식품유래 곰팡이 대사산물의 항암효과 (Inhibitory Effects of Fungal Metabolites Isolated from Foodstuffs on the Growth of Human Cancer Cell Lines)

  • 본 논문은 새로운 항암소재를 검색하기 위하여 식품으로부터 곰팡이를 분리하고 그 대사산물을 이용하여 수종의 암세포주의 생육을 대한 억제 물질을 탐색하고자 하며, 이를 바탕으로 새로운 항생물질 연구의 기초자료를 확보하고 결과물의 응용도를 모색하고자 실험을 수행하였다. 식품으로부터 총 72종의 곰팡이를 분리 하였고, 전면 및 후면의 색, 모양. 성장속도에 따라 4가지 group로 나누어 실험에 사용하였다. 그리고 그 대사산물(배양무, 균체)을 ethly acetate로 추출하여, 총 6종의 암세포주에 대하여 MTT assay을 실험하였다. 그 ruf과 A, B, C, D group 중 D group에서 가장 높은 증식 억제를 확인 하였으며, D4의 증식억제 효과가 가장 높음을 확인하였다. 위암 세포주인 KATO III, AGS, SUN-668에 대한 증식억제 효과는 30-98%로 나타났으며, 간암 세포주인 Hepa1c1c7, HepG2에는 10-98%, 유방암 세포주인 MDA-MB-231에 대해서는 25-90%의 증식억제 효과를 확인하였다. 배양물과 균체의 항암성은 개체에 따라 약간의 차이는 있으나, 배양물이 대부분 우수한 것으로 확인되었다.

AMP-activated protein kinase determines apoptotic sensitivity of cancer cells to ginsenoside-Rh2

  • Ginseng saponins exert various important pharmacological effects with regard to the control of many diseases, including cancer. In this study, the anticancer effect of ginsenosides on human cancer cells was investigated and compared. Among the tested compounds, ginsenoside-Rh2 displays the highest inhibitory effect on cell viability in HepG2 cells. Ginsenoside-Rh2, a ginseng saponin isolated from the root of Panax ginseng, has been suggested to have potential as an anticancer agent, but the underlying mechanisms remain elusive. In the present study, we have shown that cancer cells have differential sensitivity to ginsenoside-Rh2-induced apoptosis, raising questions regarding the specific mechanisms responsible for the discrepant sensitivity to ginsenoside-Rh2. In this study, we demonstrate that AMP-activated protein kinase (AMPK) is a survival factor under ginsenoside-Rh2 treatment in cancer cells. Cancer cells with acute responsiveness of AMPK display a relative resistance to ginsenoside-Rh2, but cotreatment with AMPK inhibitor resulted in a marked increase of ginsenoside-Rh2-induced apoptosis. We also observed that p38 MAPK (mitogen-activated protein kinase) acts as another survival factor under ginsenoside-Rh2 treatment, but there was no signaling crosstalk between AMPK and p38 MAPK, suggesting that combination with inhibitor of AMPK or p38 MAPK can augment the anticancer potential of ginsenoside Rh2.

Trichoplusia ni 세포에 내재하는 당 수송체에 D-fructose가 미치는 효과와 Trichoplusia ni 세포에 발현된 사람 HepG2형 포도당 수송 단백질의 photolabelling (Effect of D-Fructose on Sugar Transport Systems in Trichoplusia ni Cells and Photolabeling of the Trichoplusia ni Cell-Expressed Human HepG2 Type Glucose Transport Protein)

  • Trichoplusia ni (T. ni) 세포는 사람 당 수송체를 이성질적으로 많은 양 생산하려 할 때 유용하게 사용되는, baculovirus 발현 시스템의 숙주세포로서 이용된다. 그러나 T. ni 세포에 존재하는 내재된 당 수송체의 높은 활동은, 발현된 외재적 당 수송체의 수송활성과 같은 직접적 증거 제시에 장애가 된다. 뿐만 아니라 곤충세포에 내재하는 당 수송체계의 특성에 대해서는 밝혀진 바가 거의 없다. 그래서 본 연구에서는 baculovirus 발현 시스템을 보다 잘 활용하기 위해 T. ni 세포의 2dGlc기질 수송에 D-fructose가 미치는 영향을 살펴 보았으며, T. ni 세포에 발현된 사람 당 수송체의 생물학적 활성을 보다 용이하게 검증하기 위해 발현된 수송체를 [$^3H$] cytochalasin B를 이용하여 photolabelling 하였다. 우선 감염되지 않은 세포와 recombinant AcMPV-GTL 감염시킨 T. ni 세포의 2dGlc uptake를 300 mM D-fructose가 있을 때와 없을 때, 그리고 $20{\mu}M$ cytochalasin B가 있을 때와 없을 때의 상황에서 살펴보았다. 감염되지 않은 세포에서의 육탄당 uptake는 D-fructose에 의해 강력하게 억제 되었으나 cytochalasin B에 의해서는 단지 미미한 억제 효과만을 보여주었다. 흥미롭게도 AcMPV-GTL 바이러스 감염된 T. ni 세포에서는 비록 2dGlc uptake율은 감염되지 않은 세포와 비교해 다소 낮았지만 육탄당 수송 억제 반응은 근본적으로 동일함을 보여 주었다. 또한 [$^3H$] cytochalasin B를 이용한 발현단백질 photolabelling에서는, L-glucose가 존재하는 상황 하에만 하나의 날카롭게 표지된 peak가, 바이러스 감염된 세포에서 관찰되었다. 감염되지 않은 세포에서는 이러한 peak는 관찰되지 않았다. 게다가 D-glucose 존재 하에서는 발현된 단백질의 photolabelling이 완전히 억제되어짐을 보여주어, labelling의 입체선택성(stereoselectivity)을 입증하였다.

홍삼약침액(紅蔘藥鍼液)의 위암세포주(胃癌細胞柱) 유전자(遺傳子) 발현(發顯)에 미치는 영향(影響) (Effect of Ginseng Radix Rubra Herbal-acupuncture Solution(GRR-HAS) on Gene Expression in SNU484 carcinomar cells)

  • Objective : It has long been known about the anticancer effect of GRR-HAS, however, it has not been systemically determined the differentially regulated genes by GRR-HAS in cancer cells. The purpose of this study is to screen the GRR-HAS mediated differentially expressed genes in cancer cells such as SNU484 gastric cancer cell lines. Oligonucleotide microarray approache was employed to screen the differential expression genes. Methods : GRR-HAS was prepared by boiling and stored at $-70^{\circ}C$ until use. Cells were treated with various concentrations of GRR-HAS(0.1, 0.5, 1.5, 10, 20mg/ml) for 24 h. Cell toxicity was tested by MTT assay. To screen the differentially expressed genes in cancer cells, cells were treated with 1.5mg/ml of GRR-HAS. For oligonucleotide microarray assay, total RNA was used for gene expression analysis using oligonucleotide Genechip (Human genome Ul33 Plus 2.0., Affimatrix Co.). Results : It has no cytotoxic effects on both HepG2 and SNU484 cells in all concentrations(0.1, 0.5, 1.5, 10, 20mg/ml). In oligonucleotide microarray assay, in SNU484 cells, the number of more than twofold up-regulated genes was 346. The number of more than twofold down-regulated genes was 9. Discussion : This study showed the comprehensive gene expression analysis using oligonucleotide microarray for the screening of GRR-HAS mediated differentially regulated genes. These results will provide a better application of GRR-HAS in cancer field and drug target development.

Anti-angiogenic activity of wilfoside glycosides isolated from Cynanchum wilfordii

  • Wm was isolated from Cynanchum wilfordii (Asclepiadaceae) as a mixture form of polypregnane glycosides that included wilfoside K1N and wilfoside C1N. In the present study, we investigated the anti-angiogenic effect of wilfoside glycosides using in vivo and in vitro assay systems. We first demonstrated that concentrated conditioned media obtained from Wm-treated HepG2 human hepatoblastoma cells blocked the angiogenic activation of Wm-untreated concentrated conditioned media, suggesting that Wm may have an inhibitory effect on tumor-induced angiogenesis. Antioxidant activities of licorice-derived prenylflavonoids

  • Glycyrrhiza uralensis (or licorice) is a widely used Oriental herbal medicine from which the phenylflavonoids dehydroglyasperin C (DGC), dehydroglyasperin D (DGD), and isoangustone A (IsoA) are derived. The purpose of the present study was to evaluate the antioxidant properties of DGC, DGD, and IsoA. The three compounds showed strong ferric reducing activities and effectively scavenged DPPH, $ABTS^+$, and singlet oxygen radicals. Among the three compounds tested, DGC showed the highest free radical scavenging capacity in human hepatoma HepG2 cells as assessed by oxidant-sensitive fluorescent dyes dichlorofluorescein diacetate and dihydroethidium bromide. In addition, all three compounds effectively suppressed lipid peroxidation in rat tissues as well as $H_2O_2$-induced ROS production in hepatoma cells. This study demonstrates that among the three phenylflavonoids isolated from licorice, DGC possesses the most potent antioxidant activity, suggesting it has protective effects against chronic diseases caused by reactive oxygen species as well as potential as an antioxidant food additive.

Nrf2 활성화(活性化)를 통한 작약(芍藥)의 간보호효과(肝保護效果) (Hepatoprotective effect of Paeoniae radix via Nrf2 activation)

  • Objectives : Liver is one of the largest organs in the human, and has a function of detoxification and energy sensing to prevent severe disease. Paeoniae radix has been used to treat a variety of liver diseases such as hepatitis and chronic hepatic failure. Although P. radix has been used as an medicinal herb for a long time, the effects of P. radix on severe oxidative stress and its action mechanism on the liver was not clearly verified.Methods : This study investigated the protective effects of P. radix extract (PRE), and the underlying mechanism of its action in the liver. tert-butyl hydroperoxide (t-BHP) and carbon tetrachlroride (CCl4) were used to induce oxidative stress in the HepG2 hepatocyte cell line and Sprague-Dawley rats, respectively.Results : t-BHP significantly induced cell death and ROS production in HepG2 cell, as indicated by MTT and FACS analysis. However, pretreatment of PRE inhibited a decrease in cell viability and H2O2 production in the HepG2 cells. PRE also blocked the ability of t-BHP to damage in mitochondrial membrane transition. More importantly, PRE induced Nrf2 activation and antioxidant Phase II enzyme, which may have a role in the effects of PRE. In mice, PRE inhibited the liver damage induced by CCl4.Conclusions : PRE inhibited oxidative stress and hepatic damages as mediated with Nrf2 activation. This study unveil, in part, the effect and mechanism of old medicinal herb, P. radix.

오디 추출물(Morus alba L.)의 혈행개선 효과 (Effect of Mulberry (Morus alba L.) Extract on Blood Flow Improvement)

  • 본 연구에서는 다양한 생리활성물질을 다량 함유하고 있는 국내산(전라북도 부안군) 오디의 혈액순환장애의 개선 효과를 확인하기 위하여 in vitro 및 고지혈증 동물모델을 이용한 실험을 진행하였다. MBE가 DPPH 라디칼을 50% 소거하는 $SC_{50}$값은 $89.36{\pm}5.46{\mu}g/mL$로 정제되지 않은 복합물질임을 감안하면 다른 식물추출물에 비해 항산화능이 우수함을 알 수 있었다. RAW 264.7 세포 및 HepG2 세포에 대한 MBE의 세포독성은 2,500 ${\mu}g/mL$ 이하의 농도를 처리한 경우 세포독성을 나타내지 않았다. In vitro 실험에서 COX-2의 활성저해 효과를 측정한 결과 MBE의 $IC_{50}$값은 $215.94{\pm}18.15$이었고, human LDL의 산화억제 효과는 최종반응 농도가 200 및 400 ${\mu}g/mL$일 경우 유의적인 감소(P<0.05) 효과가 있었다. 또한 TNF-${\alpha}$에 의하여 활성화된 HUVECs에서 MBE가 증가된 ICAM-1 및 VCAM-1의 발현을 감소(P<0.05)시켰으며 LPS에 의하여 활성화된 RAW 264.7 세포에서 MBE가 증가된 COX-2 및 5-LO의 발현을 감소(P<0.05)시킴을 알 수 있었다. 고지혈증 동물모델을 이용한 실험에서 MBE의 일부 실험군(200 mg 및 400 mg/kg 체중/day, 14 days)에서 혈청 total cholesterol의 수치는 음성대조군에 비해 유의적으로 감소(P<0.05)시켰으나 혈청 C-reactive protein의 수치는 모든 실험군에서 음성대조군 대비 유의적으로 감소하지 않았다. 이러한 MBE의 항산화능, COX-2 활성억제 및 COX-2와 5-LO의 발현억제를 통한 염증억제, 혈중 total cholesterol 감소를 통한 이상지질혈증의 개선 효과 및 ICAM-1과 VCAM-1의 발현억제를 통한 혈관협착 방지의 효과들을 종합해 보면 오디추출물인 MBE가 혈액순환장애를 유발하거나 확대시키는 다양한 핵심요소들을 다각도로 개선시킴으로써 혈액순환장애의 예방과 치료에 긍정적인 효과가 있을 것으로 사료된다.

Silencing of NUF2 Inhibits Tumor Growth and Induces Apoptosis in Human Hepatocellular Carcinomas

  • Background: As an important component of the NDC80 kinetochore complex, NUF2 is essential for kinetochore-microtubule attachment and chromosome segregation. Previous studies also suggested its involvement in development of various kinds of human cancers, however, its expression and functions in human hepatocellular carcinoma (HCC) are still unclear. Materials and Methods: In the present study, we aimed to test the hypothesis that NUF2 is aberrant in human HCCs and associated with cell growth. Results: Our results showed significantly elevated expression of NUF2 in human HCC tissues compared to adjacent normal tissues, and high expression of NUF2 in HCC cell lines. Using lentivirus-mediated silencing of NUF2 in HepG2 human HCC cells, we found that NUF2 depletion markedly suppressed proliferation and colony formation capacity in vitro, and dramatically hampered tumor growth of xenografts in vivo. Moreover, NUF2 silencing could induce cell cycle arrest and trigger cell apoptosis. Additionally, altered levels of cell cycle and apoptosis related proteins including cyclin B1, Cdc25A, Cdc2, Bad and Bax were also observed. Conclusions: In conclusion, these results demonstrate that NUF2 plays a critical role in the regulation of HCC cell proliferation and apoptosis, indicating that NUF2 may serve as a potential molecular target for therapeutic approaches.

In Vitro Scolicidal Effects of Salvadora persica Root Extract against Protoscolices of Echinococcus granulosus

  • It has been known that Arak, Salvadora persica, has a number of medicinal properties. We tried to investigate in vitro scolicidal effect of root extracts of this plant against protoscolices from hydatid cysts of Echinococcus granulosus. Protoscolices were aseptically collected from sheep livers containing hydatid cysts. S. persica root extract was used in 10, 30, and 50 mg/ml concentration for 10, 20, and 30 min. The viability of protoscolices was ascertained by 0.1% eosin staining. Scolicidal activity of S. persica extract at a concentration of 10 mg/ml was 36.3%, 50.3%, and 70.8% after 10, 20, and 30 min of exposure, respectively. The scolicidal effect of this extract at a concentration of 30 mg/ml was 52.9%, 86.7%, and 100% after 10, 20, and 30 min of exposure, respectively. S. persica extract at a concentration of 50 mg/ml, meanwhile, killed 81.4%, 100%, and 100% of protoscolices after 10, 20, and 30 min, respectively. Also, the cytotoxic potential of S. persica was assessed on human liver cells (HepG2) using trypan blue exclusion test. No cytotoxic effect was observed on HepG2 cell line. The present study confirmed for the first time that the ethanolic extract of S. persica has high scolicidal power in vitro. However, in vivo effect of this material remains to be studied for treatment of echinococcosis in humans and herbivorous animals.
