Effects of Leonurus Sibiricus L. Extract on Diet-induced Hypercholesterolemia in Rats (익모초 추출물이 흰쥐의 식이성 고콜레스테롤혈증에 미치는 영향)

  • This study was performed to investigate the cholesterol-lowering effect of liquid extract from Leonurus sibiricus Linne (LS). SD rats were fed atherogenic diet for 6 weeks. During the experimental period, LS liquid extract (3 g/kg and 6 g/kg) was orally given to the rats. The liver weight and levels of total cholesterol, LDL-cholesterol, and triglycerides in serum were significantly increased in rats fed with atherogenic diets. Oral treatment of LS liquid extract to the rats fed atherogenic diet significantly reduced the liver weights and significantly decreased the levels of total cholesterol, LDL-cholesterol, and triglycerides. The level of serum HDL-cholesterol was significantly increased at a high dose group compared with rats fed normal diet chow. These results indicate that LS liquid extract has the potential to restore altered lipid profiles induced by cholesterol rich diets and prevent the development of atheroclerosis.

Isolation of Hyperlipidemic Substances from Methanol Extract of Paeoniae Radix (작약의 메탄올 추출물로부터 항고지혈 활성성분의 분리)

  • We previously showed that a methanol extract of Paeoniae radix decreased total cholesterol level in rats with hyperlipidemia. In order to isolate the active ingredient(s), the methanol extract of Paeoniae radix was fractionated with chloroform/methanol(4:1) solution and isolate into soluble part and insoluble part of the the methanol extract. Above two parts were tested on the experimentally induced hypercholesterolemia in rats for the lowering effect of serum lipoprotein contents. Hyperlipidemia was induced on male Wistar rats by feeding high choleserol diet for 7 days. After oral administration of above samples for 4 weeks, serum lipid profile was verified on these rats by measuring total cholesterol, triglyceride, high density lipoprotein cholesterol and low density lipoprotein cholesterol. The chloroform/methanol(4:1) soluble part and insolule part showed lowering activity of total cholesterol level and triglyceride level at 4 week point significantly(p<0.01 and p<0.05) compare with the control group and the soluble part was more effective.

Cholesterol-lowering Effect of Rice Protein by Enhancing Fecal Excretion of Lipids in Rats

  • The aim of this study was to investigate the effects of isolated protein from white rice on lipid metabolism in a hypercholesterolemic animal model. Male Sprague-Dawley rats were divided into three groups and fed either a normal diet or a high-cholesterol diet (HCD) containing either casein or isolated rice protein for 4 weeks. Compared with rats fed a HCD with casein, the total cholesterol (TC) level in the plasma was significantly reduced in the rats fed rice protein. However, no significant differences were observed in the triglycerides, high-density lipoprotein (HDL), and glucose levels among the experimental groups. Hepatic total lipids and TC levels were significantly decreased by supplementation with rice protein. In addition, rice protein significantly increased the levels of TC and bile acids in the feces. These results suggest that rice protein may improve HCD-induced hypercholesterolemia by enhancing fecal excretion of cholesterol.

Effect of Several Solvent Extracts from Paeoniae Radix on Experimental Hyperlipidemia in Rats (고지혈증 랫트를 이용한 작약의 수종 용매 추출물에 의한 항고지혈 효과)

  • Hexane, chloroform, methanol and water extracts of Paeoniae Radix were tested on the experimentally induced hypercholesterolemia in rats for lowering effect of serum lipoprotein contents. Hyperlipidemia was induced on male Wistar rats by feeding high cholestetrol diet for 7 days. Serum lipid profile was verified on these rats by measuring total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL), and low density lipoprotein cholesterol (LDL). Then, the diet was changed to normal. At the same time, hexane, chloroform, methanol and water extract of Paeoniae Radix were given orally on daily basis, and the changes in the serum lipid profile were assessed for 4 weeks. Methanol extract of Paeoniae radix decreased TC level at 1, 2, and 4 week point significantly, and water extract decreased TC level at 4 week point significantly comparing with the control group.

Hypocholesterol Effect of Opuntia humifusa Extract on High Cholesterol Diet-induced Hypercholesterolemic Rats (고콜레스테롤혈증을 유도한 쥐를 대상으로 천년초 추출물의 콜레스테롤 저하 효과)

  • The aim of this study was to investigate the hypocholesterol effects of Opuntia humifusa extract in hypercholesterolemic rats. Rats (8-week-old, male) were randomly divided into four groups (n=4) as follows: N-control, normal diet; H-control, hypercholesterolemic diet; OH-1, 2% O. humifusa extract-supplemented hypercholesterolemic diet; OH-2, 4% O. humifusa extract-supplemented hypercholesterolemic diet. After 4 weeks, we observed that hypercholesterolemia induced significant increases in serum lipids (total cholesterol and LDL-cholesterol), hepatic lipids (total cholesterol and triglyceride), and hepatic function parameters (ALT, alanine aminotransferase; AST, aspartate aminotransferase) (P<0.05). Treatment with O. humifusa extract for 2 weeks normalized these indexes up to the levels of normal rats. O. humifusa extract tended to increase fecal lipid (H-control: 142.74 mg/day vs. OH-1: 214.05 mg/day; OH-2: 200.95 mg/day, P<0.05) and bile acid (H-control: 37.07 ${\mu}M/day$ vs. OH-1: 47.23 ${\mu}M/day$, OH-2: 47.93 ${\mu}M/day$, P<0.05) contents of hypercholesterolemic rats. We concluded that oral administration of O. humifusa extract effectively improved cholesterol metabolism in a hypercholesterolemic animal model induced by hypercholesterol diet.

Studies on the Hypocholesterolemic Activities of Glycyrrhizin (Glycyrrhizin의 Hypocholesteroleinic Action에 관(關)한 연구(硏究))

  • Glycyrrhizin, an active constituent of Glycyrrhiza, was chemically extracted and its hypocholesterolemic activities were studied in rats. Following were the results: 1. The Sprague Dowley rats fed with the standard diet were divided into 4 groups, $50{\sim}60$ in each, and glycyrrhizin was given in doses of 0 (control), 0.5, 1.0 and 2.0mg per 100gm body weight daily for 10, 20, 30, and 60 days. Glycyrrhizin showed more or less the hypocholesterolemic activities in all animals. All of the animal groups to which 1. 0mg and 2.0mg of glycyrrhizin were given died before 60 days, while no mortality cases was noted in the group to which 0.5mg of glycyrrhizin was given. 2. Cholesterol administration induced hypercholesterolemia in all experimental animals, however, simultaneous administration of cholesterol with 0.5mg of glycyrrhizin suppressed the rise of the total serum cholesterol levels. The former group did not survive until 56 days, while the latter did. 3. Glycyrrhizin also Suppressed the cholesterol biosynthesis and incorporation of cholesterol in the liver tissues. 4. Glycyrrhizin induced the increase of the fecal excretion of the sterol compounds.

Effects of Varied Mushroom on Lipid Compositions in Dietary Hypercholesterolemic Rats (고콜레스테롤혈증 유발 흰쥐에 있어서 버섯류가 지질 성분에 미치는 영향)

  • For the investigation of the effects of mushrooms on prevention of hypercholesterolemia, dietary hyperc-holesterolemic rats were fed for 2 weeks with basal diet containing 5% G. lucidum, 5% L. edodes, 5% A. judae and 10% G. lucidum, 10% L. ededos or 10% A. judae mushroom. Concentrations of total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, phospholipid, free-cholesterol, cholesteryl ester, VLDL and chylomicron in serum were analyzed. The result obtained are as follows : Concentrations of total cholesterol, LDL, LDL-cholesterol in serum were lowest in the group 6 (10% L. edodes mushroom) and HDL-cholesterol concentration in serum was significantly higher than the concentrations of the control group. Concentrations of triglyceride, phospholipid, free-cholesterol, cholesteryl ester in serum were lower than the concentration of the control group, and the concentrations of triglyceride, cholesteryl ester in group 6 were lower than those of the other groups. Concentrations of phospholipid and free-cholesterol in group 3 were lower than those of the other groups.

Effects of cholesterol and Lactobacillus acidophilus on testicular function

  • Objective: In this study, the effects of Lactobacillus acidophilus on testosterone (TES), follicle-stimulating hormone (FSH), luteinizing hormone (LH), androgen-binding protein (ABP), factor-associated apoptosis (FAS), and total cholesterol (TC), as well as histopathological changes, were investigated in male rats fed a high-cholesterol diet. Methods: The study included three groups. The control (C) group was fed standard-diet for 8 weeks. The hypercholesterolemia (HC) group was fed a 2% cholesterol-diet for 8 weeks. The therapeutic group (HCL) was fed a 2% cholesterol-diet for 8 weeks and administered L. acidophilus for the last 4 weeks. FSH, TES, and FAS levels in testicular tissue were determined using an enzyme-linked immunosorbent assay (ELISA), while another sample was examined histopathologically. LH and ABP levels were determined using ELISA, and serum TC levels were assessed via an autoanalyzer. Results: In the HC group, the TC levels were significantly higher and the LH levels were lower (p<0.05) than in the C group. The ABP levels were lower (p>0.05). In the HCL group, the LH and ABP levels were higher (p>0.05) and the TC level significantly lower (p<0.05) than in the HC group. The TES and FSH levels were lower, and the FAS levels were higher, in the HC than in the C group (p<0.05). In the HCL group, levels of all three resembled control levels. Histologically, in the testicular tissue of the HC group, the cells in the tubular wall exhibited atrophy, vacuolization, and reduced wall structure integrity. However, in the HCL group, these deteriorations were largely reversed. Conclusion: Supplementary dietary administration of an L. acidophilus to hypercholesterolemic male rats positively impacted testicular tissue and male fertility hormone levels.

Effects of Callophyllis japonica Extracts on the Glucide and Lipid Metabolism in Hypercholesterolemic Rats (고콜레스테롤혈증 흰쥐의 당질 및 지질 대사에 볏 붉은잎(Callophyllis japonica)추출액이 미치는 영향)

  • This study was designed to observe the effects of the feeding Callophyllis japonica extracts on the improvement of the blood glucose, lipid in the serum of dietary hypercholesterolemic rats(S.D. strain, ?) fed the experimental diets for 4 weeks. Concentrations of total cholesterol and LDL-cholesterol in serum were significantly lower in the Callophylis japonica extracts group than in the choloesterol supplemented diet group. Concentrations of HDL-cholesterol in serum were higher in the Callophyllis japonica extracts group than in the cholesterol supplemented diet group. Concentrations of free cholesterol, triglyceride, phospholipid and blood glucose in serum were significantly lower in the Callophyllis japonica extracts group than in the cholesterol supplemented diet group. The activities of aspatate aminotransferae(AST), alanine aminotransferase(ALT), lactate dehydrogenase(LDH) and alkaline phosphatase(ALP) in serum were lower in the CAllophyllis japonica extracts supplemented group than in the cholesterol diet group. From the above research, the physiological activity substances in Callophylis japonica extracts were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hypercholesterolemic rats.
