  • Alkaline bacterium producing a high pro-tease activity at low temperature was isolated by using enrichment culture from soil samples and identified as Bacil-lus sp. WRD-1 Cell growth was maximal at 10 hours and the optimal initial pH and culture time of culture condition for enzyme production was pH 7 and 10 hours, respectively. Temperature range of high enzyme activity were $10~40^{\circ}C$. The optimal pH and temperature for the enzyme activity were pH9 and $30^{\circ}C$.

  • Among microorganisms isolated from soil, YJ-66 strain was the best producer of flocculant and was examined for flocculating ability in the active carbon and CaCl2. YJ-66 strain was the best producer of flocculant and was examined for flocculating ability in the active carbon and CaCl2. YJ-66 strain was identified to be a species belonging to the genus Achromobacter. The optimum culture condition for production of bioflocculant with the isolated strain was for 72hrs at 3$0^{\circ}C$ and pH7.5. The favorable carbon, nitrogen sources and inorganic salts for production of the flocculant were sucrose, peptone, MgSO4 and KH2PO4, whose optimal concentrations were 2%. 0.067%, 0.1% and 0.1%, respectively. Addition of the carbon and inorganic salts significantly increased the production of flocculant. Compositions of optimized culture medium for bioflocculant production by Achromobacter sp. YJ-66 were 2% sucrose, 0.067% peptone, 0.1% MgSO4 and 0.1% KH2PO4 in initial pH 7.5 during at 3$0^{\circ}C$ for 72hrs.

Effect of Low Intensity Pulsed Electric Field on Ethanol Fermentation and Chemical Component Variation in a Winemaking Culture

  • Electric polarity of working electrode and counter electrode was periodically switched at the intervals of 30 sec. Electric current generated by anodic and cathodic reaction of working electrode was reached to +30 and -12 mA in low intensity pulsed electric field (LIPEF). The yeast growth, ethanol production, and malate consumption in the initial cultivation time were more activated in the LIPEF than the conventional condition (CC). Polyphenol, total phenolic contents (TPC), and total flavonols (TF) were gradually decreased in all cultivation conditions during incubation for 2 weeks but antioxidation activity was not. TF was significantly lower in 3 and 4 V of LIPEF than CC and 2 V of LIPEF; however, the polyphenol, TPC, and antioxidation activity were a little influenced by the LIPEF. After ripening of the winemaking culture for 15 days, polyphenol, TPC, and TF were a little increased but the antioxidation activity was not.

고체배양에 의한 Monascus anka의 적색색소 생성의 최적 조건 (The Optimal Condition for Production of Red Pigment by Monascus anka on Solid Culture)

  • Monascus anka KCTC 6121을 이용하여 백미에 의한 고체 배양을 통한 실험균의 적색 색소의 생산을 위한 배양조건의 최적화에 관한 연구 결과는 다음과 같이 요약된다. 실험균주가 생산하는 적색색소 생산의 최적 배양조건은 초기 수분함량 25%, 2.5$\times$$10^{6}$/mL의 실험균주의 포자, 배양온도3$0^{\circ}C$, 배양습도 90%에서 배양 12일째가 가장 양호했다. 생산된 적 색 색소의 추출은 80% 에탄올에서 2일간 추출이 가장 양호했으며 2일 이후의 색소 추출은 색소 추출량에 거의 변화가 없음을 알 수 있었다. 실험균주로부터 추출한 색소의 빛에 대한 안정성은 반 직사광선에서 색소의 색상이 30일간 8% 미만으로 감소하여 햇빛에 대한 안정성은 비교적 높았다.

동치미에서 분리한 Lactobacillus sp. FF-3가 생산하는 bacteriocin의 최적 생산조건 (The Optimal Producing Conditions of Bacteriocin Produced by Lactobacillus sp. FF-3 Isolated from Korean Dongchimi)

  • 동치미로부터 분리된 Latobacillus sp. FF-3의 Bacteriocin 생산에 대한 최적 배양조건과 배양 특성을 조사하였다. Bacteriocin 생산을 위한 최적 생산조건은 MRS(proteose peptone 1%, beef extract 1%, yeast extract 0.5%, glucose 2%, polysorbate 80 0.1%, ammonium citrate 0.2%, sodium acetate 0.5%, magnesium sulfate 0.01%, manganese sulfate 0.005%, dipotassium phosphate 0.2%) 배지를 기본으로 하여 21시간 배양한 결과, 배양온도는 30∼37$^{\circ}C$, 초기 pH는 7.0 으로 확인되었다. 최적 생산배지조성으로 탄소원은 glucose 3%, 질소원은 tryptone 4%, 무기염류는 manganese sulfate 0.005%가 bacteriocin 생성에 최적 조성으로 나타났으며, 최적 배지상에서 항균활성은 최대 484 BUf/mL로 나타났다.

  • A biosurfactant-producing microorganism was isolated from activated sludge by enrichment culture when grown on a minimal salt medium containing n-hexadecane as a sole carbon source. This microorganism was identified as Pseudomonas sp. and it was named Pseudomonas sp. EL-G527. It's optimal culture condition is 2% n-hexadecane, 0.2% NH$_4$NO$_3$, 0.3% KH$_2$PO$_4$, 0.3% $K_2$HPO$_4$, 0.02% MgSO$_4$ㆍ7$H_2O$, 0.0025% CaCk$_2$ㆍ6$H_2O$, 0.0015% FeSO$_4$ㆍ7$H_2O$ in 1$\ell$ distilled water and initial pH 7.0. Cultivation was initiated with a 2% inoculum obtained from starter cultures grown in 30 $m\ell$ of the same medium in 250 $m\ell$ flask. They were cultivated at 3$0^{\circ}C$ in reciprocal shaking incubator and the highest biosurfactant production was observed after 4 days.

  • A screening was performed to isolate exoploysaccharide-producing microorganisms, which synthesized specific exopolysaccharide for the substitutive of commercial polysaccharides, from natural sources. Soil bacterium, one of 378 mucoid isolates, was finally selected as potential producer of polysaccharides which made the culture broth very viscous and thus examined in detail for optimal medium composition. Isolated strain was identified as Xanthomonas sp. EPS- 1 from the results of morphological and biochemical characteristics. The composition of optimal medium for exopolysaccharide production was as follows: 50 g sucrose, 1.5 g peptone, 2 g KH$_{2}$PO$_{4}$, 2 g MgSO$_{4}$, -7H$_{2}$O, 3 g NaCl, 0.05 g CaCO$_{3}$, 0.07 g FeSO$_{4}$-7H$_{2}$O and 0.05 g MnSO$_{4}$-7H$_{2}$O in 1 liter of distilled water. From the experiments of temperature and pH dependence, the optimal conditions for exopolysaccharide biosynthesis seemed to be 30$\circ$C and 8.0, respectively. About 14.9 gram of maximum exopolysaccharide per liter was obtained at the initial pH 8.0, 30$\circ$C and 250 rpm in a flask culture. The exopolysaccharide EPS-1 had such potential as an emulsifying agent and a gelling agent in comparision with commercial exopolysaccharide.

Submerged Culture of Phellinus linteus for Mass Production of Polysaccharides

  • In order to increase the mycelial production of Phellinus linteus, which exhibits potent anticancer activity, some ingredients of the medium used to culture P. linteus were investigated. The optimal medium composition for the production of Phellinus linteus was determined to be as follows: fructose, 40 g/l; yeast extract, 20 g/l; $K_2HPO_4$, 0.46 g/l; $K_2HPO_4$, 1.00 g/l; M$MgSO_4\cdot7H_2SO$, 0.50 g/l; $FeCl_2\cdot6_2O$, 0.01 g/l; $MnCl_2\cdot4H_2O$, 0.036 g/l; $ZnCl_2$, 0.03 g/l; and $SuSO_4\cdot7H_2O$, 0.005 g/l. The optimal culture conditions were determined to be as follows: temperature, 28$^{\circ}C$; initial pH, 5.5; aeration, 0.6 vvm; and agitation, 100 rpm, respectively. Under optimal composition and conditions, the maximum mycelial biomass achieved in a 5 l jar fermentor was 29.9 g/l.

디프테리아 toxin 생산을 위한 발효조건 최적화 (Optimization of Culture Conditions for the Production of Diphtheria Toxin)

  • 디프테리아는 Corynebacterium diphtheriae에 의해 발생되는 호흡기 질병으로 C. diphtheriae의 exo-toxin을 불활성화 시킨 toxoid 백신을 사용하여 예방해 왔다. 현재까지 국내에서는 정치배양 방법으로 디프테리아 toxin을 생산해 왔기 때문에 생산성과 품질에 한계가 있었으며, 이를 극복하기 위해 발효조를 이용한 발효조건 최적화에 대한 연구를 수행하였다. 디프테리아 toxin을 보다 효율적을 얻기 위한 배지로서 beef antigen이 함유되어 있지 않은 casein 유래의 NZ-Case를 선택하였다. 발효조에 의한 toxin 생성은 세포성장과 함께 증가하는 growh-associated form으로 나타났다. 최대의 세포성장과 toxin 생성은 초기 pH가 7.0인 배지에서 0.22vvm의 통기와 400rpm의 교반조건에서 얻을 수 있었다. 또한 최대의 toxin 생성을 위한 최적의 iron 이온의 농도는 0.3mg/L 이었으며, morgamcphospale의 첨가에 의한 calcium-phosphate 침전이 배지 내에 iron 이온의 농도조절을 위하여 요구되었다. 면역원성을 확인하기 위한 역가시험 결과 발효조 배양에서 얻은 toxoid는 4단위에서 모든 guinea-pigs가 생존하여 2단위인 정치배양 toxin에비해 월등히 우수한 것으로 판단되었다. 결국 발효조 배양으로 toxin을 생산할 경우 부작용이 적고 수율과 생산성 및 면역원성 이 우수한 toxid의 생산이 가능하였다.

  • 기내에서 성장한 배추의 자엽, 하배축과 계대배양한 지 2~3주 되는 어린잎이 원형질체 분리재료로 사용되었다. 배추의 원형질체 분리에 가장 적합한 효소의 조합은 0.4M mannitol 을 삼투조절제로 한 1% Cellulysin과 0.5% Macerozyme의 효소조합이 배추 원형질체 분리에 가장 적합한 것으로 나타났으며, 잎조직을 27$\pm$1$^{\circ}C$에서 30rpm의 속도로 12~16시간 동안 효소와 반응시켰을 때 7.6$\times$$10^{5}$ protoplast/g의 가장 많은 원형질체를 얻었다. 세포분열을 유기시키기 위하여 K8p 배지에 5 mg/L 2,4-D와 2 mg/L에 첨가하였을 때 배양 7~10일에 자엽과 하배축에서 분리한 원형질체에서 세포군들이 형성되었다. 분열한 세포가 8~10 세포크기 단계에 이르면 0.2% agarose 반고체 배지에 고정시켜 배양하였다. 얻어진 calli들을 100가지 다양한 재분화 배지에 옮겼으나 식물체의 재분화는= 이뤄지지 않았지만, 간혹 캘러스에서 뿌리가 분화되는 것은 관찰할 수 있었다.
