우량 수박종자 생산을 위한 적정 성숙일수 및 후숙일수 확립 (Establishment of Days after Anthesis(DAA) and Fruit After-ripening Period(FAP) for High-Quality Seed Production of Watermelon)

  • This study was conducted to investigate days after anthesis (DAA) and fruit after-ripening period (FAP) for seed-harvesting of high quality watermelon seeds. Fruit weight and number of seed per fruit increased according to DAA, while those did not significant about FAP. Ratio of cotyledon at whole seed was higher about 2 to 4% compared to seed coat irrespective of DAA and FAP. Germinability of watermelon was not a significant effect by DAA, however, it had differences by FAP. Percent of germination (PB) was below 50%, when 30 days maturated fruits after anthesis was omitted ripening, while PB was increased to 92% by ripening. In addition, seeds at DAA 40 and FAP 20 were higher general seedling vigors (hypocotyl length, diameter etc.) in BP test. Results indicated that considering seed productivity, it had maximized seed viability at DAA 40 and FAP 20.

  • Many accidents occur on the oceans. Accidents involving ships can cause enormous damage. When an accident occurs, the cost of environmental restoration can bankrupt even a top-ranked international company. The potential damages resulting from ship accidents have resulted in many international agreements such as MARPOL, SOLAS, ISM Code, etc. The viability of damaged ships has become more and more important. A damaged ship's viability and cargo safety can be improved by a safety assessment design procedure. We consider th£ shipbuilding industry's distributed characteristics to define a safety assessment system with software engineering-based requirements analysis. We used a distributed network-based simulation method (HLA - High level architecture), because of ease of reuse and expansion of existing components to other situations, and because it is a military standard. HLA is the standard for distributed network-based simulation in many countries including the United States, Canada, Israel, and Korea. The paper describes research to develop a prototype of a network-based safety assessment simulation system by software engineering based analysis.

Protective Effect of Padina arborescens Extract against High Glucose-induced Oxidative Damage in Human Umbilical Vein Endothelial Cells

  • Dysfunction of endothelial cells is considered a major cause of vascular complications in diabetes. In the present study, we investigated the protective effect of Padina arborescens extract against high glucose-induced oxidative damage in human umbilical vein endothelial cells (HUVECs). High-concentration of glucose (30 mM) treatment induced cytotoxicity whereas Padina arborescens extract protected the cells from high glucose-induced damage and significantly restored cell viability. In addition, lipid peroxidation, intracellular reactive oxygen species (ROS), and nitric oxide (NO) levels induced by high glucose treatment were effectively inhibited by treatment of Padina arborescens extract in a dose-dependent manner. High glucose treatment also induced the overexpressions of inducible nitric oxide synthase (iNOS), cyclooxygenase- 2 (COX-2) and NF-${\kappa}B$ proteins in HUVECs, but Padina arborescens extract treatment reduced the over-expressions of these proteins. These findings indicate the potential benefits of Padina arborescens extract as a valuable source in reducing the oxidative damage induced by high glucose.

Aspergillus sp. PS-104의 분생포자 생활력에 미치는 첨가제 효과 (Effect of Additives on the Conidial Viability of Aspergillus sp. PS-104)

  • 포자의 장기보존 풍 생활력 유지를 위하여 계면활성제(Tween 80, SDS, Triton X-100) 및 2가 금속이온 $Ca^{2+}$, $Fe^{2+}$, $Cu^{2+}$, $Zn^{2+}$ 및 3가 금속이온 $Mo^{3+}$을 각각 0, 0.001, 0.01, 0.1, 1.0%(v/v) 농도로 첨가하여 생활력에 미치는 영향을 조사하였다. 또한 각종 유기물(PEG 200, glycerol, 식용유, 종합 vitamin, 설탕, 당밀)도 각각 0, 0.01, 0.1, 1.0, 5.0%(v/v) 농도로 첨가하여 2주, 4주, 2개월, 4개월, 6개월간 보관하면서 생활력을 검정하였다. 계면활성제 중 Tween 80을 $0.01%\sim1.0%$ 농도로 첨가하여 6개월 저장시 상대생존율이 80%에 이르렀으며, 이에 비해 대조구는 약 50% 수준이었다. 이때 분생포자수는 대조구에 비해서 약 100배 높은 수준이었다. 계면활성제 처리 후 저장기간에 따른 균의 생활력은 Tween $80{\geq}SDS>Triton$ X-100 순으로 감소하였다. 유기물 중에서 1.0% 맥반석, 5.0% glucose 및 설탕을 첨가하였을 때가 장기보존시 우수함을 확인하였으며 6개월 경과시 대조구는 초기에 비해 50%의 상대생존율을 보였으나 1.0% 맥반석 첨가시 상대생존율은 약 80%의 높은 수준을 유지하였다. 각종 금속이온을 0.1% 농도로 처리 후 저장기간에 따른 균의 생활력은 $Cu^{2+}>Ca^{2+}>Mo^{3+}>Zn^{2+}>Fe^{2+}$ 순으로 감소하였다.

미니 돼지 동결정액의 융해 온도가 정자성상에 미치는 영향 (Effect of Thawing Temperature on Sperm Characteristics of Frozen Semen in Miniature Pig)

  • 본 연구는 0.5ml straw를 이용한 정자 동결융해 시 응해 온도가 동결정자의 성상에 미치는 영향을 파악하고 미니 돼지의 동결정액에 최적화된 적정 응해 조건을 찾기 위하여 실시하였다. 정액동결 straw를 37, 50 및 $70^{\circ}C$에서 각각 5초, 10초 및 45초간 융해하여 생존율(SYBR-14/PI staining), 정자원형질막기능검사 (Hypoosmotic Swelling Test) 및 첨체반응율(CTC : chlorotetracycline staining)을 검사 한 결과, $70^{\circ}C$에서 5초간 융해한 정자의 생존율과 CTC 결과가 $37^{\circ}C$$50^{\circ}C$에서 10초 및 45초간 융해한 처리구보다 유의적(p<0.05)으로 높은 생존율과 낮은 비율의 첨체 반응율을 얻었다. 따라서 미니돼지의 동결 정액은 고온에서 단시간 융해를 하는 것이 정자의 성상에 유리한 것으로 사료된다.

닭 생식반월의 Busulfan 가온 주입방법에 의한 원시생식세포 제거 효과 (Depletion Effects of Chick Germinal Crescent's Primordial Germ Cells by Heat Activated Busulfan Injection)

  • 본 연구는 생식선 키메라 생산효율을 높이기 위한 방법으로 busulfan 가온 주입법을 이용하여 효과적인 원시생식 세포의 이동능력을 검증하였다. 효율적인 생식선 키메라 닭 생산에서 중요한 요건 중 하나인 공여체 원시생식세포의 생존율을 측정한 실험에서는 시간이 지남에 따라 생존율에 변화를 보였으나, 평균 $70{\sim}80%$을 유지하고 있었으며, busulfan 처리 유무에 따른 공여체 원시생식세포 이동능력은 형광염색 후 주입한 실험에서 대조구가 4.8%인 반면 실험구는 23.5%을 나타냈다. 이식전 원시생식세포 배양 조건에 따라, 96시간과 118시간 배양 처리구에서 높은 이동능력을 보여 주었다. 원시생식세포의 형태학적, 생리학적 특징을 응용한 이식방법은 매우 효과적일 것이다. 그리고 본 연구에서는 생식반월의 발달단계 별 busulfan 처리 효과는 48시간이 가장 높은 53.4%였으며, 그러나 본 연구에서는 생식반월 유래 원시생식세포 이식은 48시간 이전, 혈관계가 발달하기 직전으로 가장 높은 효율을 보였다. 결론적으로 생식선 키메라 방법을 통한 형질전환 닭 생산 연구의 가장 큰 관건은 최대한 많은 수의 공여체 원시생식세포가 수용체의 저해작용 없이 안정적으로 수용체 gonad로 이동하여 분화하는 것으로, 본 연구 결과를 토대로 개선된 방법을 이용하면 높은 효율의 생식선 키메라 닭이 생산될 것으로 사료된다.

Development of Sperm MTT Assay for Its Application in Boar Semen

  • The MTT assay is one of superior evaluation methods widely used to analyze the viability of metabolically active cell. It can be used to determine the percentage of viable sperm through measurement of the reduction of MTT granules at mitochondria in sperm tail. The purpose of this study is to determine the optimal condition of a simple and easy MTT assay to validate boar sperm viability and compare the accuracy of this test with microscopic examination. The MTT reduction rate for sperm viability were analyzed in microtiter plates (96 well) from 1 hr to 5 hr incubation periods at $37^{\circ}C$ using spectrophotometer (microplate reader) at 550 nm wavelength. The remainder of semen sample was simultaneously examined to compare the correlation of accuracy between MTT assay and other sperm parameters. Those sperm parameters were included the motility, survival rates, membrane integrity, mitochondria activity and acrosome integrity. The OD values of MTT assay (MTT reduction rates) did not greatly change at 1 hr to 5 hr incubation periods in different proportion of live and freeze-killed sperms (dead sperm). The MTT reduction rates or survival rates were decreased according to the different concentration of live and dead sperm. The linear regression at 1 hr and 4 hr incubation periods in sperm MTT assay was y=291.55x-72.176 and y= 180.64x-44.569, respectively. There are high correlation between 1 hr and 4 hr incubation periods (p<0.001). The results of MTT assay and other sperm parameters has a positive correlation (p<0.01 or 0.05). The correlation coefficients for MTT assay was 0.88115 for motility, 0.89868 for survival rates, 0.91722 for membrane integrity and 0.77372 for acrosome integrity, respectively. In conclusion, the MTT assay can be used as a reliable and efficient evaluation method for boar sperm viability. It can be use practical means to evaluate the quality of boar sperm by a fast, inexpensive and easy method.

향기요법에 사용하는 캐리어 오일과 에션셜 오일의 세포에 대한 독성 (A Cytotoxicity of Carrier Oil and Essential Oils on Cells by Using of Aromatherapy)

  • Essentail oils and carrier oils are generally used for Aromatherapy. Therefore the toxicity, possibilities of irritations and sensitive reactions and injury of essential oils must be considered for clients and therapists. So that, in this studies a toxicity of jojoba and 4 species essential oils (fennel, mandarine, tea tree and cedarwood) were investigated by the measurement of MTT-assay and sirius red staining. Liver, kidney and brain tell were chosen for the cell viability assay and observation of morphological change. In the result, no cytotoxicity was observed on live., kidney and brain cell at concentration of 0.01 $\mu\el/m\el$ jojoba oil. And lysis and nucleus breaking were not observed at same concentration of jojoba oil on live., kidney and brain cell. fennel oil was showed 50% of cell viability and inhibited cell growth on liver, kidney and brain cell at relatively high concentration compared with the other oils. 50% of liver, kidney and brain cell viability and delayed cell growth of tea tree and mandarine oil were revealed at lower concentration than fennel oil. In cedarwood oil, 50% of liver cell viability at concentration of 0.00067 $\mu\el/m\el$ was showed, but cell viability and cell growth of kidney and beam cell were effected at the lowest concentration compared with other oils. So that, jojoba oil as using of carrier oil may be not harmful. And 3 essential oils from the fennel, tea tree and mamdarine may have very low toxicity, but cedarwood may be used carefully for inhalation. And over dosage of concentrated cedarwood oil should be not directly touched and exposured, and absolute essential oils must be diluted with carrier oils for topical and systematic massage.

A Novel Switched-Capacitor Based High Step-Up DC/DC Converter for Renewable Energy System Applications

  • This paper presents a new high step-up dc/dc converter for renewable energy systems in which a high voltage gain is provided by using a coupled inductor. The operation of the proposed converter is based on a charging capacitor with a single power switch in its structure. A passive clamp circuit composed of capacitors and diodes is employed in the proposed converter for lowering the voltage stress on the power switch as well as increasing the voltage gain of the converter. Since the voltage stress is low in the provided topology, a switch with a small ON-state resistance can be used. As a result, the losses are decreased and the efficiency is increased. The operating principle and steady-states analyses are discussed in detail. To confirm the viability and accurate performance of the proposed high step-up dc-dc converter, several simulation and experimental results obtained through PSCAD/EMTDC software and a built prototype are provided.

Suppressive Effects of an Ishige okamurae extract on 3T3-L1 Preadipocyte Differentiation

  • The biological activity of tissue specific stem cell is under the control of their specific microenvironment and the exogenous chemicals derived from digestive tract can be one of the constructing factors of that. It is suggested that the extract of brown algae Ishige okamurae has antioxidant-, apoptosis induction-, and antiinflammatory-effects. On the other hand, a few studies have shown that antioxidant assist inhibition of accumulation of fat. So we studied the effect of the extract of I. okamura on the cellular activity and differentiation of 3T3-L1 preadipocyte to adipose cell. The viability of cell was analyzed using 3-[4,5-dimethylthiazo-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) assay. Adipogenesis of 3T3-L1 cell was analyzed after induction in the induction medium containing the I. okamurae extract. The cellular activity was high compared with the vehicle and 0.05 mM caffeine in all groups of I. okamurae extract treated cells. The extract of I. okamura inhibited accumulation of lipids in 10 and $50{\mu}g/ml$. The expression of the marker genes for adipocyte differentiation coincided with cytochemical results. These results suggest that the extract of I. okamurae increases the cellular viability of adipose precursor cells. On the other hand, it suppresses the differentiation of preadipocyte to adipocyte and accumulation of lipids in concentration-dependent manners. It may be possible that the major component of the extract can be applied in the control of adipose tissuegenesis.
