천마분말이 고콜레스테롤혈증 흰쥐 혈청의 지질성분에 미치는 영향 (Effects of Gastrodia rhizoma on Lipid Components of Serum in Hypercholesterolemic Rats)

  • This study was designed to investigate the optimum recommendation level on Gatrodia rhizoma and the effects on the improvement of the lipids in the dietary hypercholesterterolemic rats. Experimental diets mixed with 5% Dioscorea batatas(Group 2), 10% Dioscorea batatas(Group 3), 15% Dioscorea batatas (Group 4) 5% Gastrodia rhizoma (Group 5), 10% Gastrodia rhizoma (Group 6), and 15% Gastrodia rhizoma (Group 7), were administered to the male rats of the Sprague Dawley for 3 weeks. Concentration of total cholesterol in serum was lower in the Gastrodia rhizoma groups than in the other groups, especially total cholesterol concentration of 10% Gastrodia rhizoma(Group 6) was the lowest in the Gastrodia rhizoma groups. Concentration of HDL-cholesterol in serum was higher in the 10% Gastrodia rhizoma and 15% Gastrodia rhizoma than in the other groups. Concentrations of cholesteryl ester, LDL, LDL-cholesterol in serum were the lowest in the 10% Gastrodia rhizoma. Concentration of glucose and activity of GPT in serum were the lowest in the 10% Gastrodia rhizoma group. The activity of GOT in serum was lower in the 10% Gastrodia rhizoma group and 15% Gastrodia rhizoma group than in the other groups. Therefore, we consider that there are effects on the improvement of the lipids in the Gastrodia rhizoma and the optimum recommendation quantity of Gastrodia rhizoma is 10% to quantity of food composition.

식이칼슘 수준이 콜레스테롤 첨가 또는 무첨가 고지방식 섭취 흰쥐의 지질대사에 미치는 영향 (Effects of Dietary Calcium Levels on Lipid Metabolism in Rats Fed High Fat Diet with or without Supplemental Cholesterol)

  • 본 연구에서는 성숙한 흰쥐에게 동물성 포화지방(식이중 beef tallow로서 18%)과 식이에 콜레스테롤을 첨가하지 않은 조건과 첨가한 조건(1%) 하에서 식이칼슘을 수준별로(식이에 0.1%, 0.5%, 1.5% 첨가) 4주간 동안 급여한 후 혈액과 조직의 지질함량, 소장내용물 중 지질의 분포 및 분중 지질배설량 등이 측정되었다. 그 결과, 콜레스테롤 무첨가의 고지방식을 4주간 급여한 경우에는 고콜레스테롤혈증은 유발되지 않았으며, 식이칼슘의 투여효과도 인정되지 않았다. 한편 콜레스테롤 첨가 고지방식을 급여한 경우에는 고콜레스테롤혈증의 유발이 가능하였다. 이때 식이칼슘의 투여효과는 식이칼슘의 수준 증가에 따라 혈청 콜레스테롤 농도가 저하하는 경향을 보였다. 즉 식이칼슘 섭취 부족에 따른 혈중 콜레스테롤 상승효과(hypercholesterolemic effect)와 칼슘섭취 증가에 따른 혈중 콜레스테롤 저하효과(hypocholesterolemic effect)가 인정되었다. 또 칼슘 섭취가 체내 지질대사에 미치는 효과에 대한 하나의 작용메카니즘으로서 콜레스테롤의 소화율과 장관내 흡수양상이 검토되었는데, 이때 분중 지질성분의 배설량은 고칼슘식이군에서 유의적으로 높았고 콜레스테롤의 소화율은 저칼슘식이군에서 유의적으로 높았다. 이것은 칼슘이 지질 성분의 배설을 촉진하므로서 지질 성분의 흡수를 저하시킨다고 하는 장관내 지질 흡수 메카니즘에 대한 “지질-칼슘 soap 형성”으로 칼슘의 역할이 설명될 수 있다. 본 과제에서 처음으로 시도된 소장내용물의 지질과 칼슘성분의 존재양상은 지질 흡수와 칼슘의 상호작용을 보다 직접적인 방법으로 측정할 수 있었으나, 얻어진 결과에 대한 해석에는 아직 어려움이 있었다.

고 콜레스테롤 식이 투여 흰쥐에 있어서 녹차의 콜레스테롤 저하 효과 (Hypocholesterolemic Effects of Green Tea in Cholesterol-Fed Rats)

  • 본 연구는 녹차의 혈중 콜레스테롤 저하 효과 및 녹차 추출물인 catechin과 EGCG가 hypocholesterolemic 효과에 미치는 정도를 식이로 고콜레스테롤혈증을 유도한 흰쥐에서 평가하고자 수행하였다. 4주령의 수컷 SD계 흰쥐를 난괴법에 따라 각 군당 5마리씩 무녹차섭취군(대조군), 4% 녹차분말군(GTP), 1% 녹차 catechin군(Catechin),0.5% 녹차 EGCG군(EGCG)의 4군으로 7주간 사육을 하였다. 녹차 첨가군은 모두 0.5% EGCG를 함유하도록 녹차 첨가량을 결정하였으며 모든 식이는 고콜레스테롤혈증을 유도하기 위하여 1% cholesterol과 0.5% cholic acid를 첨가하였다. 식이섭취량은 각 군간에 유의적인 차이를 나타내지 않았다. 혈청 총 및 LDL-콜레스테롤 수준은 Catechin군이 189.2 mg/dL, 166 mg/dL, EGCG군에서는 161.4 mg/dL, 138 mg/dL로 나타나 대조군(245 mg/dL, 222 mg/dL)에 비하여 유의적으로 감소되었고 (p<0.05), EGCG군에서 그 효과는 더욱 크게 나타났다. 혈청의 중성지방농도는 녹차섭취군이 대조군보다 낮았으며 EGCG군에서 가장 낮게 나타났다. HDL-콜레스테롤 수준은 유의적이지는 않았지만 녹차섭취군이 대조군보다 높은 경향을 보였고, HDL/LDL-콜레스테롤 비율은 대조군에 비하여 녹차섭취군에서 증가하여 EGCG군에서 가장 높았다. 이와 같은 결과는 녹차의 섭취가 혈중 콜레스테롤 및 중성지방을 감소하고, HDL-콜레스테롤은 증가시킴으로써 동맥경화 위험을 줄일 수 있는 가능성을 반영하고 있다. 간에서의 CYP7Al활성과 유전자 발현정도는 Catechin군, EGCG군이 대조군에 비하여 높게 나타났으며, EGCG군에서 가장 많이 증가하여 혈청에서의 지질수준 감소 경향과 일치하였다. 이상의 본 연구 결과들은 녹차 섭취군에서의 혈중 콜레스테롤 저하작용 특히 EGCG군에서의 현저한 hypocholesterolemic 효과는 CYP7Al 유전자 발현 및 효소의 활성증가에 따른 콜레스테롤의 담즙산으로의 배설 증가에 기인하는 것으로 추측된다.

Anti-atherosclerotic Effect of the Methanol Extract of Sorbus commixta Cortex in the High Cholesterol-Diet Rats

  • Hypercholesterolemia is a pivotal pathogenic factor for the development and maintenance of atherosclerosis. The present study was designed to evaluate whether the methanol extract of Sorbus commixta cortex (MSC) restores vascular dysfunction in association with the aortic expressions of proinflarnmatory and adhesion molecules in high cholesterol (HC) diet-rats. Chronic treatment with low (100 mg/kg/day) or high doses (200 mg/kg/day) of MSC lowered the increase in plasma levels of triglyceride (TG) and low-density lipoprotein (LDL) cholesterol induced by a cholesterol-enriched diet without affecting on the plasma level of high density lipoprotein (HDL)-cholesterol. Vascular tone attenuated in the HC-diet rats was restored by administration with MSC. Treatment with MSC also suppressed the HC-induced increase in the monocyte chemoattractant protein-1 (MCP-1) and nuclear factor-$_K$B (NF-$_K$B) p65 expressions as well as expressions levels of adhesion molecules including intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (ICAM-1), and E-selectin in aorta. The present study also showed that MSC inhibited the HC-mediated induction of ET-1 and ACE expression. In histopathological examination, aortic segments in the HC-diet rat revealed thickening intima and media, which were blocked by administration with MSC. Taken together, MSC could suppress the development of atherosclerosis in the HC-diet rat model through the inhibition of the aortic expression levels of pro-inflammatory and adhesion molecules.

식이지방의 종류와 L-arginine 보충이 당뇨쥐의 ENdothelium 기능 및 체내 지질대사에 미치는 영향 (Effect of Dietary Fat Sources and L-arginine Supplementation on Endothelial function and Lipid Metabolism in Streptozotocin-Induced Diabetic Rats)

  • Alteration in the syntesis or enhanced inactivation of nitric oxide(NO) can induce impairment of endothelial cell function. Insulin dependent diabetes mellitus(IDDM) is characterized by impaired endothelial function and vascular disease. NO is produced through L-arginine pathway To elucidate the hypothesis that the decreased production on NO in IDDM reflects vascular damage and the NO production can be manipulated by either dietary fat(7% of kg diet) or the oral supplementation with L-arginine(2g/kg bw), plasma markers for vascular endothelial damage and plasma lipid profiles were measured in streptozotocin(STZ)-induced diabetic rats. Diabetic or normal Sprague-Dawley rats were fed 6 different experimental diets for 4 weeks(SO : soybean oil, SOA: soybean oil + L-arginine supplementation, BT : beef tallow, BTA_ beef tallow + L-arginine supplementation, OV olive oil, OVA : olive oil + L-arginine supplementation). Plasma glucose, total cholesterel, HDL-cholesterol, LDL-cholesterol and triglyceride were measured. Endothelial markers, plasma von Willebrand factor(vWf), thromboxane B$_2$, and 6-keto PGF1$\alpha$ of aorta were measured by ELISA. Plasma NO production was evaluated through the measurement of nitrite by EIA. Feeding saturated fatty acid(SFA, BT) increased relative liver size(RLS) in diabetic rats compared to either polyunsatunted fatty acid(PUFA, SO) or monounsaturated fatty acid(MUFA, OV) The supplementation of L-arginine inhibited the liver and kidney enlargement in olive oil find diabetic rats. Plasma glucose was lower in diabetic animal find the olive oil compared to fed beef tallow and the supplementation L-arginine decreased it in diabetic rats find beef tallow significantly(p < 0.05). Plasma TXB$_2$ levels were increased due to diabetes and the value of beef tallow group showed highest value. Plasma vWf concentration of beef tallow group was higher value in normal rats and was elevated more in diabetes. In diabetic groups, the vWf concentration of olive oil group was lower than beef tallow or soybean oil group. The supplementation of L-arginine in diabetic rats decreased plasma TXB$_2$ and vWf levels significantly(p < 0.05). NO production was higher in normal olive oil fed rats and was tend to be decreased in diabetic rats and the supplementation of L-arginine recovered to normal value(p < 0.05), Olive oil supplemented with L-arginine tended to lower plasma total cholesterol and LDL-cholesterol after 4 week treatment. These results suggest that generalized vascular endothelial changes based on plasma TXB$_2$and vWf occurs in diabetic rats. and olive oil with L-arginine supplementation contributes to a better control of the hyperglycemia, endothelial changes and hypercholesterolemia accompanying diabetes as compared with beef tallow or soy bean oil in this rat model.

Fruiting Body Extracts of Paecilomyces tenuipes Ameliorate Lipid and Antioxidant Metabolism in Rats Fed a High Fat-Cholesterol Diet

  • The present study examined the lipid lowering and antioxidant activity of Paecilomyces tenuipes. Eight week-old male Sprague-Dawley rats were fed one of the three diets, a reference diet without cholesterol addition (NC), a high fat (17 g/ 100 g)-high cholesterol (1 g/100 g) diet (HC) and a HC diet supplemented with 3% P. tenuipes (PT) for 30 days. Total lipid and total cholesterol were reduced significantly by 33 and 37%, respectively, in the PT diet group compared with controls. A similar reduction was found for low density lipoprotein (LDL) cholesterol levels, while plasma high density lipoprotein (HDL) cholesterol and triglyceride (TG) concentrations were not significantly different among groups. Hepatic total lipid and total cholesterol levels, but not hepatic TG levels, were significantly decreased in the PT group compared to the HC group. The administration of P. tenuipes increased the plasma total antioxidant potential and decreased the levels of lipid peroxidation. These results suggest that P. tenuipes exerts significant health benefits through the modulation of physiological functions including a variety of atherogenic lipid profiles and antioxidants in hypercholesterolemia.

고지혈증 랫트를 이용한 수종 전통 한약제의 항고지혈 효과 (Antihyperlipidemic Activity of Several Traditional Herbal Medicines on Experimental Hyperlipidemia in Rats)

  • Three herbal medicine recipes were tried on the animal model of diet induced hypercholesterolemia to screen for the lipid-lowering effect. The recipes adopted were based on the prescription for atherosclerosis-related symptoms by folk-medicine practioners. Hyperlipidemia was induced in rats by giving the high lipid/cholesterol diet for 7 days. Then, the recipes started with the normal diet. Blood was sampled at 1,2,3 and 5 week-points of the treatment, and levels of total cholesterol, triglyceride, high density lipoprotein and lower density lipoprotein were measured. Dae-shiho-tang decreased total cholesterol level significantly. Hwangryun-haedok-tang and Samhwang-sasim-tang slightly decreased total cholesterol level, although it was not statistically significant.

Effect of St. John's Wort (Hypericum perforatum) on obesity, lipid metabolism and uterine epithelial proliferation in ovariectomized rats

  • BACKGROUND/OBJECTIVES: This study was conducted to assess the potential of St. John's Wort (Hypericum perforatum) to prevent obesity and abnormalities in lipid metabolism induced by ovariectomy in a rat model without stimulatory activity on uterus. MATERIALS/METHODS: Ovariectomized (OVX) rats were treated for 6 weeks with 70% ethanol extracts of Hypericum perforatum [HPEs: whole plant (WHPE) and flower and leaves (FLHPE)], ${\beta}$-estradiol-3-benzoate at a dose of $50{\mu}g/kg/day$ (E2) or vehicle (distilled water). RESULTS: As expected, OVX increased body weight gain and adiposity and showed higher food efficacy ratio. OVX also increased the serum cholesterol as well as insulin resistance, while reducing uterus weight and uterine epithelial proliferation rate. HPEs (WHPE and FLHPE) showed estrogen-like effect on body weight gain, adipose tissue weight and food efficacy ratio in OVX rats. HPEs prevented hypercholesterolemia induced by OVX more effectively than E2. E2 increased uterus weight and epithelial proliferation rate in OVX rats, while HPEs maintained them at the level of the sham-operated animals. CONCLUSIONS: Our finding demonstrates that HPEs can be considered as an effective agent to prevent OVX-induced obesity without stimulatory activity on uterus.

Red Wine Prevents Brain Oxidative Stress and Nephropathy in Streptozotocin-induced Diabetic Rats

  • We have studied the effects of red wine on brain oxidative stress and nephropathy in streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in Wistar rats with a single intraperitonally injection of STZ (50 mg/kg). Two weeks before and four weeks after injection, red wine was given orally in both normal and diabetic rats. Blood samples were taken from the neck vascular trunk in order to determine the glucose, triglycerides, total cholesterol, HDL-cholesterol (HDL-c), atherogenic index (AI), total protein, blood urea nitrogen (BUN), creatinine, insulin, lipid peroxidation products, reduced glutathione (GSH) and superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities. As well, we estimated the lipid peroxidtion, GSH and SOD, GSH-Px and catalase activities in brain and renal homogenates, and the excretion of albumin, proteins and glucose in urine over 24 h period. The administration of STZ caused significant increases in levels of glycosuria, proteinuria, albuminuria, glycemia, total cholesterol and AI, as well as in lipid peroxidation products in the brain, plasma and kidney, whereas it decreased the GSH content and SOD, GSH-Px and catalase activities. Treatment with red wine significantly prevented the changes induced by STZ. These data suggested that red wine has a protective effect against brain oxidative stress, diabetic nephropathy and diabetes induced by STZ, as well as it protects against hypercholesterolemia and atherogenic risk.

Effect of Dietary Inclusion of Lactobacillus acidophilus ATCC 43121 on Cholesterol Metabolism in Rats

  • This study examined the effects of Lactobacillus acidophilus ATCC 43121 (LAB) on cholesterol metabolism in hypercholesterolemia-induced rats. Four treatment groups of rats (n=9) were fed experimental diets: normal diet, normal $diet+LAB(2{\times}10^6\;CFU/day)$, hypercholesterol diet (0.5% cholesterol, w/w), and hypercholesterol diet+LAB. Body weight, feed intake, and feed efficiency did not differ among the four groups. Supplementation with LAB reduced total serum cholesterol (25%) and VLDL+IDL+LDL cholesterol (42%) in hypercholesterol diet groups, although hepatic tissue cholesterol and lipid contents were not changed. In the normal diet group, cholesterol synthesis (HMG-CoA reductase expression), absorption (LDL receptor expression), and excretion via bile acids (cholesterol $7{\alpha}-hydroxylase$ expression) were increased by supplementation with LAB, and increased cholesterol absorption and decreased excretion were found in the hypercholesterol diet group. Total fecal acid sterols excretion was increased by supplementation with LAB. With proportional changes in both normal and hypercholesterol diet groups, primary bile acids (cholic and chenodeoxycholic acids) were reduced, and secondary bile acids (deoxycholic and lithocholic acids) were increased. Fecal neutral sterol excretion was not changed by LAB. In this experiment, the increase in insoluble bile acid (lithocholic acid) reduced blood cholesterol level in rats fed hypercholesterol diets supplemented with LAB. Thus, in the rat, L. acidophilus ATCC 43121 is more likely to affect deconjugation and dehydroxylation during cholesterol metabolism than the assimilation of cholesterol into cell membranes.
