An histological and immunohistochemical study of endocrine cells in the gastrointestinal tract of the Amur lizard (Takydromus amurensis) (아무르장지뱀의 위장관 내분비세포에 관한 조직화학적 및 면역조직화학적 연구)

  • The distribution and the frequency of endocrine cells in the GIT of the Amur lizard(Takydromus amurensis) were investigated using silver techniques and immunohistochemical method. Only argyrophil cells stained by the Grimelius technique were found numerous in the pyloric glands and moderate in the duodenum. Two types of immunoreactive endocrine cells were identified by immunohistochemical method. Bovine CG-immunoreactive cells were demonstrated the entire GIT. BPP-immunoreactive cells were restricted in the duodenum and the ileum. The results showed that: the number of argyrophil cells was lower than the number of cells stained with bovine CG antiserum. Therefore, bovine CG-immunostaining and the Grimelius silver technique did not correspond with various endocrine cells in the Amur lizard.

  • The present investigation was undertaken to confirm of clinical effects of Sagnating. So, experimental studies were done, especially, to ascertain whether Sagantang had pharmacological effect of analgesia, antipyresis, diuresis and decreasing blood pressure and so on. The results of the studies were obtained as follows: 1. The analgesic effects of Sagantang were obtained in mice. 2. Sagantang prolonged the duration of hypnosis which is induced thiopental-Na in mice, but it was not effective in rotor rod method. 3. Antipyretic effect of Sagantang was known on the typhoid vaccine induced fever in rats. 4. Sagantang inhibited automatic movement on the isolated ileum of mice, and antiacetylcholine effect and antibarium chloride effect of Sangantang were known on that. 5. Sangantang decreased blood pressure due to vasodilatation in anesthetized mice. 6. The effect of diuresis were noted in mice. According to the above results, clinical effects of Sagantang on oriental medical references were similar to the actual experimental results.

  • KUNLITANG(建理湯) have been widely used in the disease of digestive system, which was based on the oriental references. In order to investigate the clinical effect of it, the experimental works were carried out with mice and rats. The following results have been obtained : 1. The analgesic effect in mice was recognized in acetic acid method. 2. The inhibitory effect on transfortation activity in small intestine of mice was recognized. 3. In the experiment to see the effects of KUNLITANG on the gastric juice in rats, volumn, free HCL and total acidity were decreased, pepsin activity was inhibited. Whi1e pH was increased by the administration of KUNLITANG, respectably. 4. In the experiment to see the effects of KUNLITANG on the gastric ulcers in rats, anti-ulcerative effects were recognized in Shay rats and induced by histamine or aspirine. 5. Spontaneous contraction in the isolated ileum in rats was inhibited. According to the above experimental results, it can be concluded that KUNLITANG are very effective on many gastrointestinal diseases of biwihehanheung(脾胃虛寒型)

Effect of BODUOPAESAN(BOS) on Gastrointestinal Tract (오패산(烏貝散)과 보두(寶豆)의 병용투여(倂用投與)가 위장관(胃腸管)에 미치는 영향(影響))

  • In order to investigate the effect of BODUOPAE-SAN(BOS), experiments were performed on analgesic effect induced by acetic acid, duration of hypnosis induced by pentobarbital-Na in mice and gastric ucler in Shay rats and indomethacin induced gastric ulcer, HCI-ethanol induced gastrci lesion in rats and gastric juice secretion in Shay rats. The results are as followings. 1. BOS showed significantly an analgesic effect induced by acetic acid. 2. BOS prolonged the duration of hypnosis induced by pentobarbital-Na in rats 3. BOS rised the spontaneous motility of isolatied ileum of mice temporarily. 4. BOS depressed the gastric motility of rats significantly. 5. BOS showed an anti-ulcer effect in Shay rats and indometacine-induced ulcer rats significantly. 6. BOS reduced the ulcer index of the HCl-ethanol induced gastric lesion in rats. 7. BOS reduced the gastric juice secretion in Shay rats. From above results think BOS is more effective in comparison to the OPAESAN alone on gastrointestinal disorder.

Neonatal Gastrointestinal Perforation (신생아 위장관천공)

  • Perforation of the gastrointestinal tract in neonatal period has been associated with a grim prognosis. Recently there has been some improvement in survival. To evaluate the remaining pitfalls in management, 19 neonatal gastrointestinal perforation cases from May 1989 to July 1996 were analysed retrospectively. Seven patients were premature and low birth weight infants. Perforation was most common in the ileum(56.3%). Mechanical or functional obstruction distal to the perforation site was identified in 7 cases; Hirschsprung's disease 3, small bowel atresia 3, and anorectal malformation 1. These lesions were often not diagnosed until operation. Five cases of necrotizing enterocolitis and 1 of muscular defect were the other causes of perforation. In six cases, the cause of the perforation was not identified. Perinatal ischemic episodes were associated in five cases. Overall mortality was 15.1%. Because a considerable number of gastrointestinal perforations resulted from distal obstruction, pediatric surgeon should be alert for early identification and intervention of gastrointestinal obstruction, particularly in patients that are premature and have a history of ischemia.

Pathological changes on naturally occuring necrotic enteritis in chicken (닭의 괴사성 장염에 대한 병리학적 연구)

  • From January of 1991 to December of 1992, 42 chickens collected from 21 poultry farms and also diagnosed as necrotic enteritis(NE) was examined clinical signs, gross and histopathological findings. Main clinical signs were characterized by decreased appetite, mild to severe depression, reductance to move, ruffled feathers, greenish to yellow-browinish diarrhea sometimes including blood. As progressed, diseased chickens showed feces mixed with necrotic debris which detached from the intestinal mucosa and mostly resulted in the death. In chronic cases, there were dirty feathers around cloaca due to diarrhea and notably retarded growth. Principle gross lesions were usually confined to the jejunum and ileum, especially toward the lower part of Meckel's diverticulum. The part of small intestine was frequently distended with gas, and also showed mucosal congestion and hemorrhages with varying degrees. Sometimes, the intestinal mucosa was thickened, and also covered with fibronecrotic psuedomembrane. In addition, there were focal necrosis and severely multifocal ulcreation in the mucosa of small intestine. Major histopathological findings included villous necrosis and erosion of the small intestine covering with lots of bacterial colonies, inflammatory cell infiltration in the lamina propria, and dilatation and hyperplasia of crypts. Luminal exudate contained bacterial colonies, fibrin, erythrocytes, and desquamated epithelium. Thirteen(61.9%) out of 21 NE-occurring farms were complicated with intestinal coccidiosis.

Immunohistochemical study of causative agents for the porcine viral diarrhea disease on newborn piglets in Cheju (제주지역 양돈장에서 발생한 자돈 설사형 바이러스의 면역조직화학적 확인)

  • In an outbreak of acute porcine diarrhea in newborn piglets, an etiological study was carried out using piglets submitted in Cheju Province Institute for Livestock Promotion(Cheju Veterinary Service for the disease diagnosis). Sixteen piglets(2-7 days old) were collected from 4 farms during outbreaks of diarrhea disease(from January to April 2000). Specimens were taken after necropsy and examined by immunohistochemistry using of monoclonal antibodies for porcine epidemic diarrhea(PED) virus, transmissible gastroenteritis(TGE) virus, and porcine rotavirus. Immunohistochemistry showed that PED virus antigens, but both TGE virus and rota virus antigens not, were localized in the some epithelial cells of the intestines of 14 animals among 16 piglets examined. PEB virus antigens were mainly detected in the cytoplasm of enterocytes. Infected cells, which were most abundant in the villous epithelial cells of the jejunum and ileum, were uncommon in the crypt, epithelial cells, the lamina propria and Peyer's patches of piglets examined. The results suggest that PED virus is one of the most prevailing agents in an outbreak of fatal diarrhea in newborn piglets on Cheju island and PED virus was need to further study to prevent this disease.

Changes in the Serotonin-, Somatostatin- and Motilin-Immunoreactive Cells in the Gastrointestinal Tract of the Bullfrog, Rana catesbeiana, at Various Developmental Stages

  • In order to investigate the regional distribution and relative frequency of the serotonin-, somatostatin- and motilin-immunoreactive cells in the gastrointestinal tract GIT) of the bullfrog (Rana catesbeiana) with developmental stages, group of bullfrogs subdivided into the tadpole with hindlegs, metamorphosed bullfrog with tail, 2 wk after metamorphosed bullfrog and adult bullfrog, were stained by immunohistochemical methods (PAP methods). Multifarious changes of the regional distribution and relative frequency of the gastrointestinal endocrine cells were observed as developmental stages but motilin-immunoreactive cell ere not detected in this study. Serotonin-immunoreactive cells were detected from 2 wk after metamorphosed bullfrog in the whole GIT. The positive cells were predominant in the pylorus regions. In the adult bullfrog, these cells increased in fundus. On the other hand, somatostatin-immunoreactive cells were observed from the tadpole with hindlegs in the stomach regions. In the metamorphosed bullfrog with tail, they were found in the whole GIT except for the ileum, and increased with developmental stages except for rectum. In the rectum, these cells were not detected after that period. Our results show that most of the immunoreactive cells appear from completed metamorphosed periods when the histological structure of GIT is completely differentiated.

Immunohistochemistry of Endocrine Cells in the Alimentary Tract of the Tree Frog, Hyla arborea japonica

  • The regional distribution and relative frequencies of endocrine cells were studied immunogistochemically (PAP methods) in the alimentary tract of the tree frog, Hyla areorea japonica, using specific antisera against serotonin, somatostatin, bovine Sp-1/chromogranin (BCG), cholecystokinin (CCK)-8, vasoactive intestinal polypeptide (VIP), gastrin, bombesin, secretin and pancreatic polypeptide (PP). Six kinds of endocrine cells were identified in this study, These immunoreactive cells were located in the gastric glands of stomach regions and in the basal portion of the epithelium of the intestinal tract or esophagus with variable frequencies. They were spherical or spindle-shaped. Serotonin-immunoreactive cells were observed in the whole alimentary tract including the esophagus. Somatostatin-immunoreactive cells were also detected throughout the alimentary tract except the rectum. CCK-8-immunoreactive cells were observed from the pylorus to ileum. Vip-immunoreactive cells were restricted to the rectum. Bombesin-immunoreactive cells were restricted to the fundic gastric regions and gastrin-immunoreactive cells were restricted to the pylorus. However, no BCG-, secretin and PP-immunoreactive cells were demonstrated in this study. In conclusion, the regional distribution and relative frequency of the endocrine cells in the alimentary tract of the tree frog were similar to other anuran species, but some differences which may be caused by feeding habits and species specification were also observed.

Chemistry and Anti-inflammatory Activity of Prunus davidiana Stems (산복사 줄기의 성분과 항염증 작용)

  • The possibility as an anti-inflammatory drug of Prunus davidiana which have been used in Korean folk medicine for treatment of neuritis and rheumatism was investigated on carrageenin-induced paw edema in rats. The MeOH extract exhibited potent inhibitory effect on carrageenin-edema rats when topically applicated. On the other hand, it did not show any effect when orally tested. It also had no influences on isolated ileum of guinea pig and anti-platelet aggregating activities. Thus, it appears from the present findings that the MeOH extract of P. davidiana may be utilizable only for the external treatment of inflammed sites. This study was also conducted to further isolation of flavonoids from the EtOAc soluble fraction and characterized as persiconin and isosakuranin by spectrometric analysis.
