A STUDY ON THE ANTIMICROBIAL EFFECT OF FLUORIDE-RELEASING REMOVABLE ORTHODONTIC APPLIANCES (불소유리 가철성 교정장치의 항균효과에 관한 연구)

  • To study the antimicrobial effect of fluoride-releasing removable orthodontic appliances, 0 wt%(Group 1), 5 wt%(Group 2) and 10 wt%(Group 3) of $CaF_2$ were added to orthodontic resin. The specimens were then put in media in which S. mutans and Lactobacillus, known cariogenic bacteria, were cultured. Inhibiting zones were measured 24, 48 and 72 hours later. The following conclusions were reached: 1 In the Lactobacillus media, no zone of inhibition was observed regardless of the time passed. 2. In the S. mutans media, zones of inhibition were observed after 48 hours. 3. S. mutans showed greater inhibition in Group 3 compared to Group 2, after 72 hours rather than 48 hours(p<0.01).

Antioxidative Activity and Tyrosinase Inhibition Effect of Fractions from the Extract of Inonotus obliquus (차가버섯 용매별 분획물의 항산화 활성과 Tyrosinase 저해 활성)

  • This study was conducted to investigate the antioxidative activities and tyrosinase inhibition effects of fractions from the distilled water extract of Inonotus obliquus. Moreover, GC-MS based analysis with trimethylsilyl (TMS) derivatization was carried out for active compound in the extract of I. obliquus. In DPPH radical scavenging ability, $SC_{50}$ values of the ethyl acetate fraction was 0.393 mg/ml as a result of the most effective than other fractions. Meanwhile, aqueous fraction showed higher effect in tyrosinase inhibitoty activity. In GC-MS based analysis with TMS derivatization, 7 compounds including syringic acid, vanillic acid and protocatechuic acid were observed in ethyl acetate fraction, and oxalic acid is the main compound in aqueous fraction. As a result, it was confirmed that oxalic acid in aqueous fraction from the distilled water extract of I. obliquus was a compound showing tyrosinase inhibition effect.

  • A series of in vitro experiments was conducted to investigate response of rumen bacterial deamination to the ionophore salinomycin. Addition of salinomycin to the inoculum, strained rumen fluid, depressed ammonia production from casein, while increased accumulation of ${\alpha}$-amino acids. This suggests an inhibitory effect of salinomycin on ruminal deamination. When the effect in washed bacterial suspension was monitored with individual amino acid, aspartic acid degradation was markedly inhibited by salinomycin. This inhibition was not observed when the mixed rumen bacteria were ultrasonically disrupted and used as the enzyme source. Extent of the inhibition tended to be higher in the bacteria source from sheep on a high roughage diet. From these results it was speculated that the inhibition of deamination with salinomycin is caused by a decreased transport of amino acid into the bacterial cells as well as a decreased proportion of deaminating bacteria in the rumen.

In vitro Screening of UVA Phototoxicity Inhibitors using the Natural Products (In vitro 실험법에 의한 천연물 중의 UVA 광독성 억제제 검색)

  • The phototoxicity inhibitory activity of 15 natural products having antiinflammatory effect was screened by three in vitro methods: yeast growth inhibition test with Candida albicans, RBC photohemolysis and MTT assay. We induced phototoxic reaction by irradiating UVA (365 nm) on chlorpromazine (CPZ) that has been widely documented as phototoxic agent in clinical and experimental studies and then observed the effects of the natural products after treating them with CPZ. In yeast growth inhibition test, X. stramonium showed the inhibitory effect on the UVA phototoxicity and E. officinalis, Yeast, P. suffruticosa showed phototoxicity inhibitory effect in that their % hemolysis compared with control were 36.14${\pm}$ 2.69, 42.82${\pm}$1.35, 36.41${\pm}$0.48 on UVA. In MTT assay, all tested natural products increased cell viability compared with the control.

Inhibition Effect of Testosterone Metabolism of Some Natural Products Containing Yacon and their Ameliorative Effect of Benign Prostatic Hyperplasia Symptom (야콘을 포함한 수종 천연물의 남성호르몬 대사 억제효과 및 전립선비대 증상 개선효과)

  • We reported previously that the extracts of Yacon tuber and leaf have spermatogenic activity and testosterone level increasing effect in men. The aim of this study was to investigate the mechanism of testosterone level increasing effect of Yacon tuber and leaf extracts. In this study, we found that Yacon leaf extracts and chlorogenic acid, a constituent of Yacon extracts showed significantly inhibition effect of testosterone degradation in rat liver homogenate. We suggest that this could be a part of direct mechanism about increasing effect of testosterone level. Futhermore, we evaluated the inhibition effect of testosterone metabolism of Torilis japonica and Coix lacryma-jobi have been reported as $5{\alpha}$-reductase inhibitor. The inhibition ratio of testosterone metabolism of these were Torilis japonica>Coix lacryma-jobi>Yacon. We also evaluated the benign prostatic hyperplasia (BPH) score with the preparation composed of these natural product, Yacon pill. It showed the decreased the BPH score in men. Rats with BPH showed significantly increased dihydrotestosterone (DHT) levels in the serum; however, Yacon pill extracts treated rats showed significant reduction of DHT levels compared with the BPH group. Conclusively, Yacon pill showed the possibility as ameliorable agents of BPH symptoms.

Studies on the Effect of korean Ginseng Components on Acetic acid Fermentation. [I] (인삼성분이 초산발효에 미치는 영향에 관한 연구(제1보))

  • In order to study the effect of Korean ginseng (Panax ginseng C. A. Meyer) components on acetic acid fermentation, ginseng extracts, sucrose, total can de saponins were added to the basal niedium respectively and surface culture was carried out at 30$^{\circ}C$. Lag ime, total acidity of the fermentation broth inhibitors and the degrees of inhibition were determined in tile course of fermentation . 1. Acetic acid fermentation was not inhibited by the addition of less than 1.93% of sucrose but the degree of inhibition was increased slightly by the addition of sucrose more than that. 2. Ginseng extract inhibited acetic acid fermentation slightly, and the degree of inhibition was similar to that of sucrose. Lag time was about 72 hours when a 20% of ginseng extract was added to the basal medium while that of the control was 22hours. 3. The free saponins inhibited acetic acid fermentation considerably, and the degree of inhibition of the saponins was about 400 folds of that of ginseng extracts. An increase of total acidity of the broth which contained 2.905% of the saponins was not observed even after one month. 4. It was presumed that some other components except saponins and sucrose in ginseng extracts counter the inbition effect of saponins on acetic acid fermentation

Hypoglycemic and Angiotension Converting Enzyme Inhibitory Effect of Water and Ethanol Extracts from Haesongi Mushroom (Hypsizigus marmoreus)

  • Water and ethanol extracts were prepared from the haesongi mushroom (Hypsizigus marmoreus) to measure functional components. The ability of the extracts to inhibit angiotensin-converting enzyme (ACE) and their hypoglycemic effects were also determined; the latter was measured by $\alpha$-amylase and glucosidase inhibition. Extraction yield, protein content, total phenol, and $\beta$-glucan in the water extracts were 55.86, 17.71, 1.89, and 21.93%, respectively. The respective values for the ethanol extracts were lower than those for water extracts. Both water and ethanol extracts showed dosedependent ACE inhibition, the effect of the former being greater. The water extract inhibited ACE activity by 95.34% at 40 mg/mL. The $IC_{50}$ values of the water extracts were 63.32 and 0.41 mg/mL for $\alpha$-amylase and glucosidase, respectively. Thus, the water extracts had a greater hypoglycemic effect than the ethanol extracts. From these results, water is a better solvent than ethanol to extract from the haesongi mushroom functional components that show ACE inhibition and have hypoglycemic effects.

Antimicrobial test of Antagonistic Microbes for Biological Control of Large patch of Zoysiagrass (잔디 Large patch의 생물학적 방제를 위한 길항 미생물의 선발과 항균력 검정)

  • A large patch disease caused by Rhizoctonia solani AG2-2(IV) is a serious problem in turfgrass sites including golf courses and sports fields in Korea. The objectives of this study were to isolate some antagonistic microorganisms and to explain some involving mechanisms. Initially single colonies which were formed from the filtrates of various soil samples were obtained from LB culture and then co-cultured with R.solani AG2-2(IV) on PDA plate to explore some antagonistic microbes against for large patch fungus, Rhizoctonia solani AG2-2(IV). Out of total 82 antagonistic isolates which commonly had inhibition effect on Rhizoctonia solani AG2-2(IV) mycelial growth, one candidate (YPIN22) showed the most antifungal effect, which was confirmed by the longest distance from the edge of bacterial colony to the mycelial edge of the Rhizoctonia solani AG2-2(IV) in the dual culture. A succeeding investigation was to test any potential effect of the isolate on growth inhibition of 5 other turfgrass pathogens including R. solani solani AG2-2(IIIB), P. ultimum, C. caudatum, C. lunata, and F.oxysporum. Preliminary result indicated that the new isolate YPIN22 was also found to have antagonistic potential on the growth inhibition of those turfgrass pathogenic fungi, which was explained by inhibition zones ranging from 8 to 22mm. A further explanation of some characteristics of the isolate YPIN22 will be discussed in detail.

Effect of ivermectin and doramectin for larval(Anisakis spp) migratory inhibition in rats (랫드에서 아니사키스 유충에 대한 이버멕틴과 도라멕틴의 운동성 억제효과)

  • This experiment has been investigated in order to examine the effect of ivermectin and doramectin for laval migratory inhibition in rats. Anisakis spp larvae were obtained from the mackerels in fish market in Cheongju. A total of 225 rats were classified with 3 groups with the same number ($3{\times}75$ rats); group I was control divided into 3 subgroups including 25 rats in each one, others (test groups) were treated subcutaneously with ivermectin and doramectin by 3 kinds of concentrations ($200{\mu}g/kg$, 400, 800), respectively. At 15 minutes after treating, 10 larvae were taken orally into each rat of 6 subgroups in test, and 5 rats were necropsied at 0.5, 1, 2, 4 and 8 hours after oral administration. The recovered larvae were used in larval migration inhibition (LMI). Ivermectin is significantly more effective than doramectin. In rats, the inhibitory effect of ivermectin and doramectin is slightly less than in vitro.

Beneficial Effect of Scutellaria baicalensis Georgi Extract on Mercury Chloride-Induced Membrane Transport Dysfunction in Rabbit Renal Cortical Slices (황금약침액(黃芩藥鍼液)이 가토(家兎) 신피질절편(腎皮質切片)에서 수은(水銀)에 의한 세포막(細胞膜) 물질이동(物質移動) 기능장애(機能障碍)에 미치는 영향(影響))

  • This study was undertaken to determine whether Scutellaria baicalensis Georgi (SbG) extract exerts the protective effect against $HgCl_2$-induced alterations in membrane transport function in rabbit renal cortical slices. The slices were treated with 0.1 mM $HgCl_2$ for 60 min at $37^{\circ}C$. $HgCl_2$ caused an inhibition in PAH uptake by renal cortical slices. Such an effect was accompanied by depressed $Na^+-K^+$-ATPase activity and ATP depletion. SbG prevented $HgCl_2$-induced inhibition of PAH uptake in a dose-dependent manner at the concentration ranges of 0.01-0.1%. $HgCl_2$-induced inhibition of $Na^+-K^+$-ATPase activity and ATP depletion were significantly prevented by 0.05% SbG. These results suggest that SbG prevents $HgCl_2$-induced alterations in membrane transport function in rabbit renal cortical slices. Such protective effects of SbG may be attributed to inhibition of peroxidation of membrane lipid.
