Synbiotics (mixture of probiotics and prebiotics) ameliorates DSS-induced ulcerative colitis in vivo.

  • Ulcerative colitis (UC) is one of inflammatory bowel disease (IBD), characterized by chronic inflammatory response and dysregulation of immune function. The severity of US has been influenced by environmental factors and food habit. The immune modulatory, anti-inflammatory and steroidal medicine have been used for the treatment of UC. However, long-term administration of those medicine is accompanied with side-effect. So, it is necessary to develop the non side-effect medicine using natural product. Prebiotics influences intestinal condition and food consumption. The heredity, immunity and environmental condition are related with occurrence of UC. In recent study, UC patients had lower level of prebiotics such as Lactobacillus and Bifidobacterium compared with healthy people. Also, previous study announced that imbalance of enteric flora aggravates the severity of UC. The effectiveness of probiotics might affect colon ability and viable bacteria also could promote the proliferation of beneficial intestinal bacteria. Prebiotics, such as herbal medicine, could lead to balance of intestinal bacteria or increase beneficial bacteria. So, proper choice of herbal medicine could control the intestinal condition. This study aimed to investigate the effect of mixture of probiotics and prebiotics (synbiotics) on dextran sulfate sodium (DSS)-induced UC in vivo. The synbiotics consist of Lactobacillus buchneri, Polymnia sonchifolia and Glycine max Merr. in this study. To evaluate the effect of synbiotics, 3% DSS was administered in BALB/c mice and synbiotics was daily administered for experimental days. The administration of synbiotics regulated colon length shortening, body weight change and disease activity index effectively. Also, extract of synbiotics upregulated survival ability of Lactobacillus buchneri in gut condition. These results suggest that mixture of probiotics and prebiotics, called as synbiotics, could influence intestinal condition also regulate the colon disease. Synbiotics might be a therapeutic agent for treatment of UC.

  • Carthamus tinctorius L. and Eucommia umoides Oliver are often used in traditional herbal medicines for reducing damage to the liver, kidney, bone and muscle. In the present study, we investigated cell viability and alkaline phosphatase activity in the human osteoblast-like MG-63 cell line with methanol extracts of Carthami Semen (CS) and Eucommiae Cortex (EC) alone or in a mixture (CS+EC). Osteoblast cell viability was evaluated using the MTS assay and alkaline phosphatase activity assays. The cell viability and alkaline phosphatase activity significantly increased in MG-63 osteoblast cells treated with the CS+EC mixture. These findings suggest the CS+EC mixture may have beneficial effects on bone health through the proliferation of osteoblast cells.

  • 본 연구는 방사선 육종 차조기와 백출 복합물의 조골세포 분화 활성 및 파골세포 형성 억제를 조사하였다. 차조기와 백출 복합물은 MG-63 세포에서 ALP 활성 및 arlizarin red 염색을 확인하였고 조골세포 형성의 영향은 RAW 264.7 세포에서 TRAP 활성과 TRAP 염색을 진행하였다. 세포 독성시험에서 차조기와 백출 복합물은 50 ㎍/㎖ 농도 이하에서 안전한 것으로 확인되었다. ALP 활성 및 골석회화 형성 능력은 대조군보다 활성이 낮았으나, 파골세포에서 TRAP 활성을 유의적으로 감소시켰으며, 효과적으로 TRAP(+) 다핵세포를 억제하였다. 따라서 차조기와 백출 복합물은 골 흡수 억제 활성을 향상시켜 뼈 관련 질환의 예방 및 치료에 효과적인 것으로 보여진다.

  • New herbal formula (NHF) is the ethanol extract mixture of Puerariae radix, Artemisia capillaries and Perilla frutescens. We have Investigated the effects on anti-inflammation by NHF and attempted acetic acid induced writhing to verify the analgesic effect. Macrophages and chondrocytes were obtained from mouse and rabbit. Inflammation was induced bγ interleukin-1, tumor necrosis $factor-{\alpha}$, $interferon-{\gamma}$, and lipopolysaccharide. NHF showed strong inhibitory efficacy against cytokine-induced proteoglycan degradation, $PGE_2$ production, NO production, and MMP-9 expression in rabbit articular chondrocyte. In the writhing test, NHF exhibited a dose-dependent inhibition of writhing. Futhermore, NHF increased the activity of SOD. NHF have anti-inflammatory and analgesic activities, and could be a good herbal medicine candidate for curing of osteoarthritis.

지황(地黃)과 발효(醱酵) 지황(地黃)의 생리활성 비교 연구 (Comparison of Biological Activities on Rehmanniae Radix and Fermented Rehmanniae Radix)

  • Herbal medicines are medicinal products containing a single or a mixture of two or more different herbal substances or herbal preparations as active principles. Recently, much attention has been paid to developing various kinds of fermented herbal extracts, a new type of traditional herbal medicine in the field of Korean traditional medicine. The fermentation of medicinal herbs is intended to exert a favorable influence on bioestability, bioavaliablilty and pharmacological activity of herbal extract in the gastrointestinal tract as well as intensifying the nutritional and pharmacological aspects of the medicinal herbs. The purpose of this study was to investigate biological activities of fermented Rehmanniae Radix by lactic acid bacteria at $30^{\circ}C$ for 3 days in comparison with those for Rehmanniae Radix The fermented Rehmanniae Radix exhibited different chemical profile to Rehmanniae Radix generated with HPLC, indicating production of new ingredients during fermentation. Rehmanniae Radix served as good nutritional sources for the growth of lactic acid bacteria showing increased number of bacteria during fermentation. Toxic effect of the fermented Rehmanniae Radix to cells were not seen judged by the MTT assay. The fermented Rehmanniae Radix exhibited better antioxidant effect than non-fermented Rehmanniae Radix analyzed by a SOD-likely assay. Both hypoglycemic and hypotensive effects of the fermented Rehmanniae Radix were also detected and better than those for Rehmanniae Radix in showing dose-dependent inhibitory effects on alpha-glucosidase and ACE, respectively. In conclusion, fermented Rehmanniae Radix appears to have more biological activities than non-feremented Rehmanniae Radix showing not only antioxidant effect but also cardiovascular protection.

Subacute Oral Toxicity and Bacterial Mutagenicity Study of a Mixture of Korean Red Ginseng (Panax ginseng C.A. Meyer) and Salvia plebeia R. Br. Extracts

  • As various populations are rapidly becoming an aging society worldwide and interest in health issues has increased, demand for functional foods including herbal products has increased markedly to maintain a healthy state which has led to safety issues about their intake as an inevitable result. The objective of this study was to identify the safety profile of a Korean red ginseng and Salvia plebeia R. Br. extract mixture (KGC-03-PS) which is a valuable ingredient that can be used as a functional food. In the present study, the subacute oral toxicity and bacterial reverse mutagenicity of KGC-03-PS were evaluated. Sprague Dawley rats were administered KGC-03-PS orally for 28 days by gavage. Daily KGC-03-PS dose concentrations were 0, 500, 1,000, or 2,000 mg/kg body weight (bw) per day. Bacterial reverse mutation test with KGC-03-PS dose levels ranging from 312.5 to $5,000{\mu}g/plate$ was carried out by OECD test guideline No. 471. Five bacterial strains (Salmonella typhimurium TA98, TA100, TA1535, TA1537, and Escherichia coli WP2) were tested in the presence or absence of metabolic activation by plate incorporation method. There were no toxicological effects related with test substance in the clinical evaluation of subacute oral toxicity test including clinical signs, body weight, and food consumption. Moreover, no toxicological changes related to KGC-03-PS were observed in the hematological and serum biochemical characteristics as well as in the pathological examinations, which included organ weight measurements and in the gross- or histopathological findings. KGC-03-PS did not induce an increase in the number of revertant colonies in all bacterial strains of the bacterial reverse mutation test. The no-observed-adverse-effect level of KGC-03-PS is greater than 2,000 mg/kg bw/day, and KGC-03-PS did not induce genotoxicity related to bacterial reverse mutations under the conditions used in this study.

건조방법과 향신료 추출물 첨가가 육포의 품질특성에 미치는 영향 (The Effects of Drying Method and Spice Extracts Added to Beef Jerky on the Quality Characteristics of Beef Jerky)

  • To develope Hanbang beef jerky as health food, six kinds of beef jerky were prepared by adding sugar (A), licorice (B) and three kinds of spice extracts (clove: C, fennel fruit: D and Chungyang green pepper extract: E) and mixture of all extracts (F). The effects of the drying method and added spice extracts on the quality characteristics of the beef jerky were evaluated. In general, the air-blast dried beef jerky contained $31{\sim}33%$ moisture, $50.0{\sim}51.2%$ crude protein, $7.2{\sim}7.8%$ crude lipid and $3.0{\sim}3.3%$ crude ash. For the mineral content of the air-blast dried jerky, the most prevalent mineral was Na $(1540.08{\sim}1838.17$ mg%), followed by K, P, Mg, and Ca. The Ca content of the beef jerky was highest in the mixed extract group (88.53 mg%), and the lowest content was in sugar-added group (53.12 mg%). For the color properties, the L-value (lightness), a-value (redness) and b-value (yellowness) were higher in the air-blast dried beef jerky than in the hot air dried samples. The drying methods showed their greatest affect on the redness (a) for all six jerkies (p<0.001). For the preference by sensory evaluation, the beef jerky samples with added sugar (A) and licorice extract (B) had significantly higher scores than the beef jerky samples with the added spice extracts, for both of air-blast drying and hot air drying (p<0.05). Preference for the air-blast dried beef jerkies with added clove (C) and fennel fruits (D) were significantly higher, in terms of taste, color, softness and aftertaste as compared to the respective hot air dried jerkies (p<0.01). Considering all the obtained results, we concluded that licorice and spice extracts can be used as natural preservatives in the development of health foods and the air-blast drying method is recommended to improve the quality characteristics of beef jerky.

족부백선의 한의학 논문에 대한 고찰 (Review on Studies of Korean Medicine about Tinea Pedis)

  • Objectives : The purpose of this study is to analyze research trends on tinea pedis in studies of Korean medicine.Methods : We searched papers using NDSL, KISS, RISS and KTKP(Korean Traditional Knowledge Portal). The first search used the keyword "Tinea pedis" in NDSL, KISS, RISS and KTKP. Used searching duration was not specified.Results : Studies found in NDSL, KISS and RISS were 122 and 118 studies were excluded. Studies found in KTKP were five papers and four studies of them were excluded. Finally five studies were selected and analyzed. Two studies of five selected ones were experimental researches and three studies were clinical researches. Among 2 researches of experimental researches, one of them was about antifungal efficacy of herbal medicines and ethahol extract of the mixture of Sophorae Subprostratae Radix, Aconiti Radix and Hibisci Syriaci Cortex and hot water extract of Phellodendri Cortex were effective. The other was about antifungal effect of the medicinal herb extraction method and vinegar extract was effective. Among 3 researches of clinical researches, there were one clinical study and two case studies. Functional soap containing herbal medicines and bee venom therapy were effective.Conclusions : As we looked for five researches, which were two experimental studies, one clinical study and two case studies. It is possible to treat tinea pedis with korean medical approach by conclusions of 5 researches. We expect that further researches will be proceeded and following results can be actively used as clinical treatments.

건조방법과 한약재 추출물 첨가가 육포의 미세구조에 미치는 영향 (Effects of Drying Method and Medicinal Herb Extract Addition on the Microstructure of Beef Jerky)

  • 본 연구의 목적은 육포의 제조방법이 품질특성에 미치는 영향을 규명하기 위하여 기본 양념(간장, 물엿, 참기름, 정종, 마늘, 양파, 배즙)에 설탕(A), 감초(B), 3종류의 향신료(정향;C, 회향;D, 청양고추;E) 추출물과 여러 추출물을 혼합 첨가(F)한 6군으로 나누어 쇠고기를 절인후, 열풍건조와 송풍건조하는 육포의 제조과정에서 근육의 미세구조 변화를 주사전자현미경(SEM)과 투과전자현미경(TEM)으로 관찰하였다. 투과전자현미경(TEM)으로 관찰한 결과, 쇠고기는 액틴과 미오신의 근육라인과 근원섬유가 평행으로 발달되어 있었고 세포의 미토콘드리아와 내막을 볼 수 있었으나 절임과정에서 근섬유가 절단되고 근육라인의 분해 및 세포 구조물들의 퇴행이 일어났다. 주사전자현미경(SEM)으로 관찰한 결과, 자연건조한 한약재 추출물 첨가 육포는 근원 섬유와 공간구조가 남아있고 육질이 보존되어 있었으나 열풍건조한 육포는 근원섬유의 배열과 공간구조가 전혀 관찰되지 않았다. 이상의 주사전자현미경과 투과전자현미경으로 육포의 미세구조를 관찰한 결과에서 한약재 추출물은 근섬유의 결을 보존하는데 도움이 되며 송풍건조는 열풍 건조에 비하여 육포의 물성을 좋게 함으로써 품질을 향상시킬 수 있는 건조방법으로 생각된다.

고지방식이로 비만을 유도한 C57BL/6 마우스에서 SM17의 항산화 및 항비만 효과 (Antioxidant and Anti-Obesity Effect of SM17 in High-Fat Diet Induced C57BL/6 Mice)

  • Objectives : Obesity is caused by the excess accumulation of fat in the body due to energy imbalance, and it causes various diseases. The aim of this study was to investigate an anti-obesity efficacy and an antioxidant activity of water from herbal mixture extract (SM17). Methods : The antioxidant activities were evaluated through radical scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radicals. To evaluated anti-obesity effect of SM17, we used a high fat diet fed mouse model. The SM17 (150 mg/kg body weight/day, p.o.) was treated every day for 6 weeks to C57BL/6 mice. Body weight and food intake were measured every day. The changes of reactive oxygen species (ROS), alanine aminotransferanse (ALT), aspartate aminotransferase (AST), triglycerids (TG) and total cholesterol (TC) in serum were analyzed after experiment. Also, expression of lipid metabolism related proteins were investigated by western blot analysis. Results : It was effective in antioxidant measurements, SM17 administration inhibited the biomarkers of lipid metaboism in serum and tissues. The administration of SM17 showed a significant reduction of body and tissue weight. Morever, it decreased ROS, ALT, AST, TG and TC in serum, compared with those of the obese mice. Adipogenesis-related protein expressions increased in obese mice compared to normal mice. However, SM17 group exhibited the down-expression of these proteins. Conclusion : A SM17 aqueous extract has a great effect on the stimulation (AMPK) activation, and may have a benefit to reduce a fatty acid metabolism through inhibition of lipid accumulation.
