번식기, 비번식기 및 손상상태에 따른 한국꿩 고환의 면역조기화학적 연구 (An Immunohistochemical Study of Pheasant Testis in Active, Inactive and Damaged States)

  • In order to achieve optimal reproductive performance, reliable morphological and physiological basic data on the reproductive organs are desirable. Adult male Korean ring-necked pheasant in inactive(mid of January) and active state (end of April) were used in this study. In addition, five active state pheasants were received a single dose of 60Co-ray 500 rads each to damage the testes. The objective of this study was to investigate the distribution pattern of protein gene product (PGP) 9.5 and ${\alpha}$-tubulin in the pheasant testes of the active, inactive and ${\gamma}$-ray irradiated active states. The results obtained were summarized as follows 1. The seminiferous tubules collected in inactive states( mid of Jan) showed narrow lumen, and the spermatogonia and the Sertoli cell were well preserved. The PGP 9.5 immunoreactivity of these tubules showed a positive reaction in paranucleus area of the spermatogonia, and a positive reaction in a small number of the Leydig cells in the interstitium of the seminiferous tubules. 2. The seminiferous tubules were dilated in active state(end of April) as compared with the inactive state. The PGP 9.5 reactivity in these tubules showed a positive reaction in many Leydig cells in the interstitium of the seminiferous tubules, and the testes of ${\gamma}$-ray irradiated group showed partially weak reaction in the interstitium of the seminiferous tubules. 3. The ${\alpha}$-tubulin reactivity in the seminiferous tubules of the inactive testes was strongly positive in the cytoplasmic process of the Sertoli cell from the basal stem region to the apical ex-tension. From the broad part of the stem region to the luminal space, the active testes showed a strong positive reaction. The ${\gamma}$-ray irradiated groups showed diminished reaction in the basal region.

  • ALP (alkaline phosphatase) is a membrane-bound metalloenzyme that is expressed in osteoblasts, hepatocytes, lung, kidney, endothelial cells, leukocytes and other cells. Normal soft tissue and skin show little tissue nonspecific ALP (TN-AP), However, scar tissue contains high levels of TN-AP activity, and in fact, TN-AP is expressed intensely in regenerating connective tissue after the wounding. The purpose of this study was to evaluate the change of ALP expression in hypertrophic scar model in rabbits and the effect of triamcinonolone on ALP expression. Adult male New Zealand white rabbits, weighing about 2.5 kg, were used. After full-thickeness wounding over the ventral surface of each ear, either saline (control ear) or triamcinolone (contralateral ear) was injected on day 16. Rabbits were sacrificed on day 3, 7, 15, 17, 19, 23, and the specimens were retrieved en bloc. Histologic and immunohistochemical examinations of tissue samples were done. The results obtained were as follows: On day 3, ALP reaction was observed on fibroblasts and inflammatory cells in wound margin. On day 7, ALP reaction was more intense than day p in capillaries, inflammtory cells, and fibroblasts behind newly formed epithelium. On day 15, ALP reaction was lessened in both groups and appeared mainly in subepidermal capillary network, Since day 17, ALP reaction was lessened in both groups and weaker in triamcinolone-injected group than in saline-injected group. These results suggest that ALP reaction isn't increased in triamcinolone-injected scar and triamcinolone reduces scar not by increasing TN-AP expression but other mechanism.

전통된장의 면역증강 효과 (Immunostimulatory Effects of Traditional Doenjang)

  • 된장의 면역증강 효과에 미치는 영향을 위장관도에서 관찰하여 다음과 같은 결과를 얻었다. $CD4^+/CD8^+$ T 림프구의 면역조직화학적 염색반응에서 된장식이를 첨가한 모든 실험군에서 $CD4^+$ T 세포는 공장의 점막 고유층과 음와 아래의 고유층에서 강한 면역 반응을 나타내었고, 결장에서는 점막 하층과 외막층의 혈관 주위에서 강한 면역반응을 나타내었다. 반면, $CD8^+$ T 세포는 Group III에서 결장의 점막상피, 점막 고유층, 점막하층 및 외막층에서 강한 면역반응을 나타내었고, uNOS에 대한 면역 반응에서는 된장 식이를 첨가한 모든 실험군에서 결장 점막하층과 근육층신경얼기에서 강한 면역반응을 나타내었다. Protein kinase C-${\alpha}$에 대한 면역반응은 Group II와 Group III에서 점막상피와 근육층에서 강한 면역반응을 나타내었고, stem cell factor에 대한 면역반응은 된장식이를 첨가한 모든 실험군의 점막상피와 Group I의 근육층에서 강한 면역반응을 나타내었다. 이상의 실험결과로 대조군에 비하여 농도별로 된장식이를 첨가한 실험군에서 $CD4^+/CD^+8$에 대하여 강한 면역반응을 보인 것으로 보아 위장관에서 면역능을 증가시킬 것으로 사료되며, uNOS의 증가에 따른 NO의 방출이 위장관의 운동과 혈관운동을 촉진하여 위비움과 결장의 운동을 촉진할 것으로 사료되었다. 또한 농도별로 된장 식이를 첨가한 실험군에서 protein kinase C-${\alpha}$와 stem cell factor에 대한 면역반응이 위장관의 점막상피에서 강하게 나타난 것으로 보아 점막 상피세포의 증식과 분화를 촉진시켜 여러 가지 물질의 흡수와 전달에 관여할 것으로 사료되었다.

법랑모세포종의 재발과 p53, Ki-67, PCNA 및 cytokeratin 17, cytokeratin 18의 발현과의 상관관계에 관한 연구 (EXPRESSIONS OF P53, KI-67, PCNA AND CYTOKERATIN 17, CYTOKERATIN 18 IN RECURRED AND NON-RECURRED AMELOBLASTOMA)

  • Ameloblastoma is a common odontogenic benign tumor of the jaw bone. However, it might be albe to infiltrate into the adjacent tissue, causing bony destruction and high recurrent rate. The aim of the study is to understand the biologic behavior of recurred ameloblastoma through immunohistochemical study. The PCNA, Ki-67, p53 and cytokeratin 17, cytokeratin 18 antibody staining were used. There was significant difference of positive reaction between non-recurred ameloblastoma and recurred ameloblastoma in PCNA and cytokeratin 17. There were no significant difference of positive reaction between non-recurred ameloblastoma and recurred ameloblastoma in p53, Ki-67 and cytokeratin 18. From the above results, it is suggested that the recurrence of ameloblastoma is related to positive reactions of PCNA and cytokeratin 17 and the progonsis of the recurrence of ameloblastoma is able to be predicted by using PCNA and cytokeratin 17.

Suitability of denervated muscle flaps as recipient sites for pancreatic islet cell transplantation

  • Background Extensive research has been conducted on islet transplantation as a possible cure for diabetes. Islet transplantation in the liver via the portal vein has shown remarkable results, but numerous other recipient sites are currently being investigated. We aimed to show the effectiveness of using a muscle flap as a recipient site for islet transplantation. Methods Islet cells were harvested from 12 isogenic Lewis rats, and then diabetes was induced in another 12 isogenic Lewis rats by streptozotocin injection. In six rats, 3,000 islets were transplanted into gastrocnemius muscle flaps, and in the other six rats, the same number of islets were transplanted into the gastrocnemius muscle. The transplanted islet cell function between the two groups was compared by means of blood glucose tests, glucose tolerance tests, immunohistochemistry, and real-time reverse transcription polymerase chain reaction. Results In the muscle flap group, blood glucose levels significantly decreased after islet transplantation. Blood glucose levels were significantly different between the two groups at 3 weeks after transplantation. The muscle flap group showed nearly normoglycemic results upon the glucose tolerance test, whereas the muscle group was hyperglycemic. Immunohistochemical evaluation showed positive results against insulin and glucagon in biopsies of both groups, and the islet cell density was higher in the muscle flap group. There were no statistically significant differences between the two groups in real-time reverse transcription polymerase chain reaction results. Conclusions Our results suggest that muscle flaps are promising candidates for islet cell transplantation.

PLGA의 염증완화에 대한 피브린의 효과 ; In vivo 연구 (Reduction of Inflammatory Reaction of PLGA Using Fibrin; in vivo Study)

  • PLGA는 미국 식품의약품안전청(FDA)의 승인을 받은 합성고분자로서 생체재료로 널리 쓰이고 있다. 하지만, 분해산물인 산으로 인하여 염증반응을 일으키고, 독성물을 생산하여 세포증식률의 감소를 야기시킨다고 보고된바 있다. 이러한 PLGA의 단점을 보완하고자 생체재료인 피브린을 사용하였는데, 피브린은 피브리노겐의 풍부함과 이들의 정제가 비교적 용이하다는 장점을 가지고 있다. 본 연구에서는 PLGA의 염증완화에 대한 피브린의 효과를 알아보았다. 피브린 첨가에 따른 PLGA 다공성 지지체에 염증의 발현정도를 확인하기 위해 RT-PCR을 수행하였고 지지체와 조직 간의 상호작용을 통한 염증세포의 침윤과 대식세포 발생 정도를 확인하기 위해 H&E와 ED-1 염색을 수행하였다. 위 실험결과 천연재료인 피브린이 PLGA의 이물반응을 감소시키는데 긍정적인 영향을 미치는 것으로 확인되었다.

양서류 피부 상처회복과정 중 각질화세포 cytokeratin의 분포 (The Temporal and Spatial Expression of the Cytokeratin in Keratinocytes during Cutaneous Wound Healing on the Amphibian(Bombina orientalis))

  • 상처회복은 염증, 재상피화 그리고 기질의 재형성등이 관여하는 복잡한 과정이다. 이중 재상피화에 관여하는 각질화세포의 이동경로를 분석하기 위하여 무당개구리 피부 상처유도 후 투과전자현미경과 cytokeratin에 대한 조직면역화학법을 이용하였다. 정상조직의 cytokeratin발현은 기저층의 세포들과 선상피에서 확인되었다. 상처 유도후 3시간 조직에서, 기저세포층에서 강한 반응이 관찰되었고, 1일과 2일 사이에서는 재생되는 각질화세포에서 강한 면역반응이 확인되었다. 상처반응 중기인 7일부터 10일 사이에서도 재생된 세포의 기저층세포에서 강한 반응이 일어났다. 19일경과의 조직에서는 기저층과 유극층의 세포들에서 cytokeratin의 발현이 증가하였다. 따라서, 재상피화에 관여하는 각질화세포는 기저층의 세포로부터 시작하여 상처부위로 이동하여 과립층과 유극층으로 분화됨으로서 재상피가 진행되었다.

비염증성 치은증식증의 상피 및 상피하조직내 세포변화에 관한 면역조직화학적 연구 (AN IMMUNOHISTOCHEMICAL STUDY ON THE CELLULAR CHANGE IN EPITHELIUM AND SUBEPITHELIAL TISSUE OF NON-INFLAMMATORY GINGIVAL HYPERPLASIA)

  • The gingival hyperplasia refers to an increase in the size of the gingival tissue produced by an increase in the number of its component cells. In order to investigate the cellular change in epithelium and subepithelial tissue of noninflammatory gingival hyperplasia, the gingival tissues were surgically obtained from the patients with dilantin gingival hyperplasia and idiopathic gingival hyperplasia. The excised tissue samples were fixed in neutral formalin for 6-24 hours, embedded with paraffin, sectioned at $4-6{\mu}m$ in thickness, mounted on glass slides coated with 3-aminopropyltriethoxysilane(Sigma Chemical Co., St. Louis, MO, U.S.A.) and immunocytochemically processed by Avidin-Biotin peroxidase complex method for detecting proliferating cell nuclear antigen, tenascin and collagen type IV. Monoclonal mouse anti-human PCNA antibody(Oncogene Science, Uniondale, NY, U.S.A., 1 : 250,000), monoclonal mouse anti-human tenascin antibody(Chemicon-International Inc., Temecula, CA, U.S.A., 1:5,000), and monoclonal mouse anti-human collagen type IV(Dakopatts, Glostrup, Denmark, 1: 50) were used as primary antibodies. The results were as follows: 1. In non-inflammatory gingival hyperplasia, the positive reaction to proliferating cell nuclear antigen was localized in the basal cell layer of gingival epithelium and well-developed rete pegs. 2. The positive reaction to tenascin was shown in the connective tissue subjacent to basament membrane of gingival tissue, and especially strong positive reaction was noted in the tip portion of connective tissue projections. 3. The positive reaction to collagen type IV was localized along the basement membranes of gingival epithelium and blood vessels. The results suggest that connective tissue enlargement may affect the proliferation of gingival epithelium.

Extensive inflammatory reaction in facioscapulohumeral muscular dystrophy

  • In facioscapulohumeral muscular dystrophy (FSHD), prominent inflammatory cellular infiltrates mimicking inflammatory myopathies are often observed in muscle biopsies. We report extensive inflammatory changes in a 16-year-old girl who was genetically confirmed as to have FSHD. Immunohistochemical staining revealed that this could be clearly distinguished from inflammatory myopathies, both in terms of cell subsets and the expression of antigenic targets. Our observations strongly suggest that the inflammatory cellular infiltrates in FSHD differ from those observed in inflammatory myopathies.

Peroxidase-antiperoxidase(PAP) 복합체법을 이용한 돼지콜레라의 면역조직화학적 진단 (Immunohistochemical diagnosis of hog cholera with peroxidase-antiperoxidase(PAP) complex method)

  • The present study was intended to use the peroxidase-antiperoxidase method for the identification of hog cholera virus(HCV) in the lymphatic organs of HCV-infected pigs. Sections were incubated with primary antibody (rabbit anti-HCV polyclonal or mouse anti-HCV monoclonal), followed by incubation with linkserum (goat anti-rabbit IgG) in excess and rabbit or mouse PAP complex. The viral antigen was localized mainly in the cytoplasms of lymphoid cells and macrophages. Positive reaction cells were frequently detected in the marginal areas of the germinal centers of the spleens, and also found in the tensils and lymph nodes. The method approved to be highly specific for the identification of the virus and allowed a precise localization of the viral antigen in infected cells.
