Increasing Prevalence of the Sensitization to Cat/Dog Allergens in Korea

  • Background: Recently, the number of domestic pets has increased. As a consequence, sensitization to animal allergens, such as cat or dog allergens, has become a problem. Objective: We studied the annual trends of sensitization to cats or dogs, and the characteristics of the patients. Methods: We retrospectively analyzed the medical records of 7,469 patients who visited a dermatology clinic and underwent an allergic profile test, from January 2011 to December 2015. Specific immunoglobulin E (IgE) levels to cat or dog antibody greater than 3.50 IU/ml were regarded as positive results. Results: In all, 274 patients showed significant increase in levels of specific IgE antibody to dog, and 307 revealed increase in levels of of specific IgE antibody to cat. The prevalence of these specific IgEs increased from 2011 to 2015. Independent risks for sensitization to cat allergens were sensitization to dog, but not to house dust, Dermatophagoides pteronyssinus, and D. farinae. Independent risks for sensitization to dog allergens were sensitization to cat, but not to house dust, D. pteronyssinus, and D. farinae. Total IgE level was not related to specific IgE level against either cats or dogs. Conclusion: In conclusion, the prevalence of sensitization to cat or dog has increased. Sensitization to cat or dog is related to each other, but is irrelevant to the total IgE level.

배발효 음료가 천식이 유발된 생쥐의 기도 과민성 및 면역 글로불린 분비에 미치는 영향 (Effects of Pear Alcoholic Fermentation Beverage on Airway Hyperresponsiveness and Immunoglobulin Production in Asthmatic Mice)

  • Objectives : This study was designed to investigate the effects of alcoholic fermentation beverage using pear, Bae Ro Mi In (BRMI) on airway hyperresponsiveness and immunoglobulin production in asthmatic mice Methods : We investigated the effects of BRMI on airway hyperresponsiveness by measurement of enhanced pause (Penh), and also investigated the effects on production levels of antigen specific antibody and subclasses such as IgG1, IgG2a and IgE by using ELISA methods. Prednisolone (PD, 5 mg/kg) was used as positive control. Results : Treatment with BRMI did not lowered airway hyperresponsiveness, but PD lowered significantly. Oral administration of BRMI lowered production level of ovalbumin (OVA) specific total antibody significantly. Especially, BRMI decreased IgE levels compared to non-treated control effectively. Treatment with PD lowered production levels of total antibody, IgG1 and IgE. Conclusions : These result suggest that BRMI can lower production levels of antigen specific total antibody and IgE in asthmatic mice. We also suggest that BRMI has the possibility to prevent or cure asthma through regulation of antigen specific antibody production.

인간 비만세포에서 그라비올라 잎 추출물의 항알러지 효과 (Anti-allergic Effect of Graviola Leaf Extract in Human Mast Cells)

  • 본 연구는 그라비올라(Annona muricata)가 인간 비만세포(LUVA 세포)에서 immunoglobulin(Ig) E-매개 알러지 반응에 어떠한 효과를 나타내는지 확인하는데 목적이 있다. 그라비올라 잎 추출물(GLE)의 항알러지 효과를 알아보기 위해 IgE로 감작된 비만세포에 다양한 농도의 추출물을 처리하였다. 비만세포의 IgE-매개 탈과립 실험 결과, GLE가 LUVA 세포에서 histamine, β-hexosaminidase, TNF-α, IL-4 및 IL-6의 방출을 유의하게 억제하였다. 또한 GLE는 IgE 유도에 의한 Lyn과 Syk의 인산화를 억제하였고 그 하위 경로에 있는 MAPK의 활성을 억제하였다. 이상의 결과는 GLE가 비만세포 활성화 및 알러지 반응을 억제할 수 있음을 보여주고, 이는 GLE가 항알러지 효과를 통하여 기능성 화장품 소재로서의 활용이 가능함을 시사한다.

닭고기 중 알레르기 유발성분의 동정 (Identification of the Chicken Meat Allergens)

  • The chicken meat has been reported as one of the food causing allergic reactions predominantly to Korean. At present, several in vitro tests for immunoglobulinG (IgG)-mediated as well as IgE-mediated food allergy are available. 13 clinically chicken meat-allergic patients were investigated together with 4control subjects for identification of chicken meat-specific reactivity by ELISA. Also, protein profile and IgE, IgGtotal and IgG4-reacting allergens were detected by means of sodium dodecyl sulfate-polyacrylamide gel electro-phoresis (SDS-PAGE)and immunoblotting. Chicken meat extracts were prepared as raw, heated, heat and simulated gastric fluid (SGF) treated samples to characterize the stability of allergen to physicochemical treatment. SDS-PAGE revealed 9~200 kDa bands. And in immunoblotting 7 sera were identified most major bands between 10 and 78 kDa. In case of IgE, six proteins (17, 26, 35, 40, 78 kDa) were predominant in heat-treated extract, and the one (35 kDa) was present in SGF-treated preparations. In case of IgG$_{total}$ and IgG4, most of them showed a patters simmilar to IgE. There were significant differences (P<0.05) in IgE, IgG$_{total}$ , IgG4 Abs to chicken meat between the allergic and control subjects in ELISA. In addition, the concentration of IgG4Abs in the challenge-positive subjects was significantly higher than that of control subjects. It is considered that the specific IgE response to chicken meat was rarely prevalent to Koreans. However, the specific IgG4 response play an important role in the development of allergic symptoms.

Effect of Salvia plebeia on IgE antibody mediated allergic reaction in rats

  • The effect of aqueous extract of Salvia plebeia R. Br. (Labiatae) (AESP) on immunoglobulin (lgE) antibody mediated allergic reactions in rats was investigated. AESP inhibited passive cutaneous anaphylaxis (PCA) when intravenously, intraperitoneally, and orally administered. AESP dose-dependently inhibited histamine release from rat peritoneal mast cells activated by anti-DNP IgE antibody. Moreover, AESP had an inhibitory effect on anti-DNP IgE antibody induced $TNF-{\alpha}$ production from RPMC. These results suggest that AESP inhibit the IgE-mediated allergic reaction in rats.

NC/Nga 마우스의 아토피 피부염에 미치는 카모마일, 라벤더, 샌달우드 혼합오일 도포의 치유효과 (The Effect of Mixed Aroma Oil with Chamomile, Lavender and Sandalwood on Atopic Dermatitis in NC/Nga Mice)

  • 본 연구에서는 카모마일, 라벤더, 샌달우드를 혼합한 아로마 혼합오일(CLS)과 면역억제제(FK506)의 아토피 피부염 치료효과를 알아보기 위하여 혈청 중 IgE(Immunoglobulin E)와 IgG1(Immunoglobulin G1)의 수준의 감소효과, 피부 임상지수를 이용한 육안평가 등을 실시하였다. 그 결과 IgE 수준을 낮추는데는 CLS가 더 효과적이었고, IgG1 수준을 감소시키는 데에는 FK506이 더 효과적인 것으로 나타났다. 육안평가 결과를 통하여 아토피 피부염을 가진 마우스에 FK506과 CLS를 도포한 후 2주까지는 아토피 증상을 더 악화시켰으며 4주후부터는 증상완화가 현저하게 나타남을 확인할 수 있었다. 특히 FK506의 효과가 두드러졌는데 이는 IgG1 수준감소와 관련이 있는 것으로 사료되며 이와 관련하여 보다 심도있는 실험 조사가 필요하겠다. 이상의 결과로부터 카모마일 저먼, 라벤더, 샌달우드의 아로마 혼합오일이 아토피 피부염의 개선에 효능이 있음이 인정되었고 이와 같이 장기간 피부에 도포하여 치료하는 경우 CLS가 아토피 치료의 한 방법으로서 가능성이 보임을 확인하였음에 본 연구의 의의가 있다.

Achyranthes japonica Nakai Water Extract Suppresses Binding of IgE Antibody to Cell Surface FcεRI.

  • Achyranthes japonica Nakai (AJN) water extract has a variety of physiological properties, including anti-diabetic, anti-cancer, anti-inflammatory, anti-microbial, and anti-oxidative activities. In the present study, the inhibitory effects of AJN extract were investigated in high affinity immunoglobulin E receptor ($Fc{\varepsilon}RI$)-mediated KU812F cells activation. AJN extract showed suppressive effects on histamine release and intracellular calcium [$Ca^{2+}$]i elevation from anti$Fc{\varepsilon}RI$ antibody (CRA-1)-stimulated cells in a dose-dependent manner. Flow cytometric analysis showed that AJN extract treatment caused a dose-dependent decrease in the cell surface $Fc{\varepsilon}RI$ expression and the binding between the cell surface $Fc{\varepsilon}RI$ and the IgE antibody. Moreover, reverse transcription-polymerase chain reaction analysis showed that levels of the mRNA for the $Fc{\varepsilon}RI$ ${\alpha}$ chain was decreased by treatment with AJN extract. These results indicate that AJN extract may exert anti-allergic effects via the inhibition of calcium influx and histamine release, which occurs as a result from the downregulation of the binding of IgE antibody to cell surface $Fc{\varepsilon}RI$. This mechanism may occur through $Fc{\varepsilon}RI$ expression inhibition.

치근단 병소에서 면역글로불린의 분포에 관한 연구 (A QUANTITATIVE ANALYSIS OF THE IMMUNOGLOBULIN CONTAINING CELLS IN PERIAPICAL LESIONS OF THE HUMAN TEETH)

  • Periapical lesions develop as a result of immunopathologic response to irritants from infected root canal systems. Removal of these irritants from the root canal system and sealing the root canal space may induce he31ing of the periapical lesions. 83 periapical lesions diagnosed as periapical abscess, periapical granuloma, chronic nonspecific inflammation, fibrosis and periapical Cyst were evaluated for the distribution of immunoglobulin containing cells. The influence of the state of root canal treatment on the distribution of immunoglobulin containing cells has evaluated. All lesions were divided into a group with no treatment, a group with canal enlargement, a group filled with gutta percha, and a group filled with Vitapex(calcium hydroxide). The distribution of immunoglobulin-containing cells according to the presence of pain and fistula was also evaluated. The following results were obtained. 1. Statistically significant difference in the distribution of immunoglobulin-containing cells among periapical abscess, periapical granuloma, chronic nonspecific inflammation/fibrosis and periapical cyst were found.(Kruskal-Wallis analysis, P<0.05) The number of immunoglobulin-containing cells in fibrosis was remarkably lower than that of periapical abscess, granuloma and cyst. 2. IgM and IgA containing cells were predominantly observed in periapical abscesses and periapical cysts, respectively. 3. All periapical lesions showed a large number of IgG containing cells followed by IgM, IgA and IgE containing cells. 4. There was a decrease in all Ig-containing cells in the group with canal filling compared to groups without treatment or with enlargement. That is, there is a decrease in Ig-containing cells as treatment progresses. 5. No significant correlation existed between the presence of pain and fistula and the distribution of immunoglobulin containing cells in periapical lesions.(t-test) Results appear to support that immune response are actively involved in the development and progress in periapical lesions. The fact that distribution of immunoglobulins differ according to the state of endodontic treatment suggests that root canal treatment may alter the humoral immune response of the periapical lesions.

Evaluation of serum immunoglobulin G4 concentrations in canine pancreatitis

  • The goal of this study was to measure immunoglobulin G4 (IgG4) concentrations and to evaluate the significance of these values in the management of canine pancreatitis. The medical records of 24 dogs that visited the Veterinary Medical Teaching Hospital between December 2016 and June 2018 were retrospectively reviewed to identify dogs that had been diagnosed with pancreatitis. The serum C-reactive protein and serum IgG4 concentration in the affected dogs were highly increased compared to the healthy group. Particularly, serum IgG4 measured significantly higher in dogs with pancreatitis and concurrent immune-mediated disease (p < 0.05). In conclusion, increased serum IgG4 concentrations are a characteristic finding in dogs with pancreatitis. The results of this research indicate that an elevation in IgG4 has the potential of being used as a tool for the diagnosis of pancreatitis and concurrent immune-mediated disease.

Effect of Terminalia chebula on Immediate Hypersensitivity Reaction in Mice and Rats

  • We investigated the effect of aqueous extract of Terminalia chebula (Combretaceae)(TCAE) on the immediate hypersensitivity reaction in vivo and in vivo. TCAE (0.01 to 1 g/kg) dose-dependently inhibited compound 48/80 induced systemic anaphylaxis in mice. When TCAE was pretreated at concentrations ranging from 0.01 to 1 g/kg, the plasma histamine levels were reduced in a dose-dependent manner. TCAE (0.1 and 1 g/kg) significantly inhibited local immunoglobulin E (IgE)-mediated passive cutaneous anaphylactic reaction. TCAE (0.001 to 1 mg/ml) also dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-dinitrophenyl (DNP) IgE. TCAE (0.01 to 1 mg/ml) had a significant inhibitory effect on anti-DNP IgE-induced tumor necrosis $factor-{\alpha}$ production from RPMC. These results indicate that TCAE inhibits immediate hypersensitivity reaction in vivo and in vitro.
