Botanical Characteristics and Effect of Weed Control by Herbicide in Barley (제초제 처리에 의한 보리의 작물학적 특성)

  • This study was carried out to find the botanical characters and effect of weed control for herbicides treatment after barley sowing. Barley variety used in study was "Yuyeon", hood-type awn for whole crop forage use, which developed at RDA in 2004. Sowing date was on 10th October, 2007, and was drilled as $60{\times}20cm$. Herbicides were treated on 2th day after barley sowing. Measured items were number and kinds of weeds at each treatment and growth characteristics and grain yield in barley. Cerastium holosteoides var. hallaisanense among weeds surveyed at tillering stages in Butachlor treatment were highly appeared, but Alopecurus aequalis var. amurensis did not appeared while Alopecurus aequalis var. amurensis was 39 plants and Capeslla bursa-pastoris was 37 plants in check. Occurrence of the Capeslla bursa-pastoris at primary survey and Cerastium holosteoides var. hallaisanense in ripening stage were high, while Alopecurus aequalis var. amurensis did not emerged. Botanical characteristics of barley each as plant height, number of tillers and spiklets per plant. surveyed at ripening stage were higher than check. Grain yield per 10a was the highest at Alachlor among treatments, while dry matter of leaf and culm were similar among treatments. Therefore, effect of yield increasing of barley by herbicide after sowing was reconfirmed through this study. Especially, effect of Alachlor was lightly high compared with others.

  • For weeding on the after-barley cropping soybean, the tank-mix use of postemergence herbicides acifluorfen and haloxyfop-methyl was tested. Acifluorfen has shown the transient leaf-burn on the newly formed soybean leaves, but no growth inhibitions. Under the treatment of acifluorfen only, grassy weeds occurred successively and explosively. However, under the combinated spraying of both herbicides, the grassy weeds could be controlled in a slight antagonistic, the broadleaved weeds in additive, and the total weed species including sedges in synergic tendencies, respectively. Also, to achieve the full-season weeding efficacies for the prominent crop yields, both herbicides are suggested to be mixed with more dosages than 0.245 kg ai/ha of acifluorfen and 0.15 kg ae/ha of haloxyfop-methyl, respectively.

  • Recent development in Japan on jumbo formulation of pesticides for paddy application by hand throw was reviewed. In addition, the diffusion of liquid formulations in water was examined to establish the research strategy for the jumbo formulation. Research on jumbo formulation in Japan has been focused on gas-generating formulations and self-emulsifiable formulations. Although continuous efforts to minimize the problem of pesticide deposit on the treated site have been made for the gas-generating formulations, much work is still needed to establish a generally acceptable formulation method and to commercialize a herbicide formulation. The self-emulsifiable jumbo formulations have recently been investigated. These formulations could simply be processed and showed relatively high biological efficacy. The emulsifiable concentrate was more diffusible than the suspension concentrate in water. The diffusion of the emulsifiable concentrate was not greatly disturbed by floating obstacles. And the diffusion rate was high when the specific grabity was lower than one.

  • In order to establish labor-saving culture technology in Tartary buckwheat, three cultural practices: hand planting, drill sowing machine and soil cover direct seeding machine, were compared.The highest grain yield was found in soil cover direct seeding plot with a value of 3.4 g per plant. As a result, grain yield may be estimated to be 113kg in soil cover direct seeding and 80kg in hand scattering. In addition, for the weed control, three herbicide treatments: single use of Alachlor, mixture and combination of Alachlor and Paraquat dichloride were conducted. the mixture showed over 90% weed control value, and the highest grain yield was found in the combination treatment. Combine machine was effective to reduce the ratio of grain loss and working hour by enhancing the working efficiency to 15${\sim}$20min/10a.

Functional Characterization of aroA from Rhizobium leguminosarum with Significant Glyphosate Tolerance in Transgenic Arabidopsis

  • Glyphosate is the active component of the top-selling herbicide, the phytotoxicity of which is due to its inhibition of the shikimic acid pathway. 5-Enolpyruvylshikimate-3-phosphate synthase (EPSPS) is a key enzyme in the shikimic acid pathway. Glyphosate tolerance in plants can be achieved by the expression of a glyphosate-insensitive aroA gene (EPSPS). In this study, we used a PCR-based two-step DNA synthesis method to synthesize a new aroA gene ($aroA_{R.\;leguminosarum}$) from Rhizobium leguminosarum. In vitro glyphosate sensitivity assays showed that $aroA_{R.\;leguminosarum}$ is glyphosate tolerant. The new gene was then expressed in E. coli and key kinetic values of the purified enzyme were determined. Furthermore, we transformed the aroA gene into Arabidopsis thaliana by the floral dip method. Transgenic Arabidopsis with the $aroA_{R.\;leguminosarum}$ gene was obtained to prove its potential use in developing glyphosate-resistant crops.

Cultivation Environment in Relation to Good Agricultural Practices in the Major Cultivation Area of Disocorea batatas (우리나라 주요 산약 재배지 GAP와 관련된 재배환경 실태)

  • This study was carried out to investigate the quality of soil, irrigation water, and status of agrochemicals application in relation to good agriculture practices system in the major cultivation area of Disocorea batatas. The concentrations of heavy metals as Cd, Pb, Cu, and Zn in soils and irrigation waters were lower than those of standard level for Environmental Conservation Act of Soil and Water in Korea. The dominant weed have been appeared Digitaria sanguinalis and Portulaca oleraceamite. The dominant insect pests and plant pathogens have been appeared aphid and anthracnose. Average yearly application of pesticide was 2 to 4 times for herbicide and 4 to 6 times for plant pathogens and insect pests. In order to safety production of medicinal crops could be achieved by proper cultivation management such as minimum application of agro-chemicals, effective use of by-product fertilizer, and technology development of organic farming.

  • Allelopathic compounds as naturally occurring herbicide have originally reported from local vegetation since B.C. 300. These compounds are known as secondary plant metabolites which released from plants into the environment often attract or repel, nourish or poison other organisms. In recent, many natural plant allelochemicals be used to attempt to biologically or ecologically control weed among worldwide weed scientists. Some allelochemicals have also used as fungicides, insecticides, and nematodicides, and were less than man-made agrochemicals to damage the global ecosystem. It makes efficient use of resources internal to the farm, relies on a minimum of purchased inputs. Some scientists selected for allelopathic activity when breeding weed-controlling cultivars of rice, sorghum, cucumber, surflower etc. Thus, this paper is focused on allelopathic compounds isolated from cultivated crop with the high potential of prospective herbicides. The most environmentally acceptable and sustainable approach to utilization of allelopathy for weed control is to develop plant cultivars with proven allelopathic characteristics. In rice accessions, there are 60 cultivars/lines which have known as allelopathic activity and some of these cultivars control weed more less 90% within certain radius of activity. These accessions are originated from 15 countries including Korea, Japan, USA, India, Philippines, Indonesia, Laos, Taiwan, Afghanistan, Mali, Pakistan, Colombia, Egypt, China, and Dom. Rep. From these cultivars, the most common allelopathic compounds identified in rice are p-Hydroxybenzoic, Vanillic, p-Coumaric, and Ferulic acids. In addition, allelopathic lines of the following crop have shown inhibition of weed growth : beet (Beta vulgaris), lupin(Lupinus spp.), com(Zea mays), Wheat(Triticum aestivum), oats(Avena spp.) peas(Pisum sativum), barley(Hordeum vulgare), rye(Secale cereale), and cucumber(Cucumis sativus). Thus, future allelopathy research must be designed its potentially phytotoxic propertices and the ecotoxic features of the allelochemicals from release to degradation ; its ecological sustainability, its allelopathic effect in early growth. stages, and selectivity properties in combination with chemical stages, and selectivity properties in combination with chemical concentrations. Also, research approach in allelopathy might be screened for highly allelopathic germplasm collection of crops, the idea being to ultimately transfer this agronomic character into improved cultivars by either conventional breeding or other genetic transfer techniques.

Influence of insect pollinators on gene transfer from GM to non-GM soybeans (GM 콩의 도입유전자 이동에 미치는 화분 매개충의 영향)

  • The cultivation area and use of genetically modified (GM) crops have been increased continuously over the world and concerns about the potential risks of GM crops are also increasing. One of the major concern in risk assessment is the possible development of hybrids through interspecific and intergeneric crosses with related species. This study was conducted to investigate the pollinator have an influence on insect-mediated gene transfer from GM soybeans. Hybrid was induced from GM soybeans by honeybee and western flower thrips, and non-GM soybeans were used as pollen receptor. The analysis for gene-flow was conducted by herbicide selection, immunostrip test, and PCR analysis. In the result of the analysis, three hybrids were detected on the distance 15, 75, 105 cm from pollen source in western flower thrips treatment. In honeybee treatment, one hybrid was detected in the farthest distance (300 cm). These results suggested honeybee and western flower thrips have a possibility they can transfer the introduced gene from GM soybeans to non-GM soybeans.

Expression in Escherichia coli of a Putative Human Acetohydroxyacid Synthase

  • A human gene has been reported that may encode the enzyme acetohydroxyacid synthase. Previously this enzyme was thought to be absent from animals although it is present in plants and many microorganisms. In plants, this enzyme is the target of a number of commercial herbicides and the use of these compounds may need to be reassessed if the human enzyme exists and proves to be susceptible to inhibition. Here we report the construction of several plasmid vectors containing the cDNA sequence for this protein, and their expression in Escherichia coli. High levels of expression were observed, but most of the protein proved to be insoluble. The small amounts of soluble protein contained little or no acetohydroxyacid synthase activity. Attempts to refold the insoluble protein were successful insofar as the protein became soluble. However, the refolded protein did not gain any acetohydroxyacid synthase activity. In vivo complementation tests of an E. coli mutant produced no evidence that the protein is active. Incorrect folding, or the lack of another subunit, may explain the data but we favor the interpretation that this gene does not encode an acetohydroxyacid synthase.

Effect of Crop Establishment and Weed Control Method on Productivity of Transplanted aman Rice

  • This experiment was conducted to find the most suitable crop establishment method and weed management practices for transplanted aman rice in Bangladesh. Rice variety Bangladesh Rice Research Institute (BRRI) dhan44 was used as planting materials where three crop establishment methods ($T_1$: direct wet seeding by drum seeder; $T_2$: hand broadcasting; $T_3$: transplanting) and four weeding options ($W_1$: Hand weeding (HW); $W_2$: BRRI weeder+HW; $W_3$: Herbicide+HW; and $W_4$: no weeding) were tested. Among the crop establishment methods, the highest grain yield ($5.12t\;ha^{-1}$) was obtained with the $T_3$, while the highest benefit cost ratio (BCR) of 2.08 was found in $T_2$. In case of the weed management method, $W_1$ showed superior results on the plant $density/m^2$ (139.66) at 60 days after transplanting (DAT), grain yield ($4.97t\;ha^{-1}$), and BCR (2.03). On the other hand, the highest plant dry matter (36.20 g) at 60 DAT and the highest yield ($6.10t\;ha^{-1}$) were obtained in a $T_3W_1$ combination. The results of this study show that the productivity of rice during aman season could be most significantly increased with the use of transplanting ($T_3$) alone, hand weeding ($W_1$) alone, or a combination of the two methods ($T_3W_1$).
