Relationship Between Acrylamide Concentration and Enzymatic Activity in An Improved Single Fibrin Zymogram Gel System

  • Based on the zymography analysis, Bacillus sp. DJ-4 (screened from Doen-Jang, a Korean traditional fermented food) secretes seven extracellular fibrinolytic enzymes (EFEs; 68, 64, 55, 45, 33, 27, and 13 kDa) in culture broth. These seven EFEs were analyzed by newly applied SDS-fibrin zymography combined with gradient polyacrylamide (SDS-FZGP). This improved gel system was used with a 5-20% acrylamide gradient in a fibrin zymogram gel for the separation of proteins with molecular masses from below 10kDa to over 100kDa on one gel plate. Using this system, high molecular weight bands (HMWBs) were clearly and sharply resolved. We also examined the relationship between an acrylamide concentration and the enzymatic activity of EFE using densitometric analysis.

  • Background: Colleges of Pharmacy in Korea have increased the number of years required for the degree from 4 years to 6 years in 2009. These changes are demanding revolution of education in pharmacology. The center of gravity is moving from medicine to application, theory to work. Problem-Based Learning is "learn to learn" working cooperatively in groups to seek solutions to real problems. PBL process cheers students to think critically to find optimal solution using appropriate learning resources. Method: Eighteen students in college of pharmacy are selected to participate in PBL lecture and take part in the survey about the effect of PBL in clinical pharmacy education. Results: General satisfaction score was high as four on a five-point scale, and students have improved on anxious about communication since the PBL classes compared to before of the lecture (P=0.037). Conclusion: PBL could be applied in clinical pharmacy education to progress student's participation and satisfaction. In addition this is improved communication skill in medical team during pharmacy practice.

Change Analysis of Forest Area and Canopy Conditions in Kaesung, North Korea Using Landsat, SPOT and KOMPSAT Data

  • The forest conditions of North Korea has been a great concern since it was known to be closely related to many environmental problems of the disastrous flooding, soil erosion, and food shortage. To assess the long-term changes of forest area as well as the canopy conditions, several sources of multitemporal satellite data were applied to the study area near Kaesung. KOMPSAT-1 EOC data were overlaid with 1981 topographic map showing the boundaries of forest to assess the deforestation area. Delineation of the cleared forest was performed by both visual interpretation and unsupervised classification. For analyzing the change of forest canopy condition, multiple scenes of Landsat and SPOT data were selected. After preprocessing of the multitemporal satellite data, such as image registration and normalization, the normalized difference vegetation index (NDVI) was derived as a representation of forest canopy conditions. Although the panchromatic EOC data had radiometric limitation to classify diverse cover types, they can be effectively used t detect and delineate the deforested area. The results showed that a large portion of forest land has been cleared for the urban and agricultural uses during the last twenty years. It was also found that the canopy condition of remaining forests has not been improved for the last twenty years.   • The modern market is highly competitive. It has progressed from traditional competition between enterprises to competition between supply chains. To ensure that enterprise can form the best strategy consistently, it is necessary to evaluate the trust of other enterprises in the supply chain. First, this paper analyzes the background and significance of supply chain trust research, analyzes and expounds on the qualitative and quantitative methods of supply chain trust evaluation, and summarizes the research in this field. Analytic hierarchy process (AHP) is the most frequently used method in the literature to evaluate and rank criteria through data analysis. However, the input data for AHP analysis is based on human judgment, and hence there is every possibility that the data may be vague to some extent. Therefore, in view of the above problems, this study improves the global trust method based on chain iteration. The improved global trust evaluation method based on chain iteration is more flexible and practical, hence, it can more accurately evaluate supply chain trust. Finally, combined with an actual case of Zhaoxian Chengji Food Co. Ltd., the paper qualitatively analyzes the current situation of supply chain trust management and effectively strengthens the supervision of enterprises to cooperative enterprises. Thus, the company can identify problems on time and strategic adjustments can be implemented accordingly. The effectiveness of the evaluation method proposed in this paper is demonstrated through a quantitative evaluation of its trust in downstream enterprise A. Results suggest that the subjective preferences of and historical transactions together affect the final evaluation of trust.

Taurine-enriched Rotifers Improve the Growth and Swim Bladder Inflation of Yellowtail Seriola quinqueradiata Larvae (타우린 영양 강화 로티퍼 공급이 방어(Seriola quinqueradiata) 부화 자어의 성장 및 부레 팽창에 미치는 영향)

  • We investigated the effects of taurine-enriched rotifers on larval growth and swim bladder inflation of yellowtail Seriola quinqueradiata. Rotifers were enriched with a commercial taurine supplement at two levels (0 and 800 mg/L). The larvae (initial notochord length=3.98±0.24 mm) were fed the enriched rotifers in triplicate from two days post-hatch for five days. The average taurine contents of the taurine non-enriched and enriched rotifers were 0.35±0.01 and 4.77±0.05 mg/g dry matter, respectively. The weight gain and specific growth rate of the fish fed enriched rotifers with the taurine supplement at 800 mg/L significantly improved compared with those of fish fed rotifers without taurine enrichment (P<0.05). The swim bladder inflation rate of larvae fed taurine enriched rotifers significantly (P<0.05). The results of the present study indicate that yellowtail larvae benefit from taurine concentrations compared with those typically reported to feed on non-taurine supplemented rotifers. Furthermore, taurine-enriched food for fish larval effectively improved the growth performance and swim bladder inflation of yellowtail larvae.

Application of the Combination of Soybean Lecithin and Whey Protein Concentrate 80 to Improve the Bile Salt and Acid Tolerance of Probiotics

  • To improve the bile salt and acid tolerance of probiotics against gastrointestinal stresses, we investigated the effects of soybean lecithin and whey protein concentrate (WPC) 80 on the bile salt tolerance of Lacticaseibacillus paracasei L9 using a single-factor methodology, which was optimized using response surface methodology (RSM). The survival rate of L. paracasei L9 treated with 0.3% (w/v) bile salt for 2.5 h, and combined with soybean lecithin or WPC 80, was lower than 1%. After optimization, the survival rate of L. paracasei L9 incubated in 0.3% bile salt for 2.5 h reached 52.5% at a ratio of 0.74% soybean lecithin and 2.54% WPC 80. Moreover, this optimized method improved the survival rate of L. paracasei L9 in low pH condition and can be applied to other lactic acid bacteria (LAB) strains. Conclusively, the combination of soybean lecithin and WPC 80 significantly improved the bile salt and acid tolerance of LAB. Our study provides a novel approach for enhancing the gastrointestinal tolerance of LAB by combining food-derived components that have different properties.

A Study on Visitors' Satisfaction of International Peony Festival in Heze, China through IPA (IPA를 통한 중국 허쩌 국제 모란축제관광객의 만족도에 관한 연구)

  • Purpose - This study is a study on China's national flower, the Peony Festival', with a focus on the Heze International Peony Festival, which is called the "City of Peonies, to investigate the difference in the importance and achievement of festival items". Design/methodology/approach - IPA is used to identify items that need to be improved for festival satisfaction attributes, and to suggest the direction of the annual peony festival in the future. Findings - The difference in importance and achievement for each of the 17 items of the Heze International Peony Festival, there was a statistically significant difference between importance and satisfaction, except for the peony admission ticket. It was found that satisfaction was relatively low in these items as it was higher than the average value. Second, the first quadrant showed information on traffic communication, food taste, convenient facilities (parking lot, restroom, shelter), service delivery speed, service provider attitude, natural scenery at the festival site, road signs, and information boards. Third, the diversity of cultural performances appeared in the fourth quadrant. Finally, the second quadrant is an area of high importance but low achievement. Two items were derived: the environment of the entertainment venue and the diversity of the entertainment venue. Research implications or Originality - These are the items that led to the dissatisfaction of visitors to the festival, and seem to be the items that need to be improved the most urgently.

Improvement of the Quality of Dried Wild Vegetables by Micro Oil Sprayed Thermal Air Technique

  • Wild vegetables, such as Cirsium setidens, and Aster scaber, are commonly distributed as dried materials because the wild vegetables lose their freshness quickly after harvest and distribution. Dried wild vegetables require rehydration to use as a food ingredient, and the quality of rehydrated wild vegetables is affected by pre-drying and drying methods. Here, we introduce the newly developed pre-drying and drying method, termed "micro oil sprayed thermal air (MOTA) technique". The three wild vegetables processed by the MOTA technique showed improved rehydration rate and reduced time to achieve maximum rehydration rate. Color characteristics were also improved in C. setidens. These results indicate that the MOTA technique improves the overall quality of rehydrated wild vegetables. It is expected that our findings could enhance the marketability of dried wild vegetables by improving overall quality and reducing preparation time.

Dietary effects of melatonin on growth performance by modulation of protein bioavailability and behavior in early weaned rats and pigs

  • Melatonin, which is produced from tryptophan, exerts various biological functions, including the regulation of circadian rhythm, sedative agents, and antioxidant ability. Therefore, we conducted two experiments with early-weaned rats and pigs to investigate the antioxidant and sedative effects of melatonin. In the rat experiment, a total of 42 rats (21 days old) were used, and the antioxidant capacity was determined. Next, we used 120 early-weaned piglets (21 days old) to conduct a 5-week experiment to evaluate the reductive effect of melatonin on energy-wasting movement, including roaming and fight states. Dietary melatonin supplementation significantly improved growth in both rats and pigs compared to the control groups. Additionally, rats fed a melatonin-supplemented diet showed advanced antioxidant capacity with a decrease in hepatic malondialdehyde concentration compared to rats fed a basal diet. Moreover, dietary melatonin ingestion increased resting and feeding behaviors and reduced roaming and fight behaviors during Days 8-21 compared to the control diet group. Collectively, early weaned animals given dietary melatonin supplementation showed improved growth through upregulation of hepatic antioxidant capacity and minimization of energy-wasting behavior, including roaming and fight states, after pigs' social hierarchy establishment.

Productivity effects of Hanwoo genetic improvement program

  • A genetic improvement program in Korea was implemented to improve the performance of Hanwoo cattle by generating livestock with genetically desirable economic characteristics. In particular, in response to external changes, such as the expansion of Free Trade Agreement (FTA), the livestock genetic improvement program has increased farm income by improving the productivity and quality of Hanwoo cattle. Using production cost data from Statistics Korea, the total input and output indices of Hanwoo feeding cattle from 2008 - 2021 were estimated and the growth and productivity changes were analyzed. The productivity change measures results were used to estimate the cumulative effects of the Hanwoo genetic improvement program on quality improvement, another purpose of the program, using a finite distributed lag model. The average annual increase in output (market weight) of Hanwoo was 0.9%. However, total input increased by 1.6%, resulting in a 0.6% decline in total factor productivity. In contrast, the Hanwoo genetic improvement program contributed significantly to the production of high quality beef, rather than contributing to improved productivity of the cattle. Hanwoo carcass weight, which is used as a performance indicator for the livestock genetic improvement program, has significantly improved and is projected to increase at a slower rate. The collective findings indicate the need for new performance indicators that can comprehensively indicate the performance of the genetic improvement of Hanwoo.
