Effects of garlic on the cadmium accumulation in the tissue on the hematological picture in dogs (마늘이 개의 조직내 카드뮴축적 및 혈액상에 미치는 영향)

  • The environmental pollution by heavy metals has become a significant hygenic problem. Recently it is said the garlic supplementation in diet have protective effect on the heavy metal accumulation in tissue of rat by a researcher. In this study two experiments were undertaken. Firstly, dogs were fed diet supplemented with cadmium of cadmium plus 7% of raw garlic for 50 days. on the 50th day of experiment all dogs were euthanasied and the cadmium content of various tissue were measured by atomic absorption spectrophotometeric method. Secondly, in order to find out the toxic effect of garlic dogs were fed diet supplemented with 7% of raw garlic for 50 days and hematological or hematochemical observations were undertaken at 10 day intervals. The results obtained from these experiments were summarized as follows; 1. In dogs treated with cadmium and cadmium plus garlic cadmium was accumulated in renal cortex, liver, and muscle. However the concentrations of cadmium showed no significant between the two group. 2. In dogs fed diets supplemented with 7% of raw garlic significant decrease of erythrocyte count, packed cell volume and hemoglobin content and significant increase of Heinz body erythrocyte and reticulocyte count were recognized. 3. The garlic-treated dogs and control dogs had normal asparate aminotransferase, alanine aminotransferase, alkaline phosphotase, blood urea nitrogen value. The value showed no significant difference between the two group. 4. From these results it may be concluded that the garlic have protective effect for cadmium accmulation in tissue and excess garlic in diet could induce hemolytic anemia in dogs.

The Correlation of Racing Performance with Pre-Race Blood Values in Thoroughbred Horses

  • Recently, the horse racing industry in Korea has developed rapidly with good betting turnover. One of the most important factors in enhancing the performance of racehorses is the study of equine exercise physiology. This study was performed to analyze the results of pre-race blood test on race day and evaluate its correlation with the race performance in Thoroughbred racehorses. Twenty-one, healthy 3- to 6-year-old Thoroughbred horses were used to collect blood samples and hematological and biochemical measurements were performed. Results were analyzed to identify its correlation with racing performance. Analysis of hematological profile of pre-race blood of the racehorses revealed that red blood cell indices including RBC, Hb, and PCV levels were elevated. Additionally, levels of AP, AST, and GGT were also elevated in pre-race blood of horses on race day. RBC, Hb, and PCV levels of high performance racehorses were significantly lower than those of low class racehorses. Moreover, the analysis of correlation between blood test results and racing ability-related categories have shown negative correlations for rating, placing strike rate, prize money per race, RBC, Hb, and PCV and positive correlations for G3F and PCV. Additionally, AST value was positively correlated to prize money per race. Thus, the results of this study show that red blood cell indices and AST are good parameters for evaluating racing performance of horses before the race.

Single and Four-week Intravenous Toxicity Studies of DA-3585, a Recombinant Human Erythropoietin, in Rats (재조합 사람 적혈구 조혈인자 DA-3585의 랫드에 대한 단회 및 4주반복 정맥투여 독성시험)

  • DA-3585 is a recombinant human erythropoietin produced by Dong-A pharmaceutical Co. Ltd. using recombinant DNA technique. Recently, recombinant human erythropoietin (rHu-EPO) has been used to treat various types of anemia. In this study, we examined acute and subacute toxicity of DA-3585 in rats. DA-3585 was intravenously administered to rats at dose levels of 0, 6,250, 12,500 and 25,000 lU/kg for single dose toxicity study and at dose levels of 0,100,500 and 2,500IU/kg daily for 4 week-repeated dose toxicity study. In the single dose toxicity study, there were no death, clinical signs and changes in body weight gain related to the treatment. Necropsy revealed no evidence of toxicity related to DA-3585, In the repeated dose toxicity study, all the rats survived throughout the study. There were no treatment-related changes in clinical signs, food and water intake, and body weight. Hematological examination showed increases in the number of erythrocytes, hemoglobin concentration, hematocrit value and mean corpuscular volume, and decrease in the number of platelet in 500 and 2,500 lU/kg dosed groups. Extramedullary hematopoiesis in the spleen and erythroid hyperplasia in the bone marrow were noted as treatment-related histological changes. Toxicologically significant changes were not observed in blood biochemistry, urinalysis, organ weights and in any other examinations. The treatment-related changes observed in this study were hematological or histological changes associated with pharmacological effects of DA-3585. On the basis of the results of this study, LD5n value of DA-3585 was above 25,000 lU/kg and the no-observed-adverse-effect-level was estimated to be 100 lU/kg.

Changes of hematological parameters and plasma components in the hybrid grouper(Epinephelus fuscoguttatus ♀×E. lanceolatus ♂) by acute ammonia exposure (암모니아 급성노출에 따른 대왕범바리(Epinephelus fuscoguttatus ♀×E. lanceolatus ♂)의 혈액성상 및 혈장성분의 변화)

  • Hybrid grouper(Epinephelus fuscoguttatus ♀×E. lanceolatus ♂) (mean weight 25.7±3.5g, mean length 11.2±0.9cm) were exposed to different ammonia concentrations of 0, 5, 10, 20, and 40mg L-1 for 96 hours. The hematological hematocrit and hemoglobin parameters of the hybrid grouper were significantly decreased by 20 mg L-1 ammonia exposure. In the organic plasma components, calcium was significantly decreased, whereas there was no change in magnesium. In the organic plasma components, the glucose and cholesterol values of the hybrid grouper were significantly increased by ammonia exposure. In the enzymatic plasma components, the ALP(Alkaline phosphatase) value of the hybrid grouper was also significantly increased by ammonia exposure. The results of this study demonstrate that acute ammonia exposure to hybrid grouper induced changes in hematological parameters and plasma components. Therefore, acute ammonia exposure over 20 mg L-1 appears to be toxic to hybrid grouper and the results can be used as a major indicator in breeding hybrid grouper.

Studies on the Reproductive Toxicant and Blood Metabolite in Pups Born After Bisphenol A Administration in Pregnant Mice (임신중인 생쥐에 Bisphenol A 투여 후 태어난 차산자의 생식독성과 혈액성분에 관한 연구)

  • Bisphenol A (BPA) is a monomer used in the manufacture of a multitude of chemical products, including epoxy resins and polycarbonate. The objective of this study was to evaluate the effects of BPA administration on reproductive characteristics and blood hematological and chemical values in offspring of pregnant dams treated with BPA. BPA was administrated to pregnant mice by intraperitoneally injection with 0, 0.05, 0.5 and 5.0 mg/kg B.W. for 5 times at 3 days interval on gestation days 1-16. There were no treatment-related effects of BPA on reproductive organ weight in male offsprings at 45 days-of-age, but body weight was the lowest in 5.0mg BPA group when compared to other groups (P<0.05). No differences in semen characteristics (sperm concentration, viability, motility and abnormality) were observed between the control and BPA treatment groups. The WBC, HB, HT, MCV, MCH, MCHC, albumin, BUN and total protein of blood hematological and chemical values in male offsprings were not difference for any treatment groups, but RBC value in BPA groups was significantly increased comparing to the control group (P<0.05). The PLT value was slightly higher in 5.0mg BPA groups than in any other group, but not significantly difference among the experimental groups. In female offsprings, the effects of BPA didn't affect to the body and ovary weight, but the uterus weight in 5.0mg BPA group was slightly heavier than that of control group (P>0.05). No statically significant difference in blood hematological values in female offsprings were observed between the control group and BPA groups, but the concentration of albumin and BUN were significantly higher in 0.5mg BPA group when compared to control and other BPA treatment groups (P<0.05). The histological evaluation of testis and ovary in growing offspring at 45 days-of-age was not difference between the control group and BPA groups, but endometriosis of the uterus in female offspring was dramatically increased in 0.5 and 5.0mg BPA groups. These founding suggest that low concentration of BPA might not have a important role on reproductive ability or blood metabolite in offspring of pregnant dams treated with BPA.

Comparison of hematological values of conventional pigs and transgenic pigs supressed in immune rejection response (일반돼지와 면역 거부반응이 억제된 형질전환돼지의 혈액 성상 비교)

  • Blood test is a useful tool in establishing medical treatment for livestock. It provides information such as disease diagnosis, treatment effects, prognostic judgment, and health status. This study compared the value of erythrocytes and leukocytes among conventional, transgenic miniature, and transgenic conventional pigs aged six months to 24 months. Further, it analyzed the aspects of hematological value changes according to the pigs' ages. As a result, the number of red blood cells (RBC), which include hemoglobin, and hematocrit, and the number of white blood cells (WBC), which include neutrophils, and lymphocyte, were high among transgenic miniature pigs, compared with the conventional and transgenic conventional pigs. Conventional pigs showed similar values of RBC and WBC regardless of transgenesis. In comparing their age, the RBC decreased as the age increased compared with the pigs among all the three groups aged of 6~12 months. On the other hand, WBC and neutrophils showed no significant difference regardless of different ages among all the three groups. However, various counts in RBC and WBC were mostly found to be higher in each age in transgenic miniature pigs than in conventional and transgenic conventional pigs. The results of this study show that the values of RBC and WBC were generally higher in transgenic miniature pigs than in conventional and transgenic conventional pigs. Based on this research, hematological values can be widely used in diagnosing diseases or checking the health status of transgenic pigs that are used as disease models, organ transplant source and alike.

Prognostic and Predictive Value of Hematologic Parameters in Patients with Metastatic Renal Cell Carcinoma: Second Line Sunitinib Treatment Following IFN-alpha

  • Background: Long-term survival is a problem with locally advanced and metastatic renal cell carcinomas. Sunitinib malate is an oral multitargeted tyrosine kinase inhibitor, but data on sunitinib use as a second line treatment in metastatic renal cell carcinoma (mRCC) are limited. Prognostic and predictive value of peripheral blood markers has been shown for many cancers. Materials and Methods: Efficacy and safety profiles of sunitinib after interferon alpha (IFN-${\alpha}$) were evaluated based on retrospective data for 23 patients with mRCC. Hematological parameters (neutrophils, lymphocytes, platelets, mean platelet volume, neutrophil/lymphocyte ratio, platelet/lymphocyte ratio) were recorded at the time of metastasis. It was evaluated whether hematological parameters were prognostic and predictive factors. Results: Median progression-free survival (PFS) time was 16.5 months (95%CI: 0-34.5). Median overall survival (OS) time was 25.7 months (95%CI: 10.8-40.0). Most common side effects were neutropenia (52.2%), stomatitis (26.1%) and hand-food syndrome (26.1%). PFS was found 3.13 vs 17.1 months in patients with neutrophil / lymphocyte ratio (NLR)>3 vs $NLR{\leq}3$ (p:0.012). Median OS was 6.96 vs 27.1 months in patients with NLR>3 vs $NLR{\leq}3$ (p:0.001).While 75% of patients who responded to sunitinib had $NLR{\leq}3$, in 72% of patients with no response to sunitinib NLR>3 was detected (p:0.036). The association between the Memorial Sloan-Kettering Cancer Center (MSKCC) criteria and NLR was statistically significant (p:0.022). Conclusions: Data on second line sunitinib treatment following cytokine in mRCC are limited. In our study, we observed second line sunitinib treatment following IFN-${\alpha}$ to be effective and tolerable. NLRappeared to have prognostic and predictive value.

Study on Menstrual Blood Loss and Iron Nutrition in Korean Women (한국인여성(韓國人女性)의 월경중(月經中) 혈액손실(血液損失)과 체내철분영양상태(體內鐵分營養狀態)에 관(關)한 연구(硏究))

  • The aim of this study was to define the range of the menstrual blood loss(MBL), and the upper limit of the MBL associated with iron deficiency anemia in Korean women, For this purpose, healthy unmarried women between the age of 15 to 26 years were selected from July to September, 1979 and results obtained were used for determining baseline MBL and its relationship to iron nutriture. One houndred forteen women were tested for their MBL and hematological and biochemical data such as hemoglobin(Hb) concentration, hematocrit, mean corpuscular hemoglobin concentration(MCHC), and serum iron and ferritin concentration. This study was partly supported by World Health Organization. The results of this study are summarized as follows: 1) The mean values and standard error of mean of MBL were $30.7{\pm}2.19ml$ for total subjects, $30.4{\pm}3.14ml$ for sutjects below 20 years of age and $30.3{\pm}2.49ml$ for subjects aged more than 20 years. No significant variation was noted for amount of MBL between two age groups. 2) The median value and 95th percentile value of MBL were 26.5ml and 69.0ml respectively for total subjects. Maximum number (24.6%) of the subjects fell in group with 10-20ml of MBL while 10.5% showed MBL above 50m1. 3) Mean value of the duration of menstruation for total subjects was 4.34 days. 4) It was observed that mean values of Hb concentration, hematocrit and MCHC were decreased slightly in subjects showing MBL more than 40m1. Serum iron and ferritin levels were markedly decreased in women with MBL above 40m1 and 20-30ml, respectively. 5) In subject population showing MBL above 50ml, the frequency of subjects with hematological and biochemical data below anemia criteria were remarkably increased. 6) Although the number of subjects tested was small and the subject selection was not done by random stratified sampling from a population group, this study suggested that the upper normal limit of MBL of Korean women seems to be in the range of 40-50ml.

A Comparative Analysis of Deficiency-Excess Pattern Identification with Sputum Cytokines and the Characteristics of Asthma Patients (천식환자 허실변증과 객담 cytokine 및 특성 비교분석)

  • Objective: The aims of this study were to analyze the deficiency-excess pattern identification (虛實辨證) and compare it to the sputum cytokines of asthma patients. Method: 50 asthma patients who met the inclusion and exclusion criteria were included in this study. They were divided into two groups: deficiency and excess syndrome groups. Sputum examinations were performed including $TNF-{\alpha}$, Interleukin (IL)-4, IL-5, IL-10, and IL-13. The Quality of Life Questionnaire for Adult Korean Asthmatics (QLQAKA), the Visual Analog Scale(VAS), and heart rate variability (HRV) were also measured. We also conducted laboratory tests, including the hematological indexes. Results: Based on the pattern identification, 50 asthma patients can be divided into two categories of groups: the deficiency syndrome group (N=24) and the excess syndrome group (N=26). In the analysis of sputum cytokines, although the $TNF-{\alpha}$, IL-4 and IL-13 were at a higher level in the deficient pattern group than in the excess pattern group, it was insignificantly different. There was a negative correlation in the analysis of QLQAKA and VAS. In the analysis of HRV, although the mean value of VLF, LF, and HF in the deficiency syndrome group was higher than in the excess syndrome group, it was insignificantly different. There was no significant difference in the hematological tests between the deficiency and the excess syndrome group. The mean value of the IgE in the blood tests was five times greater than the reference value. Conclusion: The cytokines of sputum including $TNF-{\alpha}$, IL-4, IL-5, IL-10, and IL-13 were indifferent statistically. Reinforcing the healthy and eliminating the pathogenic factors should be considered.

Therapeutic Effects of Acupuncture and Herbal External Preparation on Healing of Deep Partial Thickness Burn Wound in Rats (심재성 2도 화상이 유발된 흰쥐의 조직 회복에 대한 침 및 한약외용제의 효과)

  • Objectives The present study aimed to investigate the effects of acupuncture and herbal external preparation on wound healing procedure in a deep partial thickness burn model in rats. Methods A total of 40 male Sprague-Dawley rats were allocated into four groups with an equal number in each group: acupuncture treatment group, herbal external perparation group, Silver sulfadiazine dressing group and control group. We describe the effect of acupuncture and herbal external preparation on morphologic and histologic changes, epithelial growth factor (EGF), hematological value of the deep partial thickness burn wound in rats. Results At 21th day after wounding the wound size in acupuncture treat group and herbal external preparation group were decreased more significantly compared to control group. In addition, epidermal regeneration on acupuncture treatment was than other treatment and control group in histological finding. Hematological findings revealed that acupuncture treatment group and herbal external prepartion group was more effective than control group in reducing inflammation response induced by burn. In acupuncture treated group, neutrophil and leukocyte level were significantly decreased compared to other treatment group. Also, this study showed that EGF was obviously expressed in nascent tissue when wounds were treated with the acupuncture and herbal external preparation after injury. In particular, acupuncture treatment group had a significant increase of EGF expression in burn wound healing area when compared with the other treatment groups. Conclusions These findings suggest that acupuncture and herbal external prepration may improve burn wound healing through decreasing inflammatory reaction, increasing tissue regeneration and expression of EGF. Moreover, acupuncture treatment could be more effective in comparison with Silver sulfadiazine dressing.
