Effects of Small Molecular Antioxidants on Cerulein-induced Acute Pancreatitis in Rat

  • It has been suggested that oxygen free radicals are involved in the initiation process of acute pancreatitis, although its pathogenesis is not clear. This study evaluates the roles of oxygen radicals and the effects of small molecular antioxidants (rebamipide, N-acetyl-cysteine, allopurinol, ${\beta}-carotene)$ on the development of cerulein-induced acute pancreatitis. Acute edematous pancreatitis was induced by the intravenous infusion of cerulein at supramaximal dose of 10 ${\mu}g/kg/hour$ for 3.5 hours. The effects of antioxidants, rebamipide (100 mg/kg, i.p.), N-acetyl-cysteine (200 mg/kg, i.v.), allopurinol (20 mg/kg/hour), ${\beta}-carotene$ (50 mg/kg, i.p.), were examined. Cerulein administration resulted in a significant increase in serum amylase activity and pancreatic malondialdehyde (MDA), but not glutathione peroxidase (GSHpx). The glutathione (GSH) content in pancreatic tissue decreased dramatically. Pretreatment of N-acetyl-cysteine significantly decreased the cerulein-induced hyperamylasemia and maintained GSH content in pancreas, but MDA was slightly decreased. In addition, N-acetyl-cysteine ameliorated histological damage. Allopurinol and ${\beta}-carotene$ attenuated cerulein-induced hyperamylasemia, but histologically there was no difference from control. These results indicate that oxygen free radicals play an important role in the initiation of experimental acute pancreatitis. N-acetyl-cysteine is an effective antioxidant that ameliorates the cerulein-induced acute pancreatitis, and the possible therapeutic application of antioxidants against acute pancreatitis needs a further evaluation.

The Effect of SoSikCheongWool-Tang on White Rats' Hepatocyte Damages Induced by Radix Aconitine (소소식청울탕(消消食淸鬱湯)이 부자독성(附子毒性)으로 유발(誘發)된 흰쥐의 간조직(肝組織)에 미치는 영향(影響))

  • The purpose of this study is to investigate the effect of SoSikCheongWool-Tang on white rats' hepatocyte damages induced by Radix Aconitine. For the study, Radix Aconitine was administered to white rats, damaging their hepatocytes, and then SoSikCheongWool-Tang was given to the rats. As a result, the following histological changes of the hepatocytes were observed through light and electron microscope: 1. Radix Aconitine damaged the congestion, necrosis and hemolysis in the liver. 2. In the six-hour control group, the cisternae of rough and smooth endoplasmic reticula were dilated or disoriented, and the attached ribosomes of rough endoplasmic reticulum were hardly observed. 3. In the three-day control group, the destruction of the nucleus as well as the damage of all the cell-organelles was observed. 4. In all the experiment groups, the rats, after given SoSikCheongWool-Tang, quickly recovered from the cytotoxicity induced by aconitine extracts. From the above results, this study has come to the conclusion that SoSikCheongWool-Tang is efficacious against the cytotoxicity induced by Radix Aconitine.

The injured conditions of Eriophyes kuko $K_{ISHIDA}$ (구기자 혹응애(Eriophyes kuko $K_{ISHIDA}$의 가해상태)

  • This study was intended to know the histological damage of the gall on the leaves, peduncles and calyx of Lycium chinense $M_{ILL}$ formed Eriophyes kuko $K_{ISHIDA}$ during 1962 to 1965. The results are summarized as follows; 1. Eriophyes kuko $K_{ISHIDA}$ fed on only the Palisade layers of the loaves of Lycium chinense $M_{ILL}$ were injured by boring. 2. From the 1st to 10th day, some parts of the spongy layer of the Lycium chinense $M_{ILL}$ leaves were injured, but variation were not significant in this period. 3. From the 15th day, Eriophyes kuko $K_{ISHIDA}$ injured not only the spongy layer of Lycium chinense $M_{ILL}$. leaves but also the great part of e chloroplast of the loaves, and the sponginess caves were appeared inside of the galls.

Effects of Acupuncture at SP6 on Reflux Esophagitis in Rats

  • Objectives : The purpose of this study was to evaluate whether acupuncture at $SP_6$ attenuates esophageal inflammation on refluxed-induced esophagitis. Methods : Acupuncture at $SP_6$ was stimulated by acupuncture torsion technique for 30 seconds four times every hour after an operation induced reflux esophagitis(RE), and its effects were assessed in comparison with RE rats without acupuncture, and normal rats. Results : $SP_6$ acupuncture stimulation markedly ameliorated mucosal damage in the histological evaluation. Reflux-induced esophagitis rats exhibited the down-regulation of antioxidant-related protein expression levels such as heme oxygenase-1(HO-1) in the esophagitis; however, the associated levels with $SP_6$ acupuncture stimulation were significantly higher than those in RE rats without acupuncture stimulation. Moreover, $SP_6$ acupuncture stimulation significantly reduced the expression of inflammatory proteins through mitogen-activated protein kinase(MAPK)-related signaling pathways. The increased protein expressions of inflammatory mediators, cyclooxygenase-2(COX-2) and inducible nitric oxide synthase(iNOS), by nuclear factor-kappa B(NF-kB) activation were significantly suppressed through $SP_6$ acupuncture stimulation. Conclusions : Our findings support the therapeutic evidence for $SP_6$ acupuncture stimulation alleviating the development of esophagitis via regulating inflammation through the activation of the antioxidant pathway.

Effects of Zinc and Aluminum Hot-dip Galvanized Sheet Steel on the Gill and Hepatopancreas of the Abalone Haliotis discus hannai (아연 및 알루미늄 용융도금 처리된 강판이 북방전복(Haliotis discus hannai)의 아가미와 간췌장에 미치는 영향)

  • We investigated the toxicity of zinc and aluminum hot-dip galvanized sheet steel to abalone Haliotis discus hannai via changes in the gill and hepatopancreas using histological and transmission electron microscopy analysis. Experimental groups were composed of one control and four exposure conditions (direct or indirect exposure to zinc and aluminum hot-dip galvanized sheet steel). In the control group, aluminum exposure groups (direct and indirect), and indirect zinc exposure group, abalone mortality was not observed until the end of the experiment, and no histopathological changes were observed in the gill and hepatopancreas. However, the direct zinc exposure group exhibited 100% mortality. Ultrastructural analysis of the cytoplasm of ciliated and microvilli-bearing epithelial cells from gill filaments revealed electron-dense vesicles near the cell membrane and disruption of the nuclear membrane. We also observed swollen mitochondria and a loss of mitochondrial cristae. The hepatopancreas showed similar changes, and we detected highly electron-dense particles within the vesicles. These results suggest that abalone exposed directly to zinc hot-dip galvanized sheet steel experience acute toxicity, causing damage to cell organelles in the gill and hepatopancreas and, finally, inducing mortality.

Pharmacological Actions of New Woohwangchungsimwon Liquid on Cerebral Ischemia and Central Nervous System (신우황청심원액의 뇌허혈 및 중추신경계에 관한 약효연구)

  • ln order to investigate pharmacological properties of New Woohwangchungsimwon Liquid (NCL) and Woohwangchungsimwon Liquid (CL), effects of NCL and CL on cerebral ischemia and central nervous system were compared. Cerebral ischemia insult was performed using unilateral carotid artery occlusion in Mongolian gerbils. The histological observations showed a preventive effect of NCL and CL treatments with ischemia-induced brain damage. The ATP in brain tissue was decreased in vehicle-treated ischemic gerbils. This decrease was prevented by CL treatment. In contrast to what was seen with ATP, the lactate and lipid peroxide were both elevated in vehicle-treated ischemic gerbils. This elevation was inhibited by NCL and CL treatments. While NCL and CL had no effects on the hexobarbital-induced sleeping time, they prevented the seizures induced by electric shock and pentetrazol. NCL and CL showed sedative effect in rotarod and spontaneous activity test. Respiration rate and depth were increased at the high dose of NCL and CL. Furthermore, NCL and CL showed anti-stress effect. Our findings suggest that the pharmacological profile of NCL on cerebral ischemia and central nervous system are similar to that of CL.

  • To evaluate radioprotective effects of Kamisagoonjatang(KST), Kamijihwangtang (KJT) and Kamigoonjajihwangtang(KKJT), studies were done experimentally. The results were obtained as follows: 1. By FACS analysis after exposure to radiation by Liniac, T cell and T-helper cell were significantly increased in KST treated group and also B cell and macrophage in KJT treated group while splenocytes were significantly decreased in control group. 2. WBC, PLT were significantly increased in KKJT treated group as compared with control group after exposure to radiation by Liniac. 3. In histological changes of jejunum of $BALB{\backslash}C$ mice after after exposure to radiation by Liniac, exclusion and fusion of villi were decreased in all groups as compared with control group. 4. In the observation of morphological changes by SEM and TEM after radiation by Liniac, KKJT, KJT and KST inhibited demage of internal structures such as mitochondria, ESR and golgi of jejunum cells in order as compared with control group. From above results it was concluded that KJT, KST and KKJT could be usefully applied for protection from damage by radiotherapy to cancer.

The anti-oxidant activities of processed fruits and vegetables in APAP induced oxidative stress in BALB/c mice

  • There is a strong connection between the diet rich in antioxidants and the decreased incidence of cardiovascular and cancerous diseases. Diets that are rich in anti-oxidants particularly include fruits and vegetables containing the high amounts of vitamin A-E, carotenoids, and minerals. Different processing conditions applied for vegetables and plants results in the alteration of the nutrients present in them. Therefore the rationale of our study was to compare the antioxidant effects of different processed vegetables and plants and to see that which one of them showed best anti-oxidant activity. For this purpose, we have used acetaminophen induced oxidative stress model in mice to check the effects of processed apple, pear, carrot, cabbage, broccoli and radish. Our results have shown that the administration of these samples effectively decreased the expression of parameters related with oxidative stress like ALT, AST, catalase, superoxide dismutase, GPx and 8-OHdG. Moreover they also significantly protected the mice livers from APAP induced damage as shown by histological changes. Therefore our results have demonstrated the effects of processed fruits and vegetables in mice model of oxidative stress.

The role of testosterone on nephrotoxicity of mercuric chloride (Testosterone에 의한 승홍의 신독성 유발에 의한 연구)

  • Mercuric chloride, inorganic compound, is one of the most important drugs that has been used in the field of argriculture, antisyphilitica and anticeptics, but it is not used clinically at present. We have studied the effect of testosterone on the mercuric chloride-induced nephrotoxicity. Renal lipid peroxide concentration of male rat treated with mercuric chloride was significantly increased in comparison with that of the female rat, it showed similar effects on testosterone pretreatment. Changes in renal catalase and gluta- thione peroxidase activities were not siginificantly different in testosterone-treated groups. But, renal xanthine oxidase and aldehyde oxidase activities of testrosterone-treated group given mercuric chloride significantly increased in comparison with that of the testoste- rone-treated alone. Animals treated with testosterone prior to mercuric chloride showed more severe damage on histological observations than those treated with testosterone only. Consequently, we suggest that the mercuric chloride-induced nephrotoxicity might be renal lipid peroxide generating enzyme system by testosterone.

Anti-oxidant and Hepatoprotective Activities of Alcoholic Extract of Terminalia arjuna

  • Alcoholic extract of Terminalia arjuna [TA] was evaluated far its hepatoprotective activity against carbon tetrachloride (CCl$_4$)-induced hepatic damage in rats. The hepatoprotective activity of TA was evaluated by measuring levels of serum marker enzymes like serum glutamate oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT), serum alkaline phosphatase (SALP). The serum levels of total proteins(TP), total albumins (TAL) and bilirubin (BILN) were also estimated. The histological studies were also carried out to support the above parameters. Silymarin (SM) was used as standard drug. Administration of TA (250 and 500 mg/kg/po) markedly prevented CCl$_4$-induced elevation of levels of SGOT, SGPT, SALP, TP, TAL and BILN. These biochemical observations were supplemented by histopathological examination of liver sections. Alcoholic extract of TA also shown significant in-vitro free radical scavenging activity against 1, 1-diphenyl-2-picryl hydrazyl (DPPH) and nitric oxide (NO) radicals. Thus, the present study provides a scientific rationale for the traditional use of this plant in the management of liver diseases.
