누에에서 곰팡이(Aspergillus niger) 감염에 의해 유도 발현되는 유전자의 클로닝과 동정 (Cloning and Identification of Differentially Expressed Genes Induced by Fungal Infection from Silkworm, Bombyx mori)

  • 본 연구는 곤충으로부터 새로운 항 진균 단백질을 발굴하기 하기 위한 목적으로 누에를 대상으로 Aspergillus niger의 감염을 유도하였을 때 발현되는 유전자의 특성을 분석한 것이다. Annealing control primer 법에 기초한 GeneFishing Kit를 사용하여 A. niger를 약 $6{\times}10^8$ colony per unit로 5령기 누에 유충의 체강에 감염시킨 후, 6시간 경과한 다음에 유도 발현되는 유전자(differentially expressed genes, DEGs)를 분석 한 결과, 10개의 유도 발현되는 유전자를 분리하였고 RT-PCR을 통해서 lysozyme, enbocin 그리고 한 개의 기능이 알려지지 않는 유전자등 3개의 유전자가 A. niger의 감염에 의해서 유의하게 과 발현된다는 것을 검증하였다. 일반적으로 그람 음성 및 양성 세균의 감염에 의해 유도된다고 알려진 enbocin 유전자가 A. niger의 감염에서도 과 발현이 유도되는 본 연구의 결과는 앞으로 enbocin 유전자의 항 진균 활성 연구에 중요한 기초 자료로 활용될 수 있을 것이다.

건축공간에서 공기 감염균 확산을 해석하기 위한 추적가스 고찰과 농도에 따른 감염 위험성 예측 연구 (A Inquiry of Tracer Gas for Analysis of Dispersion and Prediction of Infection Possibility according to Airborne Viral Contaminants)

  • The SARS virus began to appear and spread in North America and Southeast Asia in the early 2000' s, infecting and harming many people. In the process of examining the causes for the virus, studies on the airborne SARS virus and the way it spread were carried out mainly in the medical field. In the field of architecture, studies were done on the diffusion of air pollutants in buildings using gases such as $CO_2$, $N_2O$, or $SF_6$, but research on virus diffusion was limited. There were also explanations of only the diffusion process without accurate information and discussion on virus characteristics. The aim of this study is to analyze the physical characteristics of airborne virus, consider the possibility of using coupled analysis model and tracer gas for analyzing virus diffusion in building space and, based on reports of how the infection spread in a hospital where SARS patients were discovered, analyze infection risk using tracer gas density and also diffusion patterns according to the location, shape, and volume of supply diffusers and exhaust grilles. This paper can provide standards and logical principles for evaluating various alternatives for making decisions on vertical or horizontal ward placement, air supply and exhaust installation and air volumes in medium or high story medical facilities.

Expression Analysis of Sweetpotato Sporamin Genes in Response to Infection with the Root-Knot Nematode Meloidogyne incognita

  • Sweetpotato (Ipomoea batatas [L.]) is a globally important root crop cultivated for food and industrial processes. The crop is susceptible to the root-knot nematode (RKN) Meloidogyne incognita, a major plant-parasitic RKN that reduces the yield and quality of sweetpotato. Previous transcriptomic and proteomic analyses identified several genes that displayed differential expression patterns in susceptible and resistant cultivars in response to M. incognita infection. Among these, several sporamin genes were identified for RKN resilience. Sporamin is a storage protein primarily found in sweetpotato and morning glory (Ipomoea nil). In this study, transcriptional analysis was employed to investigate the role of sporamin genes in the defense response of sweetpotato against RKN infection in three susceptible and three resistant cultivars. Twenty-three sporamin genes were identified in sweetpotato and classified as group A or group B sporamin genes based on comparisons with characterized sweetpotato and Japanese morning glory sporamins. Two group A sporamin genes showed significantly elevated levels of expression in resistant but not in susceptible cultivars. These results suggest that the elevated expression of specific sporamin genes may play a crucial role in protecting sweetpotato roots from RKN infection.

경기도 광주 여름재배 시설토마토의 가루이 매개 바이러스 TYLCV, ToCV 발생현황 (Patterns of the Occurrence of TYLCV and ToCV with Whitefly on Summer-Cultivated Tomato in Greenhouse in Gwangju, Gyeonggi Province)

  • 경기 광주시 완숙토마토 시설재배단지에서 가루이가 매개하는 바이러스 2종 tomato yellow leaf curl virus (TYLCV)와 tomato chlorosis virus (ToCV)의 발생양상을 조사하였다. 2020년 여름재배 작형에서 TYLCV와 ToCV의 감염률은 수확기에 급격히 증가하였고, 단독 감염률은 각각 30.9%, 5.0%였고, 복합감염률은 52.2%로 TYLCV 단독감염과 2종 바이러스 복합감염이 83.1%로 대부분을 차지하였다. 2종 바이러스를 매개하는 가루이의 발생분포 및 보독률을 조사한 결과, 작물 재배기간 내내 담배가루이가 높은 비율로 발생하였고, 조사한 담배가루이는 모두 Mediterranean (MED)으로 확인되었다. 담배가루이 MED는 TYLCV를 단독으로 보독하거나 ToCV와 복합 보독하는 경우가 48.5%로 많았으며, ToCV의 보독률은 29.1%로 온실가루이(7.2%)에 비해 높았다. 담배가루이 MED의 2종 바이러스 보독률이 토마토 정식 전부터 47.2%로 높았고, 이후 계속 증가하여 수확기에는 73.8%로 가장 높았다. 수확기 토마토 재배포장별 2종 바이러스의 감염률에 따라 수확량을 비교 분석해본 결과, TYLCV와 ToCV의 복합감염률 증가가 수확량을 감소시키는 것을 확인하였고, 복합감염률이 10% 증가함에 따라 405.4 kg/10a 수량감소를 보였다.

소아 급성 하기도 감염의 바이러스 원인 및 임상 양상 (2002년 부산 지역) (Viral Etiology and Clinical Pattern of Acute Lower Respiratory Tract Infection in Children(Busan Area in 2002))

  • 목 적 : 호흡기 바이러스는 인간에게 가장 전염성이 강한 병원체의 하나로 급성 호흡기 감염 특히 하기도 감염증은 소아에서의 유병률과 사망률의 주요 원인이 되고 있다. 저자들은 하기도 감염증에 이환된 환아들을 대상으로 influenza A, B virus, adenovirus, enterovirus의 감염 연령 분포와 계절적 유행 양상, 바이러스별 임상 양상을 알기 위하여 이 연구를 시작하였다. 방 법 : 2002년 1월부터 2002년 12월까지 부산 메리놀병원에 입원하거나 외래 방문한 하기도 감염증 소아들을 대상으로 인후 가검물, 콧물, 타액 분비물 등을 채취하여 바이러스를 동정하였고, 검사를 시행한 568명 중 바이러스가 동정된 54명을 대상으로 호발 연령, 유행 시기, 임상 증상을 후향적으로 조사하였다. 결 과 : 1) 대상 환아의 연령 분포는 2개월부터 14세 사이였고, 남녀비는 1.8 : 1이었다. 3~6세가 29.6%로 가장 많았고, 2~3세 사이가 14.8%, 12세 이상이 13% 순이었다. 2) 동정된 바이러스는 influenza A virus가 59.3%로 가장 많았고, enterovirus가 33.3%, adenovirus 5.6%, influenza B virus 1.9%였고, parainfluenza virus와 RSV는 동정되지 않았다. 3) 임상 양상은 폐렴이 51.9%로 가장 많았고, 기관지염이 31.5%, 크루프 9.3%, 모세기관지염 7.4%의 순이었다. 4) 호흡기 바이러스는 각각의 유행 시기가 있어, influenza A virus는 3~5월, 11월에 2차례의 유행이 있어 주로 겨울에 유행하는 양상을 보였으며, influenza B virus는 3월에, adenovirus는 3월과 7월에, enterovirus는 3~7월에 유행하여 주로 봄부터 여름까지 유행하는 양상을 보였다. 결 론 : 2002년 부산 지역 호흡기 바이러스 감염의 주된 원인은 influenza virus였다. 이러한 결과는 influenza virus의 유행을 보여주는 것으로 이에 대한 지속적인 감시와 예방, 조기 진단과 치료가 필요하다고 하겠다.

Recurrent Pseudomonas aeruginosa Infection in Chronic Lung Diseases: Relapse or Reinfection?

  • Background: Pseudomonas aeruginosa infection is particularly associated with progressive and ultimately chronic recurrent respiratory infections in chronic obstructive pulmonary disease, bronchiectasis, chronic destroyed lung disease, and cystic fibrosis. Its treatment is also very complex because of drug resistance and recurrence. Methods: Forty eight cultures from 18 patients with recurrent P. aeruginosa pneumonia from 1998 to 2002 were included in this study. Two or more pairs of sputum cultures were performed during 2 or more different periods of recurrences. The comparison of strains was made according to the phenotypic patterns of antibiotic resistance and chromosomal fingerprinting by pulsed field gel electrophoresis (PFGE) using the genomic DNA of P. aeruginosa from the sputum culture. Results: Phenotypic patterns of antibiotic resistance of P. aeruginosa were not correlated with their prior antibiotic exposition. Fifteen of 18 patients (83.3%) had recurrent P. aeruginosa pneumonia caused by the strains with same PFGE pattern. Conclusion: These data suggest that the most of the recurrent P. aeruginosa infections in chronic lung disease occurred due to the relapse of prior infections. Further investigations should be performed for assessing the molecular mechanisms of the persistent colonization and for determining how to eradicate clonal persistence of P. aeruginosa.

Dynamic Patterns of Systemic Innate Immunity and Inflammatory Associated Factors in Experimental Caprine Coccidiosis

  • The present study was designed to assess the dynamic patterns of pro-inflammatory cytokines, including $IFN-{\gamma}$, $TNF-{\alpha}$, IL-4, IL-6, acute phase protein (${\alpha}1$-acid-glycoprotein, AGP), and an inflammation associated factor (adenosine deaminase; ADA) following experimental caprine coccidiosis. Ten kids aging from 2 to 4 months were infected orally with $5{\times}10^4$ sporulated oocysts and 10 animals served as controls. Blood samples were collected in both groups before infection and at days 3, 7, 14, 21, 28, and 35 post-infection (PI), and the levels of above-mentioned factors were measured. $IFN-{\gamma}$, $TNF-{\alpha}$, IL-4, IL-6, AGP, and ADA activities were significantly higher in infected animals from day 7 PI (P<0.05). In conclusion, the circulatory levels of most systemic inflammatory markers, including pro-inflammatory cytokines ($IFN-{\gamma}$, $TNF-{\alpha}$, IL-4, IL-6), AGP, and ADA increased significantly starting from day 3 to day 7 PI in caprine coccidiosis.

생분해성 iodophor microsphere 함유 유두침지 소독제의 건유기 젖소에서의 유방내 신규감염 예방 효과 (Effects of teat dipping disinfectant containing biodegradable iodophor microspheres on preventing dry period intramammary infection in dairy cows)

  • 1% iodophor loaded microspheres of PLGA (Poly[DL-Lactide-co-Glycolide]) were prepared by solvent evaporation method and were applied to the cows on dry period for evaluating it's preventive effects on intramammary infections. The morphology of the microspheres were evaluated using scanning electron microscopy and their releasing patterns were investigated. On investigating idophor releasing patterns of the microsphere, burst releasing pattern was detected until 2 days after in vitro incubation and sustained releasing was observed until 4 weeks. In field trial of teat dipping solution containing idophor loaded microspheres in dry cows showed significant preventive effects of intramammary infection caused by S. aureus, S. agalactiae, coagulase negative Staphylococci and coliform bacteria (p<0.05).

Paleoparasitological Studies on Mummies of the Joseon Dynasty, Korea

  • Paleoparasitology is the application of conventional or molecular investigative techniques to archeological samples in order to reveal parasitic infection patterns among past populations. Although pioneering studies already have reported key paleoparasitological findings around the world, the same sorts of studies had not, until very recently, been conducted in sufficient numbers in Korea. Mummified remains of individuals dating to the Korean Joseon Dynasty actually have proved very meaningful to concerned researchers, owing particularly to their superb preservation status, which makes them ideal subjects for paleoparasitological studies. Over the past several years, our study series on Korean mummies has yielded very pertinent data on parasitic infection patterns prevailing among certain Joseon Dynasty populations. In this short review, we summarized the findings and achievements of our recent paleoparasitological examinations of Joseon mummies and discussed about the prospects for future research in this vein.

Expression Analyses of MicroRNAs in Hamster Lung Tissues Infected by SARS-CoV-2

  • Coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), is an infectious disease with multiple severe symptoms, such as fever over 37.5℃, cough, dyspnea, and pneumonia. In our research, microRNAs (miRNAs) binding to the genome sequences of severe acute respiratory syndrome coronavirus (SARS-CoV), Middle East respiratory-related coronavirus (MERS-CoV), and SARS-CoV-2 were identified by bioinformatic tools. Five miRNAs (hsa-miR-15a-5p, hsa-miR-15b-5p, hsa-miR-195-5p, hsa-miR-16-5p, and hsa-miR-196a-1-3p) were found to commonly bind to SARS-CoV, MERS-CoV, and SARS-CoV-2. We also identified miRNAs that bind to receptor proteins, such as ACE2, ADAM17, and TMPRSS2, which are important for understanding the infection mechanism of SARS-CoV-2. The expression patterns of those miRNAs were examined in hamster lung samples infected by SARS-CoV-2. Five miRNAs (hsa-miR-15b-5p, hsa-miR-195-5p, hsa-miR-221-3p, hsa-miR-140-3p, and hsa-miR-422a) showed differential expression patterns in lung tissues before and after infection. Especially, hsa-miR-15b-5p and hsa-miR-195-5p showed a large difference in expression, indicating that they may potentially be diagnostic biomarkers for SARS-CoV-2 infection.
