  • pH- and thermo-sensitive hydrogels containing maleilated chitosan(MC) and N-isopropylacrylamide(NIPAAm) were prepared and characterized for their swelling behavior, biodegradability and drug release profiles. The hydrogels exhibited a typical pH-sensitivity due to the carboxylic acid groups of maleilated chitosan. The change of ratio of NIPAAm to MC in weight did not affect on either lower critical solution temperature(LCST) or EWC significantly. The pH sensitivity of the hydrogel, however, depended on the amounts of carboxylic acid groups of MC. MC was degradable up to 80% weight reduction in 2 hours. The in vitro drug relase profiles were established both in buffer solution pH 1.4 and pH 7.4. Only a negligible amount of indomethacin was released at pH 1.4 in 6 hours, while at pH 7.4 more than 90% of the total drug in the hydrogel was gradually released over ca. 5 hours.

  • These studies were carried out to investigate the effect of Jungchuntang water extract on the inhibitory contractile action of acetylcholine in rat. The results of these studies were as follows; 1. Contractile force of acetylcholine from trachea smooth muscle in rat was significantly inhibited by Jungchuntang. 2. Dose-response of acetylcholine pretreated Jungchuntang in rat was not significantly changed. 3. Inhibitory contractile action of acetylcholine pretreated propranolol in rat was decreased. 4. Inhibitory contractile action of acetylcholine pretreated indomethacin was not significantly changed by Jungchuntang. 5. Inhibitory contractile action of acetylcholine pretreated methylene blue was significantly changed by Jungchuntang. 6. Effects of Yupoongyangyeong-tang(YYT) on the Changes of Regional Cerebral Blood Flow and Mean Arterial Blood Pressure in Rats (유풍양영탕(愈風養營湯)이 정상 흰쥐의 국소 뇌혈류량 및 평균혈압에 미치는 영향(影響))

  • Objectives: The present study was carried out to investigate the effects of Yupoongyangyeong-tang(YYT) on changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in rats. In addition, the author also investigated action mechanisms of YYT on changes in rCBF and MABP. Methods and Results : In this study, treatment with YYT elevated rCBF in a dose-dependent manner, but MABP levels were elevated only in the 10 mg/ml treatment group. Pre-treatment with indomethacin. an inhibitor of cyclooxygenase, inhibited increase of rCBF effectively. Pre-treatment with methylene blue, an inhibitor of guanylate cyclase, inhibited increase of rCBF induced by YYT, too. In addition. pre-treatment with indomethacin also inhibited increase of MABP. However, pre-treatment with methylene blue did not affect MABP levels. Conclusion : These results suggest that YYT is useful to treat patients with diseases related to cerebral ischemia, because YYT can increase rCBF. In addition, the mechanisms are thought to be related to both cyclooxygenase and guanylate cyclase.

The Mechanism Study of Prescription for Treatment Abundant Expectoration due to Deficiency of Qi on Brain Disease in Rats (기허담성치방이 뇌병환에 미치는 기전연구)

  • This Study was designed to investigate the mechanism of Prescription for Treatment Abundant Expectoration due to Deficiency of Qi(Yukgunja-Tang, YGT) on cerebral hemodynamics [regional cerebral blood f1ow(rCBF) and pial arterial diameter(PAD)] in cerebral ischemia rats. The results were as follows: Both rCBF and PAD were significantly and stably decreased by YGT (10㎎/㎏, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in Control group. Pretreatment with indomethacin(1㎎/㎏, i.p.), an inhibitor of cyclooxygenase and methylene blue(10㎍/㎏, i.p.), an inhibitor of guanylate cyclase significantly but unstably increased the YGT-induced increases in rCBF during the period of cerebral reperfusion. Pretreatment with indomethacin significantly and stably decreased the YGT-induced increases in PAD during the period of cerebral reperfusion, but pretreatment with methylene blue increased unstably the YGT-induced increases in PAD during the period of cerebral reperfusion. In conclusion, the present authors thought that mechanism of YGT on cerebral hemodynamics was connected with guanylate cyclase in cerebral ischemia rats.

  • This study was attempted to investigate the effect of Machili Cortex extract(Machilus thunbergii Sieb. et Zucc. ) on digestive system in experimental animals. EtOH and MeOH extracts(E.E. and M.E.) were found to inhibit the charcoal transport rate in the small intestine of mice. E.E. exhibited the inhibition of spontaneous movement(motility) and tension in the ileum and colon strips of rabbit, and these actions were inhibited by action of acetylcholine. E.E. and M.E. given intraduodenaly(i.d.) exhibitied the significant decrease of gastric acid secretion in pylorus-ligated rats. E.E. and M.E. inhibited the formation of some experimental gastric ulcers(pylorus ligation-ulcer i.d., indomethacin-induced ulcer p.o. and aspirin-induced ulcer p.o. ) in rats, which are considered to relate to a protective action. E.E. and M.E. caused remarkable increase of bile excretion, compared with normal-control group, when adminstered through i.d., i.v. and p.o. The antibacterial activity against several intestinal microorganisms and other bacteria in vitro test was observed in the administration of E.E. and M.E.

  • This study was carried out to investigate the effects of Gooboeum extract on the inhibitory contractile action of acetylcholine in the control and sensitized rat. The results were obtained as follows: 1. The acetylcholine contractile force of the trachea smooth muscle with epithelium was significantly relaxed by Gooboeum. 2. Dose-response of acetylcholine from the trachea smooth muscle pretreated Gooboeum was not changed. 3. Effect of Gooboeum on the inhibitory contractile action of trachea smooth muscle pretreated propranolol was not significantly inhibited. 4. The inhibitory contractile action of acetylcholine of trachea smooth muscle pretreated indomethacin was not significantly changed by Gooboeum. 5. Effects of Gooboeum extract on the inhibitory contractile action of acetylcholine of trachea smooth muscle pretreated methylene blue was not significant. 6. The contractile force of acetylcholine of the trachea smooth muscle without epithelium was significantly inhibited by Gooboeum. 7. Dose-response of acetylcholine of the trachea smooth muscle pretreated Gooboeum was not significant. 8. Effects of Gooboeum extract on the inhibitory contractile action of acetylcholine of the trachea smooth muscle pretreated propranolol was significantly inhibited. 9. Effects of Gooboeum extract on the inhibitory contractile action of acetylcholine of the trachea smooth muscle pretreated indomethacin decreased. 10. Effects of Gooboeum extract on the inhibitory contractile action of acetylcholine of the trachea smooth muscle pretreated methylene blue was not significant.

  • An experimental studies were done to investigate the spontaneous movements to the isolated ileum with the liquid extracts of Wahngkigunchunghapleechungtang. Then the effects of isolated ileum, pylorus-ligated and indomethacin-induced ulcer, gastric juice secretion, and transport abilities of the small and large intestine were studied. The following results have been obtained; 1. Wahngkigunchunghapleechungtang showed to have an inhibitory effects on the smooth muscle contraction of the isolated ileum by acetylcholine chloride and barium chloride. 2. Wahngkigunchunghapleechungtang showed to have an inhibitory effect on the contraction of the gastric fundus-strip by acetylcholine chloride and barium chloride. 3. The preventive effects were not recognized on the pylorus-ligated ulcer, by administration of Wahngkigunchunghapleechungtang. 4. The preventive effects were recognized on the indomethacin-induced ulcer by administration of Wahngkigunchunghapleechungtang. 5. The Inhibitory effects on the secretion gastric juice, free & total acidity, and pepsin output were recognized by administration of Wahngkigunchunghapleechungtang. 6. The transport abilities in the small intestine were not recognized by administration of Wahngkigunchunghapleechungtang. 7. The transport abilities in the large intestine were increased by administration of Wahngkigunchunghapleechungtang. According to the above results, it is considered that the experimental effects of Wahngkigunchunghapleechungtang has correspond to the oriental medical literatures.

Anti-pyretic and anti-inflammatory activity of chloroform extract of Croton roxburghii in standard animal models

  • The chloroform extract of Croton roxburghii (Family: Euphorbiaceae) was evaluated for its antipyretic effects in Brewer's yeast induced hyperthermia in rats. The anti-inflammatory effect of the Croton roxburghii was also evaluated by using carrageenan, dextran, histamine, serotonin induced rat paw oedema and cotton pellet induced granuloma (chronic) models in rats. The chloroform extract of Croton roxburghii (CECR) exhibited significant anti-pyretic and anti-inflammatory effect at the dose 50, 100 and 200 mg/kg. Maximum inhibition (55.32%) was notedat the dose of 200 mg/kg after 3 h of drug treatment in carrageenan induced paw oedema, whereas the Indomethacin (standard drug) produced 61.33% of inhibition. The extract exhibited significant anti-inflammatory activity in dextran induced paw edema in a dose dependent manner. In the chronic model (cotton pellet induced granuloma) the CECR (200 mg/kg) and Indomethacin (10 mg/kg) showed decreased formation of granuloma tissue by 52.32% and 56.32% respectively. The extract also exhibited a significant antipyretic response in Brewer's yeast induced pyrexia in rats. Thus, the present study revealed that the CECR exhibited significant antipyretic and anti-inflammatory activity in the tested animal models.

Inhibition of Experimental Gastric Ulcer by Potato Tubers and the Starch

  • In an attempt to establish anti-ulcerogenic activity of potato tubers, inhibitory activity against ethanol- and indomethacin-induced gastric ulcer models in rats was evaluated for the first time. From several varieties of potato tubers including Solanum tuberosum L. cv. Superior (white skin and fresh potato) and two new varieties of (Bora valley and Gogu valley), raw potato juice was prepared and the starch was obtained from each juice by filtration and drying. Upon oral administration to rats, raw potato juice showed more or less inhibitory activity. The starch showed higher and dose-dependent inhibitory activity, suggesting that the active ingredient in raw potato juice may be the starch. Particularly, the starch obtained from the tubers of new potato variety, "Bora valley," with purple color, showed the highest inhibitory activity (62.4% and 37.1% inhibition of ulcer index at 500 mg/kg), while omeprazole (proton pump inhibitor) used as a reference drug showed 74.4% and 75.7% inhibition at 20 mg/kg against ethanol- and indomethacin-induced ulcer formation, respectively. The present study provides a first evidence of anti-ulcerogenicity of raw potato juice and the starch. Especially, the starch from "Bora valley" strongly inhibited ulcer formation in rats. Considering that these are food components, they may be safely used for anti-ulcerogenic nutraceuticals.

Gastric Protective Effects of Banhasasim-tang on Indomethacin-treated Rats

  • Purpose : Banhasasim-tang (BHSST) has been applied for treating the symptom of gastric stuffiness, which is similar to dyspepsia. The object of this study was to observe the healing effect of BHSST on the indomethacin (IND)-induced gastric ulcer in rats. Methods : Three different dosages of BHSST (400, 200 and 100 mg/kg) were orally administered 30 min before IND treatment; 6 hrs after IND treatment, the changes on the gross lesion scores, fundic histopathology, myeloperoxidase (MPO) activity, lipid peroxidation and antioxidant defense system (glutathione contents, catalase (CAT) and superoxide dismutase (SOD) activities) were observed, and compared with the activity of the synthetic anti-ulcer drug, a representative proton pump inhibitor omeprazole (OME) 10 mg/kg. Results : All three different dosages of BHSST treatment in the IND-induced gastric ulcer rats, significant and dose dependent decreased gastric damages - hemorrhagic gross lesions, gastric mucosa MPO levels and histopathological gastric ulcerative lesions - were detected as compared with the IND treated control rats. BHSST also strengthened the antioxidant defense systems - decreased the level of lipid peroxidation and CAT activity but increased the level of GSH and SOD activity, and BHSST 200 mg/kg showed similar anti-ulcerative effect as compared with OME 10 mg/kg. Conclusions : The results obtained in this study suggest that BHSST has favorable effects against IND-induced gastric damages, through significant and dose-dependent decreasing gastric damages and the strengthening of the body's antioxidant defense systems with direct anti-inflammatory effects.
