This study investigated the anti-obesity effects of Eriobotrya japonica leaves extract (EJE) in a high-fat diet-induced obese mice model. For the analysis, 40 mice were randomly divided into five groups: normal diet (ND), high-fat diet group (HD), high-fat diet containing EJE 100 mg/kg b.w. (EJE-L), 500 mg/kg b.w. (EJE-H), and Garcinia cambogia extract 100 mg/kg b.w. in each AIN 93G supplement for 8 weeks as positive control. During the study period, food intake and body weight were measured separately. Body weight, serum triglyceride level, and total cholesterol levels of the EJE group were significantly lower than those of the HD and ND. Moreover, mice fed an EJE-containing high-fat diet showed decreased leptin levels compared with high-fat diet-induced obese mice. These results suggest that EJE has potential as an anti-obesity agent by reducing body weight and improving levels of triglycerides, total cholesterol, low density lipoprotein-cholesterol, and leptin in serum.

This study investigated the effect of young Phragmites communis (Pc) leaves on lipid profiles, lipid metabolism and erythrocyte antioxidant defense system in high-fat diet fed mice. Three groups of mice were fed different diets for 8 weeks: normal diet (Normal), high-fat diet (High-fat; 37% calories from fat) and high-fat diet supplemented with 1% Pc (wt/wt, HF-Pc). Body weight, daily food intake and energy intake tended to decrease by Pc supplement in high-fat fed mice. Pc supplementation significantly lowered plasma triglyceride and total cholesterol concentrations compared to the high-fat control group. Pc also lowered hepatic and heart cholesterol contents, whereas it significantly increased fecal excretion of triglyceride and cholesterol compared to the high-fat control group. Pc significantly inhibited fatty acid synthase, 3-hydroxy-3-methylglutaryl CoA reductase and acyl-CoA:cholesterol acyltransferase activities compared to the high-fat control group. Erythrocyte superoxide dismutase and catalase activities were also significantly higher in the high-fat group than in the normal group, however Pc supplementation reversed these changes. The Pc supplementation significantly lowered erythrocyte lipid peroxidation level compared to the high-fat control group. Accordingly, these results suggest that Pc improves lipid metabolism and erythrocyte antioxidant defense system in high-fat diet fed mice.

The intake, dry matter (DM) digestibility, total mean retention time (TMRT) and weight changes were investigated in four fat tailed and four Dorper sheep fed on a maize stover crop residue. Animals were subjected to five dietary treatments, which were: 90, 135 and $180g\;DM/kgBW^{0.75}/day$ of chopped maize stover offered with urea, and in subsequent dietary treatments $180g\;DM/kgBW^{0.75}/day$ of stover supplemented with either dried bean husks or poultry manure. Although not statistically different (p>0.05) fat tailed sheep had higher maize stover intake than the Dorper sheep when expressed per metabolic body weight. The DM intake and digestibility increased significantly (p<0.001) in both sheep breeds when the maize stover offer levels were increased from 90 to $135g/kgBW^{0.75}/day$. Dorper sheep had significantly (p<0.001) higher DM digestibility and longer total mean retention times for both the liquid and solid matter than the fat-tailed sheep. Fat-tailed sheep gained 4.75 kg while Dorper sheep gained 3.75 kg over the entire experimental period. The weight change per dietary treatment was affected by the amount of maize stover and the type and amount of supplement consumed. The higher DM digestibility and longer total mean retention time in Dorper sheep shows that the breed has adapted to utilize the low quality roughage diets. An indigenous fat tailed, has the potential to ingest high volumes of fibrous feed and has higher voluntary intakes relative to its body size. The two sheep breeds are a valuable genetic resource in sub Saharan Africa that is characterized by long dry season with no readily available and affordable conventional feeds for small ruminants.
This study an analyzes the effects of the P/S ratio of dietary lipids and antioxidant vitamin supplements on serum lipids level and fatty acid profile, the degree of lipid peroxidation, and the antioxidant enzyme activities in the liver of rats treated with 7,12-dimethylbenz($\alpha$) anthracene(DMBA). P/S ratio of dietary lipids was made into 0.5, 1 and 2 by mixing palm oil, soybean oil, sesame oil and perilla oil at 10%(w/w) fat level and n-6/n-3 ratio was fixed to 4. Antioxidant vitamin of $\alpha$-tocopherol or $\beta$-carotene was supplemented in addition to vitamin mixture which was given at 1 % of the standard diet. female Sprague-Dawley strain rats, about 60 days old, were divided into three groups(LP : low P/S ratio(0.5), MP : medium P/S ratio (1.0), HP , high P/S ratio(2.0)) and each group was sub-divided into three groups(S ; standard, T ; tocopherol supplemented, C : carotene supplemented): Two weeks after feeding experimental diets, all groups were treated with a single dose of DMBA(2mg/100g BW) by gastric intubation and fed experimental diet for 9 week. The results were as follows ; 1) Serum total cholesterol(TC) level was not significantly influenced by diet but tended to be lower in HP groups compared to LP and MP groups. Triglyceride level was the highest in LP groups and the lowest in $\alpha$-tocopherol supplemented groups. 2) Thiobarbituric acid reactive substance(TBARS) level, representing lipid peroxidation in hepatic microsome, tended to be increased as the unsaturation of dietary lipids increases. $\alpha$-Tocopherol supplement significantly decreased TBARS level. 3) The activities of superoxide dismutase(SOD) and glutathione peroxidase(GSHPx) in hepatic cytosol showed the tendency to be high with increasing P/S ratio of dietary lipids. SOD activity was not significantly influenced by antioxidant vitamin, but GSHPx activity was significantly increased in $\alpha$-tocopherol supplemented groups. In summary, high polyunsaturated fat diet was effective on reducing the serum level of total cholesterol and triglyceride, while it increased unsaturation and peroxidizability of serum fatty acid. With increasing P/S ratio of dietary lipids, lipid peroxidation was increased in the liver and antioxidant enzyme system was induced to inhibit lipid peroxidation against oxidative damage. $\alpha$-Tocopherol supplement was effective in lowering lipid peoxidation, but $\beta$-carotene supplement did not exhibit antioxidant effect. Supplement of dietary fibers (DF) is regarded as one of the most effective way to prevent and relieve chronic diseases caused by long-term intake of a high-fat diet in the current society. The health benefits of soluble dietary fibers (SDF) have been widely researched and applied, whereas the insoluble dietary fibers (IDF), which represent a higher proportion in plant food, were mistakenly thought to have effects only in fecal bulking. In this article, we proved the anti-obesity and glucose homeostasis improvement effects of IDF from pear pomace at first, and then the mechanisms responsible for these effects were analyzed. The preliminary study by real-time PCR and ELISA showed that this kind of IDF caused more changes in the gut microbiota compared with in satiety hormone or in hepatic metabolism. Further analysis of the gut microbiota by high-throughput amplicon sequencing showed IDF from pear pomace obviously improved the structure of the gut microbiota. Specifically, it promoted the growth of Bacteroidetes and inhibited the growth of Firmicutes. These results are coincident with previous hypothesis that the ratio of Bacteroidetes/Firmicutes is negatively related with obesity. In conclusion, our results demonstrated IDF from pear pomace could prevent high-fat diet-induced obesity in rats mainly by improving the structure of the gut microbiota.
The aim of this study investigated the fluctuations of 3 characters from 3 ecotypes [early ripening (ER), middle ripening (MR), and late ripening (LR)] of 20 Korean brown rice cultivars in different storage systems [time: 12 and 24 weeks, temperature: low ($10^{\circ}C$) and room ($25^{\circ}C$)]. With increase of storage time and temperature, lipoxygenase activity, and fat acidity increased, whereas germination rate was reduced. ER cultivars exhibited the highest lipoxygenase activity of $35.49{\pm}2.46$ unit/mg protein during 24 weeks storage at $25^{\circ}C$, followed by LR ($32.73{\pm}1.33$) and MR ($32.66{\pm}1.62$) cultivars. The amounts of fat acidity also were observed by the same order (ER: $20.40{\pm}2.12$>LR: $19.68{\pm}1.86$>MR: $19.64{\pm}1.35$ mg KOH/100 g). Germination rate slightly decreased with increase of time and temperature (MR>LR>ER), but MR and LR cultivars showed the most significant changes (ER: $60.90{\pm}23.47%$, MR: $32.66{\pm}13.95%$, and LR: $32.53{\pm}5.87%$). On the basis of above results, MR cultivars were evaluated the highest quality, because high lipoxygenase activity, high fat acidity, and low germination rate have deteriorated in quality and generated off-odor. Thus, MR cultivars might be very important sources in food processing and stored dietary supplement aspects.
Ginseng and probiotics have anti-obesity effects in mice fed a high-fat diet (HFD). Absorption of ginsenoside and colonization of probiotics occur in the intestine. In this study, a mixture of fermented ginseng and two probiotics, Bifidobacterium longum BORI and Lactobacillus paracasei CH88, was administered to HFD-fed mice for 9 weeks. The mixture significantly suppressed weight gain (p < 0.05, n = 8) and lipid deposition in the liver and adipose tissues as well as increased the mice's food intake. The adipocyte size of the adipose tissue was significantly decreased in the mixture-fed group, especially when 0.5% fermented ginseng and $5{\times}10^8/ml$ of the two probiotics were used (p < 0.05, n = 10). The expression of TNF-${\alpha}$ in adipose tissue was efficiently downregulated in the mixture-fed group (p < 0.05, n = 4). The supplement also improved the mice's fasting blood glucose levels (p < 0.05, n = 8) and total cholesterol feces excretion (p < 0.05, n = 8). The mixture of fermented ginseng and B. longum BORI and L. paracasei CH88 could have an anti-obesity effect and suppress lipid deposit in the liver and adipose tissues.
Obesity-induced disorders contribute to the development of metabolic diseases such as insulin resistance, fatty liver diseases, and type 2 diabetes (T2D). In this study, we evaluated whether the Aloe QDM complex could improve metabolic disorders related to blood glucose levels and insulin resistance. Male C57BL/6 obese mice fed a high-fat diet for 54 days received a supplement of Aloe QDM complex or pioglitazone (PGZ) or metformin (Met) and were compared with unsupplemented controls (high-fat diet; HFD) or mice fed a regular diet (RD). RT-PCR and western blot analysis were used to quantify the expression of obesity-induced inflammation. Dietary Aloe QDM complex lowered body weight, fasting blood glucose, plasma insulin, and leptin levels, and markedly reduced the impairment of glucose tolerance in obese mice. Also, Aloe QDM complex significantly enhanced plasma adiponectin levels and insulin sensitivity via AMPK activity in muscles. At the same time, Aloe QDM decreased the mRNA and protein of $PPAR{\gamma}/LXR{\alpha}$ and scavenger receptors in white adipose tissue (WAT). Dietary Aloe QDM complex reduces obesity-induced glucose tolerance not only by suppressing $PPAR{\gamma}/LXR{\alpha}$ but also by enhancing AMPK activity in the WAT and muscles, both of which are important peripheral tissues affecting insulin resistance. The Aloe QDM complex could be used as a nutritional intervention against T2D.
The hepatoprotective and nephroprotective effects of Allium victorialis (AV) methanol extracts were investigated on high fat diet (HFD) supplied mice. Treatment of AV extracts (62.5, 125, 250 mg/kg) once a day for 12 weeks markedly decreased the liver steatohepatitis and kidney damages. AV extracts were inhibited the serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), blood urea nitrogen (BUN) and creatinine elevations and reduced the histopathological changes of livers induced by HFD supply. In addition, AV extracts strengthened the antioxidative defense system with an increased activity of superoxide dismutase (SOD), glutathione (GSH), catalase and reduced malondialdehyde (MDA) levels. The 125 mg/kg of AV extract showed similar favorable effects as compared with silymarin 100 mg/kg. It is suggested that AV methanol extract administration is beneficial to the improvement of the alleviation of liver and kidney damages in HFD supplement.

In many cases, obesity is associated with metabolic disorders. Recently, natural compounds that may be beneficial for improving obesity have received increasing attention. Bitter melon has received attention as a diabetes treatment. $NAD^+$-dependent deacetylase (Sirtuin 1, SIRT1) has emerged as a novel therapeutic target for metabolic diseases. In this study, ethanol extract of bitter melon (BME) suppressed adipocyte differentiation and significantly increased the expression of SIRT1 in fully differentiated 3T3-L1 cells. Moreover, it enhanced the activation of AMP-activated protein kinase (AMPK). In high-fat diet (HFD)-fed induced-obesity mice, BME suppressed HFD-induced increases in body weight and white adipose tissue (WAT) weight. BME also increased the expression of SIRT1 and suppressed peroxisome proliferator-activated receptor and sterol regulatory element binding protein 1 expressions of WAT from HFD-fed mice. These findings suggest that BME prevents obesity by activating the SIRT1 and AMPK pathway and that it may be a useful dietary supplement for preventing obesity.
