Characteristics of Cosmetic with Whitening Compounds from Phellodendron amurense (Phellodendron amurense의 미백물질을 이용한 화장품 특성)

  • The tyrosinase inhibitory activity of extracts from Phellodendron amurense was examined. Tyrosinase inhibitory activity of 60% ethanol extracts was determined as 25% and the inhibitory activity of 60% ethanol extracts against melanin biosynthesis in melanoma cell (B16F10) was 31.2%. The purified inhibitory compounds against tyrosinase by Sephadex LH-20, MCI-gel CHP-20 column chromatography from P. amurense was confirmed as obacunone by $^1H$-NMR, $^{13}C$-NMR and Fast atom bombardment (FAB)-Mass spectrum. The tyrosinase inhibitory activities of purified obacunone was respectively as 35.1%. The safety of essence with tyrosinase inhibitory compounds from P. amurense was also assayed by various safety profiles. First, pH and viscosity change of essence for 60 days were not detected. The essence also showed the stability against temperature and light for 60 days. All these findings suggest that extracts from P. amurense has a great potential as a cosmeceutical ingredient, which has a potent whitening effect.

Separation of $H_2$/$N_2$ Gas Mixture by PTMSP-PEI and PDMS-PEI Composite Membranes (PTMSP-PEI와 PDMS-PEI 복합막에 의한 수소/질소 혼합기체 분리)

  • Polymer membranes such as poly(1-trimethylsilyl-1-propyne)-polyetherimide (PTMSP-PEI) and poly(dimethylsiloxane)- polyetherimide (PDMS-PEI) composite membrane were prepared by solution casting method. To investigate the characteristics of these membranes, the analytical methods such as FT-IR, $^1H-NMR,$ DSC, TGA, GPC, and SEM have been utilized. The number-average (equation omitted) and weight-average (equation omitted) molecular weight of PTMSP were 477,920 and 673,329 respectively. The glass transition temperature ($T_g$) of PTMSP was $224^{\circ}C.$ The separation of the gas mixture ($H_2/N_2$) through the composite membranes were studied as a function of pressure. The separation factor (${\alpha}, {\beta},$ quation omitted) of the composite membranes used in this work increased as the pressure of permeation cell increased. The real separation factor (${\alpha}$), head separation factor (${\beta}$), and tail separation factor (equation omitted) of PTMSP-PEI composite membrane were 2.28, 1.17, and 1.96 respectively at ${\Delta}P$ 30psi and $25^{\circ}C.$ (${\alpha}, {\beta}$ and equation omitted of PDMS-PEI composite membrane were 3.70, 1.53, and 2.42 respectively at ${\Delta}P$ 30psi and $25^{\circ}C$.

Preparation and Characterization of Highly Pured Water-soluble Chitosan Oligosaccharides as Biomaterials (생체재료로서의 고순도 수용성 키토산 올리고당의 제조와 특성)

  • To develop water-soluble chitosan as an effient gene delivery carrier, chitosan oligosaccharides (COSs) with various molecular weights (MW) were studied for gene transfection agents. MWs of COSs fractionated by ultrafiltration techniques were identified as narrow MW distributions with the average MW ranging from 1 to 10 kDa through gel permeation chromatography (GPC) measurement depending on the applied ultrafiltration membranes. Their structural characterizations were analyzed by FTIR spectrophotometer and $^1H$ NMR. The degree of deacetylation was determined by UV spectroscopy showing the degree of deacetylation above 90%. The relative cell viabilities were maintained over 100% (10 mg/mL), independent of the MW of the fractionated COSs. The fractionated COSs of 10 mg/mL concentration with narrow MW distributions showed non-cytotoxicity in Caco-2 cells.

Upregulation of Mir-34a in AGS Gastric Cancer Cells by a PLGA-PEG-PLGA Chrysin Nano Formulation

  • Background: Nano-therapy has the potential to revolutionize cancer therapy. Chrysin, a natural flavonoid, was recently recognized as having important biological roles in chemical defenses and nitrogen fixation, with anti-inflammatory and anti-oxidant effects but the poor water solubility of flavonoids limitstheir bioavailability and biomedical applications. Objective: Chrysin loaded PLGA-PEG-PLGA was assessed for improvement of solubility, drug tolerance and adverse effects and accumulation in a gastric cancer cell line (AGS). Materials and Methods: Chrysin loaded PLGA-PEG copolymers were prepared using the double emulsion method (W/O/W). The morphology and size distributions of the prepared PLGA-PEG nanospheres were investigated by 1H NMR, FT-IR and SEM. The in vitro cytotoxicity of pure and nano-chrysin was tested by MTT assay and miR-34a was measured by real-time PCR. Results: 1H NMR, FT-IR and SEM confirmed the PLGA-PEG structure and chrysin loaded on nanoparticles. The MTT results for different concentrations of chrysin at different times for the treatment of AGS cell line showed IC50 values of 68.2, 56.2 and $42.3{\mu}M$ and 58.2, 44.2, $36.8{\mu}M$ after 24, 48, and 72 hours of treatment, respectively for chrysin itslef and chrysin-loaded nanoparticles. The results of real time PCR showed that expression of miR-34a was upregulated to a greater extent via nano chrysin rather than free chrysin. Conclusions: Our study demonstrates chrysin loaded PLGA-PEG promises a natural and efficient system for anticancer drug delivery to fight gastric cancer.

CD34 Monoclonal Antibody-Immobilization on Polyurethane Surface by Poly(PEGA-co-BMA) Coating (PEGA/BMA 공중합체의 코팅을 통해 CD34 단일클론항체가 고정화된 폴리우레탄 표면)

  • A polyurethane (PU) surface enabling in vivo endothelialization via endothelial progenitor cell (EPC) capture was prepared for cardiovascular applications. To introduce CD34 monoclonal antibody (mAb) inducing EPC adhesion onto a surface, poly (poly (ethylene glycol) acrylate-co-butyl methacrylate) and poly (PEGA-co-BMA) were synthesized and then coated on a surface of PU, followed by immobilizing CD34 mAb. $^1H$-NMR analysis demonstrated that poly(PEGA-co-BMA) copolymers with a desired composition were synthesized. Poly(PEGA-co-BMA)-coated PU was much more effective for the immobilization of CD34 mAb, comparing with PEG-grafted PU prepared in our previous study, as demonstrated by that surface density and activity of CD34 mAb increased over 32 times. Physico-chemical properties of modified PU surfaces were characterized by X-ray photoelectron spectroscopy (XPS), water contact angle, and atomic force microscopy (AFM). The results demonstrated that the poly(PEGA-co-BMA) coating was effective for CD34 mAb immobilization and feasible for applying to cardiovascular biomaterials.

Preparation and Characterization of Chitosan-coated PLGA Nanoparticle (키토산이 코팅된 PLGA 나노입자의 제조 및 특성)

  • In this study, poly lactic-co-glycolic acid (PLGA) nanoparticles (PNP) were prepared through double (w/o/w) emlusion and emulsifying solvent-evaporation technique using PLGA, which has biocompatibility and biodegradability. To maximize stability and bioavailability of the particles, chitosan-coated PLGA nanoparticles (CPNP) were prepared by charge interaction between PNP and chitosan. We demonstrated that CPNP can be utilized as a drug carrier of oral administration. The chemical structure of CPNP was analyzed by 1H-NMR and FT-IR, and all characteristic peaks appeared, confirming that it was successfully prepared. In addition, particle size and zeta potential of CPNP were analyzed using dynamic light scattering (DLS) while morphological images were obtained using transmission electron microscope (TEM). Thermal decomposition behavior of CPNP was observed through thermogravimetric analysis (TGA). In addition, the cytotoxicity of CPNP was confirmed by MTT assay at HEK293 and L929 cell lines, and it was proved that there is no toxicity confirmed by the cell viability of above 70% at all concentrations. These results suggest that the CPNP developed in this study may be used as an oral drug delivery carrier.

Synthesis and Photovoltaic Properties of a Low Band Gap Polymer for Organic Solar Cell (유기태양전지를 위한 작은 밴드갭 고분자의 합성과 광전특성)

  • We synthesized a low band gap alternating copolymer containing electron-rich units (i.e. dithienosiloles and benzodithiophenes) and electron-deficient units (i.e. difluorobenzothiadiazoles) for high performance organic solar cells. The polymer was prepared by the Stille coupling reaction and characterized using $^1H$ NMR, GPC, TGA, UV-visible absorption spectroscopy, and cyclic voltammetry. Solar cells were fabricated in a structure of ITO/PEDOT:PSS/polymer: $PC_{70}BM/Al$ with five different blending ratios of polymer and $PC_{70}BM$ (1:1.5, 1:2, 1:3, 1:3.5 and 1:4 by weight ratio). The best efficiency was achieved from the 1:3 ratio of polymer and $PC_{70}BM$ in the photoactive layer, and TEM revealed that there is an optimal nanoscale phase separation between polymer and $PC_{70}BM$ in the 1:3 ratio blend film.

Isolation of Growth Inhibition Substance on Food borne Microorganisms from Hypericum ascyron L. and Application to Food Preservation (물레나물(Hypericum ascyron L.)의 식중독 미생물 증식 억제 물질의 분리 및 식품적용)

  • The ethanol extract and n-hexane fraction from Hypericum ascyron L. showed strong growth inhibition at 25 ppm on 5 strains of Listeria monocytogenes for 72 hr at $32^{\circ}C$. The purified substance, H2-5-2 fraction, was isolated by silica gel column and preparative thin layer chromatography from n-hexane fraction of Hypericum ascyron L. The H2-5-2 fraction showed a strong bacteriostatic activity on 5 strains of L. monocytogenes at 10 ppm in tryptic soy broth, and the viable cell was reduced 1 log cycle compared to initial cell number. The n-hexane fraction of Hypericum ascyron L. showed strong growth inhibition at 25 ppm on Bacillus cereus and Staphylococcus aureus, and at 50 ppm on Vibrio parahaemolyticus for 72 hr. The purified antimicrobial substance, the H2-5-2 fraction, was assumed as high unsaturated sterol by $^1H-NMR$ and $^{13}C-NMR$. On application test using minced Alaska pollack and ground beef, the n-hexane fraction of Hypericum ascyron L. at the level of 250 ppm was applied at $32^{\circ}C$ and $5^{\circ}C$. At $32^{\circ}C$ storage condition, the antimicrobial substances did not reduced L. monocytogenes ATCC 19113, meanwhile at $5^{\circ}C$ storage condition, L. monocytogenes ATCC 19113 was reduced in viable number.

  • In this study, 1-(2-methoxyethyl)-3-methylimidazolium methanesulfonate ionic liquid has been synthesized, characterized and tested with respect to carbon dioxide absorption with the aim to use it as advanced absorbent materials in fossil fuel processing. The ionic liquid was synthesized by a one step method, low cost. The thermal and chemical stability of selected ionic liquid has been investigated by DSC, TGA and the structure was verified by $^1H$-NMR spectroscopy. The solubility of carbon dioxide in the methanesulfonate-based ionic liquids were measured using a high-pressure equilibrium apparatus equipped with a variable-volume view cell at 30, 50 and $70^{\circ}C$ and pressure up to 195 bar. The results show that carbon dioxide solubilities of 1-(2-methoxyethyl)-3-methylimidazolium methanesulfonate increased with pressure increasing and temperature decreasing, and the carbon dioxide absorption capacity showed 27.6 $CO_2/IL$(g/kg) at $30^{\circ}C$, 13 bar.

Mitochondrial Damage and Metabolic Compensatory Mechanisms Induced by Hyperoxia in the U-937 Cell Line

  • Experimental hyperoxia represents a suitable in vitro model to study some pathogenic mechanisms related to oxidative stress. Moreover, it allows the investigation of the molecular pathophysiology underlying oxygen therapy and toxicity. In this study, a modified experimental set up was adopted to accomplish a model of moderate hyperoxia (50% $O_2$, 96 h culture) to induce oxidative stress in the human leukemia cell line, U-937. Spectrophotometric measurements of mitochondrial respiratory enzyme activities, NMR spectroscopy of culture media, determination of antioxidant enzyme activities, and cell proliferation and differentiation assays were performed. The data showed that moderate hyperoxia in this myeloid cell line causes: i) intriguing alterations in the mitochondrial activities at the levels of succinate dehydrogenase and succinate-cytochrome c reductase; ii) induction of metabolic compensatory adaptations, with significant shift to glycolysis; iii) induction of different antioxidant enzyme activities; iv) significant cell growth inhibition and v) no significant apoptosis. This work will permit better characterization the mitochondrial damage induced by hyperoxia. In particular, the data showed a large increase in the succinate cytochrome c reductase activity, which could be a fundamental pathogenic mechanism at the basis of oxygen toxicity.
