Studies on the Host Range of Rice Stripe Virus (벼 줄무의잎마름병 바이러스의 기주범위에 관한 연구)

  • An investigation was carried out to know the host range of the rice stripe virus as a basis of the disease control. 1. In order to investigate the host range of the rice stripe virus 30 species including gramineaceae and forage crop and weeds were inoculated with viruliferous Laodelphax sriatellus Fallen. As a result, 21 out of 30 species showed typical symptoms. 2. Cyperus amuricus Maximowicz var. laxus., Pycreus sanguinolentus Nees. and Eriocaulon robustius Makino, not belonging to gramineaceae were known to be new host plants of rice stripe virus. The highest infection occurred on Pycreus sanguinolentus grown widly in the wet paddy fold. 3. The number of vectors inoculated on the plants was decreased after 48 hours than that of 24 hours.

  • 벼, 바랭이, 톨페이스큐로부터 분리한 벼 도열병균에 대한 기주범위를 벼, 밀, 보리, 옥수수, 호밀, 귀리, 톨페스큐 등의 작물과 피, 왕바랭이, 바랭이, 강아지풀, 개기장 등의 잡초를 대상으로 실험하였다. 대부분의 균주들은 공시작물 모두와 잡초인 강아지풀에대하여 병원성을 나타내었다. 이러한 결과는 논 주면의 강아지풀이 벼 도열병균의 기주로 작용할 수 있음을 나타내었다.

Developing screening system for resistance to anthracnose in grapes by using culture filtrates from Elsinoe ampelina

  • It was investigated whether culture filtrates produced by X. fastiduosa could be used to determine varietal susceptibility in grape cultivars to anthracnose as a substitute for pathogen inoculation or field screening. Bioassay of grape leaves with culture filtrates showed that their phytotoxicities were active and host-selective. Ethyl acetate extracts from those also showed the toxicities and host selectivity among grape cultivars. The sensitive range of plants to culture filtrates and their ethyl acetate extracts was consistent with the host range to the pathogen. Susceptible cultivars were sensitive to even highly diluted culture filtrates but resistant cultivars were not affected even at original culture filtrates. Susceptible cultivars were sensitive to the undiluted culture filtrates than highly diluted culture filtrates and the younger leaves were the more sensitive to the culture filtrates and their ethyl acetate extracts in grapes.

GPU Acceleration of Range Doppler Algorithm for Real-Time SAR Image Generation (실시간 SAR 영상 생성을 위한 Range Doppler Algorithm의 GPU 가속)

  • In this paper, a GPU-accelerated kernel of range Doppler algorithm (RDA) was developed for real-time image formation based on frequency modulated continuous wave (FMCW) synthetic aperture radar (SAR). A pinned memory was used to minimize the data transfer time between the host and the GPU device, and the kernel was configured to perform all RDA operations on the GPU to minimize the number of data transfers. The dataset was obtained through the FMCW drone SAR experiment, and the GPU acceleration effect was measured in an intel i7-9700K CPU, 32GB RAM, and Nvidia RTX 3090 GPU environment. Including the data transfer time between host and devices, it was measured to be accelerated up to 3.41 times compared to the CPU, and when only the acceleration effect of operation was measured without including the data transfer time, it was confirmed that it could be accelerated up to 156 times.

  • Inorganic-organic composite was made through sol-gel process. Possible range for hosting organic dye was investigated with various composition. The transmittance of the composite was the most important condition for a host was increased to 90% with decreasing HCl as a catalyst. Clear absorbance peak was obtained by decreasing concentration of organic dye, fluorescent intensity was increased and transmittance was decreased with rising gelling temperature.

A Series of IncQ-Based Reporter Plasmids for Use in a Range of Gram-Negative Genera

  • Many studies require expression analysis of the same gene/promoter across a range of bacterial genera. However, there is currently a lack of availability of reporters based on the broad-host-range IncQ replicon, which is compatible with a popular improved IncP transfer system that is self-transfer defective. We report IncQ lacZ reporter plasmids with features including (1) compatibility with IncP, IncW, and pBHR/pBBR replicons, (2) a variety of antibiotic markers (Sp-r, Sm-r, Km-r, Cm-r), (3) convenient mobilization via a novel self-transfer-defective IncP conjugation system, and (4) GenBank DNA sequences. Utility is demonstrated using three different promoters in different Gram-negative genera.

Studies on the RK-temperate phage of bacillus cereus (Bacollis cereis의 RK-용원파아지에 관한 연구)

  • The RK-temperate phage which infected with Bacillus cereus was isolated and the characters were investigated. The induction of RK-temperate phage from host bacterium attained by ultraviolet light irradiation (15W, 30cm, 30-120sec) and mitomycin C treatment (0.2-2 ug/ml). The host range of RK-temperate phage was not revealed with lysogenic and related strains of B. cereus. But B. cereus(PS) 352 which obtained by N-nitrosoguanidine treatment (1,000{$\mu}g/ml)$ to phage infected with host bacteria was sensitive bacteria of RK-temperate phage. RK-temperate phage was stabilized at the condition of nutrient broth (pH 7-8), Tris-buffer (pH 7-8) and ammonium buffer (pH 8-9) and Sorensen's phosphate buffer (pH 6-7), but unstabilized at other salt solutions and pH range. Also, thermostability was to $45^{\circ}C$ but unstabilized at above $50^{\circ}C$. At RK-temperate phage, the measurment values of head, neck, mid tail and end tail were 59nm, $9{\times}16nm,\;10{\times}189nm,\;and\;10{\times}14nm$ respectively. The morphology of head was regular polyhedron, and the end tail was coneate form. On the one hand, the number of capsid protein layer of tail were consist of 4, 35, and 1 at neck, mid tail, and end tail, respectively. RK-temperate phage was identified with DNA phage and G+C contents were 38.63. The latent time of RK-temperate phage was 30 minutes and the burst size was 70-80. And the host bacteria was lysed in case of multi-infection, above moi 1.

Virulence Factors and Stability of Coliphages Specific to Escherichia coli O157:H7 and to Various E. coli Infection

  • Characteristics of E. coli O157:H7-specific infection bacteriophages (O157 coliphages) and broad-host-range bacteriophages for other E. coli serotypes (broad-host coliphages) were compared. The burst sizes of the two groups ranged from 40 to 176 PFU/infected cell. Distributions of the virulence factors stx1, stx2, ehxA, and saa between the two groups were not differentiated. Broad-host-range coliphages showed lower stability at $70^{\circ}C$, in relation to O157 coliphages. However, O157 coliphages showed high acid and ethanol tolerance by reduction of only 22% and 11% phages, respectively, under pH 3 and 70% ethanol for 1 h exposure. Therefore, these results revealed that the O157 coliphages might be more stable under harsh environments, which might explain their effective infection of the acid-tolerant E. coli O157:H7.

Petunia Asteroid Mosaic Virus Isolated from Petunia hybrida Vilm. (폐츄니아에서 분리한 Petunia Asteroid Mosaic Virus)

  • A virus was isolated from petunia (Petunia hybrida Vilm.) plants showing chlorotic ring spots on the leaves and color breaking on the flowers, and was identified as petunia asteroid mosaic virus (PAMV). Identification of the PAMV was established by host range test, electron microscopy, serological reaction, and physical properties of the virus. In the host range test, Nicotiana glutinosa, N. rustica, N. clevelandii, P. hybrida, Gomphrena globosa, and Chenopodium amaranticolor were systemically infected with the virus. The virus produced local lesions on inoculated leaves of N. tabacum‘Samsun’, N. tabacum‘Xanthi nc’, Datura stramonium, Vigna unguiculata‘White eye’, C. quinoa, Capsicum annuum, Vicia faba, and Lycopersicon esculentum‘Rutgers’. However, Cucurbita sativus and C. moschata did not show any symptoms. PAMV particles were isometric with 30 nm in diameter. The crude sap from G. globosa infected with the virus reacted positively with antiserum to tomato bushy stunt virus (TBSV) in agar gel double diffusion test. Thermal inactivation point of the virus was 8$0^{\circ}C$ and the virus retained its infectivity at the dilution of 10-4. Longevity in vitro of the virus was estimated longer than 35 days.
