Evaluation of Intrinsic Bioremediation of Methyl Tert-butyl Ether (MTBE) Contaminated Groundwater

  • This paper reported the use of real-time polymerase chain reaction (PCR), denaturing gradient gel electrophoresis (DGGE), and the culture-based method in the intrinsic bioremediation study at a petroleum contaminated site. The study showed that phenol hydroxylase gene was detected in groundwater contaminated with benzene, toluene, ethylbenzene, xylene isomers (BTEX) and methyl tert-butyl ether (MTBE). This indicated that intrinsic bioremediation occurred at the site. DGGE analyses revealed that the petroleum-hydrocarbon plume caused the variation in microbial communities. MTBE degraders including Pseudomonas sp. NKNU01, Bacillus sp. NKNU01, Klebsiella sp. NKNU01, Enterobacter sp. NKNU01, and Enterobacter sp. NKNU02 were isolated from the contaminated groundwater using the cultured-based method. Among these five strains, Enterobacter sp. NKNU02 is the most effective stain at degrading MTBE without the addition of pentane. The MTBE biodegradation experiment indicated that the isolated bacteria were affected by propane. Biodegradation of MTBE was decreased but not totally inhibited in the mixtures of BTEX. Enterobacter sp. NKNU02 degraded about 60% of MTBE in the bioreactor study. Tert-butyl alcohol (TBA), acetic acid, 2-propanol, and propenoic acid were detected using gas chromatography/mass spectrometry during MTBE degraded by the rest cells of Enterobacter sp. NKNU02. The effectiveness of bioremediation of MTBE was assessed for potential field-scale application.

  • Bioremediation has been recognized as one of the best tools for hydrocarbon contaminated soil and nearby groundwater which had been heavily polluted in industrial areas. Degradation of PAHs in PAH-polluted loam soil were investigated under polyurethane foam environment with adsorbed bacteria Pseudomonas sp. (KCCM 40055) in order to acquire vital data for the environmentally-friendly process and material. macroporous commercial polyurethane foam that is widely used for microbial attachment in such as sewage treatment was selected for experiments. We also examined the microbial adherence upon the media. SR9-35C/G among the PU samples showed the highest degree of attachment and bioconversion. The conversion efficiency increased with moisture content of soil.

Characterization of biphenyl biodegradation, and regulation of iphenyl catabolism in alcaligenes xylosoxydans

  • Alcaligenes xylosoxydans strain SMN3 capable of utilizing biphenyl grew not only on phenol, and benzoate, but also on salicylate. Catabolisms of biphenyl and salicylate appear to be interrelated since benzoate is a common metabolic intermediate of these compounds. Enzyme levels in the excatechol 2. 3-dioxygenas which is meta-cleavage enzyme of catechol, but did not induce catechol 1, 2-dioxygenase. All the oxidative enzymes of biphenyl and 2, 3,-dihydroxybiphenyl (23DHBP) were induced when the cells were grown on biphenyl and salicylate, respectively. Biphenyl and salicylate could be a good inducer in the oxidation of biphenyl and 2, 3-dihydroxybiphenyl. The two enzymes for the degradation of biphenyl and salicylate were induced after growth on either biphenyl or salicylate, suggesting the presence of a common regulatory element. However, benzoate could not induce the enzymes responsible for the oxidation of these compounds. Biphenyl and salicylate were good inducers for indigo formation due to the activity of biphenyl dioxygenase. These results suggested that indole oxidation is a property of bacterial dioxygenase that form cis-dihydrodiols from aromatic hydrocarbon including biphenyl.

Emulsification Activity of Acinetobacter sp. 2-3A Isolated from Petroleum Oil-Contaminated Soil (유류오염 토양에서 분리한 Acinetobacter sp. 2-3A의 유화활성)

  • Fifty hydrocarbon-metabolizing microorganisms were isolated from soil samples polluted by the petroleum oils in Gamman-dong, Busan. Among them, strain 2-3A, showing strong emulsification activity, was selected by oil film-collapsing method. This bacterium was identified as Acinetobacter sp. and designated as Acinetobacter sp. 2-3A. The optimum temperature and pH on the growth of Acinetobacter sp. 2-3A were $25^{\circ}C$ and pH 7.0, respectively. The carbon and nitrogen sources for the most effective emulsification activity were 3.0% olive oil and 0.5% peptone, respectively. The 0.15% potassium phosphate was the most effective emulsification activity as a phosphate source. The optimum emulsification activity condition was $20^{\circ}C$, pH 7.0, and 2.0% NaCl. The optimum time for the best production of biosurfactant was 27 hrs. The emulsification stability was maintained at the temperature range from $4^{\circ}C$ to $100^{\circ}C$, pH range from 6.0 to 10.0, and NaCl range from 0% to 10%. For the oil resolvability of the biosurfactant, the residual oils were investigated by gas chromatography. As a result, it was verified that the biosurfactant decreased and decomposed crude oils from $_nC_{10}$ to $_nC_{32}$.

  • Sr0.92Y0.08TiO3-δ (SYT) was investigated as an alternative anode in humidified CH4 fuel for SOFCs at low temperatures (650 ℃-750 ℃) and compared with the conventional Ni/yttria-stabilized zirconia (Ni/YSZ) anode. The goal of the study was to directly use a hydrocarbon fuel in a SOFC without a reforming process. The cell performance of the SYT anode was relatively low compared with that of the Ni/YSZ anode because of the poor electrochemical catalytic activity of SYT. In the presence of CH4 fuel, however, the cell performance with the SYT anode decreased by 20%, in contrast to the 58% decrease in the case of the Ni/YSZ anode. The severe degradation of cell performance observed with the Ni/YSZ anode was caused by carbon deposition that resulted from methane thermal cracking. Carbon was much less detected in the SYT anode due to the catalytic oxidation. Otherwise, a significant amount of bulk carbon was detected in the Ni/YSZ anode.

Emulsification of Crude Oil by Acinetobacter sp. SH-14

  • As basic study to evaluate the treatability of oil-contaminated environment with bacteria, isolation and characterization of crude oil-degrading bacterium were carried out. A bacterial strain SH-14 capable of degrading crude oil was isolated from contaminated soils by enrichment culture technique and identified as Acinetobacter sp. by morphological, cultural and biochemical characteristics, and so named Acinetobacter sp. SH-14. The optimal medium composition and cultural conditions for the growth and emulsification of crude oil by Acinetobacter sp. SH-14 used were crude oil of 2.0%, $KNO_3$ of 0.2%, $K_2HPO_4$ of 0.05%, and $MgSO_4\;{\cdot}\;7H_2O$ of 1.0%, along with initial pH 7.0 at $30^{\circ}C$. Acinetobacter sp. SH-14 showed to be resistant to chloramphenicol and utilized various hydrocarbons such as dodecane, hexadecane, isooctane, cyclo-hexane etc., as a sole carbon source. Acinetobacter sp. SH-14 harbored a single plasmid. By agarose gel electrophoresis and curing experiment it was found that the genes for crude oil components degradation were encoded on the plasmid.

Photocatalytic degradation of a polycyclic aromatic sulfur hydrocarbon ($TiO_2$를 이용한 다환 방향족 유기황 탄화수소의 광분해효율 연구)

  • [ $TiO_2$ ] 광촉매에 의한 분해 반응의 활성을 높이기 위한 다양한 연구가 진행되었다. 광촉매 반응은 1차 반응을 따랐으며 초기농도가 높을수록 분해효율이 감소하는 경향을 보였다. 본 연구에서는 산화제로 과산화수소가 주입되었을 경우 분해효율을 조사하였으며, 과산화수소를 주입하였을 경우가 그렇지 않은 경우보다 더 높은 분해효율을 보였다. 또한 과산화수소 주입량을 달리했을 때, 주입량이 증가할수록 효율이 높아지다가 일정량 이상에서는 오히려 효율이 감소하는 것으로 나타났다. 따라서 과산화수소 최적첨가량이 존재함을 알 수 있었다. 한편 $TiO_2$에 전이금속을 첨가하여 전이금속이 $TiO_2$ 촉매의 분해효율에 미치는 영향을 알아보았다. Pt(0.5%)-$TiO_2$가 가장 높은 분해효을을 보였으며, Pt첨가함량이 더 큰 Pt(2%)-$TiO_2$는 함량이 증가했음에도 불구하고 큰 차이는 아니지만 오히려 효율이 감소하였다. 따라서 촉매표면에서 전자와 정공이 생성되었을 때, Pt가 전자를 포획함으로써 전자와 정공의 재결합율을 감소시켜 OH라디칼을 생성할 수 있는 정공이 많아져 반응효율을 증가되는 것을 알 수 있었고, 금속에 따른 최적 첨가함량이 존재함을 알 수 있다. 반면에 Pd를 첨가했을 경우는 첨가 함량에 관계없이 모두 분해효율이 오히려 감소하는 경향을 나타냈으며 이는 전이금속 고유의 성질이나, 또는 대상물질에 따라 각기 다른 경향이 존재함을 나타내며 추가적인 연구가 필요하다고 사료된다.

Degradation characteristics of acenaphthene or acenaphthylene by Fenton oxidation (펜톤산화에 의한 acenaphthene 또는 acenaphthylene 분해특성)

  • The acenaphthene(ACE) or acenaphthylene(ACEL) is one of the most frequently found compound in polycyclic aromatic hydrocarbon (PAH)-contaminated soil. In this study, we make 10mg/L ACE or ACEL in ethanol which is the model washing solvent for contaminated soil. This was followed by Fenton treatment in which 0.2 or 0.3mL of 30% $H_2O_2$ and 0.2 ml of 0.5 M $Fe^{2+}$ were added. The results showed more than 88 or 99% of ACE or ACEL removal efficiency, respectively. Additionally, we employed GC-MS to identify the main oxidation product generated by the optimized Fenton oxidation [i.e., ACE or ACEL degraded in to 21, 34 % 1,8-naphthalic anhydride(NAPAN), repectively]. It is expected that biodegradability of NAPAN is enhanced because NAPAN has three oxygens compared with ACE and ACEL. Therefore the results suggest that the hybrid treatment system (i.e., ethanol washing -Fenton oxidation treatment) can be effectively applied to remove ACE or ACEL from soil..

Hydrocarbon-Organic Composite Membranes for Improved Oxidative Stability for PEMFC Applications (연료전지용 탄화수소 전해질 막의 산화안정성 향상을 위한 유기물 복합막의 제조 및 특성 분석)

  • In order to mitigate oxidative degradation of polymer membrane during fuel cell operation, an organic radical quencher was introduced. Rutin was selected as a radical quencher and mixed with sulfonated poly(arylene ether sulfone) to prepare composite membrane. Physicochemical properties of the composite membranes such as water uptake and proton conductivity were characterized. Hydrogen peroxide exposure experiment, which can mimic accelerated oxidative stability test during fuel cell operation, was adopted to evaluate the oxidative stability of the membranes. The composite membranes containing Rutin showed similar proton conductivity and enhanced oxidative stability compared to pristine ones.

A DFT Study for the Reaction Pathway(s) of Polycyclic Aromatic Hydrocarbons I: Phenanthrene Degradation with two OH Radicals (다고리 방향족 탄화수소의 반응 경로에 대한 DFT 연구 I: 2개의 OH 라디칼에 의한 페난트렌의 분해 반응)

  • In this study, the DFT calculation was performed using the B3LYP/6-31G(d,p) basis sets for the reaction process in which phenanthrene decomposes due to the chain reaction of two OH radicals on phenanthrene in the gaseous state of 298 K at 1 atm. As a result of the calculation, even when two OH radicals act on phenanthrene in a chain, the reaction for producing phenanthren-9-ol is predicted to be more advantageous than the reaction for producing phenanthren-1-ol. On the other hand, it was predicted that the OH addition process at room temperature would be advantageous for the priority of the OH addition and H abstraction process.
