Association of Total Sugar Intakes and Metabolic Syndrome from Korean National Health and Nutrition Examination Survey 2001-2002 (한국 성인의 총당류 섭취와 대사증후군과의 관계 -2001년과 2002년도 국민건강영양조사자료를 이용하여-)

  • The purpose of this study was to establish an association between the percent of energy from total sugar and disease prevalence of obesity, hypertension, dyslipidemia, insulin resistance, and metabolic syndrome with the context of the current population dietary practice in Korea. The Korean National Health and Nutrition Survey, 2001 and 2002 dataset were used as the source of data for this research. Usual nutritional intakes for over 20 years old people were calculated from the two non-consecutive dietary intake data from KNHANES 2001 and 2002 dataset. SAS and SUDAAN were used for statistical analyses. Sample weighted means, standard errors, and population percentages were calculated, and multiple logistic regression model with adjustment for covariates were used to determine the odds ratios(ORs) and 95% confidence intervals. Subjects were categorized as 3 ways and compared the LS means and ORs for heath factors. First, subjects excluding pregnant women, were categorized according to percent of energy from the usual total sugar intakes as ${\leq}10%$, 11-15%, 16-20%, 21-25%, >25%. Risk of LDL cholesterol showed a tendency to increase in the '>25%' group compared to the '<10%' group. The risks of the other health effects did not show any significant differences. Second, the subjects were categorized considering both Acceptable Macronutrient Distribution Range(AMDR) from carbohydrate and %Energy from total sugar as 'CHO<55% & Total sugar ${\leq}10%$', 'CHO 55-70% & Total sugar 11-25%', and '$CHO{\geq}70%$ & Total sugar ${\geq}25%$'. The risk of obesity tended to increase in the '$CHO{\geq}70%$ & Total sugar ${\geq}25%$' group compared to the 'CHO<55% & Total sugar ${\leq}10%$'. Third, the subjects were categorized as 'CHO<55% & Total sugar ${\leq}10%$', 'CHO 55-70% & Total sugar 11-20%', and '$CHO{\geq}70%$ & Total sugar ${\geq}20%$'. The risk of obesity also tended to increase in the '$CHO{\geq}70%$ & Total sugar ${\geq}20%$' group compared to the 'CHO<55% & Total sugar ${\geq}20%$' group. In conclusion, risk of LDL cholesterol showed a tendency to increase in the over 25% total sugar intake group, and the risk of obesity tended to increase in the 20-25% total sugar intake and high carbohydrate intake group. The risks of hypertension, hyperlipidemia, insulin resistance, and metabolic syndrome were not associated with total sugar intakes. More research to elucidate the association for Korean between the intakes of total sugar, added sugar, glucose, fructose, and sweeteners and diseases prevalences shoud be excuted in the future.

effects of Vitamin E on the Antioxidative Defense System of Kidney in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐 신장조직의 항산화계에 미치는 Vitamin E의 영향)

  • The purpose of this study was to investigate the effects of vitamin E on the antioxidative defense system of kidney in streptozotocin induced diabetic rats. Sprague Dawley male rats weighing 100$\pm$10g were randomly assigned to one normal and three STZ induced diabetic groups, which were subdivided into vitamin E free diet(DM 0E group), 40mg vitamin E per kg diet(DM 40E group) and 400mg vitamin E per kg diet(DM 400E group). Vitamin E level of normal group was 40 mg per kg diet. Diabetes was experimentally induced by intravenous injection of 55mg/kg of body weight of streptozotocin(STZ) in citrate buffer(pH 4.3) after 4 weeks feeding of experimental diets. Animals were sacrificed at the 6th day of diabetic states. There were no significant on body weights, food intakes, and food efficiency ratio before the diabetic occurrence. But after the injection of STZ, body weights and food efficiency ratios were significantly decreased and the food intakes was increased. Kidney weights were significantly increased in diabetic groups compared to normal group. However, there were no significant differences among the diabetic groups. Plasma insulin levels of diabetic groups were significantly decreased, whereas, blood sugar levels were increased compared to that of normal group. There were no significant differences among diabetic groups in plasma insulin and glucose levels. Activities of superoxide dismutase(SOD) in DM 0E and DM 40E groups were signi ficantly decreased by 33% and 27%, respectively, compared to normal group. But that of DM 400E group was increased by 35% compared to DM 0E group. Activity of glutathione peroxidase(GSHpx) in DM 0E group was decreased by 20% compared with normal group. GSHpx activity in DM 400E group was increased by 29% compared to normal group. The contents of vitamin E in kidney were 58% and 49% lower in DM 0E and DM 40E group, respectively, than normal group. There was no significant difference in renal vitamin E contents between DM-400E group and normal group. The contents of superoxide radical(O2 ) in kidney were 150% and 98%, respectively, higher in DM 0E and DM 40E groups than normalgroup. DM 400E and normal groups were similar levels in their superoxide radical contents of kidneys. These results indicate that vitamin E functioned as chain breaking antioxidant in kidney such as in other tissues.

Effects of a Weight Loss Program on Body Composition and Resting Energy Expenditure according to UCP 2 Genotype in Overweight Subjects

  • The objective of this study was to examine the effects of a weight loss program on the degree of obesity and levels of resting energy expenditure (REE) in overweight subjects according to their mitochondrial uncoupling protein 2 (UCP 2) genotype. Twenty-three subjects with a body mass index (BMI) greater than 27 were recruited from the Obesity Clinic of the Kyung-Hee University Hospital during the period of December 2000 - August 2001. The subjects were genotyped for the exon 8 allele; 15 subjects were found to be of del/del genotype, 8 were del/ins, and none were of ins/ins genotype. No significant association was found between the different UCP 2 genotypes and the initial levels of weight, fat mass (FM), lean body mess (LBM), BMI, REE, and REE/LBM ratio. After 12 weeks of a weight loss program, body weight and FM were significantly decreased, while LBM, total body water (TBW), and REE were not changed, irrespective of UCP 2 genotype. Initial fasting plasma levels of albumin, glucose, triglyceride, lipoprotein cholesterol, insulin, free triiodo-thyronine (T3), free fatty acid (FFA), and leptin were not different according to the UCP 2 genotype; furthermore, these blood parameters were not changed after the 12-week weight loss program. However, plasma levels of leptin decreased in both the del/del and ins/del genotypes, from 18.7 ng/ml to 13.4 ng/ml (p<.05), and from 18.1 ng/ml to 13.9 ng/ml (p<.05), respectively, after the weight loss program. In conclusion, this study found no significant association between the del/del or del/ins UCP 2 genotypes and differing levels of REE or differing degrees of obesity, either before or after a weight loss program. This study provided evidence that a well- managed weight loss program could maintain levels of REE, which plays an important role in the maintenance of energy balance.

A study on the effect on obesity and lipid metabolism in liver hypofunction animal-experimental model induced by Acetaminophen(AAP) injection (Acetaminophen(AAP)으로 유발한 간기능 저하 동물 모델에서의 비만 및 지질대사에 대한 영향 평가)

  • Objectives: The objective of this research is to develop new animal-experimental model for Sasang Constitutional Medicine, especially for partial Taeyangin(one of four constitution which has good pulmonary function and poor hepatic function) by AAP intraperitoneal injection, and to estimate from the viewpoint of obesity and lipid metabolism. Methods: The C57bl/6J mice was divided into 4 groups ; Normal group, AAP group, High-Fat-Diet(HFD) group, and HFD+AAP group. 200mg AAP was injected intraperitoneally to the AAP group twice a week for six weeks, and HFD group was fed with 60%-High-fat Diet for six weeks. HFD+AAP group got both AAP injection and 60%-High-fat Diet at the same time for the same period. In this period, We measured the weight and Food Efficiency Ratio(FER, %) once a week. After six weeks, We conducted the blood chemical test from the groups, and extracted the fat tissue to measure weight. Results & conclusion: In the liver function test, two AAP groups had higher AST and ALP, and normal LDH. The blood level of creatinine from all groups were normal. The rate in weight was lesser by 7.8% in HFD+AAP group, and had lesser FER than HFD group. Also They had lesser Total cholesterol and LDL cholesterol, and had more HDL cholesterol than HFD group. HFD+AAP group hadmore glucose in serum and lesser Insulin-like Growth Factor 1(IGF-1) than HFD group.

Comparison of Anthropometric Measurements, Dietary Quality and Blood Lipid Levels in Ssireum Players, Regular Moderate and Intermittent Light Exercisers (씨름 선수와 규칙적 중등도 및 간헐적 저강도 운동실천자의 체격지수, 영양소 섭취 및 혈중 지질의 비교 연구)

  • Ssireum, the traditional Korean sport, is very popular at both amateur and professional levels. Ssireum players are prone to be obese which related to the chronic disease in their later life. The purpose of the study was to compare the anthropometric measurements, quality of diet, and blood parameters of Ssireum players with those of subjects who were matched body mass index, gender and age. Participants in the 3 groups, Ssireum players (SP, n=15), regular moderate exerciser (RME, n=15, >3 times /wk, >20 min/time) and intermittent light exerciser (ILE, n=14) groups. Anthropometric measurements included height, weight, fat mass (by Dual energy X-ray absorptiometry), lean body mass, triceps and thigh skin-fold thickness, mid-arm, waist, hip and thigh circumference. Dietary assessments were accomplished using 3-days food records, diet quality index (DQI), dietary variety score (DVS). Blood levels of lipids, leptin and insulin were analyzed. As a results lean body mass and mid-arm circumference were significantly higher in Ssireum players than those of other groups (p<0.01). Total body fat, trunk fat, abdominal skin fold thickness and waist-hip ratio were significantly lower in SP group than those of other groups (p<0.01). DVS were higher however, dietary quality was low in Ssireum players than in other groups. There were no differences among the 3 groups in regard to blood total cholesterol, LDL-cholesterol and glucose levels however, leptin level was low in Ssireum players. These results indicate that Ssireum players had significantly higher lean body mess and lower body fat when these were compared with regular moderate exerciser and intermittent light exerciser. Blood leptin levels of Ssireum players were low but blood lipid profiles were not significantly different.

  • Animal cell cultures generally require a nutrient-rich medium supplemented with animal serum. Adult bovine serum contains a variety of nutrients including inorganic minerals, vitamins, salts, proteins and lipids as well as growth factors that promote animal cell growth. To evaluate the potential use of gender-specific bovine serum (GSBS) for cell culture, the biochemical properties of male serum (MS), female serum (FS) and castrated-male serum (CMS) were investigated. Overall, the chemical profile of GSBS was similar to that of bovine references except for glucose, creatine kinase, lactate dehydrogenase and potassium. FS showed elevated total protein and sodium concentrations compared to MS and CMS. Proteins present in MS, FS and CMS but absent in fetal bovine serum (FBS) were selected by two-dimensional gel electrophoresis and identified by peptide mass fingerprinting. Some of the identified proteins are known to be involved in immune responses and the others have unknown physiological roles. Moreover, it was found that some proteins such as alpha-2-macroglobulin appeared to be gender-specific with higher contents in FS. Insulin and testosterone was significantly higher in MS, and $17{\beta}$-estradiol and estrone were higher in FS, as compared to the other sera. Taken together, the results indicate that each GSBS has a different ratio of components. Differences in serum constituents may affect cell cultures in a different manner and could be beneficial, depending on the specific aim of cell cultures.

Anti-obesity Effects of Galgeun-tang in High Fat Diet Induced Obese Mice Model (갈근탕이 고지방 식이 유발 백서 모델에 미치는 항비만 효과)

  • Objectives To investigate anti-obesity effects of Galgeun-tang, an herbal formula, in high fat diet induced obese mice model. Methods 24 Male C57Bl/6J mice were randomly assigned to normal group fed with normal research diet (NOR, n=6), high fat diet control group treated with water (HFD, n=6), high fat diet group treated with Orlistat (ORL, n=6, Orlistat 10 mg/kg), and high fat diet group treated with Galgeun-tang (GGT, n=6, Galgeun-tang 700 mg/kg). 12 weeks later, body weight, fat weight, liver weight, blood glucose, total cholesterol, triglyceride, HDL, ALT, AST, obesity related neuropeptides and adipokines, ratio of gut microbiota, and histopathology of liver were evaluated. Results In the GGT group, 1. body weight gain, liver weight gain, and total fat weight gain were significantly less than those in the HFD group. 2. blood glucose level was significantly lower and insulin level was significantly higher than in the HFD group. 3. total cholesterol level and triglyceride (TG) level were significantly lower and high density lipoprotein (HDL) level was significantly higher than in the HFD group. 4. appetite-promoting ARC neuropeptides such as Agrp and Npy were significantly less and appetite-inhibiting ARC neuropeptide, Cart was significantly more than in the HFD group in qRT-PCR analysis. 5. adiponectin level and visfatin level were significantly higher, and resistin level and leptin level was significantly lower than in the HFD group. 6. the relative level of Bacteroidetes was significantly higher, and the relative level of Firmicutes was significantly lower than in the HFD group. 7. the increase of adipose tissue was significantly more inhibited than in the HFD group. Conclusions The present study showed that Glageun-tang exerts anti-obesity effects in that it. 1. inhibited the increase in body weight, liver weight, and total fat weight. 2. decreased the level of TG, and increased the level of HDL. 3. influenced neuropeptides and adipokines that are important in regulating food intake and changes of body weight. 4. modified the beneficial quantitative changes in gut microbiota suppressing the tendency toward obesity.

Hypoglycemic Effect of Extracts of Soybean Paste Containing Mycelia of Mushrooms in Streptozotocin- Induced Diabetic Rats (버섯된장 추출물의 당뇨 유발 흰쥐에 대한 혈당강하 효과)

  • The hypoglycemic effects of the hot water extract of soybean paste containing mycelia of Phellinus linteus, Cordyceps militaris, Ganoderma lucidum were evaluated in streptozotocin-induced diabetic rats. All the diabetic groups were not significantly affected in food intake and food efficiency ratio. Significant decrease in blood glucose was observed in all the soybean paste containing mushroom mycelia (SPM) groups. Especially, administration of soybean paste containing mycelia of Ganoderma lucidum, blood glucose level was significantly lowered up to 45% compared to that of diabetic control group. The GPT and GOT activities indicated lower values in all the SPM groups than in the diabetic control group.

Effects of Dietary Food on Body Weight, Plasma Lipids and Glucose in Rats (다이어트 식품이 흰쥐의 체중, 혈중지질 및 혈당농도에 미치는 영향)

  • The diet food used in this test was composed of the total 16 kinds, such as refined water, herb extract, pomegranate extract, polydextrose and isomalto-oligosaccharide etc. Edible fiber as a major component in dietary food was contained to 2.60%(w/w) level. But the calorie was 104.57kca1/100g, contrast of control domestic saengshick product was 368.06kca1. As a result, calorie of dietary food sample was very low level. Vitamin and mineral as a minor component were determined 5, 6 kinds, respectively. On the other hand, dietary effect of body weight loss was determined about 14%(w/w) at animal test result used for rats during 4 weeks. The reason of dietary effect could confirmed from change of plasma neutral lipids, total cholesterol and HDL-cholesterol concentration. Increasing ratio of these in the general diet and dietary diet feeding groups during 4 weeks were 10.55, 6.90, 9.82% and 6.11, 3.85, 6.11%, respectively. from these results, decrease reason of body weight could confirmed the relative relationship with plasma lipids and cholesterol concentration. But plasma plasma glucose and insulin concentration have not positive relativity.

Cardiovascular Autonomic Neuropathy Predicts Higher HbA1c Variability in Subjects with Type 2 Diabetes Mellitus

  • Background: This study aimed to investigate the association between the presence and severity of cardiovascular autonomic neuropathy (CAN) and development of long-term glucose fluctuation in subjects with type 2 diabetes mellitus. Methods: In this retrospective cohort study, subjects with type 2 diabetes mellitus who received cardiovascular autonomic reflex tests (CARTs) at baseline and at least 4-year of follow-up with ${\geq}6$ measures of glycosylated hemoglobin (HbA1c) were included. The severity of CAN was categorized as normal, early, or severe CAN according to the CARTs score. HbA1c variability was measured as the standard deviation (SD), coefficient of variation, and adjusted SD of serial HbA1c measurements. Results: A total of 681 subjects were analyzed (294 normal, 318 early, and 69 severe CAN). The HbA1c variability index values showed a positive relationship with the severity of CAN. Multivariable logistic regression analysis showed that CAN was significantly associated with the risk of developing higher HbA1c variability (SD) after adjusting for age, sex, body mass index, diabetes duration, mean HbA1c, heart rate, glomerular filtration rate, diabetic retinopathy, coronary artery disease, insulin use, and anti-hypertensive medication (early CAN: odds ratio [OR], 1.65; 95% confidence interval [CI], 1.12 to 2.43) (severe CAN: OR, 2.86; 95% CI, 1.47 to 5.56). This association was more prominent in subjects who had a longer duration of diabetes (>10 years) and lower mean HbA1c (<7%). Conclusion: CAN is an independent risk factor for future higher HbA1c variability in subjects with type 2 diabetes mellitus. Tailored therapy for stabilizing glucose fluctuation should be emphasized in subjects with CAN.
