Enhancement of Anticancer Activities from Lithospermum erythrorhizon Extracts by Ultra High Pressure Process (초고압 가공 공정을 통한 지치 추출물의 항암 활성 증진)

  • This study was performed to enhance anticancer activities of Lithospermum erythrorhizon by eluting high amount of shikonin through ultra high pressure process. Extraction yield was increased up to 5~10% by ultra high pressure process, compare to the normal extraction processes such as water solvent extraction, 70% ethyl alcohol solvent extraction. The cytotoxicity of the extracts ($1.0{\mu}g/m{\ell}$) from ultra high pressure process was showed the lowest cytotoxicity 13.4% for human lung cell (HEL299). The anticancer activities showed 80~85% by adding $1.0{\mu}g/m{\ell}$ of the extracts from ultra high pressure process in several cancer cell lines such as AGS, Hep3B, MCF-7 and HeLa cells. Among them, MCF-7 cell of the endocrine system was highest inhibited than other cells. The anticancer activities of the extracts from ultra high pressure extraction process showed 10~15%, which was higher than the extracts from normal extraction processes. From HPLC analysis of the extracts, the contents of shikonin in the extracts from ultra high pressure process was 11.42% (w/w), which was 20% higher than others. This results indicate that ultra high pressure process could increase the extraction yield of shikonin and other contents, which resulted in higher anticancer activities.

  • Safflower seed extract was prepared by a high-pressure extraction technology and its quality characteristics were compared to that of other conventional extraction techniques, such ultrasonic and reflux extractions. Safflower seeds were extracted with 80% aqueous ethanol by three above extraction methods, and further fractionated with Diaion HP-20 column chromatography to obtain a partially purified safflower seed extract (PPSSE). Among the three extraction techniques examined, the reflux extraction showed the higher yields of EtOH extract and PPE than the ultrasonic and high-pressure extractions. Levels of most phenolic compounds in the EtOH extract of safflower seed are higher in reflux and ultrasonic extractions than the high pressure extraction, but levels of two serotonin aglycones, N-(p-coumaroyl)serotonin (CS) and N-feruloylserotonin (FS), in PPSSE were higher in the high pressure extraction than the reflux and ultrasonic extractions. In addition, color values (L and a) of the PPSSE were higher in the high-pressure extraction than the reflux and ultrasonic extractions, although there were no significant differences in pH and UV maxima absorption spectra among three extraction techniques. These results indicate that the high-pressure extraction technology is a simple and effective extraction for preparation of a high quality of safflower seed extract containing CS and FS with anti-wrinkle activity.

Enhancement of Antioxidant Activities of Curcuma longa Leaves by Ultra High Pressure Extraction (초고압 공정을 이용한 강황 잎 추출물의 항산화 활성 증진)

  • This study was designed to evaluate antioxidant activity of Curcuma longa L. leaves treated by ultra high pressure extraction. Curcuma longa L. leaves was subjected to 5,000 bar for 5 and 15 min at $25^{\circ}C$ The highest phenolics and flavonoids content was observed in the treatment of high pressure extraction for 15 min ($308.28{\mu}g/mg$, $124.33{\mu}g/mg$). The DPPH scavenging activity was 82.34% at $1.0mg/m{\ell}$ of Curcuma longa L. leaves treated by ultra high pressure process for 15 min. The highest SOD-like acitivity of Curcuma longa L. leaves ($1.0mg/m{\ell}$) was observed at ultra high pressure extraction for 15 min (67.54%). The high pressure extraction significantly increased the contents of phenolics and flavonoids and also enhanced the antioxidant activity. These results provide useful information for enhancing biological properties of Curcuma longa L. leaves.

Quality Characteristics of Citrus Fruit by Cyclic Low Pressure Drying and High Hydrostatic Pressure Extraction (초고압 처리에 의한 감귤의 추출률 및 특성변화)

  • We developed a method for improving the antioxidant activities of Citrus extracts through cyclic low pressure drying(CLPD) and a high hydrostatic pressure extraction (HPE) process. Citrus fruits were prepared for water extraction at $60^{\circ}C$ and 300 MPa for 5 min (high hydrostatic pressure extraction, HPE5) and 15 min (high hydrostatic pressure extraction, HPE15) after cyclic low pressure drying method. Extraction yields obtained by cyclic low pressure drying and high hydrostatic pressure extraction process were 20.41, 23.47, and 28.19%, respectively. Total polyphenol contents were increased by combined process. Generally, CLPD and HPE resulted in higher yields than the conventional extraction process. Further, HPE15 showed 48.21% DPPH radical scavenging activity (EDA, %) at $1,000{\mu}g/mL$. In general, antioxidant activities of Citrus increased by CLPD and HPE. Therfore, CLPD and HPE of Citrus resulted in higher antioxidant activity than conventional water extraction.

Effect of Immune Activity on Berberis koreana Palibin by Ultra High Pressure Low Temperature Process (초고압 저온처리에 의한 매자나무의 면역 활성)

  • This study was performed to investigate a methodology of improving immune activities of Berberis koreana Palibin by ultra high pressure on low temperature extraction process. Extraction yield was enhanced up to more than 35% by above process, compare to the control and extraction average. The cytotoxicity on human kidney cell (HEK293) was showed below 20.4%, 21.6% in adding 1.0 mg/ml of the highest concentration. Generally, the extracts by ultra high pressure extraction process showed less toxicity about 5% than the other processes. It could tell that toxic materials that existing in the plant could be reduced or broken by ultra high pressure process due to can be broken bond such the hydrogen bond, the electrostatic bond, the van der Waals bond, and the hydrophobic bond, can be broken by high pressure. The result could be employed to develop a new type of functional food from B. koreana Palibin by low temperature high pressure process.

Effects of Antioxidant Activities of Small Colored Potatoes (Solanum tuberosum L.) by using Ultra High Pressure Extraction Process (초고압 처리가 꼬마칼라감자의 항산화 증진에 미치는 영향)

  • We investigated a method to improve antioxidant activities of colored potato extracts by ultra high pressure extraction process. The colored potato was extracted by water at $60^{\circ}C$(WE) and 300 MPa for 15 min (High Pressure Extraction, $HPE_{15}$) and 30 min (High Pressure Extraction, $HPE_{30}$). The extractions yielded by different extraction processes were 1.73(WE), 2.10($HPE_{15}$), and 2.41($HPE_{30}$)%. Total phenolic acid contents of different extraction processes were estimated as 48.21(WE), 50.20($HPE_{15}$) and 51.34($HPE_{30}$) GAL mg/g, respectively. The flavonoids contents of different extraction processes were measured as 13.12(WE), 14.35($HPE_{15}$) and 15.17($HPE_{30}$) RE mg/g, respectively. Generally, for the contents of phenolic acid and flavonoids, the samples from HPE were higher than those from conventional extraction process. $HPE_{30}$ showed 76.21% of DPPH radical scavenging activity (EDA, %) in 1,000 ug/mL. The reducing power of $HPE_{30}$ also showed the high activity as 0.42. In generally, antioxidant activities of colored potato were increased by high pressure extraction process. We could tell that the HPE extracts of colored potato had a higher antioxidant activity than those from conventional water extraction. The results of HPE showed obvious advantages in higher efficiency, shorter extraction time.

Anticancer Activity of Acer mono Wood Extracted by Ultra High Pressure Extraction Process (초고압 추출 공정을 통한 고로쇠 목부 추출물의 항암활성 증진)

  • We investigated a method to improve anticancer activities of Acer mono wood extracts by ultra high pressure extraction process. The A. mono was extracted by water at $40^{\circ}C$ and 300 MPa for 15 min (High Pressure Extraction, HPE). The extraction yield by ultra high pressure extraction process was 5.42%. The cytotoxicity on human normal lung cell (HEL299) of the extracts from HPE showed 21.54% lower than that from conventional water extraction at $100^{\circ}C$ in adding the maximum concentration of 1.0 mg/$m{\ell}$. Ultra high pressure extracts process for 15 minutes extracts (HPE15) showed more potent scavenging effect than the control, BHA. On SOD-like test, the HPE15 showed highest activity as 32.4% at 1.0 mg/$m{\ell}$ concentration. Human stomach adenocarcinoma, liver adenocarcinoma, breast adenocarcinoma and lung adenocarcinoma cell growth were inhibited up to about 67~79%, in adding 1.0 mg/$m{\ell}$ of extracts from HPE. HPE was 20~25% higher than conventional water extraction. It was interesting that, among several cancer cell lines (stomach adenocarcinoma, liver adenocarcinoma), the growth of digestive related cancer cells were most effectively inhibited as about 75~79%. On in vivo experiment using ICR mice, the variation of body weight of mice group treated A. mono wood extracts from HPE of 100 mg/kg/day concentration was very lower than control and other group. The survival times of group treated this extracts was 61.96% longer than that of the control group and this extracts showed the lower tumor weight, which were 10.49 g than positive control as 16.17 g. Based on these results, we could tell that the HPE wood extracts of A. mono had higher anticancer activity than conventional water extraction. The results of HPE showed obvious advantages in higher efficiency, shorter extraction time, at lower energy costs.

Enhancement of Skin Immune Activities of Curcuma longa L. Leaf Extract by Ultra High Pressure Process (초고압 공정을 이용한 강황 잎 추출물의 피부 면역 활성 증진)

  • Curcuma longa L. leaf was extracted by water at $60^{\circ}C$ for 12 hours after being treatment of Ultra High Pressure under 500 MPa for 5-15 minute. The high pressure extraction for 15 minute (HPE15) was measured the highest extraction yield of 25.48% (w/w), compared to those from conventional extraction methods. The HPE15 showed the lowest cytotoxicity as 11.97% in adding $1.0mg/m{\ell}$ of concentration. Also, HPE15 was measured the highest inhibition of hyaluronidase as 44.48% in adding $1.0mg/m{\ell}$. In addition, The production of NO from macrophages was measured as $7.06{\mu}M$ in adding $1.0mg/m{\ell}$ of HPE15, which was lower than the those from others processes. Finally, HPE15 significantly reduced up to $649.44pg/m{\ell}$ of $ProstaglandinE_2$ production from UV-irradiation. These results suggest that the Curcuma longa Linne leaf extract from high pressure process might enhance the skin immune activities possibly by high elution of active components than other processes.

Comparison of Biological Activities of Acer mono and A. okamotoanum Extracts by Water Extracton and Low Temperature High Pressure Extraction (고로쇠와 우산고로쇠 저온고압 수용성 추출물의 유용생리활성 비교)

  • Hepatoprotective and antioxidant activities of Acer mono and A. okamotoanum were compared after being extracted by low temperature and high pressure (LTHP) at 20 MPa and $60^{\circ}C$ for 15 minutes. Extraction yield of both A. mono and A. okamotoanum was increased about 40~43% by this extraction process. On scavenging activities, the bark of A. okamotoanum from this extraction process showed the highest activity as about 97%. This value was higher than that from conventional water extraction and A. mono extracts. Both of A. mono and A. okamotoanum showed high ability on nitrite scavenging, but decreasing tendency according to increasing of pH. On SOD-like test, A. okamotoanum had the highest activity as 46.28% at $1.0\;mg/m{\ell}$ concentration. A. okamotoanum extracted by LTHP also showed the highest activity as 197.38% in adding $1.0\;mg/m{\ell}$ concentration. Generally, the extracts from low temperature and high pressure extraction process are higher hepatoprotective and antioxidant activities than that from conventional water extraction. It can conclude that the bark of A. okamotoanum has better biological activities than other parts of A. mono.

Enhancement of Skin Immune Activities of Spirulina maxima by High Pressure Extraction Process (Spirulina maxima 초고압 추출물의 피부 면역 활성 증진)

  • A marine alga, Spirulina maxima, was extracted under high pressure and low temperature conditions at 500 MPa and $60^{\circ}C$ for 5 and 10 min. A high pressure of 500 MPa was applied to improve process yields because of low temperature extraction. This method resulted in highest higher extraction yield of 26.1% (w/w) in comparison to those results obtained from conventional extraction methods which produced a yield of 17.6% (w/w) from water. The extracts from this process also showed 19% of low cytotoxicity against human normal fibroblast cells in adding 1.0 mg/ml of the highest concentration. The crude extract significantly reduced the production of Prostaglandin $E_2$ ($PGE_2$) from CCD-986sk cells and increased nitric oxide production by macrophages. These higher activities of enhancing skin immune functions were found to have high antioxidant extract properties, like a 98% increase in DPPH radical scavenging activity. The extracts from the high pressure process showed a higher elution of active components than other processes and generated new compounds based on HPLC analysis. This clearly indicates that the extracts from high pressure and low temperature conditions have higher skin immune activation properties that have not been previously reported.
