte was not clearly investigated. Author's performed this experiment to investigate the contents of TNF-$\alpha$ and IL-6 on the assumption that TNF-$\alpha$ and IL-6 may reside in nonfailing heart that has gone cardiac surgery and play some role in cardiac function. Material and Method : Right auricular tissues were sampled from 12 patients who had undergone total corrective surgery for both congenital and acquired heart diseases from January 1998 to June 1998 in Kosin Universcfy Gospel hospital. The quantitive analysis of TNF-$\alpha$ and IL-6 were assessed by ELISA method in right auricular tissue. Hemodynamic values about the pressure of ventricle atrium aorta pulmonary artery and cardiac index pulmonary and systemic vascular resistance and cardiac output were measured by echocardiography and cardiac catheterization and biochemical analyses of LDH & AST were done before operation. statistical analysis was by Paired Student t-test. Patients were divided into children(under 15 years olds) and adults groups and the data was compared beween two groups. Conclusion: Mild pulmonary hypertension and increased pulmonary vascular resistance were existed in both group. The contents of tissue TNF-$\alpha$ IL-6 in each group were independent of each data.

Fidelity of Transgene Transmission and Expression in the Transgenic Mice

  • In this study, we examined transmission efficiency and expression level of the transgenes in the transgenic mice. The transgenic lines secreting a considerable amount of human lactoferrin(LF) thrombopoietin(TPO), interleukin-10(IL-10) into their milk were subjected to access the inheritance and maintenance of transgenic phenotype. They were bred through three generations. The transmission frequency for each generations(F9, F10, F11) of 3 lines was 38.03±10.43%(13/35), 48.33±3.76%(19/39) and 31.83±8.88%(9/28) in the LF line, 51.33±18.98%(20/38), 63.70±35.71%(12/20) and 29.57± 15.05%(8/26) in the TPO line, 38.27±17.74%(15/37), 47.47±29.88%(14/28) and 50.87±5.85%(14/28) in the IL-10 line, respectively.   • Cheongpyesagan-tang (CP) is one of the traditional medicinal prescription to treat Taeumin (太陰人)'s disease. It has been commonly used for the treatment of stroke, arthritis, diabetes and obesity. In this study, we investigated an anti-inflammatory potential of CP water extract. We examined the effects of CP on the lipopolysarccharide (LPS)-induced production of nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$). We also examined the levels of protein or mRNA of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and proinflammatory cytokines in RAW 264.7 cells. CP inhibited NO and $PGE_2$ production in a dose dependent manner and decreased the protein and mRNA expression of iNOS and COX-2. Also, CP decreased the mRNA expression of interleukin-6 (IL-6), interleukin-$1{\beta}$ (IL-$1{\beta}$), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$). These results suggest that CP has potential as anti-inflammatory therapeutic medicine.

  • The intricate role of innate type-2 cytokines in immune responses is increasingly acknowledged for its dual nature, encompassing both protective and pathogenic dimensions. Ranging from defense against parasitic infections to contributing to inflammatory diseases like asthma, fibrosis, and obesity, these cytokines intricately engage with various innate immune cells. This review meticulously explores the cellular origins of innate type-2 cytokines and their intricate interactions, shedding light on factors that amplify the innate type-2 response, including TSLP, IL-25, and IL-33. Recent advancements in therapeutic strategies, specifically the utilization of biologics targeting pivotal cytokines (IL-4, IL-5, and IL-13), are discussed, offering insights into both challenges and opportunities. Acknowledging the pivotal role of innate type-2 cytokines in orchestrating immune responses positions them as promising therapeutic targets. The evolving landscape of research and development in this field not only propels immunological knowledge forward but also holds the promise of more effective treatments in the future.
