A review on the main aspects associated with yeast flocculation and its application in biotechnological processes is presented. This subject is addressed following three main aspects-the basics of yeast flocculation, the development of "new" flocculating yeast strains and bioreactor development. In what concerns the basics of yeast flocculation, the state of the art on the most relevant aspects of mechanism, physiology and genetics of yeast flocculation is reported. The construction of flocculating yeast strains includes not only the recombinant constitutive flocculent brewer's yeast, but also recombinant flocculent yeast for lactose metabolisation and ethanol production. Furthermore, recent work on the heterologous $\beta$-galactosidase production using a recombinant flocculent Saccharomyces cerevisiae is considered. As bioreactors using flocculating yeast cells have particular properties, mainly associated with a high solid phase hold-up, a section dedicated to its operation is presented. Aspects such as bioreactor productivity and culture stability as well as bioreactor hydrodynamics and mass transfer properties of flocculating cell cultures are considered. Finally, the paper concludes describing some of the applications of high cell density flocculating bioreactors and discussing potential new uses of these systems.e systems.
본 연구에서는 재조합 E. coli 시스템에 의한 재조합 cystein proteinase의 대량 생산을 위하여 4L 회분 배양 및 최적 지수 생장기에서의 IPTG- induction을 연속적으로 실시하였다. 발현된 rPwCP1 양은 진탕 배양 시스템에서의 결과와 비교해 볼때 현저하게 향상되었음을 알 수 있었으며, 더불어 metal affinity chromatography 방법으로 처리하여 전기영동을 수행한 결과, 목적 재조합 단백질만을 정제 및 농축시킬 수 있음이 확인되었다.
Gardenia callus was induced in MS medium containing $10{\;}{\mu}M$ of 2,4 diphenoxy acetic acid (2,4-D), $1{\;}{\mu}M$ kinetin, and 3% sucrose in the dark. $B_5$ medium was identified to be the most adequate medium for cell growth. Indole-3-acetic acid (IAA) was better growth regulator than 2,4-D not only for cell growth but slso for carotenoid production. Ligt also played a critical role on synthesis of carotenoid. Gardenia cells grown in $B_5$ medium could utilize a polysaccharide, soluble starch, as a carbon source. The cell growth was stimulated in $B_5$ medium fortified with 0.2% yeast extract. The optimum pH for cell growth was 5.7. High density cultures can be maintained by increasing inoculum size and medium concentration accordingly. Specific growth rate and mass doubling time were 0.095 $day^{-1}$ and 7.3 days, respectively. The cell immobilized in alginate tends to formulate more enlarged vacuoles containing yellow pigment compared with those of suspended cell. Carotenoid content of immobilized cell was about $264.4{\;}{\mu}g/g$ fresh weight (F.W.) corresponding twice of the content of suspended cell ($112.08{\;}{\mu}g/g$ F.W.). The color of gardenia cell was shifted from yellow to red when carbohydrase-secreting fungus, Trichoderma reesei, was co-cultivated with gardenia cells.
본 연구에서는 참당귀 현탁세포배양에 의한 면역 증강성 ECP의 생산을 증진시키기 위하여 배지교환식 배양을 수행하였으며, ECP의 분비를 촉진하는 초음파 처리와 Pluronic F-68의 영향을 조사하였다. 참당귀의 현탁세포배양시, 최대 세포생장은 6일째에 16.8 g DCW/L였고, ECP의 생산은 세포생장과 함께 증가하다가 8일째에 최고 0.9 g/L가 생산되었다. 식물세포 고농도배양법인 배지교환식 배양을 적용해, 초기 당 농토를 높이고 접종 후 5일부터 연속적으로 배지교환을 해주어 23.8 gDCW/L의 고농도의 현탁세포를 얻을 수 있었다. 초음파 처리 및 Pluronic F-68의 첨가는 참당귀 세포의 세포막의 투과성을 증진시켜 ECP의 생산성을 높임과 동시에 세포 내의 다당의 배지로의 배출을 유도한 것으로 판단된다.
Chlorella kessleri cultivated in artificial wastewater using diurnal illumination of 12h light/12h dark (L/D) cycles. The inoculum density was 10(sup)5 cells/mL and the irradiance in light cycle was 45$\mu$mol㎡s(sup)-1 at the culture surface. As a control culture, another set of flasks was cultivated under continuous illumination. Regardless of the illumination scheme, the total organic carbon (TOC) and chemical oxygen demand (COD) was reduced below 20% of the initial concentration within a day. However, cell concentration under the L/D lighting scheme was lower tan that under the continuous illuminating scheme. Thus the specific removal rate of organic carbon under L/D cycles was higher than that under continuous illumination. This result suggested that C. kessleri grew chemoorganotrophically in the dark periods. After 3 days, nitrate was reduced to 136.5 and 154.1mg NO$_3$-N/L from 168.1mg NO$_3$-N/L under continuous illumination and under diurnal cycles, respectively. These results indicate nitrate removal efficiency under continuous light was better than that under diurnal cycles. High-density algal cultures using optimized photobioreactors with diurnal cycles will save energy and improve organic carbon sources removal.
A thermostable $\alpha$-L-arabinofuranosidases ($\alpha$-L-AFase) is an industrially important enzyme for recovery of L-arabinose from hemicellulose. The recombinant $\alpha$-L-AFase from Thermotoga maritima was expressed in Escherichia coli by using a constitutive pHCE or an inducible pRSET vectors. In batch fermentation, the constitutive expression system resulted in slightly faster growth rate (0.78 vs. 0.74/hr) but lower enzyme activity (2,553 vs. 3,723 units/L) than those of the induction system. When fed-batch fermentation was performed, biomass and enzyme activity reached the highest levels of 36 g/L and 9,152 units/L, respectively. The fed batch cultures performed superior results than batch culture in terms of biomass yield (4.62-5.42 folds) and enzyme synthesis (3.39-4.00 folds). In addition, the fed-batch induction strategy at high cell density resulted in the best productivity in cell growth as well as enzyme activity rather than the induction method at low cell density or the constitutive expression.
Dissolved oxygen level of cell culture media has a critical effect on cellular metabolism, which governs specific productivity of recombinant proteins and mammalian cell growth However, in the cores of cell aggregates or cell-immobilized beads, oxygen level frequently goes below a critical level. Mammalian cells have a number of genes induced in the lower level of oxygen, and the genes contain a common cis-acting element (-RCGTG-), hypoxia response element (HRE). By binding of hypoxia inducible factor-l (HIF-I) to the HRE, promoters of hypoxia inducible genes are activated, which is a survival mechanism. In this work, to develop a CHO cell capable of producing recombinant proteins in immobilization and high density cell culture efficiently, mammalian expression vectors containing human tissue-type plasminogen activator (t-PA) gene controlled by HRE were constructed and stably transfected into the CHO cells. In $Ba^{2+}$ -alginate immobilization culture, CHO/pCl/dhfr/2HRE-t-PA cells produced 2 folds higher recombinant t-PA activity than CHO/pCl/dhfrlt-PA cells without $CoCl_2$ treatment. Furthermore, in repeated fed batch culture, productivity of t-PA in immobilized CHO/pCI/dhfr/2HRE-t-PA cells was 121 ng/ml/day, total production of 0.968 mg/day at 11 days culture while CHO/pCIIdhfrlt-PA cells was 22.8 ng/ml/day. All these results indicate that HRE is very useful for the enhancement of protein productivity in mammalian cell cultures.
광합성 홍색세균의 light harvesting complex II (LHC II)발현 유전자가 제거된 돌연변이종을 halogen 램프 하에서 거리에 따라 광도를 달리하며 배양하여 wild type과 생산성을 비교한 결과 낮은 광도(34 ${\mu}E/m^2/s$)에서는 wild type가 높은 광도 (376, 118 ${\mu}E/m^2/s$)에서는 mutant가 높은 세포농도를 나타내었다. 특히 118 ${\mu}E/m^2/s$에서 LHC $II^-$ mutant가 56% 높은 세포생산량을 보였다. 이는 세포내 pigment양의 감소로 mutual shading effect가 감소하였기 때문으로 판단되었다.
고정상곰팡이를 이용하여 세포내 축적 이차대사산 물인 cyclosporin A (Cy A)를 연속적으로 생산하 기 위해 perfusion 고정상연속배양을 수행하였다. 이를 위해 균사형성 생산균주인 T. inflaum을 cel lite에 고정화하는 데 필요한 공정을 획기적으로 단 순화시키는 방법을 제시하였다. 교반식 Perfusion 생물반응기 (Immobilized Perfusion Reacter S System, 이후 IPRS로 표기 내에 제한유지 된 고밀 도 고정상균체는 매우 높은 희석속도($0.1hr^{-1}$)에서 도 유리세포를 생산해내는 세포생생기의 역할을 훌 륭하게 수행하였으며, IPRS의 배출구를 통해 연속 적으로 유출되는 고농도의 유리세포(1.0g/$\ell$/hr)는 이차대사 결과 세포내에 축적되는 CyA를 연속생산 하는 데 이용될 수 있었다. 이러한 IPRS공정 운영 은 고정상균체를 배양액으로부터 효과적으로 분리시 키는 decanting column의 개발로 가능했요며, 이로 인해 기존의 연속현탁배양에서 문제시되었던 높은 희석속도에서의 wash-out현상이 극복될 수 있었다. 또한 유출되는 유리세포(relesed-free cell)의 mor-phology를 원형의 conidiospore나 잘게 부서진 myc celial cell로 구성되어 배양액의 rheology가 뉴튼유 체화될 수 있도록 IPRS공정을 운영함으로써, 기존 의 균사형성 미생물의 현탁배양시 나타나는 배양액 에서의 물질전달 감소현상이 뚜렷하게 개선될 수 있 었다. 유출되는 높은 균체생산성(1.0g/R/hr)을 감안할 때, 균주개발과 production배지의 최척화가 본 IPRS공정개발과 병행되는 경우 기존의 회분식배양 또는 연속현탁배양에 비해 훨씬 효과적 인 생산성 증 대를 기대할 수 있을 것으로 보인다.
Red beet(Beta vulagris L.) 모상근의 회분배양을 이용한 천연색소인 betacyanin 생산 최적화를 위해 광도, C/N ratio, 인산의 농도를 각각 변화시켜 세포생육과 색소 생산성에 관한 동력학적 분석을 실시했다. 광도변화에 따른 배양 결과 3 klux의 경우 0.3(1/day)의 최대 비 생육속도와 0.11(mg/g-dry cell/day)의 최대 비 생산속도 그리고 14 k1ux에서 0.242(1/day)의 최대 비 생육속도와 0.125(mg/g-dry cell/day)의 최대 비 생산속도를 나타냈다. 광도와 균체의 생육관계를 검토한 결과 광도에 따른 세포 생육은 photoinhibition model이 적용됨이 확인되었다. Red beet 모상근으로부터 betacyanin의 생산은 partially growth related process임이 입증됐다. 이에 따른 세포당 최대 betacyanin 생산을 나타내는 $\alpha$는 0.3756 (mg/cell)이며, 최대 생산속도를 나타내는 $\beta$는 0.001 (mg/g-cel1/day)로 측정됐다. C/N ratio에 따른 실험결과 42.1(w/w)에서 0.26(1/day)의 최대 비생육 속도를 나타내었으나 최대 비 생산속도는 31.6(w/w)에서 0.075(mg/g-cell/day)를 나타냈다. 인 농도에 대한 균체의 생육 및 물질 생산성의 관계를 검토한 결과 1.25mM에서 0.31(1/day)의 비생육 속도와 0.134(mg/g-dry cell/day) 비생산 속도를 나타내었다. 최적 조건을 결정하기 위한 response surface methodology(RSM)결과 세포 생육과 betacyanin의 최대 생산을 위한 최적 광도는 5.5 (klux),최적 C/N ratio와 인의 농도는 27(w/w), 1.25 (mM)로 결정됐다. 그리고 0.1 $\mu$M kinetin 첨가시 대조구에 비해 비생산성이 0.085(mg/g-dry cell/day)로 증가함이 입증됐다. Normal조건과 optimum조건의 비교결과 세포의 농도인 X(g-dry wt./L)가 8와 16, betacyanin의 생산량인 P(mg/L)가 4.48과 12.5, 그리고 optimum 조건에서 최대비 생육속도인 $\mu$$_{max}$ 가 0.375와 그리고 최대비 생산속도인 q$^{max}$$_{p}$ 는 0.134로 약 2배로서 최적화가 되었다.
