High Frequency Regeneration of Plantlets from Seedling Explants of Asteracantha longifolia (L.) NEES

  • Plantlet regeneration in Asteracantha longifolia(L.) Nees (Acanthaceae), a medicinal herb has been achieved from seedling explants on basal MS medium. Three different seedling explants including node, internode and leaf segments on used. Of these three explant, leaf explants gave better response for both callus mediated organogenesis and direct multiple shoot induction. Number of explants showing differentiation of shout buds was higher on MS media supplemented with BA compared to kinetin. MS medium fortified with BA ($2.0mgl^{-1}$) and NAA ($0.5mgl^{-1}$) was found to be most suitable for both callus mediated organogenesis and elongation of shouts. The elongated shoots were successfully routed on MS medium fortified with NAA or IBA. Among them $0.1mgl^{-1}$ NAA or $0.2mgl^{-1}$ IBA provides better response for rhizogenesis. Regenerated plantlets were successfully established in soil where 85.4% or them developed into morphologically normal and fertile plants. RAPD profiling using four decamer primers confirmed the genetic uniformity of the regenerated plantlets and substantiated the efficacy and suitability of this protocol for in vitro propagation of A. longifolia.

In Vitro Propagation of Zantedeschia spp. through Shoot Tip Culture (경정배양에 의한 Zantedeschia spp.의 기내번식)

  • This experiment was conducted to propagate Zantedeschia spp. in vitro. The frequency of adventitious bud clusters (ABC) formation from shoot tips in Z. 'Best Gold' was high at more than 65% on media with 2.0∼5.0 mg/L BA or 0.1∼1.0 mg/L thidiazuron. The highest formation rate of ABC (75%) was obtained on medium containing 2.0 mg/L BA. Comparing to treatment of BA alone, combined one of BA and NAA did not stimulate the formation of ABC and the shoot regeneration from shoot tips. The proliferation of ABC from sections (0.7∼1.0 cm) of ABC occurred effective on medium with 2.0 mg/L BA. Shoots developed from the sections (0.7∼1.0 cm) of ABC grew and rooted favorably on media containing 1.0∼2,0 mg/L IBA. The shoots were multiplicated effectively on medium with 0.5 mg/L thidiazuron in Z. 'Childsiana', on medium with 3.0 mg/L BA in 2. 'Golden Affair', and on medium with 5.0∼10.0 mg/L BA in Z. 'Pacific Pink'.

  • Protoplast-source meterial and enzyme strength had a significant influence on protoplast yield from hybrid poplar, Populus alba ${\times}$ P. grandidentata. The yield of protoplasts from in vitro culture of 1 month-old plantlets was more than that from greenhouse grown 4 month-old stock plant. In vitro cultured plantlets regulary produced more viable protoplasts with E-I enzyme solution (0.5% cellulase and 0.1% macerase) than those with E-II enzyme solution (1.0% cellulase and 0.2% macerase) after overnight incubation. The mean yield of protoplasts from in vitro cultured plantlets was $4{\times}10^6$ with E-I enzyme solution. Cell division was observed in these protoplast cultures after 7-10 days. Protoplast-derived hybrid poplar cells survived over 3 weeks in culture and some continuous cell divisions were evident. Other aspects associated with protoplasts from in vitro cultured plantlet are also discussed.

  • This study was conducted to establishment of in vitro micropropagation through induction of multiple shoots from axillary bud culture in Calanthe discolor Lindl. Shoots initiation from axillary bud was the most effective on half strength MS medium supplemented with $0.5mg{\cdot}L^{-1}$ NAA and $2.0mg{\cdot}L^{-1}$ TDZ during four weeks of dark culture followed by culture under a 16-h photoperiod. Multiple shoots (12.5 shoots per explant) were proliferated on half strength MS medium containing $4.0mg{\cdot}L^{-1}$ TDZ and $2.0mg{\cdot}L^{-1}$ IBA. On the other hands, the abnormally emerged shoots during the multiple shoot proliferation stage were recovered to normal shoots on half strength MS hormone free medium. Multiple shoots were well elongated and rooted on half strength MS medium with $0.1mg{\cdot}L^{-1}$ NAA. The plantlets were acclimatized up to 100% on TKS substrate after pretreating with $10mg{\cdot}L^{-1}$ NAA for 30 min. and these plantlets showed good growth as well.

Improvement of asymbiotic seed germination and seedling development of Cypripedium macranthos Sw. with organic additives

  • To find the optimal propagation condition for endangered Cypripedium macranthos Sw., also known as lady's slipper orchid, the effect of various organic additives on in vitro germination, protocorm formation and seedling growth was investigated during asymbiotic seed culture. When $100ml{\cdot}L^{-1}$ coconut water was added to the basal medium, the highest germination rate and protocorm formation rate were achieved, with 70.8% and 74.2% respectively. Supplementation of phloem sap from birch tree or maple tree also showed a facilitating effect to improve the germination and protocorm development. With $100ml{\cdot}L^{-1}$ birch sap or maple sap, both the germination and protocorm formation rates were roughly more than 65% and 68%. The roots and buds of the seedlings grew vigorously in the medium containing $100ml{\cdot}L^{-1}$ coconut water or phloem sap, in particular, their bud formation rates increased by more than 70%. Addition of banana powder and peptone could not create a more significantly favorable culture condition, and non-addition had the worst results. Our results demonstrated that proper organic amendments such as coconut water and phloem sap might be preferred to in vitro germination and the growth of seedlings developed from the protocorm of C. macranthos Sw. during asymbiotic seed culture.

Virus Free Stock Production by In vitro Stem Cutting of Shoot Tip Cultures of Grapes (포도 경정배양에서 얻은 유묘의 기내삽목에 의한 무병묘 생산)

  • The experiment was conducted to know the effect of plant growth regulators on axillary bud elongation from in vitro stem cutting and the possibility of virus-free stock production. Axillary buds were well elongated in 3/4 strength MS medium supplemented with 0.1 or 0.5 mg/1 BA and 0.05 mg/1 NAA. Transferred plantlets could be established well in vermiculite and peat moss mixture (3:1, v/v) compare to other mixtures. In virus indexing, all the varieties of mother plants were infected by GLRV Ⅲ. Infected percentages of the three varieties were ranged from 30% to 75%. But negative response was revealed against the other species of virus, GLRV Ⅰ, GFLV and ArMV. Plantlet of 'Schuyler' and 'Muscat of Alexandria', which were cultured in vitro, showed positive response against GLRV Ⅲ and infected percentage of the former was 37.5% but the latter, 12.5%. On the other hand, that of 'Campbell Early' negativiely responded against all the species of virus indexed.

  • Apple (Malus×domestica Borkh.; Rosaceae) is an important fruit crop grown mainly in temperate regions of the world. Tissue culture in vitro is a biotechnological technique that has been used to genetically improve cultivars (scions) and rootstocks. This could be important in the production of genetically uniform scions and rootstocks for commercial apple production. In nurseries, apple plants are produced by grafting scions onto rootstocks. The Cornell-Geneva (Geneva® series) breeding program has bred several dwarf rootstocks that are resistant to diseases and pests and are also cold hardy. This study was conducted to determine the optimal medium strength to improve sprouting shoot rate of apical meristem of the apple rootstocks of fire blight resistance. The apple rootstocks apical meristem at size (0.2 mm to 0.3 mm) with axillary buds were cultured on the MS(Murashige & Skoog) medium supplemented with plant growth regulators. The sprouting ratio and growth characteristics was evaluated after eight weeks in vitro culture. The highest rate of bud differentiation and shoot formation were 23.8% and 55.6%, respectively. After 6 weeks, shoots were regenerated from apical meristem, and their growth characteristics was significantly varied on the respective basal medium with different plant growth regulators. Our studies showed that the apple rootstocks the apple rootstocks of fire blight resistance plantlets could be successfully produced from apical meristem differentiated out of young twigs via organogenic regeneration.

Lysophosphatidylcholine Enhances Chondrogenesis by the Modulation of Protein Kinase C Isoform Expression

  • Lysophosphatidylcholine (LPC) has been reported to be responsible for the sustained activation of protein kinase C (PKC). As chondroqenesis is known to be regulated by PKC, this study was performed to investigate the effects of LPC on chondrogenesis of chick limb bud mesenchymes in vitro. LPC treatment of mesenchymes during micromass culture significantly enhanced chondrogenic differentiation. The most effective time of LPC on the stimulation of chondrogenesis was the first day of micromass culture. Analysis of LPC effects on the expression of PKC isoforms revealed that LPC treatment increased expression of PKCa, among the multiple PKC isoforms, in the membrane fraction on day one of culture. The stimulatory effect of LPC on chondrogenesis was abolished if PKCa was down regulated by the prolonged treatment of cells with phorbol ester. The results suqqest that LPC promotes chondrogenesis through the activation of PKCa at the early stage of chondrogenic differentiation.

  • Present experiment were attempted to examine in vitro multiplication throughbud culture of Cornus officinalis. Bud derived shoot formation was established successfully on Murashige and Skoog's medium supplemented with $0.5mg\;/\;{\ell}$ BAP(N-benzyl amino purine). The shoot proliferation increased on the Driver Kuniyuki Walnut medium containing $0.5mg\;/\;{\ell}$ NAA(Napthalene acetic acid) and $0.5mg\;/\;{\ell}$ BAP. Addition of 2,4-D(2,4-Dichlorophenoxy acetic acid) to the media produced excessive callus inducton. IAA(Indole-3-acetic acid) and IBA (Indole-3-bu-tyric acid) enhanced multple shooting, and NAA showed callus induction and multiple shooting. Shoot growth was enhanced supplemented with 3% sucrose, $2g\;/\;{\ell}$ activated charcoal, and 1 / 4MS in organic salts. However, root formation of proliferated shoots was low about 5%

Correlative Effect of Adenine Sulphate and Benzylaminopurine on the Regeneration Potentialily in Cotyledonary Explants of Groundnut (Arachis hypogaea L.)

  • An efficient method of shoot regeneration of peanut is described. In vitro shoot organogenesis from the callus of cotyledon explants of Arachis hypogaea L. was stimulated by addition of Adenine sulphate (Ads) along with 6 - benzylaminopurine (BAP) and - napthalene acetic acid (NAA). Ads (13 ${\mu}{\textrm}{m}$) had a stimulatory effect on shoot bud differentiation when combined with BAP (13 ${\mu}{\textrm}{m}$) and NAA (2 ${\mu}{\textrm}{m}$). Shoot organogenesis was markedly higher (92%) from callus induced on Ads, BAP and NAA combined media than from those formed by the individual supplementation of Ads or BAP with NAA. The shoots elongated on the media with GA$_3$ (1 ${\mu}{\textrm}{m}$). Elongated plantlets rooted with MS media containing IBA (9 ${\mu}{\textrm}{m}$).
