Action Characteristics of EK3143 and EK3150, New Cyclohexanedione Herbicides, and Their Effects on Acetyl CoA Carboxylase Activity (신규 cyclohexanedione계 화합물 EK3143과 EK3150의 제초작용 발현특성과 Acetyl CoA Carboxylase 활성에 미치는 효과)

  • This study was carried out to investigate the herbicidal action characteristics of EK3143 and EK3150, newly synthesized cyclohexanedione analogues, and their inhibitory effects on the acetyl-CoA carboxylase activity isolated from various plants. Sethoxydim, a typical cyclohexanedione herbicide, had a high herbicidal activity against only grasses with growth inhibition, leaf chlorosis and necrosis. EK3143 and EK3150 had a lower effect against grasses but a higher effect against broadleaf plants such as black nightshade and velvetleaf than sethoxydim had. Low rate application of EK3143 induced leaf bleaching(whitening), while high rate application induced the same symptoms as those of sethoxydim. EK3150 induced more significant whitening, additionally accompanying a curling of meristem part in broadleaf plants. A consistant tendency was observed between the growth inhibition against grasses and the senstivity of grass ACCase to these compounds, but not between herbicidal activities against broadleaf plants and the senstivity of dicotyledonous ACCase; i.e. EK3150, which had higher herbicidal activity against black nightshade and velvetleaf than EK3143 had in greenhouse, rather showed lower ACCase inhibition. In EK3150-treated barnyardgrass, growth inhibition occurred prior to decrease of photosynthetic pigments, in contrast to that of norflurazon and EK3140. These results suggest that EK3143 and EK3150 may have two target sites, ACCase and an unknown site on fatty acid synthesis related to growth inhibition and bleaching respectively, and their symptoms would be appeared in different degree by a differential binding affinity between two action sites.

Herbicidal activity of a new benzenesulfonylurea compound, KSC-13906 (신규 benzenesulfonylurea 계 화합물 KSC-13906의 제초활성)

  • Herbicidal activities of newly synthesized experimental compound, KSC-13906 [Erythro N-{(4,6-dimethoxy pyrimidin-2-yl)aminocarbgnyl}-2-(2-fluoro-1- hydroxy-n-propyl)benzenesulfonamide, US Patent 5,461,025], were investigated in greenhouse and field for development as a paddy herbicide. KSC-13906 exihibited excellent weeding effects and selectivity to rice under submerged paddy condition in geenhouse. At the application rate of $2.5{\sim}5$ g ai/ha treated on surface of water, KSC-13906 effectively controlled three annual weeds such as Echinochloa crus-galli var. oryzicola. Scirpus juncoides, and Monochoria vaginalis as well as five perennial weeds such as Cyperus serotinus, Sagittaria pygmaea, Eleocharis kuroguwai, Potamogeton distinctus, and S. trifolia. However, there was no effect on Aneilema keisak and Lindernia procumbens. Especially, KSC-13906 effectively controlled Echinochloa crus-galli var. oryzicola at growth stages ranging from pre-emergence to 3-leaf stage. Antagonistic interaction was found between pyrazosulfuron-ethyl and mefenacet, but not between KSC-13906 ana mefenacet. KSC-13906 had high selectivity to transplanted rice, but caused phytotoxic effect on direct-seeded rice when treated before 3-leaf stage. Herbicidal activity of KSC-13906 on the Echinochloa crus-galli var. oryzicola was 7-folds greater than that of pyrazosulfuron-ethyl in terms of relative dose ratio of the herbicides. These results suggest that KSC-13906 possesses selective herbicidal potency in rice.

  • Field study was conducted to evaluate safening effect of fenclorim (4,6-dichloro-2-phenyl pyrimidine) against benthiocarb, butachlor, and pretilachlor injury in rice plants under vinyl-covered subirrigated seedbed. Combinated application of benthiocarb 210, and 315 g ai/10a with fenclorim 10 g ai/10a or higher concentration, butachlor 180, and 270 g ai/10a with fenclorim 20 g ai/10a or higher concentration, and pretilachlor 60, and 90 g ai/10a with fenclorim 30 g ai/10a reduced herbicide injury of rice, and thus increased standing, plant height, tillers, and dry weight of rice plants compared to no combined application of fenclorim.

  • The experiments were conducted to evaluate safening effect of fenclorim (CGA 123'407, 4,6-dichloro-2-phenyl pyrimidine) against injuries of pretilachlor, butachlor, and benthiocarb in direct-seeded rice under flooded conditions. Pretilachlor and butachlor and benthiocarb reduced crop standing and inhibited growth of rice plants. However, premixture application of pretilachlor, butachlor or benthiocarb with fenclorim at 10-30 g ai/10a greatly reduced rice injury caused by the application of each herbicide alone and thus increased crop standing plant height and dry weight of rice plants compared to herbicide treatments alone.

Effects of Spring-sown Rye Living Mulch on Weed Suppression and Soybean Production (춘파호밀 리빙멀치가 잡초억제 및 콩 생산성에 미치는 영향)

  • Establishment of mechanized soybean production system without herbicide is required for organic soybean production in recent. Effects of mechanized soybean planting with partial tillage between growing rye living mulch, which was sown in April or early May, on weed suppression, soybean early growth and yield were investigated with different rye sowing dates and locations in 2007 and 2008, respectively. Effect of rye living mulch on weed suppression at 40~50 days after soybean planting was high as much as 70~90% regardless of rye sowing dates. In particular, a strong suppression effect on the occurrence and growth of Echinochloa crus-galli which occupied 60~80% of total weed biomass was observed. Rye living mulch sown on April 9, of which biomass was more than that sown on April 27, inhibited soybean early growth severely and reduced soybean grain yield in 2007. However, soybean grain yield was no t reduced despite suppression of early growth of soybean plant by living mulch sown during from late April to early May compared with conventional planting. Rye sowing before late April was not proper considering tendency of weed occurrence in spring because of unfavorable changes in weed suppression, early growth and grain yield of soybean according to amount of living mulch.

Effect of 1,8-NA and CGA 123'407 on Reducing Herbicide Injury of Rice Plants (1, 8-NA와 CGA 123'407 처리(處理)가 수도용(水稻用) 제초제(除草劑)의 약해경감(藥害輕減)에 미치는 영향(影響))

  • Since herbicide damage has been occurred in subirrigated rice seedbed in Korea, the use of crop safener is required to reduce crop injury. Field studies were conducted to evaluate safening effect of 1,8-naphthalic anhydride (NA) and CGA 123'407 (4, 6-dichloro-2-phenyl-pyrimidine) against four kinds of paddy herbicides applied to subirrigated vinyl-covered rice seedbed. NA as a seed dressing at 0.5% (W/W) was effective in reducing rice injury from butachlor, pretilachlor, chlormethoxynil and benthiocarb, and increased number of seedling established, plant height, and dry weight of rice seedlings, while growth inhibition and stand loss were observed at 1% NA. Pretilachlor slightly decreased number of seedlings established and inhibited growth of rice seedlings. But pre-mix combination of pretilachlor (2%) and CGA 123'407 (0.5%) reduced rice injury caused by pretilachlor alone and increased number of seedlings established and dry weight of rice seedlings. The results indicate that NA and CGA 123'407 would permit the safe application of butachlor, pretilachlor, chlormethoxynil, and benthiocarb in subirrigated rice seedbed.

Effect of Fertilization and Yearly Application of Identical Herbicides on Weed Succession and Yields of Rice (시비조건과 제초제의 연용이 잡초군락의 변화와 수도의 수량에 미치는 영향)

  • The field studies were carried out in paddy field over a four year period in order to find out the effects of butachlor, nitrofen, oxadiazon or CG 102, benthiocarb-S, and bentazon applied successively to the same paddy field for four years on weed succession and rice yields under different fertilized conditions including compost, chemical fertilizer and straw application. Total dry weight of weeds was steadily increased yearly over 4 year period regardless of fertilized conditions, and significantly higher increase of weed dry weight was observed on non-fertilized and compost plots. The most dominant weed species was Scirpus hotarui, and Potamogeton distinctus, Eleocharis kuroguwai and Cyperus serotinus were predominated on compost plot, E. kuroguwai and E. aclularis on non-fertilized plot, E. kuroguwai, C. serotinus, and Monochoria vaginalis on chemical fertifizer plot, and C. serotinus on straw plot. When the same herbicides were used continuously on the same plots, weed control effect was decreased and thus weed population was greatly increased particularly on butachlor and nitrofen treated plots. P. distinctus, E. kuroguwai, and C. serotinus were predominated on butachlor treated plot, P. distinctus and C. serotinus on nitrofen treated plot, C. serotinus on benthiocarb-S treated plot, C. serotinus and P. distinctus on oxadiazon or CG 102 treated plot and P. distinctus on bentazon treated plot. Annual weeds were slightly increased by repeated annual application of oxadiazon or CG 102 and benthiocarb-S, whereas perennial weeds were predominated by successive application of butachlor, nitrofen. and bentazon for 4 years. Yield reduction of rice became prominent by successive application of same herbicides. particularly butachlor and nitrafen. This results suggested that successive annual application of same herbicides should be limited and herbicide combination or herbicide rotation should be applied to control the remaining weed species.

Effect of Antibiotics and Herbicide on Shoot Regeneration from Cotyledon and Hypocotyl Explants of Chinese Cabbage (항생제와 제초제가 배추 자엽 및 배축 절편체로부터의 신초 형성에 미치는 영향)

  • To develop a selection system for regenerating plants from transformed tissues, effects of four antibiotics (kanamycin, hygromycin, carbenicillin, cefotaxime) and herbicide (phosphinotricin) on shoot regeneration from cotyledon and hypocotyl explants of Chinese cabbage (Brassica campestris L. ssp. pekinensis) were studied. For cotyledon, shoot induction was not significantly affected by kanamycin at $1mg{\cdot}L^{-1}$, but the number of shoots formed was significantly reduced at $2mg{\cdot}L^{-1}$, and no shoots were regenerated from any explants at $6mg{\cdot}L^{-1}$ or higher. Hypocotyl explants showed similar result as cotyledon. Kanamycin at $7mg{\cdot}L^{-1}$ may be adequate for selecting Chinese cabbage transformants. Hygromycin at $4mg{\cdot}L^{-1}$ or higher completely inhibited the growth and shoot regeneration of Chinese cabbage explants. Therefore, resistance gene to hygromycin may also be used as a selective marker for Chinese cabbage transformation. Carbenicillin and cefotaxime, the cephalosporin type of antibiotics, had little effect on shoot regeneration of Chinese cabbage explants. Since carbenicillin and cefotaxime have low toxicity to Chinese cabbage, they are suitable for use in tissue culture to eliminate Agrobacterium in transformation experiments after co-cultivation. Shoot regeneration from cotyledon and hypocotyl explants was significantly reduced in presence of $1mg{\cdot}L^{-1}$ phosphinotricin (PPT) and completely inhibited by $2mg{\cdot}L^{-1}$ or higher. PPT, same as antibiotics, may also be used to select transformed cells. Since Chinese cabbage is known to be recalcitrant to in vitro shoot regeneration compared to other Brassica species, even though lower levels of selectable markers result in more transformants but simultaneously allow more untransformed escapes to develop, lower levels of antibiotics and herbicides could be successfully used as a selectable marker to reduce selection pressure.

Herbicidal Properties of 5,8-dihydroxy-1,4-naphthoquinone and Their Possible Mode of Action (천연물 유래 5,8-dihydroxy-1,4-naphthoquinone의 살초특성과 작용기구)

  • This study was conducted to assess the possibility of 5,8-dihydroxy-1,4-naphthoquinone (DHNQ) as a environmental friendly herbicide candidate. Foliar application of DHNQ showed excellent herbicidal effect to the 3 grasses and 5 broad-leaved weeds. Among them, Digitaria sanguinalis and Solanum nigrum were completely controlled by $250{\mu}g\;mL^{-1}$ of DHNQ with main symptoms of desiccation or burndown within 24 hours. Aeschynomene indica was also sensitive to DHNQ treatment. All of the eight weed species were controlled by 90~100% at a concentration of $1000{\mu}g\;mL^{-1}$. However, soil application of DHNQ to Digitaria sanguinalis did not show any herbicidal symptoms. DHNQ strongly inhibited KAPAS activities in vitro and the $IC_{50}$ was $4.4{\mu}M$. Cellular leakage from cucumber leaf squares treated with DHNQ increased depending on the concentrations increased from 6.25 to $100{\mu}M$ after 24 hours incubation with or without light. However, chlorophyll loss in cucumber leaf squares was negligible. Biotin supplements significantly rescued the inhibition of germination rate of Arabidopsis thaliana seeds previously inhibited by the DHNQ. According to above results, DHNQ is a good natural herbicide candidate having a new target KAPAS, which is involved in biotin biosynthesis pathway, with environmental friendly.

  • Residual period and carry-over effect of some herbicides were determined using a bioassay method in five winter crops (chinese cabbage, radish, spinach, onion and garlic). The effects were measured at regular time intervals after applling different rates of the herbicides. There were no great differences in residual period and carry-over injury between the soils and kinds of crops used. However, the residual period varied with the herbicides studied and the carry-over injury was rate of the herbicide application, sampling depth of soil, and kind and seeding date of the test plant. When the residual herbicides were applied, the carry-over injury could be minimized by selecting tolerant crops, delaying seeding of the crops after application of the herbicides, and regulating the cultivation depth. Herbicides which showed no residual effect by the end of the cropping period (200-240 days for winter crops) and no carry-over effect were alachlor, trifluralin, ethalfluralin and prometryn. When pendimethalin, metolachlor, linuron and methabenthiazuron were applied at the recommended rate or less, there was no carry-over injury at harvesting time. With doubling the recommended rate, however, the carry-over effect was found in sensitive crops. Napropamide applied in winter crops at rate of 150-300g a.i./10a brought about carry-over injury for such Gramineae as Italian ryegrass, direct-seeded rice and barley, whereas the injury was not found in lowland-transplanted rice, Cruciferae, Cucurbitaceae and Solanaceae. Long residual herbicide nitralin applied at the rate of 75g a.i./10a caused the carry-over injury for Italian ryegrass, direct-seeded rice, baley and lowland-transplanted rice at 275 days in winter crops. In addition, a slight injury occurred in sesame, perilla and spinach, However, there was no injury for Cruciferae, Cucurbitaceae and Solanaceae.
