Hepatoprotective effect of Schisandra chinensis on high-fat diet-induced fatty liver in rats (고지방 식이에 의한 흰쥐의 지방간증에서 오미자 추출물의 간보호 효과)

  • The purpose of the present study is to determine whether Schisandra chinensis (SC) has a protective effect on high fat diet (HFD)-induced fatty liver including hepatic lipid accumulation in rats. The HFD-induced obese rats were weighed after SC extracts were administered through the gastrointestinal tract at a concentration of 250 mg/kg b.w/day for 5 weeks. After 5 weeks, all of the rats on a high fat-diet were 36.5% heavier compared with normal controls. In contrast, rats on a high-fat diet supplemented with SC were 23.5% lighter than rats fed only a high-fat diet. Although there was no significant difference in food intake among the groups during the experimental diet period, the body weight gain of the SC group was significantly lower than the weights of the HFD groups. SC treatment slightly decreased the liver weight. Reduction of hepatic TBARS contents by SC was observed in rats fed a diet containing SC, and antioxidant activity was markedly increased in HFD+SC group compared to those of HFD group in liver. Moreover, total-lipid and triglyceride contents in the liver of groups fed a diet containing SC were significantly lower compared to those of the HFD group. High fat feeding elevated liver cholesterol concentration, but the addition of SC to the HFD rats resulted in the significant decrease in liver cholesterol. In histological observation of liver tissues, the hepatocytes of HFD rats showed a typical fatty liver morphology showing numerous lipid droplets in cytoplasm, whereas administration of SC reduced the size and numbers of lipid droplets. These results clearly demonstrated the attenuation of SC on nonalcoholic fatty liver induced by obese rats fed HFD.

Supplementary Effect of the Giant Embryonic Rice on Serum and Heaptic Lipid Levels of Streptozotocin-induced Diabetic Rats (Streptozotocin 유발 당뇨 쥐의 거대배아미 식이에 의한 혈액 및 간조직의 지질조성)

  • This study was carried out to investigate the supplementary effects of giant embryonic rice, which has over two times of embryo size compared the normal rice, on lipid metabolism of insulin dependent diabetic rats. Streptozotocin induced diabetic rats were fed four kinds of experimental diets com starch diet as a control (C-D), a polished rice diet (R-D), a brown rice diet (BR-D) and a giant embryonic rice diet (GER-D) respectively, for 6 weeks. Diet intake body weight, organ weights and lipid levels of serum, liver and feces were measured. There was significant difference in diet intake and body weight among experimental groups. The concentrations of serum triglyceride and total serum cholesterol of BR-D and GER-D groups were lower than those of others. The total hepatic cholesterol level was the lowest in GER-D group. The contents of total lipid and total cholesterol excreted in feces of BR-D and GER-D groups were higher than those of C-D and R-D groups. These results suggested the giant embryonic rice diets can effectively reduce serum triglyceride level and total hepatic cholesterol level in insulin dependent diabetic rats, and hypolipidemic effects be due to increasing fecal lipid excretion.

Inhibitory Effect of Combination with Korean Red Gnseng and Morus Alba in High Fructose-induced Vascular Inflammation and Steatohepatitis (고과당식이 투여 랫드모델에서 홍삼과 상엽 복합투여에 대한 혈관염증 및 지방간염 억제 효과)

  • This study was designed to elucidate whether combination with Korean red ginseng and Morus alba L. (MPM), traditional treatment for diabetes, ameliorates on high fructose-induced steatohepatitis and vascular inflammation. Animals were divided into four groups; Control receiving tap water, fructose-fed, rosiglitazone-treated fructose-fed rats, and MPM-treated fructose-fed rats both receiving supplemented with 60% fructose (n=10). The MPM or rosiglitazone groups initially received a high-fructose diet alone for 8 weeks, with supplementation with MPM or rosiglitazone, peroxisome proliferators-activated receptor gamma ($PPAR{\gamma}$) agonist, occurring during the final 6 weeks. Treatment with MPM significantly prevented the increase in c-reactive protein (CRP) levels in the high fructose group. MPM suppressed high fructose diet-induced vascular inflammation marker expression such as intracellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin. MPM also reduced intima/media thickness of thoracic aorta. Histologic observation and oil red O staining demonstrated hepatic tissue damage and lipid accumulation were severe in high fructose group. Treatment with MPM ameliorated hepatic tissue morphology with minimized steatosis. In addition, MPM attenuated hepatitis by inhibition of monocyte chemoattractant protein-1 (MCP-1) expression. MPM-fed group showed lower serum GOT and GPT levels comparing with high fructose group. MPM and rosiglitazone (positive control) significantly decreased the size of epididymal adipocytes. Taken together, the administration of MPM inhibited high fructose-induced steatohepatitis and vascular inflammation. These results suggested that MPM is useful in the prevention or treatment of metabolic syndrome-related disorders such as fatty acid metabolism and vascular homeostasis.

Overexpressed Derlin-1 Inhibits ER Expansion in the Endothelial Cells Derived from Human Hepatic Cavernous Hemangioma

  • Proteins that are unfolded or misfolded in the endoplasmic reticulum (ER) must be targeted for refolding or degradation to maintain the homeostasis of the ER. Derlin-1 was reportedly implicated in the retro-translocation of misfolded proteins from the ER to the cytosol for degradation. In this report, we showed that Derlin-1 was down-regulated in the endothelial cells derived from human hepatic cavernous hemangioma (CHEC) compared with other tested cells. Electron microscopy analysis showed that ER was aberrantly enlarged in CHEC cells, but not in other tested cells. When overexpressed, Derlin-1 induced the dilated ER to return normal size. This ER dynamic was associated with the activation of unfolded protein response (UPR). In CHEC cells where Derlin-1 was down-regulated, increased expression of the immunoglobulin heavy chain-binding protein (Bip) and UPR-specific splicing of X-box DNA-binding protein 1 (XBP1) mRNA were detected, as compared with that in other tested cells, indicating that UPR was activated. After Derlin-1 overexpression, the extent of UPR activation diminished, as evidenced by decreased expression of Bip, reduced amount of the spliced form of XBP1 ($XBP1_S$), and elevated expression of the unspliced form of XBP1 ($XBP1_U$). Taken together, these findings provide another example of a single protein being able to affect ER dynamic in mammalian cells, and an insight into the possible molecular mechanism(s).

Diagnostic Imaging of Biliary Atresia (담도폐쇄증의 영상 진단)

  • Biliary atresia is a rare but significant cause of neonatal cholestasis. An early and accurate diagnosis is important for proper management and prognosis. To diagnose biliary atresia, various imaging studies using ultrasonography, MRI, hepatobiliary scans, and cholangiography can be performed, although ultrasonography is more important for initial imaging studies. In this article, we review the findings of biliary atresia from various imaging modalities, including ultrasonography, MRI, hepatobiliary scans, and cholangiography. The known key imaging features include abnormal gallbladder size and shape, periportal thickening visible as a 'triangular cord' sign, invisible common bile duct, increased hepatic arterial flow, and combined anomalies. Aside from the imaging findings of biliary atresia, we also reviewed the diagnostic difficulty in the early neonatal period and the role of imaging in predicting hepatic fibrosis. We hope that this review will aid in the diagnosis of biliary atresia.

Effects of Catechin on Lipid Composition and Adipose Tissue in Obese Rats Fed High Fat Diet (고지방 식이로 유도된 비만쥐에서 녹차 Catechin이 체지방 조성 및 지방조직에 미치는 영향)

  • The current study examined the effects of catechin on lipid composition of serum and liver and adipocyte of epididymal fat pads in obese rats fed high fat diet. Sprague-Dawley male rats weighing $100{\pm}10g$ were randomly divided into eight groups, four normal diet groups and four high fat diet groups according to the level of dietary catechin supplement. The rats were fed ad libitum experimental diets for 4 weeks and then they were sacrificed. Body weight in HF group was heavier than that of NC group, but HFCM and HFCH groups were significantly reduced compared to HF group. Relative body weight to abdominal weight and relative body weight to epididymal weight in HF group were increased to 103% and 106%, respectively, compared to NC group, but HFCM and HFCH groups were significantly reduced as compared to HF group. The levels of serum triglyceride, total cholesterol, LDL-cholesterol and atherogenic index in HFCH groups were significantly lower than those of HF group, whereas HDL-cholesterol levels were increased. Total lipid contents of liver in HF group was significantly higher than that of NC group, but HFCH group maintained the NC level. There were no significant difference in hepatic triglyceride contents of high fat diet groups. Contents of hepatic cholesterol in HF group was 29% higher than that of NC group, but HFCM and HFCH groups were significantly reduced as compared to HF group. Cell number and cell size of epididymal fat pads in HFCM and HFCH groups were significantly reduced, respectively, compared to HF group. Improved lipid metabolism observed in rats fed catechin may be caused by an alteration of number and size in epididymal fat pad and lipid composition.

Preoperative Detection of Hepatic Metastases from the colorectal Cancers: Comparison of Dual-phase CT scan, Mn-DPDP enhanced MRI, and combination of CT and MRI (대장암의 간 전이 진단: 이중시기 CT, Mn-DPDP 조영증강 MRI, 그리고 CT-MRI 종합 판독의 비교)

  • Purpose : To determine the usefulness of additional Mn-DPDP MRI for preoperative evaluation of the patients with colorectal cancers by comparison of dual-phase CT scan, Mn-DPDP enhanced MRI and combination of CT and MRI. Materials and Methods : Fifty-three colorectal cancer patients with 92 metastatic nodules underwent dualphase (arterial and portal) helical CT scan and Mn-DPDP MRI prior to surgery. The indication of MRI was presence or suspected of having metastatic lesions at CT scan and/or increased serum carcinoembryonic antigen (CEA) levels (10 ng/mL or more). The diagnosis was established by the combination of findings at surgery, intraoperative ultrasonography, and histopathologic examination. Two radiologists interpreted CT, MRI, and combination of CT-MRI at discrete sessions and evaluated each lesion for location, size, and intrinsic characteristics. The lesions were divided into three groups according to their diameter; 1cm<, 1-2 cm, and >2 cm. Diagnostic accuracy was evaluated using the alternative-free response receiver operating characteristic method. Detection and false positive rate were also evaluated. Results : In the lesions smaller than 1 cm, detection rate of combined CT-MRI was superior to CT or MRI alone (82%, p=0.036). The mean accuracy (Az values) of combined CT and MRI was significantly higher than that of CT in the lesions smaller than 2 cm (1 cm<, p=0.034; 1-2 cm, p=0.045). However, there was no significant difference between MRI and combined CT-MRI. The false positive rate of CT was higher than those of combined CT-MR in the lesions smaller than 1 cm (28%, p=0.023). Conclusion : Additional MRI using Mn-DPDP besides routine CT scan was helpful in differentiating the hepatic lesions (<2 cm) and could improve detection of the small hepatic metastases (<1 cm) from colorectal carcinoma.

Effects of Samhwangsasim-tang on obesity-related metabolic disease in mice (삼황사심탕(三黃瀉心湯)이 수컷 생쥐의 비만(肥滿) 관련 대사질환(代謝疾患)에 미치는 영향)

  • Objectives : Samhwangsasim-tang (SHSST) is a traditional Korean medication, which has been used in Korea for treatment of hypertension and chest pain. Hyperlipidemia and inflammation could influence hypertension and chest pain. This study investigated whether and how SHSST reduces the hyperlipidemia and inflammation related to high-cholesterol diet-induced obesity in rats. Methods : Mice were divided randomly into four groups: the normal diet group, high-cholesterol diet group, low dose treatment group supplemented with 30% ethanol extract of SHSST (L) and high dose treatment group supplemented with 80% ethanol extract of SHSST (H). L and H groups were orally administered with SHSST at the dose of 50mg/kg a day respectively and others were administered with the same volume of physiological saline. Results : Administration of SHSST resulted in a decrease in serum levels of total cholesterol and low-density lipoprotein. Expression of hepatic genes(SREBP2, LXR, LDLR, and HMG-CoA) related with cholesterol metabolism was also suppressed. In addition, SHSST decreased the expression of inflammation-related gene (TNF-${\alpha}$, IL-6, ICAM-1, VCAM-1, TGF-${\beta}1$ and fibronectin). Histological examinations also showed that the size of the adipocytes was smaller in the SHSST treated group than in the high-colesterol diet group. In an in vitro study, SHSST inhibited the production of nitric oxide in a concentration-dependent manner. Conclusions : This study indicates that SHSST has anti-hyperlipidemia and anti-inflammatory effects. It may also suggest that SHSST may be alternative therapy for treatment of hyperlipidemia and its complications.

Temperature distribution in VX-2 hepatoma heated with thermoseed hyperthermia (열소자 온열요법시 VX-2 hepatoma내의 온도 변화에 대한 연구)

  • It was the purpose of present study to develop a new thermoseed for heating deep-seated tumors and assessment of the effect of magnetic control on thermoseeds. Aqueous suspension of iron micro spheres (Ferropolysaccharide) was injected directly into the VX-2 hepatoma and heated with 1.2 MHz inductive radiofrequency unit. Aqueous thermoseed suspension was delivered to the tumor by simple percutaneous injection. The limitation of the thermoseed heating method is the positional change of thermoseed particles in the tumor after implantation. The thermoseed particles could enter the systemic blood circulation and cause a severe embolization of a critical organ. To minimize this limitation, we have used the magnetic control after loading the thermoseed in the tumor, W hen ferropolysaccharides were exposed to a strong magnetic field, they magnetized and subsequently exerted a magnetic force on each other, forming larger aggregates of particles. The size of aggregated Particles were too big to enter the systemic blood circulation. Thus, unlike other thermoseed method, we hold the thermoseed particles stationary in the tumor. The temperature of the injected site and immediate vicinity elevated by $4-5^{\circ}C$. The temperature of the surrounding normal hepatic tissue elevated by $1-2^{circ}C$ only. The heating effect within the tumor was variable depending on the density of ferromagnetic aqueous suspension. Our results suggest that inductive heating of tumor injected with ferropolysaccharide solution offers the possibility of effective heat delivery to the defined tumor volume, which is difficult to heat with other heating devices.

Antitumor Activity of Salvia miltiorrhiza Herbal Extract in Rat Tumor Model (흰쥐의 종양에 대한 단삼 추출물의 항종양 활성)

  • The present study was carried out to investigate the antitumor activity of Salvia miltiorrhiza (SM) herbal extract in rat tumor model. We used a new tumor animal model for the invasion and metastasis of cancer using genetically k-ras-induced rat kidney cells (RK3E-ras). We observed tumor as early as 7 days after the injection of RK3E-ras cells in subcutaneous of Sprague-Dawley rats. All of the rats developed tumor mass at the inoculated site. After 7 days, the experimental groups were divided into two: saline control and injected with SM (200 mg/kg) groups. We investigated tumor's weight, size and hepatic metastasis of each group. Injection of SM herbal extract every other day for 14 days significantly inhibited tumor growth. Histologically, the tumors were undifferentiated carcinoma showing multifactorial necrosis and hemorrhage; also, the tumor invaded into hepatoportal region. Treatment with SM herbal extract caused significant inhibition of tumor cell proliferation. Our data showed that SM herbal extract is effective in controlling the tendency of tumor cell proliferation and metastasis by injection of RK3E-ras cells. These findings provided the potential value of SM as a novel antitumor agent candidate.
