Immunoregulatory Action of OGAPI (오가피의 면역조절작용)

  • The purpose of this research was to investigate the immunoregulatory effect and the leukemia cell apoptosis of EtOH extract of OGAPI(OGP). The proliferation of cultured splenocytes, thymocytes and mesenteric lymph node cells were enhanced by the addition of OGP. Splenic and thymic T lymphocytes, especially TH and Tc cells were significantly increased in OGP-administered mice. OGP markedly increased the production of γ-interferon in mice serum and accelerated the phagocytic activity in peritoneal macrophages. OGP treatment enhanced the apoptosis of L1210 mouse leukemia and Jurkat, Molt4 human leukemia cells, and increased the expression of apoptosis-related ICE, c-myc, p53 gene in Jurkat cell. These results suggest that OGP have an immunoregulatory action and anti-cancer activity.

Effects of Schizandra chinensis fructus on the Immunoregulatory Action and Apoptosis of L1210 cells (오미자 면역조절작용 및 L1210 세포의 apoptosis 에 미치는 효과)

  • The effects of MeOH extracts of Schizandra chinensis fructus (SZX) on the immunoregulatory effect (lymphocyte proliferation, subpopulation, nitric oxide production, phagocytic activity) and apoptosis $(sub-G_1\;peak)$ of L1210 cells were examined. The proliferation of splenocytes and thymocytes were enhanced by the addition of $10\;{\mu}g/mL$ of SZX. SZX were administered p.o. once a day for 7 days in adult male BALB/c mice. SZX resulted in altering subpopulation of splenic B and/or T and thymic T lymphocytes, especially the number of $T_H$ cells were markedly increased by the treatment of SZX in vivo and in vitro. SZX treatment induced the apoptotic cell death in L1210 mouse leukemia cells. In addition, SZX accelerated the production of nitric oxide and phagocytic activity in peritoneal macrophages. These results suggest that SZX have an immunoregulatory property and anti-cancer action.

Synthesis of Thienopyrimidine Derivatives as Inhibitors of STAT3 Activation Induced by IL-6

  • A series of thienopyrimidine compounds (6Aa-g and 6Ba-d) were synthesized and characterized by NMR spectroscopy and mass spectrometry. These compounds (6Aa-g and 6Ba-d) potently inhibited STAT3 expression induced by IL-6 in a dose-dependent manner with $IC_{50}$ values of $5.73-0.32{\mu}M$. Among the prepared thienopyrimidine derivatives, 6Aa, 6Ab, 6Ba and 6Bc significantly suppressed the phosphorylation of STAT3 and ERK1/2 stimulated by IL-6 in Hep3B cells. Furthermore, the synthesized compounds might be useful remedies for the treatment of inflammatory diseases by inhibiting the action of IL-6.

  • This study was performed to evaluate the immunoregulatory effects of Gungchihwadam-Jeon(궁치화담전, GCHDJ) and Cheongsinhwadam-Jeon(청신화담전, CSHDJ) was administered(p.o.) once a day for 14 days to mice. By the treatment of GCHDJ or CSHDJ was increased the cell viability of cultured mice splenocytes, thymocytes and mesenteric lymph node cells. Administration of GCHDJ or CSHDJ was increased the splenic T lymphocyte, especially the Tc cell subpopulation was increased by the GCHDJ, on the while the TH cell was increased by the CSHDJ. The administration of GCHDJ or CSHDJ was significantly increased the apoptosis of transplanted-L1210 leukemia cells to mice peritoneal cavity. These results suggest that GCHDJ and CSHDJ have an immunoregulatory action

  • The purpose of this research was to investigate the immunoregulatory effect, apoptosis of L 1210 leukemia cells of Comus officinalis Sieb.et Zucc. The proliferation of cultured splenocytes and thymocytes were enhanced by the addition of SSY. Splenic, thymic and mesenteric lymph node-T lymphocytes, especially TH cells was significantly increased in SSY-administered (p.o. for 7 days) mice. SSY treatment induced the apoptosis of L1210 mouse leukemia cells. In addition, SSY accelerated the phagocytic activity and nitric oxide production in peritoneal macrophages. These results suggest that SSY have an immuno-regulatory property and anti-cancer effect.

  • The purpose of this research was to investigate the effects of Chungjokupye-tang(CJKPT) on the anti-carcinogenic action. The cell viability of mouse spienocytes and thymocytes were enhanced by the addition of CJKPT. CJKPT were increased of splenic and thymic T lymphocytes, such as T/sub H/ cells were markedly increased by the treatment of CJKPT in vivo. CJKPT treatment induced the apoptotic cell death of Jurkat and HL60 leukemia cells. CJKPT reduced mitochondrial transmembrane potential and increased the expression of ICE, c-myc and p53 gene in Molt-4cells dose dependant manner. These results suggest that CJKPT have an anti-carcinogenic action via immunoregulatory mechanism.

  • The purpose of this research was to investigate the effect of Maekmoondong-tang(MMDT) on the activity of immune cell. The addition of MMDT(10 ㎍/㎖) enhanced the proliferation of cultured-splenocytes and thymocytes. And administration of MMDT(250, 500 ㎎/㎏) accelerated subpopulation of splenic T lymphocytes in BALB/c mice. In contrast, the treatment of the high concentration (500 ㎎/㎏)of MMDT were decreased thymic T lymphocytes. Administration of MMDT(250, 500 ㎎/㎏) eminently enhanced the production of IFN-γ. And MMDT did not affect the cell viability of Jurkat leukemia cells. These results suggest that MMDT have a immunoregulatory effect via enhanced cell mediated immunity

  • The purpose of this research was to investigate the effects of Seungyangikkitang (SIT) on the immune cells in BALB/c mice. SIT (500mg/kg) was administerd p.o. once a day for 7 days. SIT enhanced the proliferation of thymocytes, but decreased the proliferation of splenocytes. SIT enhanced the subpopulation of cytotoxic T cells in thymocytes and helper T cells in splenocytes, but did not affect the subpopulation of B220/Thy1 cells. SIT enhanced the production of γ-interferon and interleukin-2 in thymocytes, splenocytes and serum, but did not affect the production of interleukin-4. SIT suppressed the production of nitric oxide, but enhanced the lucigenin chemiluminescence and the engulfment of FITC-conjugated E. coli particles in peritoneal macrophages. These results suggest that SIT has a potent activity on the specific immunity via the cytokine secretion of Th1 cells and the non-specific immunity via the phagocytic activity of macrophages in vivo.

Effects of Substance P on the Activities of Immune Cell (면역세포 활성에 대한 Substance P의 영향)

  • The neuropeptide substance P(SP) has been recognized to modulate immune systems, with close proximity between peptidergic sensory nerve endings and immune cells. These include the macrophage and neutrophil activation, IL-2 production in T cell, augmentation of Ig synthesis, mast cell degranulation, $PGE_2$ and collagenase secretion in synoviocytes. In this study I examined SP-induced various biological activities such as antimicrobial action, cytokine production, and mast cell degranulation in the presence or absence of other inflammatory cell activators. Antimicrobial studies showed that undifferentiated HL-60 cells were not affected by SP. However, SP significantly enhanced antimicrobial action of TPA-treated or dbcAMP-treated HL-60 cells which had been differentiated into PMN or macrophage/monocyte. I could not find synergistic relationship between SP and LPS in parallel experiments of the above. SP did not induce IL-l production from murine macrophage cell line RAW264.7 whether costimulated with LPS or not. Mast cell degranulation was occured only when stimulated with high dose ($10^{-5}M$) of SP and the degree of this activation was slightly reduced by simultaneous application of $MIP-1{\alpha}$. In addition, CGRP which is known to be a common coexisting neuropeptide with SP within specific fibers did not augment the function of SP on mast cell degranulation. These results suggest that immunoregulatory activities of SP could be mediated through direct upregulation of various functions of immune cells and also upregulation of responsiveness of immune cells to other immune activators.

  • Abnormalities of the T cell subsets have been detected in the immunologically mediated disease sites such as periodontal lesions which are attributable to the regulatory effect of cell differentiation and specific chemokinetic effect of various cytokines. Macrophage Inflammatory protein$(MIP)-1{\alpha}$ and gammain terferon$({\gamma}-IFN)$ serve as important immunoregulatory molecules through which growth and differentiation of specific T cell subsets are known to be negatively regulated. Murine cytolytic T cell line CTLL-2 were used to perform the [$^3H$]-thymidine incorporation test, by which we obtained more comprehensive view in regulatory actions of cytokines on the T cell subset proliferation. 1. $rMIP-{\alpha}$(200ng/ml) and $r{\gamma}-IFN$(100U/ml) appreared to suppress the proliferation rate to CTLL-2 by 74 and 86% respectively, and the suppressive action of two cytokines were synergisic. 2. Culture supernatant of anti-CD3 mAb-stimulated mouse splenocyte enhanced the proliferation rate of CTLL-2 up to 10-fold with dose-dependent manner. However, culture supernatant of unstimulated splenocyte showed only 2-fold increase in the proliferation rate. 3. CTLL-2 cell proliferation was strictly IL-2 dependent.
