Analysis of Stomach Contents of Marine Orgnaisms in Gwangyang Bay and Yeosu Fish Market Using DNA Metabarcoding (DNA 메타바코딩을 이용한 광양만 및 어시장 해양 생물 위 내용물 분석)

  • Gut contents analysis is essential to predict the impact of organisms on food source changes due to variations of the habitat environment. Previous studies of gut content analysis have been conducted using traditional methods, such as visual observation. However, these studies are limited in analyzing food sources because of the digestive process in gut organ. DNA metabarcoding analysis is a useful method to analyze food sources by supplementing these limitations. We sampled marine fish of Pennahia argentata, Larimichthys polyactis, Crangon affinis, Loligo beka and Sepia officinalis from Gwangyang Bay and Yeosu fisheries market for analyzing gut contents by applying DNA metabarcoding analysis. 18S rRNA v9 primer was used for analyzing food source by DNA metabarcoding. Network and two-way clustering analyses characterized the relationship between organisms and food sources. As a result of comparing metabarcoding of gut contents for P. argentata between sampled from Gwangyang Bay and the fisheries market, fish and Copepoda were analyzed as common food sources. In addition, Decapoda and Copepoda were analyzed as common food sources for L. polyactis and C. affinis, respectively. Copepoda was analyzed as the primary food source for L. beka and S. officinalis. These study results demonstrated that gut contents analysis using DNA metabarcoding reflects diverse and detailed information of biological food sources in the aquatic environment. In addition, it will be possible to provide biological information in the gut to identify key food sources by applying it to the research on the food web in the ecosystem.

Assessment of Metal Pollution of Road-Deposited Sediments and Marine Sediments Around Gwangyang Bay, Korea (광양만 내 도로축적퇴적물 및 해양퇴적물의 금속 오염 평가)

  • In this study, heavy metal in road-deposited sediments (RDS) and marine sediment around Gwangyang Bay area have been investigated to assess the pollution status of metals and to understand the environmental impact of RDS as a potential source of metal pollution. Zn concentration for <63 ㎛ size fraction was the highest (2,982 mg/kg), followed by Cr, Ni, Pb, Cu, As, Cd, and Hg. Metal concentrations in RDS increased with decreasing particle size and relatively higher concentrations were observed around the metal waste and recycling facilities. For particle size in RDS smaller than 125 ㎛, EF values indicated that Zn was very high enrichment and Cr, Cd, Pb were significant enrichment. The concentrations of metals in marine sediments were mostly below the TEL value of sediment quality guidelines of Korea. However, the Zn concentrations has increased by 30~40% compared to 2010 year. The amounts of Zn, Cd and Pb in less than 125 ㎛ fraction where heavy metals can be easily transported by stormwater runoff accounted for 54% of the total RDS. The study area was greatly affected by Zn pollution due to corrosion of Zn plating materials by traffic activity as well as artificial activities related to the container logistics at Gwangyang container terminal. The fine particles of RDS are not only easily resuspended by wind and vehicle movement, but are also transported to the surrounding environments by runoff. Therefore, further research is needed on the adverse effects on the environment and ecosystem.

  • 한반도 서ㆍ남해안은 크고 작은 만과 해빈, 삼각주, 하구, 조간대 등 다양한 연안해역이 잘 발달 되어 있다 평균조차는 서해안 남부에서 약 3m로 북쪽으로 갈수록 증가하여 군산 부근에 6m, 인천 부근에서 약 8m에 달하며, 남해안의 평균조차는 부산 1.2m, 진해 2.1m, 충무 2.6m, 삼천포 3.0m, 여수 3.3m, 완도 3.5m, 목포 3.7m로 조석 간만의 차가 크게 나타나서 연안해역에 폭넓은 조간대가 형성되어 있다. (중략)

Effects of Polycyclic Aromatic Hydrocarbons Contamination on the Community Structure of Microorganisms in Gwangyang bay Sediments (다환방향족 탄화수소가 광양만 퇴적토의 미생물 군집구조에 미치는 영향)

  • Effect of polycyclic aromatic hydrocarbons (PAHs) on the community structure of indigenous microorganisms in Gwangyang Bay sediments was investigated in Mar. & Aug.,2000. Microbial community structure was analyzed using 5'-terminal restriction fragment length polymorphism (T-RFLP) method. Microbial community structure based on T-RFLP method revealed that community differentiated by sampling period except station 1 located near the stream discharge site from Yeosu Industrial Complex. Even, microbial diversity was higher at stations showed relatively high concentrations of PAHs. The microbial community structure was severely changed during the enrichment culture with 1,000 ppm of PAHs mixture. It was also different between cultivated at 8$^{\circ}C$ and 30$^{\circ}C$. The results implied that temperature, poyosity, organic content and etc were more responsible than PAHs on the microbial community structure.
