애엽(艾葉) 약침액(藥鍼液)이 에탄올 등에 의한 마우스 대식세포의 활성변화에 미치는 영향 (Effect of ARTEMISIAE ARGI FOLIUM Acupuncture Solution on Raw 264.7 Cells Treated by Toxicants)

  • Objectives : The leaves of Artemisia argyi L. have been used for the treatment of bleeding-related diseases in traditional korean medicine. But the immunological activities with macrophage have not been sufficiently reported. This study is to investigate the immunological bioactivities of the herbal acupuncture solution obtained from leaves of Artemisia argyi L. (AAAS). Methods & Results : Against Nicotine and Acetaldehyde, AAAS increased significantly the production of hydrogen peroxide (H2O2) within mouse macrophage Raw 264.7 cells above the concentration of 10 ${\mu}g/m{\ell}$. AAAS increased significantly the production of nitric oxide (NO) in Raw 264. 7 cells above the concentration of 100 ${\mu}g/m{\ell}$ against EtOH. And AAAS increased significantly the production of nitric oxide (NO) in Raw 264. 7 cells above the concentration of 200 ${\mu}g/m{\ell}$ against Nicotine, Acetaminophen, and Acetaldehyde. Conclusions : These results suggest that AAAS could be thought to have the immunological activities related with the production of hydrogen peroxide and NO in macrophage.

키틴 ${\cdot}$키토산 및 그 올리고당의 면역작용에 의한 항종양 활성 (Antitumor Activities by lmmunological Function of Chitin, Chitosan and Their Oligosaccharides)

  • Chitin, a linked polysaccharide composed of 2-acetamido-2-deoxy-$\beta$-D-glucopytanose residues, is distributed widely in nature. It has been utilized on various application field due to the development of chitin derivatives such as chitosan, partial deacetylated chitin, carboxylmethyl chitin, sulfated chitin, and so on. Chitin and chitosan have been recently interested in antitumor and antimicrobial activities, because of a powerful tumor inhibitory effect against experimental mouse tumors. Especially, the oligosaccharides obtained by partial degradation of them exhibited a remarkable antitumor effect against sarcoma 180, MM 48 and Meth Asolid tumors and antimetastatic effect against Lewis lung carcinoma in mice. This review describes on antitumor effects of chitin, chitosan and their oligosaccharides by their mechanism of action involving enhancement of immunological system.

Proteus vulgaris RH-90에서 추출하여 감마선 조사시킨 Lipopolysaccharide(LPS)의 항암 및 면역활성에 미치는 영향 (Antitumor and Immunological Activities of ${\gamma}$-ray Irradiated Lipopolysaccharide Extracted from proteus vulgaris RH-90)

  • The aims of this study were investigated the antitumor effects and immunological activities of lipopolysaccharides (LPS) extracted from Proteus vulgaris RH-90 toward sarcoma-180 cells. LPS extracted from Proteus vulgris RH-90 was irradiated with gamma ray for detoxification. The tumor incidence of sarcoma-180 occurs all group which injected with gamma ray irradiated LPS and tumor of sarcoma-180 was necrotized with breeding in the injected group of l0$\mu\textrm{g}$ LPS. The inhibition ratio of tumor growth showed at the highest level of 60.88% when 5$\mu\textrm{g}$ gamma ray irradiated LPS was injected into mice. The prolongation ratio of life showed 20.72% when injected into mice with gamma ray irradiated LPS of 5$\mu\textrm{g}$. In the effect of immunological activity, the number of circurating leucocyte and peritoneal exudate cells were increased significantly in the treatment group than that control group, and dose-dependent response indicated by the increase of weights of immunorgans which revealed the improvement of immunity. The effect of macrophage on phagocytes, there were not found the differences between phagocytic and corrected phagocytic index.

Antioxidant and Anti-inflammatory Activities of Allium victorialis subsp. platyphyllum Extracts

  • This study was conducted to investigate antioxidant activity and anti-immunological inflammatory effect of Allium victorialis subsp. platyphyllum extracts (AVPEs). Antioxidant activities of AVPEs were determined by free radical scavenging assay and reducing power test. Leaf-part extract had comparatively better antioxidant activity than other-part extracts. Antioxidant activity of extracts had protective effect for human umbilical vein endothelial cells (HUVECs) against superoxide anions secreted from activated neutrophils. Also, we observed AVPEs had inhibitory effects on the adherence of monocytic THP-1 to HUVEC monolayer to the basal level. Inhibitory effect on cell adhesion was caused by suppression of tumor necrosis factor-${\alpha}\;(TNF-{\alpha})-upregulated$ expression of vascular cellular adhesion molecule-1 (VCAM-1) and E-selectin in HUVECs. From these results, we expect to support the evidence of anti-immunological inflammatory effects of Allium victorialis subsp. platyphyllum (AVP) as a Korean traditional pharmaceutical.

Inhibitory Effect of Ginsenoside-Rp1, a Novel Ginsenoside Derivative, on the Functional Activation of Macrophage-like Cells

  • Ginsenoside Rp1 (G-Rp1) is a ginseng saponin derivative with chemopreventive and anti-cancer activities. In this study, we examined the regulatory activity of G-Rp1 on the functional activation of macrophages. G-Rp1 remarkably inhibited TNF-$\alpha$ production, LPS-induced cell cytotoxicity, NO production, ROS generation, and phagocytic uptake from lipopolysacchride (LPS)-activated RAW264.7 cells. According to structural feature study using several G-Rp1 analogs, two carbohydrates (glucose-glucose) at R1 position were observedto be highly effective, compared to other structural derivatives. Although the inhibitory activities of G-Rp1 on macrophage functions were not remarkable, several points that G-Rp1 was known to be safe, and that this compound was orally effective, suggest that G-Rp1 may be beneficial in treating macrophage-mediated immunological diseases.

비허온습만(脾虛蘊濕万)이 아토피 피부염 유발 NC/Nga 생쥐의 혈액내 면역 관련 인자에 미치는 영향 (Effects of BHOSB on Various Immunological Factors Related to Pathogenesis of Atopic Dermatitis in DNCB Treated NC/Nga Mice)

  • Atopic dermatitis is an allergic inflammatory skin disease caused by aberrant and overreactive immune responses including overactivation of $T_H2$ immune responses, high levels of IgE as well as proinflammatory cytokines and chemokines. We examined whether BHOSB, a traditional herbal medicine, has modulatory effects on various immunological factors related to pathogenesis of atopic dermatitis in ONCB treated NC/Nga mice. Oral administration of BHOSB at the concentration of 10.8 mg/mouse/day significantly inhibited the production of IgE compared with control, and the levels of IgG2a and IgG2b, but not IgG1, were also significantly reduced. Production of IL-6 and TNF-a was greatly decreased. The results from flowcytometry of peripheral blood mononuclear cells indicated that the percentages of C03+C069+ cells and C04+ were significantly decreased by BHOSB. Theses results suggested that BHOSB has suppressive effects on aberrant and overreactive immunological activities in ONCB-induced dermatitis mice of NC/Nga.

Meyerozyma guilliermondii FM을 이용한 꽃송이버섯 발효물의 항산화 효과 및 면역 활성 (Antioxidant and Immunological Activities of Sparassis crispa Fermented with Meyerozyma guilliermondii FM)

  • 꽃송이버섯에서 분리 선발한 Meyerozyma guilliermondii FM을 이용하여 꽃송이버섯 발효물을 제조하고 항산화 활성 및 면역조절 효과를 조사하였다. 꽃송이버섯 발효물 제조 시 M. guilliermondii FM은 유산균과 비슷한 생육능력을 보였으며, 5일째에는 오히려 높은 값을 나타내었다. 발효 후 총페놀 함량과 DPPH 라디칼 소거능은 유산균보다 유의적으로 높은 값을 나타내었다. 면역저하가 유발된 rat에서 비장 무게는 면역을 저하시킨 대조군보다 꽃송이버섯 비발효 추출물과 발효물 투여군에서 모두 유의적으로 증가하였다. 또한, 꽃송이버섯 발효물 투여에 따라 TNF-${\alpha}$, IL-2, IgE의 수치는 대조군보다 감소하였으며, IL-10은 증가하였으나 꽃송이버섯 비발효 추출물과 발효물 투여 사이의 유의적인 차이는 없었다. M. guilliermondii FM을 이용한 꽃송이버섯 발효물을 투여한 실험구는 혈액 내 백혈구 함량이 대조구 및 다른 처리구에 비하여 가장 높은 수치를 나타내었다.

Carrageenan과 degraded carrageenan의 면역 보강제로서의 효능 평가 (Evaluating the Immunological Adjuvant Activities of Carrageenan and Degraded Carrageenan)

  • Carrageena은 전세계적으로 안전한 식품첨가물로 승인되어 오랜 기간 다양한 식품, 기타 가공품에 사용되어지고 있다. 다른 한편으로, 이 Carrageenan은 동물 실험에서 염증 유도 물질로 확인되어 염증 유발 실험에 현재까지도 매우 빈번히 사용 되고 있다. 또한 이 Carrageenan을 고온과 강산에서 처리하여 부분적으로 분해한 degraded Carrageenan은 염증 유도 능이 Carrageenan 보다 더 강한 것으로 알려져 있다. 면역 보강제의 중요한 특성 가운데 하나는 선천면역(대표적으로 염증반응)의 활성화 인 것이 잘 알려져 있다. 그러나 현재까지 Carrageenan이나 degraded Carrageenan의 면역 보강제로서의 효과에 관하여 상세한 비교 연구는 수행되어 지지 않았다. 본 연구의 목적은 Carrageenan과 degraded Carrageenan의 면역 보강제로서의 효과를 비교 분석하는데 있다. 실험 동물은 마우스를 사용하였으며, 난 알부민을 항원으로, 피하면역을 수행하여 각각의 면역 보강제 효과를 항체 형성 정도로 조사하였다. Carrageenan이나 degraded Carrageenan 모두 항원 단독으로 면역한 것과 비교할 때 유의적으로 높은 IgG 생성 능을 보였다. 추가적으로 항원 특이적 IgG1과 IgG2a를 조사한 결과, 이들 Carrageenan, degraded Carrageenan은 본 실험에서 양성 대조 군으로 사용한 보강제, Complete Freund's adjuvant와 비교 할 때 IgG2a 보다는 IgG1 생성 능이 높게 유도되는 것이 확인되었다. 이들 결과를 종합하면 염증 유발 능이 보다 강한 degraded carrageenan의 면역 보강제 효과는 carrageenan과 유사한 정도로 확인되었으며, 이들 모두 IgG2 보다는 IgG1 생성 효과가 강한 것으로 나타났다.

Lyophyllum decastes의 항암성분의 면역학적 연구(I) (Immunlogical Studies on Antitumor Component of Lyophyllum decastes(I))

  • To elucidate action mechanism of lyophyllan A, an antitumor polysaccharide of Lyophyllum decastes, its immunological activities were examined. Lyophyllan A increased significantly the weights of spleen and liver of mice. Lyophyllan A also restored the decreased thymic weight in tumor-bearing mice. It did not show any direct cytotoxicity against tumor cells, but showed immunopotentiating activities by increasing the number of the plaques in hemolytic plaque assays. Lyophyllan A increased the number of peritoneal exudate cells (PEC) and inhibited the growth of sarcoma 180 mixed with PEC. Moreover the macrophages from lyophyllan A-treated mice exhibited a strong cytotoxic activity towards L5178Y target cells.
