소포체 스트레스가 Ajuba 발현유도 (Expression of Ajuba, a Novel LIM Protein, is Regulated by Endorlasmic Reticulum Stress)

  • Ajuba is a number of proteins containing cytosolic LIM domain. Its function may provide a new pathway whereby cell-cell adhesive events are transmitted to the nucleus to regulate cell proliferation and differentiation decisions. Here, Ajuba gene expression was investigated its molecular properties associated with endoplasmic reticulum (ER) stresses (tunicamycin, DTT, A23187 and BFA) which induced remarkable ex-pression of Ajuba mRNA. The mRNA half life of Ajuba was also determined, its half life of Ajuba mRNA in FRTL-5 cells was approximately 2 hr after the initial translation. Although the obvious bioligical function of Ajuba is not clear, on the base of the results, Ajuba gene expression is deeply associated with ER stresses.

Effect of Glutaraldehyde Treatment on Stability of Permeabilized Ochrobactrum anthropi SY509 in Nitrate Removal

  • For practical application, the stability of permeabilized Ochrobactrum anthropi SY509 needs to be increased, as its half-life of enzymatic denitrification is only 90 days. As the cells become viable after permeabilization treatment, this can cause decreased activity in a long-term operation and induce breakage of the immobilization matrix. However, the organic solvent concentration causing zero cell viability was 50%, which is too high for industrial application. Thus, whole-cell immobilization using glutaraldehyde was performed, and 0.1% (v/v) glutaraldehyde was determined as the optimum concentration to maintain activity and increase the half-life. It was also found that 0.1% (v/v) glutaraldehyde reacted with 41.9% of the total amine residues on the surface of the cells during the treatment. As a result, the half-life of the permeabilized cells was increased from 90 to 210 days by glutaraldehyde treatment after permeabilization, and no cell viability was detected.

Thin Film Energy Storage Device with Spray-Coated Sliver Paste Current Collector

  • This paper challenges the fabrication of a thin film energy storage device on a flexible polymer substrate specifically by replacing most commonly used metal foil current collectors with coated current collectors. Mass-manufacturable spray-coating technology enables the fabrication of two different half-cell electric double layer capacitors (EDLC) with a spray-coated silver paste current collector and a Ni foil current collector. The larger specific capacitances of the half-cell EDLC with the spray-coated silver current collector are obtained as 103.86 F/g and 76.8 F/g for scan rates of 10 mV/s and 500 mV/s, respectively. Further, even though the half-cell EDLC with the spray-coated current collector is heavier than that with the Ni foil current collector, smaller Warburg impedance and contact resistance are characterized from Nyquist plots. For the applied voltages ranging from -0.5 V to 0.5 V, the spray-coated thin film energy storage device exhibits a better performance.

신비탕 및 가미신비탕이 Allergy 천식 모델 흰쥐의 BALF내 면역세포 및 혈청 IgE에 미치는 영향에 관한 연구 (The Effects of Shinbi-tang and Gamishinbi-tang on Immune Cell and Serum OA-specific IgE in BALF in Rat Asthma Model)

  • Background : Asthma is a chronic inflammatory disorder under immunological influence. Shinbi-tang and Gamishinbi-tang are herbal decoctions used for treating asthma in traditional herbal medicine. Objective : To evaluate the effects of Shinbi-tang and Gamishinbi-tang on immune cell & serum OA-specific IgE in broncho-alveolar lavage fluid (BALF) in rat asthma model. Material and Methods : Rats were sensitized with ovalbumin (OA); at day 1 sensitized group and Shinbi-tang and Gamishinbi-tang groups were systemically immunized by subcutaneous injection of 1mg OA and 300mg of Al(OH)$_3$ in a total volume of 2ml. At the same time, 1 ml of 0.9% saline containing 6 x 10$^{9}$ B. pertussis bacilli was injected by i.p. 14 days after the systemic immunization, rats received local immunization by inhaling 0.9% saline aerosol containing 2%(wt/vol) OA. A day after local immunization, HAL fluid was collected from the rats. Rats were orally administered with each of Shinbi-tang and Gamishinbi-tang extract for 14 days from the day after local immunization. Lymphocyte, CD4+ T cell CD8+ T cell counts, CD4+/CD8+ ratio in BALF, change of serum OA-specific IgE level, CD4+ T cell CD8+ T cell percentages in the peripheral blood were measured and evaluated. Results : Shinbi-tang and Gamishinbi-tang showed an alleviating effect on asthmatic responses of rats. Shinbi-tang decreased total cell, lymphocyte, CD4+ T cell in BALF, serum OA-specific IgE level as compared with the control group. Gamishinbi-tang decreased total cell, lymphocyte, CD4+ T cell, and CD4+/CD8+ ratio in HALF as compared with the control group. CD4+/CD8+ ratio in HALF from Shinbi-tang group and serum OA-specific IgE level from Gamishinbi-tang group didn't show any significant variation from control group. CD8+ T cell in HALF, CD3+CD4+ T cell and CD3+CD8+ T cell percentages in peripheral blood showed no significant variation among groups. Conclusion : Shinbi-tang and Gamishinbi-tang alleviated asthmatic hypen-eactivity of the rat immune system through CD4+ T cell and serum IgE. Further the study of immune system modulating mechanism is expected.

콘크리트구조물 중의 철근 부식 저감을 위한 FRP Hybrid Bar의 적용성 연구 (A Study on the Application of FRP Hybrid Bar to Prevent Corrosion of Reinforcing Bar in Concrete Structure)

  • 최근 해양환경의 대형 SOC구조물 증가에 따른 구조물 내구성 증진에 관심이 증가되고 있다. 내구성 증진을 위해 개발된 FRP Hybrid Bar의 구조적 성능은 검증 되었으나 부식에 대한 저항성을 평가한 연구는 미흡하여 본 연구를 수행 하였다. 본 연구에서는 콘크리트 중의 철근 종류에 따른 철근부식 저항성을 평가하였다. 평가를 위하여 철근은 일반철근과 FRP hybrid Bar을 사용하였다. 시험방법은 갈바닉전류(Galvanic)와 반전지법(Half-Cell)을 사용하였으며, 철근부식 촉진을 위하여 콘크리트 내부에 염분을 0%, 1.5%, 3%, 6% 첨가하였다. 그 결과 갈바닉전류측정에서 FRP Hybrid Bar는 부식전류가 측정되지 않았다. 반면 일반철근에서는 시험 직후 부식이 발생하는 결과를 나타내었다. Half-Cell측정 결과에서도 4단계의 염분 함유량의 차이와 광물질혼화재료를 사용한 콘크리트와 일반콘크리트에 관계없이 FRP Hybrid Bar의 부식 저항성이 우수하게 나타났다. 따라서 FRP Hybrid Bar는 해양환경 및 철근부식이 예측되는 구조물용 대체 철근으로 사용이 가능하나 부착성능, 탄성계수, 절곡부의 강선 노출에 대한 처리 방법 등이 개선되면 염해로부터 철근부식 저항성 확보를 요구하는 구조물에 사용 할 수 있는 우수한 소재로 판단된다.

  • As Utility interactive fuel cell systems are widely used, it is required for each power conditioning system(PCS) to have higher generating performance and more stable connecting characteristics. This study is focused to minimization of power losses and hence higher efficiency related to the new half bridge type 3-stage utility interactive PCS topology. The loss factor of half-bridge converter becomes only 1.2[%] under the rated load, and hence total efficiency is maintained to be higher as 91[%].

Half Hanning 윈도우 전처리를 통한 기저 세포암 자동 검출 성능 개선 (Performance Improvement of Automatic Basal Cell Carcinoma Detection Using Half Hanning Window)

  • 본 연구에서는 일반적으로 잘 알려진 기저 세포암 검출을 위한 간단한 전처리 방법을 제안하였다. 전처리 과정은 half Hanning 윈도우와 함께 데이터를 클리핑하고 PCA(principal components analysis)를 이용하여 차원을 감소하였다. Half Hanning 윈도우는 $1650cm^{-1}$ 피크 부근의 크기를 낮춤으로써 음성 오류율을 줄여 분류 성능을 향상시켰다. 이 실험에서 사용한 MAP(maximum a posteriori), KNN (k-nearest neighbor), PNN(probabilistic neural network), MLP(multilayer perceptron), SVM(support vector machine)와 MSE(minimum squared error)의 분류결과는 제안한 방법이 효과적임을 입증하고 있다. KNN 분류방법은 216개 라만 스펙트럼에 대한 분류실험에서 민감도가 약 97.3%로 제안한 윈도우를 적용한 이 실험에서 기저 세포암 검출 성능이 가장 많이 개선되었다.

The Initial Irreversible Capacity of the Lithium Ion Battery System Using by the Gradual Control of State of Charge

  • Electrochemical characteristics of a graphite/lithium and a $LiCoO_2/lithium$ half cell and a $graphite/LiCoO_2$ full cell were analyzed using a GCSOC (gradual control test of the state of charge) technique. The IIE (initial intercalation coulombic efficiency), which represents lithium intercalation property of the electrode material, and the $lIC_s$ (initial irreversible capacity by the surface), which represents irreversible reaction between the electrode surface and the electrolyte were obtained from the GCSOC analysis. Linear-fittable capacity ranges of IIE of graphite and $LiCoO_2$ electrodes were 370 and 150 mAh/g, respectively, based on material weight. The value of lIE for graphite and $LiCoO_2$ electrodes were $93-94\%$ and $94-95\%$, respectively. The value of IICs for graphite and $LiCoO_2$ electrodes were 15-17 mAh/g and 0.3-1.7 mAh/g, respectively. The value of IIE for $graphite/LiCoO_2$ full cell, used GX25 and DJG311 as a graphite, was $89-90\%$ that lower than that for the half cells. Parameters of IIE and IICs can also be used to represent not only half cell but also full cell.

토양 미생물의 2-hydroxypyridine 대사에 미치는 기아상태와 부식산의 영향 (Influence of Starvation and Humic Acid on Soil Microbial 2- Hydroxypyridine Metabolism)

  • 본 연구에서는 기아상태에 있는 토양 미생물, Arthrobacter crystallopoietes cell의 2-hydroxypyridine대사능력의 변화 와 이 미생물의 대사능력에 대한 부식산의 영향에 대해 다루어졌다. 기질 2-HP에 대한 대수기 세포(exponential phase cell)는 적응기 세포(lag phase cell)와 비교할 때 기아상태에 있어서도 더 높은 2-HP대사능력을 보여 기아상태 3일 후 대수기 세포에서 2-HP반감기는 14시간으로 나타난 반면 적응기 세포의 경우 46.5시간으로 나타났다. 부식산은 기아상태에서 이 미생물 세포의 유도효소, 2-HP monooxygenase의 안정성을 높여주어 기아시간 2일 후 표준조건에서 기아상태에 있던 미생물의 효소활성이 처음의 1.5%로 남아 있는 반면 0.2% 부식산 용액에서 기아상태로 있던 미생물의 효소활성은 12%까지 남아 있었다. 기아시간 14일 후 까지도 부식산 용액 속에서 기아상태로 있던 이 미생물 세포의 2-HP대사능력은 표준 조건의 것에 비해 월등히 높아 2-HP반감기를 비교해 보면 표준 조건의 경우 43시간인 반면 부식산의 경우 1.25시간으로 나타났다.

Pseudomonas putida에서 생산된 전세포 benzoylformate decarboxylase의 활성특성 및 고정화 캡슐 제조 (Characteristic of whole cell benzoylformate decarboxylase from Pseudomonas putida)

  • Mandelate pathway를 거치는 Pseudomonas putida(KCTC 1751)의 전세포 benzoylformate dcarboxylase를 이용하여 benzoylformate를 benzaldehyde로 변환하였고 성장배지의 조성이 cell내부에 축적되는 benzoylformate dcarboxylase의 양에 미치는 영향을 조사하였다. 전세포효소의 재사용을 위하여 calcumalginate 캡슐 고정회법을 이용하여 캡슐고정화 Pseudomonas putida를 제조하였다. 캡슐 고정화 미생물을 M3배지에서 3일간 배양한 후 M1배지에서 1일간 배양한 결과 77.75g/L의 미생물 건조중량을 얻었다. 캡슐 고정화 전세포 benzoyltormate decarboxylase의 비활생도는 자유배양에 의한 전세포효소의 비활성도에 비해 약 1/2값을 나타내었으며 캡슐 고정화 전세포 benzoyltormate decarboxylase를 20회 재사용시 20%의 실활을 보였으며 캡슐 재사용 30회 이후 미생물의 건조중량은 약 10% 감소를 보였다.
