DNA Shuffling of aprE Genes to Increase Fibrinolytic Activity and Thermostability

  • Four aprE genes encoding alkaline serine proteases from B. subtilis strains were used as template genes for family gene shuffling. Shuffled genes obtained by DNase I digestion followed by consecutive primerless and regular PCR reactions were ligated with pHY300PLK, an E. coli-Bacillus shuttle vector. The ligation mixture was introduced into B. subtilis WB600 and one transformant (FSM4) showed higher fibrinolytic activity. DNA sequencing confirmed that the shuffled gene (aprEFSM4) consisted of DNA mostly originated from either aprEJS2 or aprE176 in addition to some DNA from either aprE3-5 or aprESJ4. Mature AprEFSM4 (275 amino acids) was different from mature AprEJS2 in 4 amino acids and mature AprE176 in 2 amino acids. aprEFSM4 was overexpressed in E. coli BL21 (DE3) by using pET26b(+) and recombinant AprEFSM4 was purified. The optimal temperature and pH of AprEFSM4 were similar to those of parental enzymes. However, AprEFM4 showed better thermostability and fibrinogen hydrolytic activity than the parental enzymes. The results indicated that DNA shuffling could be used to improve fibrinolytic enzymes from Bacillus sp. for industrial applications.

효소처리에 의한 초당옥수수 추출물의 이화학적 및 항산화 특성의 변화 (Changes in Physicochemical and Antioxidant Properties of by Enzyme-Treated Super Sweet Corn Extracts)

  • To improve usability of super sweet corn, extracts were prepared with hydrolytic enzyme and changes in physicochemical and antioxidant properties were analyzed. Soluble solids and reducing sugars contents were higher in all enzyme treatment groups than in the control. When enzyme treatment time increased, contents of soluble solids and reducing sugars were also increased. There was no significant difference in lightness between treatment groups, with redness showing the highest value in the control and yellowness showing the highest value in the invertase treatment group. Free sugar content in the control was the lowest. However free sugar content in the enzyme combination treatment group was increased by more than four times compared to that in the control. Contents of ascorbic acid, flavonoids and polyphenols were higher in the enzyme treatment group than in the control. In particular, the enzyme combination treatment group showed the highest content. DPPH and ABTS radical scavenging abilities were significantly higher in all enzyme treatment groups than in the control. Radical scavenging abilities of cellulase treatment group and enzyme combination treatment group showed high activity. The activity increased when enzyme treatment time increased. The combined enzyme treatment method for super sweet corn was suitable for food processing.

공업적 이용을 위한 동물성 키틴분해효소의 탐색 (Survey on the Chitinolytic Activity from Some Animals for the Industrial Utilization)

  • 키틴을 효소적인 방법으로 분해하여 키틴올리고머를 생산할 수 있는 값싸고 안정적인 효소원을 확보하기 위하여 농어, 방어, 대구, 닭 등의 동물체로 부터 키틴분해효소를 탐색하였다. 각 효소원의 장기와 소화액 및 달팽이 ${\beta}-glucuronidase$로 부터 키틴분해활성을 측정한 결과, 대구와 닭은 키틴을 주로 중합도 $3{\sim}5$의 크기로 분해하며 endochitinase의 활성도가 exochitinase활성보다 $7{\sim}10$배 높았다. 달팽이 ${\beta}-glucuronidase$는 키틴을 모두 N-acetylglucosamine으로 분해하였고 닭의 경우 소화기조직과 내용물에서 모두 endochitinase활성이 높았다. 어류중에서 농어는 키틴분해활성을 보이지 않았으며 방어와 대구는 비슷한 수준의 키틴분해활성을 보였다. 이들 효소의 최적pH는 $4{\sim}5$, 최적온도는 $50{\sim}60^{\circ}C$였다. 한편 키토산분해능력은 닭의 소화기내용물과 조직, 대구의 위조직에서 관찰되었으며, 닭 소화기내용물이 가장 높았으나 키틴분해능의 15%에 지나지 않았으며 그 다음은 달팽이의 ${\beta}-glucuronidase$로 닭 소화기내용물의 키토산분해능력의 약 30% 활성을 보였다. 키틴올리고머의 생산을 위한 가장 적합한 효소원으로 exochitinase활성이 적고, endochitinase활성이 높으며 값이 저렴하고 안정적인 공급이 가능한 닭의 소화기가 적합한 것으로 사료되었다.

The Effects of Biological Control using the Composted Liquid Manure on Large Patch in Zoysiagrass (Zoysia japonica)

  • This study was conducted to investigate whether several composted liquid manures (CLMs) are useful for biological control of large patch on zoysiagrass and investigate the chemical and biological factors to suppress large patch in soil treated with CLMs. The CLMs were produced at 4 different facilities for livestock excretion treatments located in Korea. Field experiments were carried out at 5 golf courses located near each facility. CLM and Chemical fertilizer (CF: water soluble fertilizer, 20-20-20) were applied four and three times with N at $12g\;m^{-2}$ per year, respectively. There was significant increase of concentration of K, Na, and Cu of soil treated with CLM compared to CF treatment. Among experimental plots, CN and GG2 plot sites were shown significant higher effect of biological control 80% and 50% respectively against large patch disease. The number of bacteria, Actinomycetes, and fungi in soil at these sites significantly increased and fluorescein diacetate hydrolytic activity was enhanced, while the soil was treated with CLM. The results of this study demonstrated that CLM application has effect on soil to suppress large patch and reduce the use of fungicide in environment-friendly turf management.

모델 시스템을 이용한 Poly(l-lactide)의 분해거동 (Degradation Behaviors of Poly(l-lactide) using Model Systems)

  • The hydrolytic kinetics of biodegradable poly(l-lactide) (PLLA) have been studied by using two model systems, solution-grown single crystal (SC) and Langmuir monolayer techniques, for elucidating the mechanism for both alkaline and enzymatic degradations. The present study investigated the parameters such as degradation medium and time. The Langmuir mono layers of PLLA showed faster rates of hydrolysis when they were exposed to a basic subphase rather than they did when exposed to neutral subphase. Both degradation mediums had moderate concentrations to show a maximized activity, depending on their sizes. An alkaline degradation of SCs of PLLA showed the decrease of molecular weight of the remained crystals due to the erosion of chain-folding surface. However, the enzymatic degradation of SCs of PLLA occurred in the crystal edges thus the molecular weight of remained crystals was not changed. This behavior might be attributed to the size of enzymes which is much larger than that of alkaline ions; that is, the enzymes need larger contact area with monolayers to be activated.

Yeast cell surface display of cellobiohydrolase I

  • Recently, genetic engineering techniques have been used to display various heterologous peptides and proteins (enzyme, antibody, antigen, receptor and fluorescence protein, etc.) on the yeast cell surface. Living cells displaying various enzymes on their surface could be used repeatedly as 'whole cell biocatalysts' like immobilized enzymes. We constructed a yeast based whole cell biocatalyst displaying T. reesei cellobiohydrolase I (CBH I ) on the cell surface and endowed the yeast-cells with the ability to degrade cellulose. By using a cell surface engineering system based on ${\alpha}-agglutinin,$ CBH I was displayed on the cell surface as a fusion protein containing the N-terminal leader peptide encoding a Gly-Ser linker and the $Xpress^{TM}$ epitope. Localization of the fusion protein on the cell surface was confirmed by confocal microscopy. In this study, we report on the genetic immobilization of T. reesei CBH I on the S. cerevisiae and hydrolytic activity of cell surface displayed CBH I.

광학활성 Phenyl Oxirane 제조용 유전자 재조합 생촉매 개발 (Development of Recombinant Saccharomyces cerevisiae Expressing Epoxide Hydrolase for the Preparation of Chiral Phenyl Oxirane)

  • Aspergillus niger #33로부터 RT-PCR 및 PCR방법을 이용하여 epoxide hydrolase (EH)유전자를 클로닝 하고 염기서 열을 분석한 결과 A. niger LCP521 유래의 EB와 85%수준의 유사성을 가지고 있었다. 클로닝된 EH 유전자를 Sacrharomyces cerevisiae에 형질전환 시킨 후 galactose를 inducer로 사용하여 발현시켰다. 유전자 재조합 S. cerevisiae는 라세믹 phenyl oxirane 기질에 대하여 입체선택적 가수분해능이 있음을 확인할 수 있었으며, 이러한 유전자 재조합 EH는 광학활성 에폭사이드 제조를 위한 생촉매로 응용될 수 있을 것이다.

프로바이오틱스 함유 한방소화제의 제조 및 특성 (Production and Characterization of Herbal Digestive Medicine Containing Probiotics)

  • To investigate the possible usage as probiotics, we isolated and identified one strain of bacillus from Korean traditional food, Jeotgal. The isolate was identified as Bacillus megaterium KS12 by examining its biochemical properties and 16S rRNA sequence analysis (99% similarity). B. megaterium KS12 exhibited high viability in artificial gastric juice for 3 h and in artificial bile salts for 24 h, about 14.4 and 14%, respectively. The herbal digestive medicine containing the isolate showed high starch hydrolytic activity and proteolytic activities (about 6-fold) compared to traditional herbal digestive medicine. The viable cells in the herbal digestive medicine containing the isolate were higher than those of traditional herbal digestive medicine.

Partial characterization of a 17 kDa protein of Clonorchis sinensis

  • A 17 kDa protein from Clonorchis sinensis adults was purified by a procedure including Sephacryl S-200 HR gel filtration and Q-Sepharose anion exchange chromatography. The protein was proved to be a cysteine protease as it showed hydrolytic activity toward Cbz-Phe-Arg-AMC in the presence of dithiothreitol and was inhibited by specific inhibitors such as iodoacetic acid or trans epoxy-succinly-L-leucyl-amido(4 guanidino) butane. The polyclonal antibody raised against the protein reacted to 17 kDa proteins of trematodes such as Paragonimuf westermani, Fasciola hepatica, Opisthorchis viverrini, Gymnophalloides seoi, and Metagonimus yokogawai. The antibody recognized the 17 kDa and 16 kDa cysteine proteases purified from C. sinensis, P. westemani, and G. seoi as well. These results suggest that the 17 kDa protein may be a cysteine protease commonly present in trematodes.

Alternaria in Food: Ecophysiology, Mycotoxin Production and Toxicology

  • Alternaria species are common saprophytes or pathogens of a wide range of plants pre- and post-harvest. This review considers the relative importance of Alternaria species, their ecology, competitiveness, production of mycotoxins and the prevalence of the predominant mycotoxins in different food products. The available toxicity data on these toxins and the potential future impacts of Alternaria species and their toxicity in food products pre- and post-harvest are discussed. The growth of Alternaria species is influenced by interacting abiotic factors, especially water activity ($a_w$), temperature and pH. The boundary conditions which allow growth and toxin production have been identified in relation to different matrices including cereal grain, sorghum, cottonseed, tomato, and soya beans. The competitiveness of Alternaria species is related to their water stress tolerance, hydrolytic enzyme production and ability to produce mycotoxins. The relationship between A. tenuissima and other phyllosphere fungi has been examined and the relative competitiveness determined using both an Index of Dominance ($I_D$) and the Niche Overlap Index (NOI) based on carbon-utilisation patterns. The toxicology of some of the Alternaria mycotoxins have been studied; however, some data are still lacking. The isolation of Alternaria toxins in different food products including processed products is reviewed. The future implications of Alternaria colonization/infection and the role of their mycotoxins in food production chains pre- and post-harvest are discussed.
