Obesity is associated with a number of pathological disorders such as non-insulin-dependent diabetes, hypertension, hyperlipidemia, and cardiovascular diseases. Bangpoongtongsungsankamibang (BTSK) has been widely used in the oriental medicine for the treatment of several diseases associated with inflammatory abnormalities in cardiovascular and nervous system. The BTSK is the modified prescription of Bangpoongtongsungsankamibang in which sea tangle (Laminaria japonica) were added. This study was carried out to detemine the anti-obestic effects of BTSK. Pretreatment with the BTSK at daily dose of 100 or 200 mg/kg (p.o.) far 4 weeks reduced serum triglyceride, total cholesterol contents in rat induced by Poloxamer-407 or Triton WR-1339, respectively. Furthermore, post-treatment with BTSK far four weeks also inhibited body weight gain, adipose tissue mass and hyperlipidemia induced by the high fat diet for six weeks. The BTSK shifted serum total-, HDL- and LDL-cholesterol levels toward the values of normal group, suggesting that BTSK has hypolipldemlc effects. The rats fed BTSK reduced lipid peroxide and hydroxy radical in the rat blood and increased superoxide dismutase (SOD) activity compared to the control group. Taken together, these results superoxide that BTSK improve hyperlidemia and obesity via the upregulation of anti-oxidative mechanism.
The effects of taurine on plasma and liver cholesterol, erythrocyte ouabain sensitive Na efflux and platelet aggregation were examined in Sprague Dawley rats fed control or 0.5% cholesterol with 0.2% cholate diet. Plasma and liver levels of total cholesterol were increased significantly (p<0.05) in rats fed cholesterol diet compared to the control, and taurine significantly decreased the elevated plasma level of cholesterol in rats fed cholesterol diet (p<0.05). HDL-cholesterol was decreased in groups fed the cholesterol diet regardless of taurine supplementation and the difference between groups with and without cholesterol was significant (p<0.01). Plasma triglyceride was decreased and liver triglyceride was increased both significantly (p<0.05) in rats fed cholesterol compared to the control. Plasma and liver triglyceride in rats fed taurine was decreased significantly compared to the control (p<0.05). Intracellular Na tended to be lower in rats fed cholesterol or taurine and higher in rats fed cholesterol plus taurine compared to the control. Na efflux through Na-K ATPase and the passive leak of Na was somewhat reduced in rats fed cholesterol or taurine and was augmented in rats fed cholesterol plus taurine compared to the control, which showed a similar trend to the intracellular Na. Taurine supplementation caused a suppression of Na efflux in groups fed control diet and restored the suppressed Na efflux in groups fed cholesterol. Platelet aggregation was significantly decreased in the group fed taurine compared to the control (p<0.05) and the group fed cholesterol plus taurine was also a little lower in aggregation than the group fed cholesterol. Microscopic examination showed that taurine prevented fatty liver in rats fed cholesterol diet. Taurine known for stimulating Na-K ATPase in some cell types rather decreased erythrocyte ouabain sensitive Na-K ATPase in the present study. Taurine had hypolipidemic and hypocholesterolemic effects and inhibited platelet aggregation which may be favorable for prevention of cardiovascular diseases.
This research was designed and implemented to evaluate how the composition of yaksuncha can affect the health of individuals suffering from diet-related diseases such as obesity and hyperlipidemia by prescribing Yaksun as a nutritional supplement with daily meals. The yaksunucha was prepared as Koekac, Sansa, Heshouwu and Woolong tea and the tea's effects on serum lipids and oxidative stress were evaluated by clinical procedures. The yaksuncha significantly increased HDL-cholesterol and decrease of LDL-cholesterol concentrations in serum. It also had significant effects on decreasing oxidative stress and homocystein levels. The physical characteristics of the yaksunucha were also examined, showing. Brix, pH and titratable acidity values of 1.4, 5.50 and 0.05% respectively. It is thought that scientific and objective evaluations were completed on the components of the yaksuncha prescription. Thus one can concluded that the components could be applied not only in the form of tea, but also various food. The data derived from this study provides basic information that will aid in the application of oriental medicinal resources to other foods as well as facilitate the study of medicinal herbs within the field of functional food research, which already draws sizable attention worldwide.
To study the effect of prosomillet (Panicum milaceum) on lipid metabolism, male Sprague-Dawley rats weighing 190$\pm$8g were fed six experimental diets for four weeks. The six diets based on AIN-76 composition consisted of one cholesterol-free(normal) and five 1%(w/w) cholesterol diets, i.e. control, two diets containing additional 0.3 and 0.6%(w/w) methanol extracts of prosomillet and another two diets containing 15 and 30% (w/w) prosomillet powder. There was no difference in weight gains between the groups but relative liver weights increased under the cholestrol diets. Plasma levels of total cholesterol and triglyceride(TG) decreased by 23-27% and by 37-52%, respectively, in the four prosomillet diet groups compared to those of the normal and control groups. Whereas in the liver, only TG levels decreased in the prosomillet diet groups. Fecal excretions of bile acid and cholesterol significantly with methanol extracts of prosomillet. There was a significant increase in the activity of hepatic microsomal cholesterol 7$\alpha$-hydroxylase when feeding 1% cholesterol but prosomillet in the diet, either as in the form of powder or methanol extract, appeared to have only slight additional effects, namely increases in enzyme activity. The activity of liver cytosolic glucose-6-phophate dehydrogenase (G6PDH) tended to be reduced with high cholesterol diets and dropped markedly by 15% using additional prosomillet powder. Those of the liver cytoxolic malic enzyme had a similar tendency to those of G6PDH. The results indicate that certain active components in prosmillet other than fiber have the potential to exert hypolipidemic effects via regulating cholesterol excretions and lipogenesis.
This study was carried out to investigate the supplementary effects of soybean oil and rice gem oil compared with lard on lipid metabolism of insulin dependent diabetic mice. Streptozotocin-induced diabetic mice were fed three kinds of experimental diets with 20% lipid from lard(L), soy bean oil(SBO) and rice gem oil(RGO) for 7 weeks, respectively. Diet intake, body weight, organs weights and lipids levels of serum, liver and feces were measured. There was no significant difference in diet intake, body and organs weights among experimental groups. But the concentrations of serum triglyceride of SBO and RGO groups, and of serum total cholesterol were lower in RGO group than in the other groups. The hepatic total lipid and total cholesterol levels of RGO group were significantly lower than those of the other groups. The contents of total lipid and total cholesterol excreted in feces of SBO and RGO groups were higher than those of L group, and the significance was shown only in RGO group. These results suggested that soy bean oil and rice germ oil can reduce serum triglyceride and total cholesterol levels and hepatic total lipid concentration of insulin dependent diabetic mice compared with lard as a animal fat source by increasing fecal lipid excretion of these groups. But the significant reducing effects on serum and liver lipid levels were shown only in RGO group, and we need to investigate the hypolipidemic effect of this oil by supplementary level and period.
Objectives : In present study, therefore, possible beneficial pharmacological activities of standard potato protein extracts (SPE) were observed on the mild diabetic obese mice. Methods : After end of 12 weeks of continuous oral administrations of three different dosages of SPE 400, 200 and 100 mg/kg, or metformin 250 mg/kg, analyzed the hepatoprotective, hypolipidemic, hypoglycemic, nephroprotective and anti-obesity effects, separately. In addition, liver antioxidant defense systems were additionally measured with lipid metabolism-related genes expressions and hepatic glucose-regulating enzyme activities for action mechanism. Results : All of diabetes and related complications including obesity were significantly inhibited by treatment of SPE 400, 200 and 100 mg/kg, dose-dependently, and they also dramatically normalized the hepatic lipid peroxidation and depletion of liver endogenous antioxidant defense system, the changes of the hepatic glucose-regulating enzyme activities, also changes of the lipid metabolism-related genes expressions including hepatic $AMPK{\alpha}1$ and $AMPK{\alpha}2$ mRNA expressions, dose-dependently. Especially, SPE 200 mg/kg constantly showed favorable inhibitory activities against type II diabetes and related complications as comparable to those of metformin 250 mg/kg in HFD mice, respectively. Conclusions : The present work demonstrated that SPE 400, 200 and 100 mg/kg showed favorable anti-diabetic and related complications including obesity refinement activities in HFD mice, through AMPK upregulation mediated hepatic glucose enzyme activity and lipid metabolism-related genes expression, antioxidant defense system and pancreatic lipid digestion enzyme modulatory activities.
Opuntia humifusa contains high levels of antioxidants including vitamin C, flavonoids and polyphenols, which may provide beneficial effects such as hypolipidemic activity and the reduction of atherosclerosis in postmenopausal women. This study was conducted to determine if the intake of O. humifusa seeds powder (OHS) regulates lipid concentrations, glutamate-oxaloacetate transaminase (GOT), and glutamate-pyruvate transaminase (GPT) in the serum of ovariectomized rats. Sprague-Dawley female rats were randomly assigned to either a sham-operated group (Sham) or one of the following four ovariectomy (OVX) subgroups: OVX with vehicle (OVX), OVX with 100, 200, and 500 mg/kg/day OHS (OHS100, OHS200, OHS500). Daily oral administration of OHS was initiated one week after ovariectomy and continued for seven weeks. Upon completion of treatments, organs were weighed and GOT, GPT, total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) and high-density lipoprotein cholesterol (HDL-C) levels were determined enzymatically. No significant differences in feed intake and organ index were observed among the groups. Significant decreases in GPT, TC and LDL-C (p<0.05) were observed in all of the OHS groups (OHS100, 200 and 500), while no significant changes in HDL-C were observed. In addition, the OHS200 and OHS500 treatment groups exhibited a lower level of serum GOT compared to the OVX group. These results indicate that supplementation with O. humifusa seeds could induce favorable changes in serum lipoprotein and lipid profiles, which frequently worsen with inadequate estrogen availability.
This research was planned and executed to evaluate how the composition of Yak-sun(oriental diet therapy) can effect health conditions of people who are suffering from diet-related diseases like obesity and hyperlipidemia by taking Yak-sun in a form of nutritional supplement with our daily meals. We produced Jangswucha with Koekac, Sansa, Heshouwu, Wulong and evaluated how this tea effects on serum lipids and oxidative stress by clinical practices. Also we examined physical characteristics of Jangswucha. Brix, pH and titratable acidity of Jangswucha were 1.4, 5.50 and 0.05%. With this observation, we found out that this tea has significant effect on increase of HDL-cholesterol, decrease of LDL-cholesterol concentration in serum. Also this tea significant effect on decrease oxidative stress and homocystein content. We think that scientific and objective evaluation was done on the components of Yak-sun tea prescription. We concluded that we could apply the components not only in a form of tea, but also in other forms of various food. The information we received from this conclusion will be a basic information on how we can apply oriental medicinal resources into other food and will also be a steppingstone for medicinal herbs to step foot in the field of functional food research, which already draws sizable attention world-wide.
This study was conducted to investigate the application possibility of leaf and stem extract(LSE) extracted from mixture of leaf and stem of ginseng radix (Panax Ginseng C.A. Meyer). We conducted analysis of the ginsenoside content by HPLC. Also we investigate the effects of the LSE on the reduction of serum lipid and improvement of blood parameters in rats fed high fat diet 5 weeks. We examined by analyzing the serum total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride and atherogenic index and hematological datas and serum metabolic variables. Sprague-Dawley rat weigh $150\;g\;{\pm}\;15\;g$, were ramdomly assigned to 4 groups, basal diet only(BDG), high fat diet weithout LSE(FDCG), high fat diet and 10% LSE(FD10G), high fat diet and 20% LSE(FD20G). The result of this study were as follow. Hematological datas of 4 groups were same level, which were not significant. The activities of ALP, GOT and LDH level were significantly different. Total cholesterol, LDL-cholesterol, triglyceride contentrations in serum and atherogenic index were remarkably reduced in LSE supplemented groups as compared high fat control groups. These result imply that LSE could be used as possible for decrease of serum lipid concentration.
The effects on the radical scavenging activities and hepatic lipid levels in rats fed a high-fat diet (HFD) in the 70% ethanolic extract from Acer tegmentosum Maxim (ATM) were evaluated. Total phenol content of ATM was 168.60 mg catechin/g in the 70% ethanolic extract of Acer tegmentosum. The DPPH and ABTS radical scavenging activities were 18.32 mM TE/g and 32.25 mM TE/g, respectively. Food efficiency ratio was lower significantly in supplemented group with 150 mg/kg BW/day during 5 weeks (HFD+ATM) compared to HFD. Total cholesterol and triglyceride contents in liver tissue of HFD+ATM were lower significantly compared to those of the HFD. Supplementation of ATM significantly decreased lipid peroxide contents and increased radical scavenging activity in the liver tissue compared with that of HFD group. Moreover, the hepatocytes of HFD rats showed a typical fatty liver morphology showing the presence of cytoplasmic lipid droplets, whereas administration of ATM attenuated the number and the size of lipid droplets. In the liver tissue of ATM administrated HFD group, the mRNA levels of SREBP-1c, $PPAR{\gamma}$, and FAS were decreased. Therefore, these results suggest that Acer tegmentosum extracts could have antioxidant activities and the hypolipidemic effects in liver tissue by its phenolic compounds.
