당뇨병의 한의학적 치료에 대한 국내 실험연구 고찰 - 2013년 이후 (Review of Domestic Experimental Studies of Korean Medicine Treatment for Diabetes Mellitus Since 2013)

  • Objective: This study reviewed experimental studies on the effects of Korean medical treatment for diabetes mellitus (DM). Method: We reviewed 24 studies about DM that had been published since 2013. We searched the Korean Institute of Oriental Medicine (KIOM) database with the keyword '당뇨', '消渴', 'Diabetes'. Results: 1. The studies used various Korean medicine treatments; 15 used single medicinal herbs, 6 used herbal complexes, and 2 used acupuncture. 2. The measurement parameters used in the studies included blood glucose, body weight, histochemical change, lipid parameters, and food/water intake. Anti-diabetic effects were demonstrated in most of the studies. 3. The experimental animals used were rats or mice. There were 15 type 1 diabetes models, 7 type 2 diabetes models, and one normal model. Among them, 13 cases of chemical diabetes induced by Streptozotosin (STZ, type 1 diabetes inducer) were the most common. Conclusion: Further articles on Korean medical treatment of DM should be studied.

의이인(薏苡仁) 추출물이 DSS(dextran sulfate sodium)로 유발된 생쥐의 궤양성 대장염에 미치는 영향 (The Effects of Coicis Semen Extract (CSE) on Dextran Sulfate Sodium - Induced Colitis in Mice)

  • Objectives : This study was carried out to investigate the effects of Coicis Semen Extract (CSE) on the experimental colitis induced by dextran sulfate sodium (DSS) in mice. Methods : Experimental colitis was induced by daily treatment with 5% DSS in the drinking water for 7 days in 6-week-old male ICR mice. The colitic mice were divided into three groups: the normal (N) group consisted of mice that were not inflammation-induced. The control (C) group was composed of untreated colitis elicited mice. The sample (S) group was administered CSE after colitis elicitation. The effects on colonic mucosal ulcers were evaluated by the morphological, histological and immunohistochemical change of the large intestine. Results : Inhibition of LPS-induced NO decreased in the S group. Inhibition of LPS-induced iNOS and COX-2 mRNA noticeably decreased in the S group from 0.25 mg/ml. In the common morphological and histochemical change, the erosion and the infiltration of inflammatory cells increased in the C group, while they noticeably decreased in the S group. The length of colon was shortened more in the C group than in the S group. The distributions of MUC2 and Hsp70 treated with CSE increased noticeably more in the S group than in the C group (p<0.05). It was confirmed histochemically and immunohistochemically that the distributions of iNOS, COX-2, MAC387, serotonin, apoptosis and PCNA treated with CSE decreased in the S group more than in the C group (p<0.05). Conclusions : It is confirmed that CSE has cytoprotective effect, so can alleviate inflammation process. Therefore, it is expected to have potential protective effect on colitis.

치은증식시 세포구성과 성장인자에 관한 면역조직화학적 연구 (IMMUNOHISTOCHEMICAL STUDIES ON CELL POPULATION AND GROWTH FACTORS IN GINGIVAL HYPERPLASIA)

  • The purpose of this study was to investigate the differences of histochemical characteristics in inflammatory fibrous gingival hyperplasia (FGH), phenytoin-induced gingival hyperplasia(PIGH), idiopathic gingival hyperplasia(IDGH) and control groups (healthy and inflammatory gingiva) by immunohistochemical method with various antibodies and histomorphological analysis. In immunohistochemical finding, antibodies to inflammatory cells (T/B lymphocytes, macrophages, other monocytes), proliferating cell nuclear antigen(PCNA), epidermal growth factor(EGF), factor VIII, and type I collagen were used. 1. The inflammatory infiltrates in FGH were less than those in inflammatory gingiva. The composition of inflammatory cells of PIGH was similar with that of FGH. IDGH showed a similar histologic findings with healthy gingival tissue. 2. In FGH, the number of fibroblasts and newly-formed collagen fibers was increased. No significant increase of fibroblasts and the dense accumulation of thick collagen fibers were seen in PIGH. The increase of fibroblasts and the dense accumulation of thick collagen were seen in IDGH. 3. PCNA-positive cells were localized mainly in the area accumulated with inflammatory cells and blood vessels, significantly increased in all hyperplastic tissue groups, and distributed evenly in IDGH. 4. The distribution of EGF were not observed in healthy gingiva but detected locally in area with confluent blood vessels,without significant difference between the other tissue groups. This results suggest that inflammation plays a significant role in inducing hyperplastic change of gingival tissue. While in DIGH, drug itself as well as inflammation seems to attribute to hyperplastic change.

A histochemical study of argentaffin endocrine cells in the gastrointestinal tract of ovariectomized rats

  • The regional distributions and frequencies of argentaffin endocrine cells in gastrointestinal (GI) tract of osteoporotic Sprague-Dawley rat induced by ovariectomy were studied by Masson-Hamperl silver stain. The experimental animals were divided into two groups, one is non-ovariectomized group (Sham) and the other is ovariectomized group (OVX). Samples were collected from each part of GI tract (fundus, pylorus, duodenum, jejunum, ileum, cecum, colon and rectum) at 10th week after ovariectomy or sham operation. Argentaffin cells were detected throughout the entire GI tract with various frequencies regardless of ovariectomy except for the rectum of OVX in which no cells were detected. Most of these argentaffin cells in the mucosa of GI tract were generally spherical or spindle in shape (open type cell) while cells showing round in shape (close type cell) were rarely found in gland regions. Significant decrease of argentaffin cells was detected in OVX compared to that of Sham except for the fundus and jejunum. However, in the fundus and jejunum, argentaffin cells in OVX showed similar frequency compared to that of Sham. In conclusion, the endocrine cells are the anatomical units responsible for the production of gut hormones that regulate gut motility and digestion including absorption, and a change in their density would reflect the change in the capacity of producing these hormones and regulating gut motility and digestion. Ovariectomy induced severe quantitative changes of GI argentaffin endocrine cell density, and the abnormality in density of GI endocrine cells may contribute to the development of gastrointestinal symptoms in osteoporosis such as impairments of calcium and some lipids, frequently encountered in patients with postmenopausal osteoporosis.

식이 Vitamin E가 Streptozotocin 유발 당뇨쥐 신장조직에서의 병리조직학적 변화에 미치는 영향 (Effects of Vitamin E on the Microstructural Changes of Renal Tissue in Streptozotocin-Induced Diabetic Rats)

  • The purpose of this study was to investigate the effects of vitamin E on the histochemical change of kidney tissue in diabetic rats. Sprague Dawley male rats weighing 100$\pm$10g were randomly assigned to one normal and three STZ induced diabetic groups, which were subdivided into vitamin E free diet(DM 0E group), 40mg vitamin E per kg diet(DM 40E group) and 400mg vitamin E per kg diet(DM 400E group). Vitamin E level of normal group was 40mg per kg diet. Diabetes was exper imentally induced by intravenous injection of 55mg/kg of body weight of streptozotocin(STZ) in citrate buffer(pH 4.3) after 4 weeks feeding of experimental diets. Animals were sacrificed at the 6th day of diabetic states. The contents of thiobarbituric acid(TBARS) in kidney were increased 119%, 84% and 33% in DM 0E, DM 40E and DM 400E groups, respectively, compared to normal group. That of DM 400E group was decreased 39% compared to DM 0E group. Content of 2 microglobulin in urine in DM 0E, DM 40E, and DM 400E groups were increased by 248%, 181%, and 164%, respectively, compared to normal group. The diabetic groups showed the regressive lesion such as renal tubule, intumescence of epithelial cell, vacuolization. The results of the observation through electronic microscope showed the mitochondria shape of proximal tubule epithelial cell, irregular array, increase of ribosome, and irregular arrangement of small villosity, etc. These types of changes appeared severer in DM 0E group than in DM 400E group. These results indicate that the TBARS productions on kdney in STZ induced diabetic rats were increased, consequently those leaded to damage of renal tubule and minuteness structure. But a large quantity vitimin E supplementation was suppressed in TBARS production and improved in peroxidative damage of renal tissue so that relieved degenerative changes of renal tubule epithelial cell.

노화된 흰쥐의 뇌줄기에 분포하는 NADPH-Diaphorase 및 Neuropeptide Y양성 신경세포에 관한 연구 (A Study on NADPH-Diaphorase and Neuropeptide Y-Positive Neurons in The Brain Stem of Aged Rats)

  • This study was aimed to charily the change of NADPH-diaphorase(NADPH-d) and neuropeptide Y(NPY) associated with aging of the rots. To verily the effect of aging of NPY and NADPH-d-positive neurons in the brain stem, the neurons were stained by the immunohistochemical and histochemical method. In the aged group, the number of NADPH-d-positive neurons was significantly decreased in substantia nigra lateralis, supragenual nucleus, raphe magnus nucleus and raphe obscurus nucleus as compared with control group. The number of NADPH-d-positive neurons was not significantly decreased in superior colliculus, interpeduncular nucleus, central gray. dorsal raphe nucleus, retrorubral nucleus, pedunculopontine tegmental nucleus, laterodorsal tegmental nucleus, pontine reticular nucleus, prepositus hypoglossal nucleus and nucleus solitarius of the aged rats. The NADPH-d and NPY-positive neurons were found in the interpeduncular nucleus, central gray, substantia nigra lateralis, laterodorsal tegmental nucleus, nucleus solitarius, raphe magnus nucleus, raphe obscurus nucleus of the control and aged groups. The coexistence of NADPH-d and NPY in the some cell was not found in the brain stem of both groups.

전기자극이 흰쥐의 정상 가자미근 형태에 미치는 영향 (Effects of Electrical Stimulation on Normal Soleus Muscle in Rat)

  • 전기자극이 흰쥐의 정상근에 어떠한 영향을 미치는지를 알아보기 위하여 좌골신경의 표적근인 가자미근을 이용하여 조직계측학적, 조직화학, 전자현미경적 관찰을 하였다. 정상적으로 활동하는 흰쥐의 골격근을 매일 전기자극한 결과 전기자극군은 자극을 시작한 후 2주까지는 근섬유의 굵기와 무게가 증가하고 그 이후로는 큰 변화가 없었다. 특히 근섬유의 종류별로는 적색섬유(red muscle fiber)의 채적밀도가 증가하였고 백색섬유(white muscle fiber)는 줄어드는 양상을 보였다. 조직화학적 검사 결과 전기자극군은 근섬유가 다소비대(hypertrophy)해지고 내근주막과 외근주막 사이가 좁아져 있었다. 정상근에서는 당원이 근섬유의 뚜렷한 구별없이 양성반응을 보였으나 실험군은 특정 근섬유에만 반응이 나타났었다. 또한 NADH-TR반응 결과 전기자극군은 적색섬유가 유의하게 증가하여 생체계측학적 결과와 일치하였고 미세구조적 관찰 결과 정상근의 적색섬유는 근섬유와 평행으로 mitochondria가 형성되어 있었고 백색섬유에서는 근절부위에서 관찰되었다. 그러나 전기자극군에서는 적색섬유는 근초부위에 mitochondria가 많이 관찰되고 백색섬유도 근절부위에 작은 mitochondria의 수가 증가되는 것을 관찰할 수 있었다.

흰쥐 정소의 방사선 조사에 따른 정자변화의 영상처리에 의한 정량분석 (The quantitative analysis by the image processing of sperm changes according to the radiation irradiation of white rat testicle)

  • 본 연구는 흰쥐의 정소에 방사선을 조사하였을 때 시간이 경과함에 따라 나타나는 정소세포 및 정자의 변화를 현미경학적으로 얻은 영상을 기초로 영상처리에 의한 정량분석을 통하여 정자의 변화에 대한 결과를 정량화함으로써 더 정확하고 객관적인 결과를 얻고자 하는데 그 목적을 두고 있다. 연구에는 8주 수령의 흰쥐를 대상으로 하였으며, 6 MV의 X선을 1회 2 Gy를 전신 조사하였다. 총 30 마리의 흰쥐를 대상으로 방사선을 조사하지 않은 정상대조군과 실험군을 조사 즉시, 조사 2시간, 4시간, 8시간, 24시간 후에 각 실험군 마다 5마리의 정소를 각각 적출하였다. 적출 후 Periodic acid Schiff 염색을 시행하여 정상대조군과 실험군에서의 정소세포와 정자의 상태를 관찰하였다. 방사선조사 후 24시간까지, 정소세포수와 정자수가 점차 감소하는 것을 정성적 및 정량적으로 유의함을 확인하였다.

방사선 조사가 백서 악관절에 미치는 조직병리학적 조기변화에 관한 실험적 연구 (THE EXPERIMENTAL STUDY OF EARLY IRRADIATION EFFECTS ON THE TEMPOROMANDIBULAR JOINT IN WHITE RAT)

  • The purpose of this study was to investigate the early effects of irradiation on the temporomandibular joint in rats. Male rats were singly irradiated with the dose of 5 Gy or 10 Gy to their head and neck region by /sup 60/Co X ray. Experimental animals were sacrificed at each of the following time intervals -1, 2, 3, 5, 7 and 14 days. The specimens were examined with a light microscope, and treated with H & E staining and immuno-histochemical staining. The results were as follows, 1. By light microscopic findings, proliferative and hypertrophic zone were narrowed and hematopoietic cells were few in number at 5 days after irradiation. Repair signs were seen at 7 days after irradiation when decrease in osteoclast, increase in hematopoietic cells and increase of proliferative zone were noted. The 10 Gy irradiated group showed more severe histopathologic change than the 5 Gy group, and their repair was more slow. 2. In the S -100 antibody, positive cells were examined in the glenoid fossa. Positive cells of irradiated group showed more slight decrease in number than the control group. Low radiosensitivity and slow repair was noted in the glenoid foosa. 3. The interarticular disc was high radioresistant, and any histopathologic changes were not seen in disc. 4. Repair was examined clearly with the response to the antibodies. Especially by 5 days after irradiation 5 Gy group showed S-l00 positive cells in hypertrophic zone next to proliferative zone, chondroitin-4-sulfate positive cell in erosive zone next to hypertrophic zone, type-1 collagen positive cell in subchondral bone.

구개골에 발생한 악성 Peripheral Nerve Sheath Tumor의 증례보고 (MALIGNANT PERIPHERAL NERVE SHEATH TUMOR ON PALATE: A CASE REPORT)

  • Summary: The malignant peripheral nerve sheath tumor(MPNST) is an aggressive neoplasm and can either arise independently or result from malignant change in preexisting neurofibromatosis (von Recklinghausen's disease). Its histologic characteristics remain controversial, but currently it is believed that the schwann cell is the origin of the peripheral nerve sheath tumors. MPNST is an uncommon neoplasm of the head and neck region, and its presentation in the oral cavity is quite rare. In this study, we report a patient with a rare case of a MPNST involving the maxilla. A case report: A 29-year-old female presented with a chief complaint of painless swelling with bleeding tendency on the left maxillary tuberosity area 2 months ago. Clinical examination showed a $5.0{\times}3.0cm^2$ sized, indurative swelling on the site. Conventional radiographs showed a relatively well-defined soft tissue mass involving the left maxillary sinus, and destruction of the anterior, posterolateral walls of the left maxillary sinus. Subtotal maxillectomy and split-thickness skin graft from thigh were undertaken. In histochemical and immunohistochemical studies, the specimen revealed positive reactivities to Vimentin and S-100 protein. Final diagnosis was made as MPNST.
