Functional Implications of Transporters Under Nitrosative Stress Conditions

  • Nitrosative stress is defined as pathophysiological conditions that are related to covalent modifications of proteins by nitration/nitrosylation by forms of nitrogen oxide ($NO_x$), leading to DNA damage, ultimately, cell death. This type of stress condition appears to be associated with a number of disease states, including diabetes, inflammation and neurodegenerative diseases. Since these pathological conditions are frequently chronic in nature and, thus, require long-term treatment, changes in pharmacokinetics are likely to affect the therapy. Transporters are membrane proteins that facilitate the movement of substrates, including drugs, across plasma membranes of epithelial / endothelial cells. Since it is now increasingly evident that transporters are pharmacokinetically significant, functional alteration of transporters by this stress condition may have therapeutic relevance. In this review, experimental techniques that are used to study both in vivo and in vitro nitrosative stress are summarized and discussed, along with available literature information on the functional implication of transporters under conditions of nitrosative stress conditions. In the literature, both functional induction and impa irment were apparently present for both drug transporter families [i.e., ATP-binding cassette (ABC) and solute carrier families (SLC)]. Furthermore, a change in the function of a certain transporter appears to have temporal dependency by impairment in the early phase of nitrosative stress and induction thereafter, suggesting that the role of nitrosative stress is complex in terms of functional implications of the transporters. Although the underlying mechanisms for these alterations are not fully understood, protein nitration/nitrosylation appears to be involved in the functional impairment whereas transcript factor(s) activated by nitrosative stress may play a role, at least in part, in functional induction. Interestingly, functional induction under conditions of nitrosative stress has not been observed for SLC transporters while such impairment has been documented for both ABC and SLC transporters. Further investigations appear to be necessary to fully delineate the underlying reasons for these differences on the impact and importance of nitrosative stress conditions.

타악기 연주 프로그램이 시설노인의 우울감과 자아존중감에 미치는 영향 (The Effects of Percussion Program on Depression and Self-esteem of the Elderly Living in Residential Care)

  • This study was to investigate the effects of percussion program on depression and self-esteem for the elderly lived in residential care. 12 older adults participated in the 40-min. percussion program for 4 weeks(10 sessions). The percussion program consisted of three steps; 1) introduction for creating rapport and relationship, 2) process for mood induction and increasing interaction, and ending for increasing self-achievement and confirming the changed mood. Korean Geriatric Depression Scale (KGDS) and Self-Esteem Scale (SES) were administered before the first session and after the completion of the program. Results showed statistical significance (p<.05) in KGDS (p=.020) and SES (p=.025). Findings imply that percussion program can induce positive changes in depression and self-esteem for elderly in the residential care, and more depressed elderly group may have better outcome in percussion playing. Future studies should consider various levels of depression among elderly, and design the program to be more sensitive to the level of functioning.

Investigation of the Antioxidant Status in Multiple Myeloma Patients: Effects of Therapy

  • Background: Multiple myeloma is a malignant silent incurable plasma cell disorder. The present study aimed to assessed the activation of the oxidative stress pathway in afected patients Materials and Methods: Advanced oxidation protein products (AOPPs), malondialdehyde (MDA), adenosine deaminase (ADA), total antioxidant capacity (TAC) levels, glutathione, ascorbic acid (vitamin C), ${\alpha}$-tocopherol (vitamin E) in addition to related enzymes glutathione peroxidase (GSH-Px), glutathione reductase (GSH-R) and superoxide dismutase (SOD) were analyzed in sixty patients with multiple myeloma before and after one month treatment with induction therapy. Results: The results of the study showed a significant elevation in AOPPs, MDA, ADA levels in patients with multiple myeloma before and after treatment in comparison to healthy control samples In contrast TAC glutathione, vitamin C and E, and the antioxidant enzymes levels were decreased significantly. On comparing samples of MM patients after treatment, there was significant increase of TAC glutathione, vitamin C and E, and the antioxidant enzymes in parallel with decreasing AOPPs, MDA and ADA levels in comparison with samples of patients before treatment. Conclusions: The results indicate oxidative stress and DNA damage activity increase in MM and are alleviated in response to therapy.

  • Purpose : The purpose of this study was to investigate, The Effects of Massage and Taping on Muscle Strength and Flexibility and Pain with Delayed Onset Muscle Soreness(DOMS). DOMS is a common problem that can interfered with rehabilitation as well as activities of daily living. Methods : Thirty subjects untrained were randomly assigned into Massage group, Taping group, Massage and Taping group. Treatment were applied at 24 hours, 48 hours and 72 hours after induction DOMS. The effects of Massage and Taping and Muscle strength and flexibility on DOMS were evaluated by a goniometer and dynamometer and taping and VAS sheet. Results : The results of this study were as follow : 1) There was a significant difference among Taping group, Massage group, Taping and massage group in the case of using graduator. The interaction of group shown a significance (p<.05). 2) In the case of using goniometer, all three groups had the significant difference in period (p<.05) and the interaction of group-period shown a significance(p<.05). 3) In the case of using VAS, all three group had the significant differences in period(p<.05) and the interaction of group-period shown a significance(p<.05). Conclusions : These findings suggested that Taping and Massage had effect on DOMS.

  • Telomerase is a ribonucleoprotein complex, whose function is to add telomeric repeats $(TTAGGG)_n$ to chromosomal ends and is also known to play an important role in cellular immortalization. Telomerase is highly active in most tumor cells, yet not in normal cells. Therefore, it may have possible applications in cancer gene therapy. Telomerase consists of two essential components; a telomerase RNA template (hTR) and a catalytic subunit (hTERT). The current study attempted to inhibit the "open" part of the human telomerase RNA (hTR) with an antisense sequence-expressing adenovirus. It was found that the antisense telomerase adenovirus suppressed the telomerase activity, tumor cell growth, and survival in vitro. Furthermore, FACS analysis and TUNEL assay suggested that the reduce viability was mediated through the induction of apoptosis, indicating that this approach might be a useful method for suppressing cancer growth in targeted cancer gene therapy.

The Short-Term Effects of Difference Frequency of Transcutaneous Electrical Nerve Stimulation on Pain Relief using c-fos Expression in Spinal Cord with Knee Osteoarthritis Rats

  • PURPOSE: The purpose of this study was to determine the effect of different frequencies (4Hz and 100Hz) of transcutaneous electrical nerve simulation (TENS) on pain relief using c-fos expression in the spinal cord of rat osteoarthritis to investigate the appropriate frequency for pain relief. METHODS: Total of 30 Sprague-Dawley rats was used and randomly divided 2 groups according TENS frequency and applicate the TENS during 3 period (3 days, 7 days, 10 days). The induction of osteoarthritis by 3mg monosodium iodoacetat was injected into the right knee joint of rats. Three days later, commercially available TENS unit was used for stimulation was set to 20minutes on 3, 7, 10 days after surgery. Western blot analysis system was used to detect immunoreactive proteins. The thickness of the bands were photographically measured by Scion Image. RESULTS: When investigating the c-fos expression of TENS on spinal cord in OA knee over 10 days, between-groups differences in c-fos expression reached a significant level by day 10. For within-groups comparisons, the c-fos expression decreased significantly across days in low- and high-frequency TENS groups. CONCLUSION: Whether at low- and high-frequency, the TENS as a therapy obtained beneficial effects of pain relief and TNES at high-frequency is more beneficial effects on the pain relief when TENS applied at injury site.

  • A 7-year-old, spayed female, Yorkshire terrier dog with polyuria/polydipsia (PU/PD), interdigital edema, pruritus and abdominal enlargement was referred to the Veterinary Medical Teaching Hospital of the present university. Pituitary-dependent hyperadrenocortisim (PDH) was diagnosed by clinical signs, physical examination, laboratory examination (complete blood count, serum chemistry, urinalysis, ACTH stimulation test and high dose dexamethasone suppression test) and diagnostic imaging (radiography, ultrasonography and computed tomography). Clinical signs were improved after trilostane treatment, and maintenance therapy with trilostane still continued after successful induction therapy. Trilostane can be used as an alternative to mitotane therapy in hyperadrenocortisim in dogs.

Effect of Muscle Fatigue on the Proprioception by the Taekwondo Training Type

  • PURPOSE: This study examined the effects of muscle fatigue on proprioception in the type of taekwondo training, causing injury to taekwondo athletes. METHODS: The subjects were divided into Taekwondo Kyorugi athletes (n = 14) and Taekwondo Poomsae athletes (n = 14). After inducing muscle fatigue to the subjects, the joint position sense (JPS) and the force reproduction sense (FRS) were measured before and immediately after the induction of muscle fatigue. RESULTS: Studies have shown no significant impact on the JPS and FRS caused by muscle fatigue induced by Poomsae and Kyorugi (p > .05). The JPS, however, showed a significant difference before and after Kyorugi (p < .05). The FRS showed significant differences before and after Poomsae (p < .05). Through this study, the injuries to taekwondo athletes appear to be influenced by the joint position and force reproduction. Moreover, the injury appears to affect the joint position sense in Kyorugi athletes and the force reproduction sense in Poomsae athletes. CONCLUSION: The results will contribute to injury prevention and athlete protection. In addition, the information can be used as basic data for the development of sports injuries prevention and rehabilitation programs for Taekwondo athletes. Research is needed on the customized management of sports methods and self-care to prevent sports damage, considering the athletic characteristics of the taekwondo athletes in the future.

Change in the treatment strategy for pediatric Crohn's disease

  • Crohn's disease is characterized by chronic inflammation involving any portion of the gastrointestinal tract. Treating Crohn's disease is a major challenge for clinicians, as no curative therapy currently exists. Pediatric Crohn's disease is characterized by frequent relapses, a wide extent of disease, a high prevalence of extraintestinal manifestations, and a severe clinical course. The classic therapeutic approach is known as the 'step-up' strategy, and follows a progressive course of treatment intensification as disease severity increases. Although this approach is usually effective for symptom control, many patients become either resistant to or dependent on corticosteroids. The efficacy of infliximab suggests that, rather than a progressive course of treatment, early intense induction may reduce complications associated with conventional treatment and improve quality of life. Intensive early therapy with infliximab is known as the 'top-down' strategy. Such therapy offers the potential for altering the natural history of Crohn's disease, and is changing treatment paradigms. However, the relatively new concept of an early aggressive or 'top-down' treatment approach is not yet widely accepted, especially in pediatric patients. The results of our current study demonstrate that early and intensive treatment of pediatric Crohn's disease patients with infliximab, at initial diagnosis, was more effective for maintaining remission and reducing flares.
