Analysis of the Stability of HLA-A2 Molecules Expressed on the Cell Surface

  • Association of antigenic peptide with class I MHC is believed to be crucial for maintaining stable conformation of class I molecules. T2 cells that are defective in TAP gene function mainly express class I molecules with an unstable conformation due to little or no association with antigenic peptides, whereas T1 cells that are normal in TAP gene function mainly express the stable form of class I molecules. In this work, attempts were made to determine the molecular stability of stable and unstable class I molecules. Dissociation of HLA-A2 molecules on T1 and T2 cells was monitored by flow cytometry using anti-HLA-A2 antibody after the cells were treated with brefeldin A to shut down the transport of newly-assembled HLA-A2. Estimated dissociation rate constants for the stable and unstable forms of HLA-A2 were 0.076 $h^{-1}$ and 0.66 $h^{-1}$, respectively. It appeared that both T1 and T2 cells express stable and unstable class I complex, but with different ratios of the two forms. Furthermore, $interferon-{\gamma}$ treatment of T1 cells appeared to induce the expression of both the stable and unstable class I molecules. These results demonstrate that class I MHC molecules can be divided into two groups in terms of structural stability and that they exist on the cell surface in both forms in a certain ratio.

  • Recently we have shown based on Lyapunov theorem that the closed loop system with the constrained infinite horizon H$\infty$ optimal controller is exponentially stable. moreover the on-line feedback implementation of the constrained infinite horizon H$\infty$ optimal control based on quadratic programs has been proposed. n this paper we summarize and discuss these results.

  • This experiment were carried out to investigate the conditions of amylase produced by Candida muscorum in wheat bran cultures and the properties of its amylase (crude enzyme). The results obtained were as follows. 1. The optimum conditions for amylase production in wheat bran cultures were; water content 75 percent, temperature $25^{\circ}C$ and incubation time 4-7 days. 2. The production of amylase was increased about 20 percent in the medium added 0.5 percent of ammonium sulfate or ammonium chloride to wheat bran, but the production of those was decreased in the case of addition of nitrates. 3. No significant effect was found in the case of the addition of carbon source on the production of amylase. 4. The properties of liquefying amylase of the selected strain were; the optimum pH 4.2, the optimum temperature $60-65^{\circ}C$, the stable pH 3.2-6.8 and the stable heating (for 15 minutes heating) below $65^{\circ}C$. 5. The properties of saccharifying amylase of the selected strain were; the optimum pH 4.5, the optimum temperature $55^{\circ}C$, the stable pH 3.8-6.2 and the stable heating (for 15 minutes heating) below $45^{\circ}C$.

Theoretical Study of Acetic Acid-Sulfur Dioxide Complexes (Acetic Acid-Sulfur Dioxide 복합체에 대한 이론 연구)

  • The formation of complexes between SO2 and acetic acid was studied theoretically. The ab initio and DFT calculations were performed with MP2 and B3LYP methods using 6-311++G(d,p), aug-cc-pVDZ and aug-cc-pVTZ basis sets. Six stable complexes were identified, and three stable bidentate complexes, C1, C2 and C3, were formed between SO2 and syn-acetic acid, which is more stable form of acetic acid. Anti-acetic acid also form three complexes, C4, C5 and C6, with SO2. C4 is bidentate and C5, C6 are monodentate complexes, which are less stable. The most stable complex, C1 has S⋯O=C and O⋯H-O interactions, and the S⋯O and O⋯H distances are less than the sum of van der Waals radii. The vibrational frequencies of complexes were calculated and were compared with those of monomers. The frequency shifts after formation of complex were found, and the overall pattern of frequency shifts relative to monomers is similar among the six complexes.

A Study on the Thermal Cycling Effect on the Hydrogenation Kinetics of Mg2Cu (Mg2Cu 수소저장합금의 thermal cycling 효과에 관한 연구)

  • The effect of thermal cycling on the hydrogenation characteristics of the $Mg_2Cu-H$ system was investigated in order to study of intrinsic degradation of the system. The hydrogen storage capacity decreased with thermal cycling from $573^{\circ}K$ to $663^{\circ}K$. By the thermal analysis it is found that stable $MgH_2$ hydride is formed during thermal cycling. With a heat treatment at $693^{\circ}K$ at a hydrogen pressure of 16 atm, the hydrogenation rate drastically decreased. From these observation, it suggested that the intrinsic degradation of $Mg_2Cu$ system results from mainly the formation of stable $MgH_2$ hydride phase.

  • Polyphenol oxidase (PPO) was isolated from the flesh of Fuji apples by DEAE-Cellulose, ammonium sulfate precipitation, phenyl-Sepharose CL-4B, and Sephdex G-100 chromatography. The molecular mass of the purified PPO was estimated to be 40 kDa by SDS polyacrylamide gel electrophoresis. With regard to substrate specificity, maximum activity was achieved with chlorogenic acid as substrate, followed by catechin and catechol whereas, there was no detectable activity with hydroquinic acid, resorcinol, or tyrosine as substrate. The optimum pH and temperature with catechol as substrate were 6.5 and $35^{\circ}C$, respectively. The enzyme was most stable at pH 6.0 and unstable at acidic pH. The enzyme was stable when it was heated to $45^{\circ}C$ but heating at $50^{\circ}C$ for more than 30 min caused 50% loss of activity. Reduced $ZnSO_4$, L-cystein, epigallocatechin-3-o-gallate (EGCG), and gallocatechin gallate (GCG) also inhibited activity.

  • An Experimental study was carried out to develop a simple method of processing copper waste sludge which is produced by PBC manufacturing. The procedure is based on leaching of wet sludge in 2N H$_2$SO$_4$, and the solid / liquid ratio is controlled approximately at 1/10. The recovery of copper is 85.4%, and pH of the leachate is 3.20. Adding ammonia solution into leachate forms ammine, and hydroxide compounds derived from other impurities in leachate at pH 10. The hydroxide compound can be treated by ferrite process, and the product is a stable oxide compound. Then the ammine solution is heated to evaporate ammonia, and the copper hydroxide is formed. Heating at 8$0^{\circ}C$by aeration, copper hydroxide is transformed into copper oxide with a purity of 98.4%. This process can recover most copper from sludge and the residue can be stabilized by the formation of a stable oxide compound which is not hazardous to environment.

  • To obtain cytochrome $c_3$ labeled with a stable isotope, the conditions of cultivation and the composition of medium for DvMF were examined. The growth of DvMF was steady and reproducible under purging with $N_2$ and under pH control. DvMF was able to go on a defined medium without natural products. The composition of medium containing a small amount of $NH_4C$l as sole nitrogen source was established. Then, uniformly $^{15}N$labeled cytochrome $c_3$ was obtained during the culture of DvMF in a defined medium with $^{15}NH_4$Cl; it was confirmed by $^1H-^{15}N$ HMQC.

  • Quality evaluation of laser welded MCFC separator panels with large active area was performed. Lap joint was applied to produce stable air-tight seam weld by employing Nd:YAG laser. Results showed that surface contamination played a key role to form weld defects at the joint interface. However, there was no evidence that weld width at the lap joint changed after the stack operation time of 2,890h. Test results also revealed that the reinforcement values which were measured on the weld line after long time operation, were stable.
