포공영(蒲公英)에 의한 뇌 소교세포에서 산화질소 (NO)의 생성 (Nitric Oxide Production in Brain Microglial Cells by Taraxacum officinale)

  • Nitric oxide (NO) is now recognized as a mediator of several biological and immunological functions, but unlike classical neurotransmitters. NO simply diffuse of the postsynaptic cells and around affecting cells. Taraxacum officinale (Compositae) has been used for maintenance of vitality, and they still occupy an important place in the traditional Korean medicine. We have examined that the effect of Taraxacum officinale water extract on NO synthesis in microglial cells of murine's brain, using the Griess method. And this study was evident that Taraxacum officinale did not induce NO production without recombinant interferon gamma ($rIFN-{\gamma}$), whereas Taraxacum officinale (10-1000 g/ml) with $rIFN-{\gamma}$ effectively produced NO in microglial cells of brain. As result. NO production in microglial cells increased most significantly in dose of 100 g/ml of the Taraxacum officinale and the production of NO was dependent on the dose of Taraxacum officinale, NG-monomethyl-L-arginine, competitive inhibitor of NO synthase, reduced the NO production by Taraxacum officinale stimulation with $rIFN-{\gamma}$ in microglial cells of murine. The effect of Taraxacum officinale was mainly dependent on Taraxacum officinale-induced tumor necrosis factor- secretion. Conclusively, this study suggested that Taraxacum officinale stimulate NO production at microglial cells in brain, which may be an important factor for mediating immune and neuroendocrinologic regulation in nervous system.

석창포가 알레르기성 비염 유발 동물 모델에 미치는 영향 (The Effects of Acorus gramineus on Changes of Nasal Tissue in Allergic Rhinitis Model)

  • Background & Objectives : Rhinitis is an inflammation of nasal mucosa. The major symtoms are watery rhinorrhea, sneezing, itchy nose, and nasal obstruction. Allergic rhinitis is an immune reaction by allergen. So, we aimed to determine therapeutic effects of Acorus gramineus by observing changes in IL-4, $IFN-{\gamma}$ and the nasal mucosal tissue. Materials and Methods : Fifteen BALC/c mice were divided into three groups : m group(which ate low concentrated herbal medicine ), M group(which ate high concentrated herbal medicine) and control group. Control and experimental group were induced allergic rhinitis by Ovalbumin as the method of Levin and Vaz. Experimental group was orally administered the Acorus gramineus extract for 28days. We observed changes in IL-4, $IFN-{\gamma}$ and trans aminase(AST, ALT) in blood and nasal mucosa and submucosa. Results : There were no significant changes statistically in IL-4 and $IFN-{\gamma}$ in blood(p<0.05). And there were no hepatotoxicity with Acorus gramineus extract. Histologically, almost no inflammatory response in treatment group(m,M) against that there were inflammatory response(increased goblet cells, dilated vessels, edema of bowman's glands and injured olfactory hairs) in control group. Conclusion : According to above results, it is supposed that Acorus gramineus has no immunological effects on allergic rhinitis.

  • Cefodizime has originally been developed for treating infections as antibiotics. However. according to some of recent studies. cefodizime. a third generation cephalosporin. may potentially have the capability of stimulating chemotactic activity of neutrophils and monocytes as well as the strong immuno-modulator. In this study. we studied to learn about the expressive effect of dentritic cells and macrophage. With this background. We have studied to see if cefodizime can be a potential substance inducing an immunological function in dendritic cells and peritoneal macrophages. IL-12 activates NK cell and macrophage, and shows antiviral effect by excreting INF-${\gamma}$. In vitro. total RNAs were extracted from murine dentritic cell at 4, 8, 12, 24hr after the application of 10, 50, 100${\gamma}g$/ml of cefodizime wighout other stimulators. And we analyzed IL-12 mRNA using RT-PCR method. In conclusion. IL-12 mRNA was increased. and the results suggest that cefodizime activate TH1 cell induction, CTL differentiation as well as accelerating the increase of NK. LAK cell.

The cover of an ear thermometer probe as a split-thickness skin graft mold in external auditory canal reconstruction

  • Maintaining the patency of the external auditory canal (EAC) during reconstruction is important because of its physiological role in hearing and immunological protective functions. The curved shape of the EAC presents a challenge when performing a skin graft. One of the key points for a successful skin graft is to ensure compression on the wound bed, and many novel methods, including prefabricated ear molds, have been reported for this purpose. In this study, we present a case of a skin graft performed to reconstruct a skin defect following excision of actinic keratosis in the EAC, using the cover of an ear thermometer probe as a mold for the graft to match the curvature of the EAC. This is an economical and practical method for secure compression dressing of a skin graft in the EAC.

  • Infection with human cytomegalovirus (HCMV) is of considerable clinical relevance after placental transmission and in immunosuppressed patients such as transplant recipients or patients with AIDS. The rapid detection method of HCMV has been required to overcome the time-consuming methods such as classical plaque assay or other immunological methods. This study was performed to establish the in situ ELISA, in which human lung fibroblasts infected with HCMV were fixed and used directly as antigen in 96 well culture plate. Expressed HCMV antigens were detected with HCMV-specific monoclonal antibodies. This method could detect HCMV dose-dependently upto $3{\times}10^2\;pfu/ml$. Antiviral activity of ganciclovir could be assayed within the known range of effective dose. This result showed that HCMV could be quantitated by in situ ELISA. The chemical, which was selected on the basis of component analysis in natural product, was tested to have the anti-HCMV activity by in situ ELISA, and three among five samples were found to have anti-HCMV activity with the dose-dependent manner. Conclusively in situ ELISA could be useful method for quantitation of HCMV and screening antiviral activity of samples to HCMV.

표면 처리 방법에 따른 타이타늄의 미세 표면 거칠기, 표면 젖음성, fibronectin 흡착량에 미치는 영향 (EFFECTS OF VARIOUS SURFACE TREATMENTS FOR TITANIUM ON SURFACE MICRO ROUGHNESS, STATIC WETTABILITY, FIBRONECTIN ADSORPTION)

  • Purpose: This study aims to get the fundamental data which is necessary to the development direction of implant surface treatment hereafter, based on the understanding the surface structure and properties of titanium which is suitable for the absorption of initial tissue fluid by researching effects of additional surface treatments fir sandblasted with large git and acid-etched(SLA) titanium on surface micro-roughness, static wettability, fibronectin adsorption Materials and Method: In the Control groups, the commercial pure titanium disks which is 10mm in diameter and 2mm in thickness were treated with HCI after sandblasting with 50$\mu$m $Al_2O_3$. The experiment groups were made an experiment each by being treated with 1) 22.5% nitric acid according to SLA+ASTM F86 protocol, 2) SLA+30% peroxide, 3) SLA+NaOH, 4) SLA+ Oxalic acid, and 5) SLA+600$^{\circ}C$ heating. In each group, the value of Ra and RMS which are the gauges of surface roughness was measured, surface wettability was measured by analyzing with Sessile drop method, and fibronectin adsorption was measured with immunological assay. The significance of each group was verified by (SPSS, ver.10.0 SPSS Inc.) Kruskal-Wallis Test. (α=0.05) And the correlation significance between Surface micro-roughness and surface wettability. surface roughness and fibronectin adsorption, and surface wettability and fibronectin adsorption was tested by Spearman's correlation analysis. Result: All measure groups showed the significant differences in surface micro-roughness, surface wettability, and fibronectin adsorption. (p<0.05) There was no significance in correlation among the surface micro-roughness, surface wettability, and fibronectin adsorption. (p>0.05) Conclusion: Surface micro-roughness and surface wettability rarely affected the absorption of initial tissue fluid on the surface of titanium.

이종 조직판막 도관 혹은 판막 이식술 직후 면역 글로불린 항체 G, M의 혈중 농도 변화 (Serum Lmmunoglobulin G and M Level after Xenograft Valve or Valved Conduit Implantation)

  • 배경: 동물조직의 생체조직이식시 이종항원 물질인 알파-갈(${\alpha}$-Galactose) 항원결정인자에 대한 환자의 면역반응에 많은 관심이 높아지고 있다. 이에 이종 판막 이식 후 체내에서 ${\alpha}$-Galactose에 대하여 면역반응이 발현하는 것으로 알려져 있는 면역글로불린 G, M (IgG, IgM)의 변화를 일반적인 혈액 검사에 의한 혈중 농도를 측정하여 면역반응의 정도를 예측할 수 있는지 알아보고자 하였다. 대상 및 방법: 2006년 3월부터 2007년 1월까지, 개심술 환자를 대상으로 하여(이종 판막 이식 환자 (실험군, 10명), 이종 장기가 이식되지 않은 환자(대조군, 9명)) 전향적 연구를 시행하였다. 환자의 나이는 $10.3{\pm}9.1$세, 체중은 $31.4{\pm}20.3kg$이었다. 환자들에게서 수술 전, 수술 후 2일째, 수술 후 10일째에 일반적인 혈액 채취를 통하여 IgM, G의 혈중 농도를 측정하였다. 수술 이전에 이종 장기가 이식되어있던 환자들과 그렇지 않았던 환자들을 비교하여 면역 글로불린의 수술 전 기저치에 차이가 있는지, 그리고 이러한 이전의 상태가 수술 후 면역 글로불린의 혈중 농도 변화에 영향을 주었는지 통계학적으로 분석하였으며, 얻어진 혈중 농도로 두 군 간에 수술 후 면역 글로불린의 혈중 농도 변화와 정도에 차이가 있는지를 통계학적으로 분석하였다. 결과: 술전 이종 장기가 이식되어 있던 환자들과 그렇지 않은 환자들 사이에 면역글로불린의 혈중 농도에는 차이가 없었다(IgG; p-value=1.00, IgM; p-value=0.898). IgG, M 모두 실험군과 대조군 각 군 내에서 수술 전에 비하여 수술 후 2일째 혈중 농도가 감소하였다가 이후 다시 증가하는 일정한 변화의 경향을 보였고, 그 변화 정도가 시간에 따라 통계학적으로 의미 있게 달라졌다. 그러나 실험군과 대조군 사이에는 그 변화의 정도, 경향에 의미있는 차이가 없었다(IgG; p-value=0.393, IgM; p-value=0.193). 결론: 이종조직 판막이식 후 혈액 채취를 통해 얻은 혈중 농도 변화는 이종조직판막 이식을 하지 않는 수술을 받은 환자와 차이가 없었다. 이것은 면역 글로불린의 혈중 농도 변화 정도가 술 후 양 군 간에 차이가 없다기보다는, 일상적인 혈액 채취를 통한 면역 글로불린의 혈중 농도 측정으로는 면역 반응에 의한 면역 글로불린의 변화를 측정하기 어렵기 때문이라고 생각한다. 따라서 이종조직판막이식 후의 이종동물항원에 대한 면역반응의 체내 변화를 관찰하기 위해서는 직접적인 ${\alpha}$-Galactose에 대한 항체의 측정이 필요할 것으로 생각한다.

Identification of Bovine Lymphocyte Antigen DRB3.2 Alleles in Iranian Golpayegani Cattle by DNA Test

  • The bovine lymphocyte antigen (BoLA)-DRB3 gene encodes cell surface glycoproteins that initiate immune responses by presenting processed antigenic peptides to CD4 T helper cells. DRB3 is the most polymorphic bovine MHC class II gene which encodes the peptide-binding groove. Since different alleles favour the binding of different peptides, DRB3 has been extensively evaluated as a candidate marker for associations with various bovine diseases and immunological traits. For that reason, the genetic diversity of the bovine class II DRB3 locus was investigated by polymerase chain reaction-restriction fragment length polymorphism method (PCR-RFLP). This study describes genetic variability in the BoLA-DRB3 in Iranian Golpayegani Cattle. Iranian Golpayegani Cows (n = 50) were genotyped for bovine lymphocyte antigen (BoLA)-DRB3.2 allele by polymerase chain reaction and restriction fragment length polymorphism method. Bovine DNA was isolated from aliquots of whole blood. A two-step polymerase chain reaction followed by digestion with restriction endonucleases RsaI, HaeIII and BstYI was conducted on the DNA from Iranian Golpayegani Cattle. In the Iranian Golpayegani herd studied, we identified 19 alleles.DRB3.2${\times}$16 had the highest allelic frequency (14%), followed by DRB3.2${\times}$7 (11%). Six alleles (DRB3.2${\times}$25, ${\times}$24, ${\times}$22, ${\times}$20, ${\times}$15, ${\times}$3) had frequencies = 2%. Although additional studies are required to confirm the present findings, our results indicate that exon 2 of the BoLA-DRB3 gene is highly polymorphic in Iranian Golpayegani Cattle.

IgY 항체를 이용하여 Lactoperoxidase 정량을 측정하기 위한 Indirect ELISA 방법의 개발 (Indirect ELISA Method for Measurement of Lactoperoxidase using IgY Antibody)

  • Lactoperoxidase(LPO)를 농도를 측정하기 위한 ELISA을 개발하기 위해 LPO로 면역시킨 갈색 산란계의 계란에서 형성된 anti-LPO IgY 항체를 분리 정제하고, 분리된 anti-LPO IgY 항체의 특이성을 ELISA 와 double immunodiffusion 방법으로 조사한 후 indirect ELISA 방법을 이용한 표준곡선을 만들었다. 분리 정제된 anti-LPO IgY항체의 titer는 1:520,000이며, ELISA와 double immunodiffusion 방법 모두에서 $\alpha$-lactalbumin, $\beta$-lactoglobulin, casein 및 lysozyme하고는 교차반응을 하지 않고 LPO만 높은 특이성을 갖는 항체로 나타났다. Indirect ELISA방법에서 LPO의 coating 농도는 0.25 $\mu\textrm{g}$/mL이며 anti-LPO IgY 최적 희석배수는 1:8,000으로 나타났다. Indirect ELISA 방법으로 LPO를 측정할 수 있는 표준곡선에서 민감도의 범위는 0.0l-l $\mu\textrm{g}$/mL로 나타났다.

Listeria spp. p60 단백질에 대한 단일클론항체의 생산 (Production of Monoclonal Antibody for Listeria spp. p60 Protein Based on iap Gene)

  • 리스테리아의 p60단백질은 Listeria속 고유의 단백질로써, 주요 세포외 단백질이기에, 식품에서 이 세균의 존재를 밝혀주는 중요한 지시단백질로 사용된다. 본 연구는 재조합 DNA 방법으로 재조합-p60 단백질을 대장균에서 생산하였으며, 아밀로오스 컬럼 크로마토그라피의 방법으로 정제하여, Listeria spp.의 p60에 특이적 항체를 생산하는 단일클론을 선별하였다. 생산된 항p60 항체 1H4는 병원성 Listeria 균주인 L. monocytogenes, L. ivanovii, L. weishi meri II 특이적으로 강한 항체반응을 보였으며, Listeria속의 비병원성균인 L. innocua등과는 상대적으로 매우 약한 항체반응을 보였으며, 타 세균의 단백질과는 거의 반응하지 않았다. 1H4항체는 복수생산법에 의해 대량생산되었으며, 이 항체의 특이성은 면역학적 방법에 의한 리스테리아 검출키트의 개발에 유용하게 사용될 수 있을 것이다.
