Isolation and Characterization of a Protease-Producing Halophilic Vibrio sp. (단백질 분해효소를 생산하는 호염성 미생물 Vibrio의 분리 및 특성)

  • In this study, a halophilic protease-producing bacterium was isolated from the west seaside mud flats of Korea. The 16S rDNA nucleotide sequences of the isolate showed 99.5% sequence homology with those of Vibrio vulnificus and Vibrio fluvialis; therefore, the isolate was named Vibrio sp. YH-127. Gram staining and the carbohydrate metabolism test results supported the isolate as one from the Vibrio family. Optimum condition for the cell growth and for the protease activity were obtained when the isolate was cultured at 25$^{\circ}C$ and pH 7.0, with the salt concentration of the medium similar to that of sea water. Finally, the addition of Mg$^{++}$ ions into medium increases protease activity suggesting that the protease produced by the isolate was a metalloprotease.

  • Oceanobacillus kimchii is a member of the genus Oceanobacillus within the family Bacillaceae. Species of the Oceanobacillus possess moderate haloalkaliphilic features and originate from various alkali or salty environments. The haloalkaliphilic characteristics of Oceanobacillus advocate they may have possible uses in biotechnological and industrial applications, such as alkaline enzyme production and biodegradation. This study presents the draft genome sequence of O. kimchii X50T and its annotation. Furthermore, comparative genomic analysis of O. kimchii X50T was performed with two previously reported Oceanobacillus genome sequences. The 3,822,411 base-pair genome contains 3,792 protein-coding genes and 80 RNA genes with an average G+C content of 35.18 mol%. The strain carried 67 and 13 predicted genes annotated with transport system and osmoregulation, respectively, which support the tolerance phenotype of the strain in high-alkali and high-salt environments.

Effect of NaCl on Halomonas subglaciescola DH-1 Incapable of Growing at Non-Salinity (Halomonas subglaciescola DH-1의 생장에 미치는 염화나트륨의 영향)

  • A halophilic bacterium, H. subglaciescola DH-1, grew at 2.0 M salinity, but did not grow at 0.8 M salinity when cultivated at higher temperature ($40^{\circ}C$) than optimum ($30^{\circ}C$). When the cell extract of strain DH-1 was heated at $50^{\circ}C$ for 60 min in the absence of NaCl, isocitrate dehydrogenase and malate dehydrogenase lost their activities, but when it was heated in the presence of 2.0 M NaCl, the activity was maintained. Meanwhile, the cell extract of E. coli did not catalyze the reduction of $NAD^+$ to NADH coupled with the oxidation of isocitrate and malate at higher salinities than 1.0 M. The pH range for DH-1 was 7 to 10, and that for E. coli was 5 to 9. DH-1 was not grown in conditions with sodium salts other than NaCl.

  • A moderately halophilic bacterium, Listeria denitrificans HB-38, isolated from mud on the seashore in Sooyoung bay, Pusan, showed the requirement of $4\%$ sodium chloride for cell growth in a medium with salts typical of a marine environment, and showed that of $10\%$ in a medium with salts typical of a terrestrial environment. The optimum temperature and pH for growth were $40^{circ}C$ and pH 7.5 in the medium containing $10\%$ sodium chloride and ions typical of a terrestrial environment. Sodium chloride as a protoplast stabilizer gave more stability than sorbitol or sucrose, meanwhile the protoplast did not require higher concentration of stabilizer than that of E. coli protoplast. Succinic dehydrogenase of HB-38 had a halophilic property showing maximal activity in the presence of $9\%$ sodium chloride. The strain HB-38 did not harbor an extrach-romosomal DNA.

Characteristics of the protease from the extreme halophile, Halobacterium sp. (고도 호염성 Halobacterium sp.가 생산하는 protease의 특성)

  • The pretense from Halobacterium sp. was purified by ethanol precipitation and gel filtration on Sephadex G-75 and G-100. The purified enzyme was found to be homogeneous by polyacrylamide gel electrophoresis It's specific activity was 364units/mg protein and yield was 14% of the total activity of the culture filtrate. The Km value against casein was determined to be $4.2{\times}10^{-4}M$ by Lineweaver-Burk plot The optimal temperature and pH for the enzyme activity were $35^{\circ}C$ and pH 8.0, respectively. The enzyme was stable from 5.0 to 11.0 at relatively wide range of pH but was inactivated at the temperature above $50^{\circ}C$. $Ca^{2+}$ and $Mg^{2+}$ appeared to react as activators whereas $Fe^{3+},\;Zn^{2+},\;Cu^{2+},\;Hg^{2+}\;and\;Cd^{2+}$ as inhibitors. The enzyme activity reduced with increasing the concentration of NaCl : the apparent activity with 2M NaCl was 65% as compared with that without the salt However the enzyme was unstable without salts : the activity was lost when dialyzed against distilled water for 2hr, whereas maintained against 0.1M solution of $CaCl_2$ for 6hr.

Characteristics and Physiological Functionalities of Unrecorded Yeasts from Wild Flowers of Seonyudo in Jeollabuk-do, Korea (전라북도 선유도일대 야생화에서 분리한 국내 미기록 효모들의 특성 및 생리기능성)

  • Six kinds of newly recorded yeasts such as Rhodosporidium diobovatum SY4-2, Cryptococcus bestiolae SY7-1, Kazachstania unispora SY14-1, Kazachstania servazzii SY14-3, Pichia holstii SY20-2 and Cryptococcus tephrensis SY26-1 were screened from sixty one yeasts derived from wild flowers found in Seonyudo, Gogunsanyeoldo, Jeollabuk-do, Korea. All of them grew in 50% glucose-containing yeast extract-peptone-dextrose (YPD) broth and Pichia holstii SY20-2 was also halophile, growing in 20% NaCl-containing YPD broth. All of them, except Cryptococcus tephrensis SY26-1, were assimilated to glucose. Cell-free extract from Kazachstania servazzii SY14-3 showed the highest 98.6% of ${\alpha}$-glucosidase inhibitory activity and maximal production of the ${\alpha}$-glucosidase inhibitor was obtained with 24h incubation at $30^{\circ}C$. The antihypertensive angiotensin I-converting enzyme inhibitory activity of the unrecorded yeasts were showed 58.6-80.4% in their supernatants.

Phylogenetic diversity of bacterial communities in a gray solar saltern and isolation of extremely halophilic bacteria using culturomics (토판염전 결정지 내 세균군집의 계통학적 다양성 및 Culturomics법을 이용한 고도 호염균의 분리)

  • In this study, we investigated the phylogenetic diversity of the bacterial community and isolation of extremely halophilic bacteria using culturomics in a gray solar saltern. The number of bacterial living cells, enumerated in a gray solar saltern by direct fluorescence microscopy was three to four orders of magnitude greater than those enumerated by plate counts, suggesting the distribution of 'viable but non-culturable bacteria'. The biodiversity of bacterial communities in a gray solar saltern was investigated by pyrosequencing, 1,778 OTUs of bacteria were comprised of 18 phyla 46 classes 85 orders 140 families 243 genera with 6.16 diversity index. Archaea communities were composed of 3 phyla 6 classes 7 orders 7 families 38 genera with 4.95 diversity index from 643 OTUs. Totally 137 isolates were isolated by 59 different cultural methods based on culturomics considering culture media and conditions suitable for the growth of extremely halophilic bacteria. Phylogenetic analyses of extremely halophilic isolates based on 16S rRNA gene sequences, extremely halophilic isolates were composed of 4 phyla and 11 genera. Haloterrigena and Haloferax can be successfully isolated from culturomics. These culturomics were effective methods for collection of diversity of extremely halophilic bacteria.
