Linkage Structure Analysis of Barley and Oat $\beta$-Glucans by High Performance Anion Exchange Chromatography

  • Cereal $\beta$-glucans, linked essentially by mixed $\beta$-(1,4/1,3) glycosidic bonds, were extracted, purified, and structurally identified. Previously chemical structure of barley $\beta$-glucans was characterized from 3 varieties of 'Gang', 'Ohl', and 'Gwangan', and the (1,4)/(1,3) linkage ratio of the $\beta$-glucans was identical. In this study, $\beta$-glucans from 1 barley ('Chal') and 3 oat ('Ohl', 'Samhan', and 'Donghan') varieties were structurally scrutinized, and the linkage pattern of total 7 cereal $\beta$-glucans was compared. The amount of 2 major 3-O-$\beta$-cellobiosyl-D-glucose (DP3) and 3-O-$\beta$-cellotriosyl-D-glucose (DP4) from barley and oat accounted for only 66.6-73.3 and 68.12-81.89% of water-extractable $\beta$-glucan fractions, and the (1,4)/(1,3) linkage ratios of both barley and oat $\beta$-glucans were within very narrow range of 2.27-2.31 and 2.38-2.39, respectively, among the cultivars tested. Structural difference in the cereal $\beta$-glucans was evident when DP3:DP4 ratio in the $\beta$-glucan structure was compared. As a result, this ratio was significantly greater for barley $\beta$-glucan (2.26-2.74) than for oat (1.54-1.66). Chal-B had the greatest DP3 to DP4 ratio among the samples, which in turn reflected the least amount of (1,4)-linkages.

  • This experiment was carried out to investigate the effects of the different nitrogen rates and cutting heights on the dead stubble after cutting, and the carbohydrate reserved in stubble of sorghum-sudangrass hybrid (Sorghum bicolor (L.) Moench) Pioneer 988. The experiment was undertaken in the Experimental Livestock Farm of Agriculture Coll., Seoul Nat'l Univ. In Suweon, 1981 and 1982. The results obtained are summarized as follows: 1. The dead stubble after cutting was found to be great with high rate of nitrogen fertilizer and low cutting height. It was also learned that the rainfall during forage cutting period made the dead stubble increased. 2. A significant higher (p<0.05) carbohydrate reserves in the stubble was observed in the high stubble height at the first cutting time and 6th through 7th day after the first cutting. The results indicate that the high stubble height reserves more carbohydrate for the early regrwoth stage after the first cutting when comparing with the low stubble. 3. The content of carbohydrate reserves was influenced by climete. Drought is caused to high content of carbohydrate, whereas, rain is caused to low content of that. The critical soluble carbohydrate content causing the death of stubble supposed to be 3 to 6% at least. 4. It is suggest that carbohydrate reserves in plant do not play a distinctive role for the regrowth in a summer annual forage like sorghum-sudangrass hybrid, but it might be rather influenced by the other factors, for example, environmental conditions at harvest and new bud.

Effects of Level of Dietary Protein or Carbohydrate on the Economic Characters and Amylase Activity of the Silkworm, Bombyx mori (인공사료의 단백질 및 탄수화물 수준이 가잠의 실용형질과 Amylase활성에 미치는 영향)

  • A series of experiment was conducted to investigate the effects of the level of dietary Protein (soybean meal) and carbohydrate (sucrose) on the growth rate, feed efficiency, nutrients digestibility and amylase activity of larvae at 5th instar. The results obtained from the experimentation are summarized as follow: (1) It was found that the body weight gain, cocoon quality and feed efficiency of the 5th instar larvae were significantly affected by the level of protein and carbohydrate in the artificial diet. Present data revealed that the proper level of dietary protein and carbohydrate would be the most important factor for the optimum growth of larvae. (2) In the amount of diet consumption, the group of larvae gained more body weight consumed more diet than the group gained less amount of body weight. (3) Digestibility of nutrients was improved as the level of protein in the diet increased, although the level of dietary carbohydrate did not exert any positive results. (4) It was observed that the amylase activity in digestive juice was stimulated by the increasement of dietary carbohydrate. However, no correlation between the level of dietary protein and carbohydrate was detected in the amylase activity. (5) Analytical data indicated that the amount of protein or carbohydrate excreted through feces and urine was proportional to the content of protein or carbohydrate in artificial diet. The group o( larvae consumed the diet contained high protein excreted more protein and less nitrogen free extract than did the group received low protein diet. Likewise, the group of larvae fed high dietary carbohydrate excreted more nitrogen free extract and less protein through excreta than the group consumed low carbohydrate diet group. (6) Although the amylase activity of the larvae received the artificial diet was higher than the larvae received natural diet (mulberry leaf), the rate of body weight gam and the quality of cocoon produced from the natural diet group was better than the artificial diet group. It is, thus, concluded that feeding the larvae by the artificial diet may net be recommendable. (7) It was determined that the best level of protein (soybean meal) and carbohydrate (sucrose) in artificial diet was found to be 40g and 0g, respectively when it was fed to the 5th instar larvae. (8) It may be concluded from the results obtained that the artificial diet that could stimulate the activity of amylase nay not be recommendable for the practical larvae feeding Purpose due simply to the Poor economic return from this diet than natural diet.

  • Dietary protein is the most expensive component in flounder feed because of the high protein requirement of this species. The non-protein energy sources may also influence the protein utilization of fish. The inclusion of adequate levels of non-protein energy sources in diets can minimize use of protein as an energy source. Protein sparing effect by fat and carbohydrate has been studied in other fish. Production of Hydrolyzed Red Ginseng Residue and Its Application to Lactic Acid Bacteria Cultivation

  • Enzymatic treatment conditions for red ginseng residue (RGR) were investigated to apply RGR as a microbial medium. Polysaccharide hydrolyase and protease were screened to obtain high solid and carbohydrate yields, and a good degree of carbohydrate hydrolysis. The optimal dosage and reaction time for Viscozyme, the chosen polysaccharide hydrolyase, were found to be 1.0% (w/w) and 3 h, respectively. Of the tested proteases, Flavourzyme, whose optimal dosage was 0.5% (w/w), was selected. Co-treatment with the optimal dosages of Flavourzyme and Viscozyme increased solid yield, carbohydrate yield, and degree of carbohydrate hydrolysis by 76%, 65%, and 1,865%, respectively, over levels in non-treated RGR. The culture characteristics of Leuconostoc mesenteroides strain KACC 91459P grown in enzymatically hydrolyzed red ginseng residue (ERGR) and RGR suspensions were compared. After cultivation for 6 h, the viable cell counts of both cell suspensions rapidly increased to $1.3{\times}10^9$ colony-forming units (CFU)/g. Moreover, while the viable cell population drastically decreased to $2.4{\times}10^6\;CFU/g$ for cells grown in RGR medium, it was maintained in cells fermented in ERGR medium for 24 h.

A Study on Food and Nutrient Intakes of Weekday and Weekend Among High School Boys in Seoul (서울 일부지역 남자 고등학생의 주중과 주말의 식품 및 영양소 섭취에 관한 연구)

  • This study was conducted to examine food and nutrient intakes of weekday and weekend with 329 high school boys residing in Seoul using the 3-day food record. Frequency for breakfast skipping was the highest as compared with lunch and dinner skipping. Frequencies for breakfast and lunch skipping were significantly higher in weekend than weekday (p<0.05). The daily average food, vegetable food and animal food intakes were 1,327.6 g, 800.5 g, and 425.8 g, respectively. Total food and vegetable food intakes of weekday were significantly higher than weekend(p<0.05). The dietary variety score(DVS) was 20.5 in daily average, 23.3 in weekday and 15.1 in weekend, which showed significantly higher in weekday than weekend(p<0.05). The daily averages of energy, protein, fat and carbohydrate intake were 2244.9 kcal, 89 g, 72.6 g, and 311.2 g, respectively. Fat intake was significantly lower and carbohydrate, fiber, phosphate, iron, sodium, potassium, vitamin A, niacin, folate, and vitamin C intakes were significantly higher in weekday than weekend (p<0.05). The percentages of energy intake from carbohydrate, protein, and fat were 55.4%, 15.8%, 28.8% in daily average, 56.8%, 15.8%, 27.4% in weekday and 53.6%, 15.8%, 30.7% in weekend, respectively. The percentages of energy intake from carbohydrate in weekday and weekend were below 60%, and that from fat was above 27% in weekday and weekend. Carbohydrate intake was significantly higher and fat intake was significantly lower in weekday than weekend(p<0.05). Energy intakes of daily average, weekday and weekend were above 83% as compared with estimated energy requirement(EER). Intakes of dietary fiber, calcium, potassium, vitamin C, riboflavin and folate were below 75% as compared with adequate intake(AI) or recommended intake(RI). Mean adequacy ratios(MAR), an index of overall dietary quality were 0.78 in daily average, 0.80 in weekday and 0.75 in weekend. MAR of weekend showed significantly lower than weekday(p<0.05). This study revealed that the overall nutrient intake status was worse in weekend than weekday among high school boys.

To develop the classification method of Agricultural by-productions for biogas production

  • The objective of this study was to develop the classification method of various organic wastes. Specifically, the effects of proximate composition on the biogas production and degradation rates of agricultural by-production was investigated and a new standards for mixture of various organic wastes based on proximate composition combination was developed. Agricultural by-products (ABPs) with medium total carbohydrate, medium crude protein and low fat contents demonstrated the single step digestion process. ABPs with low total carbohydrate, high crude protein and high fat contents demonstrated the two step digestion process of Diauxic growth. The single ABP (Class No. 15) and the mixed ABPs (Class No. 12+18, 6+12+22, 9+12+18) after 10days showed the similar biogas yield pattern. We can use the classification method for the more ABPs and organic wastes from factory and municipal waste treatment plant for the high efficient biogas production.

Quantitation of fructo- and inulo-oligosaccharides by high performance liquid chromatography (High performance liquid chromatography에 의한 fructo 및 inulo올리고당의 정량)

  • High performance liquid chromatographic method using a TSK-gel amide 80 column and isocratic elution with acetonitrile-water (63 :35 ;v/v) mixture was used for the separation and the quantitation of fructo (GF2-GF7)- and inulo-oligosaccharides (F2-F4). Retention time of each standard carbohydrate was highly reproducible. Standardization curves obtained by plotting the peak areas against the amounts of each carbohydrate showed very high coefficient of determination$({\ge}0.9884)$ and similar slopes, and a wide range of y-intercepts. Our results suggest the use of each Pure oligosaccharide for its own standardization curve instead of using a certain carbohydrate as an internal standard.

  • BACKGROUND/OBJECTIVES: The prevalence of nonalcoholic fatty liver disease (NAFLD) has increased worldwide in parallel with overnutrition characterized by high-fat and high-carbohydrate intake. Our objective was to establish, in 16 weeks, a model of NAFLD in Wistar pathogen-free rats following four dietary types. MATERIALS/METHODS: Forty (6 weeks old) healthy Wistar male rats, weighing an average of 150 g were randomly divided into four groups of ten and assigned a diet with the same quantity (15 g/rat/day), but with different composition. The moderate-fat (MF) group was fed a moderate-fat diet (31.5% fat and 50% carbohydrates), the high-fat (HF) group was fed a fat-rich diet (51% fat), the high-sucrose (HS) group and the high-fructose (HFr) group were fed a carbohydrate-rich diet (61%). The carbohydrate contents of the HS group was composed of 60.3% sucrose while that of the HFr group was composed of 59.3% fructose. RESULTS: At week 16, the HF group had the highest percentage of cells enriched in fat (40%) and the highest weight and liver weight (P < 0.05). The HFr group showed significantly higher levels of serum triglycerides, alanine aminotransferase and adiponectin at week 16 as compared to week 1 (P < 0.05). CONCLUSIONS: The 15 g/rat/day diet composed of 51% fat or 61% carbohydrates enriched mainly in fructose may induce characteristics of NAFLD in rats.

Seasonal Changes of Biochemical Composition in Cultured Bivalves (양식패류의 생화학적 체성분의 계절적 변화)

  • Seasonal changes in biochemical composition of Crassostrea giggas and Scapharca broughtonii in the Kosong bay and Kangjin bay, southern Korea were studied over 12 months in relation to their habitat. In Crassostrea giggas, protein was high during summer in all experimental station, but lipid and carbohydrate were high during winter, and no clear difference was between experimental station. Glycogen content was sharply decreased during summer, and energy content was low but carbohydrate was high during summer. In Scapharca broughtonii, protein was low, but carbohydrate was high during summer. Lipid was high from February to March, glycogen content was low from June to September. Energy content was low during winter and no clear difference biochemical composition was between experimental station of Scapharca broughtonii.
