T helper (Th) 17 cells have recently been described as a third subset of T helper cells, and have provided new insights into the mechanisms that are important in the development of autoimmune diseases and the immune responses that are essential for effective antimicrobial host defense. Both protective and harmful effects of Th17 responses during infection have been described. In general, Th17 responses are critical for mucosal and epithelial host defense against extracellular bacteria and fungi. However, recent studies have reported that Th17 responses can also contribute to viral persistence and chronic inflammation associated with parasitic infection. It has become evident that the type of microorganisms and the setting in which they trigger the Th17 response determines the outcome of the delicate balancethat exists between Th17 induced protection and immunopathogenesis.
The gastrointestinal (GI) tract, including the rumen and the other intestinal segments of cattle, harbors a diverse, complex, and dynamic microbiome that drives feed digestion and fermentation in cattle, determining feed efficiency and output of pollutants. This microbiome also plays an important role in affecting host health. Research has been conducted for more than a century to understand the microbiome and its relationship to feed efficiency and host health. The traditional cultivation-based research elucidated some of the major metabolism, but studies using molecular biology techniques conducted from late 1980's to the late early 2000's greatly expanded our view of the diversity of the rumen and intestinal microbiome of cattle. Recently, metagenomics has been the primary technology to characterize the GI microbiome and its relationship with host nutrition and health. This review addresses the main methods/techniques in current use, the knowledge gained, and some of the challenges that remain. Most of the primers used in quantitative real-time polymerase chain reaction quantification and diversity analysis using metagenomics of ruminal bacteria, archaea, fungi, and protozoa were also compiled.
Phlebopus spongiosus is a well-known edible ectomycorrhizal mushroom indigenous to southern Vietnam. The mushroom specimens collected from northern Thailand in this study were identified as P. spongiosus. This identification was based on morphological characteristics and the multi-gene phylogenetic analyses. Pure cultures were isolated and the relevant suitable mycelial growth conditions were investigated. The results indicated that the fungal mycelia grew well on L-modified Melin-Norkans, and Murashige and Skoog agar all of which were adjusted to a pH of 5.0 at 30 ℃. Sclerotia-like structures were observed on cultures. The ability of this mushroom to produce fruiting bodies in the absence of a host plant was determined by employing a bag cultivation method. Fungal mycelia completely covered the cultivation substrate after 90-95 days following inoculation of mushroom spawn. Under the mushroom house conditions, the highest amount of primordial formation was observed after 10-15 days at a casing with soil:vermiculite (1:1, v/v). The primordia developed into a mature stage within one week. Moreover, identification of the cultivated fruiting bodies was confirmed by both morphological and molecular methods. This is the first record of P. spongiosus found in Thailand and its ability to form fruiting bodies without a host plant.
The contribution of plant nutrition status in arbuscular mycorrhizal (AM) plant to the nutrient acquisition by extraradical hyphae of AM fungi was investigated using cucumber colonized with Glomus intraradicies (BEG 110) focusing on the Zn. Compartmentalized pots with separated Bones for hyphal growth were used to determine the contribution of extraradical AM hyphae to Zn uptake from hyphal zones. $0.5\;{\mu}M$ Zn was supplied into the hyphal zones as nutrient solution (10 mL/day) with a form of $ZnSO_4$. Zn foliar application was made two times for one week before harvest (8 mL/plant). The colonization rate by AM were high in all of Zn treatments. The dry weight of cucumber increased by AM colonization compared to those of non-mycorrhizal counterpart. However: Zn foliar application resulted in no significant difference in dry weight between mycorrhizal- and non-mycorrhizal plant. In addition, the enhancement of Zn content in cucumber shoot by AM colonization were also reduced by Zn foliar application. These results indicate that the interaction between host plant and AM fungus for nutrient uptake might be related to plant nutritional status and nutrient contents. In consequence, higher Zn contents in host plant by foliar application of Zn could restrict the role of extraradical hyphae of AM fungus on the Zn acquisition and transfer from fungus to host plant.
This study was carried out to investigate the diversity and frequency of ectomycorrhizal fungi by cutting levels in Quercus acuta and Pinus densiflora stands from June to October, 2001. The obtained results from investigation were as follows, 1. The total of fungi found during the surveyed periods were 50 species, 30 genus, 15 families. The fungi belonged to Agaricales(37 species, 18 genus, 7 families), Aphllophorales(7 species, 6 genus, 3 families), Auriculariales(1 species, 1 genus, 1 families) and Gasteromycetes(5 species, 5 genus, 4 families), Basidiomycota. 2. The total number of putative ectomycorrhizal fruiting bodies were 137 individuals, 22 species, 10 genus, 5 families, 79 individuals, 10 species, 6 genus, 3 families at strip-cutting area of Pinus densiflora stand, 24 individuals, 9 species, 5 genus, 4 families at strip-cutting area of Quercus acuta stand and 25 individuals, 10 species, 4 genus, 3 families at spacing cutting area of Quercus acuta stand. 3. Tylropilus neofelleus had high host specificity associated with Chamaecyparis obtusa, while Amanita pseudoporphyria had relatively wide host ranges compared to other ectomycorrhizal fungi. 4. The results analyzed by Correlation coefficients showed that relative humidity and precipitation were major enviromental factors to affect the distribution of ectomycorrhizal fruiting bodies.
Endophytic fungi are present in host plants and contribute to resistance to biotic and abiotic stress. Aquatic plants are living in aquatic environment such as saltwater or freshwater and exposed more water stress than other land plants. In this study, we investigated 4 wetlands in Jeju and collected 11 aquatic plants. Exogenous microbes were removed by preprocessing of plants and endophytic fungal strains were isolated from the plants. We isolated 126 fungal strains from Namsaengi-pond, 22 fungal strains from Sujangdong-marsh, 44 fungal strains from Yongsu-reservoir and 32 fungal strains from Gangjeongcheon. The fungal strains were identified using internal transcribed spacer (ITS) region and analyzed the phylogeny and diversity. Endophytic fungi isolated from plants of Namsaengi-pond were classified to 30 genera, 19 families, 12 orders, 7 classes and 4 phyla. Endophytic fungi of Sujangdong-marsh were classified to 11 genera, 11 families, 6 orders, 5 classes and 4 phyla. Endophytic fungi of Yongsu-reservoir were classified to 13 genera, 12 families, 7 orders, 5 classes and 4 phyla. Endophytic fungi isolated from Gangjeongcheon were classified to 9 genera, 7 families, 5 orders, 2 classes and 1 phyla. Overall, they were divided 40 genera and Alternaria, Colletotrichum and Fusarium were isolated from 4 sites in common. By investigating the endophytic fungi in aquatic plants, it is for baseline data that determination of diversity and the ecological distribution of endophytic fungi.
Pear black necrotic leaf spot (PBNLS) on pear trees (Pyrus pyrifolia) is caused by a Korean isolate of Apple stem grooving virus (ASGV-K). Yellow spots were detected in Phaseolus vulgaris (kidney bean) and Chenopodium quinoa which were grown near the diseased pears in year 2000 through 2003. The ASGV-K, the causative agent of PBNLS, was detected from the symptoms of the diseased kidney bean plant and C. quinoa. ASGV-harboring fungi were also isolated from symptomatic plants and from soils surrounding the infected plants. The ASGV-harboring fungus was identified and characterized as Talaromyces flavus. Ecopathological studies showed that the number of ASGV-harboring fungi on the pear leaves was not correlated with differences in temperature or severity of symptoms. Additionally, there was no difference in fungus frequency among the orchard locations or different host plants. Although the frequency of fungi isolated from the soil was not affected by changes in temperature or location, the fungi occurred at higher densities in the rhizosphere than in the plants themselves.
This study was conducted to examine the distribution of arbuscular mycorrhizal fungi (AMF) in the soil grown tomato plants in Damyang districts. We collected twenty one soil samples from the rhizosphere of tomato plants which were grown under structure. Number of spores/g in the soil sized over $500{\mu}m,\;355{\sim}500{\mu}m,\;251{\sim}354{\mu}m,\;107{\sim}250{\mu}m\;and\;45{\sim}106{\mu}m$ were 0.01, 0.02, 0.09, 0.9, and 2.0. Total number of spores/g in the fresh soil were 3.02. Mycorrhizal root infection by vesicles, hyphae and arbuscules were 18.0%, 6.0% and 2.0%. To identify the genus of arbuscular mycorrhizal fungi, isolated mycorrhizal spores from the soil grown tomato plants were inoculated into the host plant of sudangrass and mass propagated for 4 months. As a result of identification, mycorrhizal spores were identified as Glomus sp., Gigaspora sp. and Acaulospora sp.
Wood-rotting fungi were collected in Suweon campus of Seoul National University from September 1981 to October 1982. These fungi were classified into 2 orders, 8 families, 22 genera and 34 species. Of these, Gloeoporus adustus (Willd. ex Fr.) $Pil\acute{a}t$ and G. dichrous (Fr.) Bres. were found to be new species that belong to unrecorded genus in Korea. Korean common names of these two new species were designated by the authors. List of the wood-rotting fungi was given with the generic name of host trees on which the fungus was collected.

This experiment was carried out to screen the aeroponically grown host plants suitable for the mass propagation of arbuscular mycorrhizal fungi (AMF) inoculum and clarify the effect of P levels in nutrient solution on the growth of aeroponically grown sweet potato (Ipomoea batatas L.), AMF infection, and mass propagation of mycorrhizal spores, etc. Amount of biomass of host plant became higher, as the P levels in nutrient solutions increased from 5 to 20 M. AMF infection rates in mycorrhizal roots increased in higher P levels in nutrient solution, and decreased in lower parts of mycorrhizal roots by about 18.6-26.0%. About 586 mycorrhizal spores per 1 g fresh root were formed at 16 weeks after inoculation of mycorrhizal inoculum. Total of 830,479 mycorrhizal spores were propagated in each plot.
