  • Exponentially growing cells of S. cerevisiae were cultivated in the concomitant presence of $2{\times}10^{-3}\;M$ paraquat for 12 hours. Cellular growth became slower as the time passed and then, it was completely inhibited after 6 hours of cultivation. More than 70 percent of the yeast cells were killed with in 12 hours of paraquat treatment. It was observed that size of the yeast cell varied from $4.5{\times}5.5{\um}m$ to $2.5{\times}3.5{\um}m$ in diameter, while that of control was uniform. The number of budding cells became rare, Cell wall of the paraquat treated cell was thinner$(0.15{\mu}m)$ than that of control cell$(0.20{\mu}m)$. Outer part of the cell wall had higher density than inner part. Membrane structures such as plasma membrane and mitochondria was decomposed during the paraquat treatment for 12 hours.

Electricity Generation and Microbial Community variation in Microbial Fuel Cell with various Electrode Combinations. (다양한 탄소전극조합에 따른 미생물 연료전지의 전기발생량 및 미생물 군집변화)

  • The electrode material is one of the factors affecting the power production of microbial fuel cell. In this study, effects of carbon electrode material, thickness and configuration on the power density, biofilm formation and microbial community diversity of microbial fuel cell were investigated. To optimize the anode-cathode electrode assembly, seven lab-scale reactors which had various carbon electrode constructions were operated in continuous mode. Under the steady state condition, the electrode combination of graphite felt (6 mm) with hole showed the highest cell voltage of 238 mV and the coulombic efficiency of 37%. As a result of SEM analysis, the bacteria growing on surface of knitted type of carbon cloth and graphite felt electrode ncreased significantly. The change of dominant species between seeding sludge and biofilm on the surface of anode electrode, microbial analysis with PCR-DGGE showed that the dominant species of seeding sludge are quite different from those of biofilm on the surface of each anode electrode. Especially Geobacter sp., a well known electrochemical bacteria, was found as the dominant species of the electrode combination with graphite felt.

  • Among the renewable energy resources, wind power is growing rapidly in terms of technological development and market share. Recently, onshore wind farm have been affected by limitations of terrestrial space and environmental problems. Consequently, installation sites have been moved to the sea, and the development of floating offshore wind farms that are installed at deep waters with more abundant wind conditions is actively underway. In the context of maritime traffic, the optimal site of offshore wind farms is required to minimize the interference between ships and wind turbines and to reduce the probability of accidents. In this study, genetic algorithm based AIS(Automatic Indentification System) data composed of genes and chromosomes has been used. The optimal site of floating offshore wind farm was selected by using 80 genes and by evaluating the fitness of genetic algorithm. Further, the final site was selected by aggregating the seasonal optimal site. During analysis, 11 optimal site were found, and it was verified that the final site selected usng the genetic algorithm was viable from the perspective of maritime traffic.

  • Background : Normal cell proliferation and viability is strongly depends on the availability of metabolic energy and the maintenance of the appropriate adenylate-nucleotide pools. Hypothetically, changes in adenylate kinase (AK) expression could therefore be associated with adaptation to altered growth characteristics or inversely altered growth characteristics of proliferating cells could drive the changes in the metabolic profile. This study investigated whether the expression of either AK1 or a Mycobacterium tuberculosis adenylate kinase mutant which has the same catalytic activity of AK1 could affect the growth rate of slow-growing BCG. Method : Recombinant BCGs, which were cloned the human muscle-type adenylate kinase synthetic gene (AK1) and adenylate kinase mutation gene (AKmtDM) of Mycobacterium tuberculosis into the Mycobacterium/E.coli expression vectors, were constructed. Recombinant BCGs and wild-type BCG were cultured in 7H9 media and the optical density at 600nm was measured at intervals of 2-3 days. Result : There wasn't the growth rate change induced by AK1 or AKmtDM expression in recombinant BCGs. Conclusion : The expression of AK1 or Mycobacterium tuberculosis adenylate kinase mutant in BCG does not affect the growth rate of BCG.

  • The objective of this study was developing dynamic stand growth model to predict diameter at breast height (DBH) growth by thinning intensity and cycle for major tree species of South Korea. The yield table, one of static stand growth models, constructed by Korea Forest Service was employed to prepare dynamic stand growth models for 8 tree species. In the process of model development, the thinning type was designated to thinning from below and equations for predicting the DBH change after thinning by different intensities was generated. In addition, stand density (N/ha), age and site index were adopted as explanatory variables for DBH prediction model. Thereafter, using the model, DBH growth under various silvicuture through integrating such equations considering thinning intensities, and cycles. The dynamic stand growth model of DBH developed in this study can provide understanding of effectiveness in forest growth and growing stock when thinning practice is performed in forest. Furthermore, results of this study is also applicable to quantitatively assess the carbon storage sequestration capability.

  • Silica is known as a promising osteoconductive material, and polycaprolactone is a bioactive and degradable material. The purpose of this study was to monitor the differentiation of MC3T3-E1 cells cultured on the layer-built silica/poly caprolactone non-woven fabric produced by electrospinning. Non-woven fabric (silica, polycaprolactone, PSP, SPS) was made by electrospinning and they were inserted in the 48 well cell culture plate. MC3T3-E1 cells were prepared by subculture. Cells were seeded to each well $1{\times}10^5$ concentration per well. Dulbecco's modified eagle medium with 10% FBS and 1% antibiotic-antimycotic solution was used. Confocal laser scanning microscope was taken 4 hours after incubation (95% air. 5% $CO_2$, $37^{\circ}C$). Cell proliferation was monitored by spectrophotometer on 1, 7, 14 days, and the morphology of the growing cells was observed by field emission scanning electron microscope. To monitor the differentiation of osteoblasts on the materials, MC3T3-E1 cells were incubated in 48 well culture plate after seeding with the density of $1{\times}10^5$ concentration. Then ELISA kit & EIA kit were used on to assess osteocalcin and osteopontin expression respectively. The other conditions were the same as above. MC3T3-E1 cells were proliferated well on all of the materials. There were no statistical differences among them. The osteopontin expression of silica, PSP, SPS was significantly higher than other groups on day 3 (p/0,05), but after that time, there were no statistically signigicant differences. The osteocalcin expression was significantly higher in silica and PSP than other groups on day 14. These findings show that PSP was as good as silica on the effect of osteoblast differentiation. The PSP non-woven fabric may have the possibility as bone graft materials.

Estimating the Spatial Distribution of Forest Stand Volume in Gyeonggi Province using National Forest Inventory Data and Forest Type Map (국가산림자원조사 자료와 임상도를 이용한 경기지역 산림의 임분재적 공간분포 추정)

  • Reliable forest statistics provides important information to meet the UNFCCC. In this respect, the national forest inventory has played a crucial role to provide the reliable forest statistics for several decades. However, the previous forest statistics calculated by administrative district has not provided spatial information in a small scale. Thus, this study focused on developing models to estimate an explicit spatial distribution of forest growing stock. For this, first, stand volume model by stand types was developed using National Forest Inventory(NFI) data. Second, forest type map was integrated with this model. NFI data were used to calculate plot-level stand volume and basal area. The stand types of NFI plot including the species composition, age class, DBH class and crown density class are very crucial data to be connected with forest type map. Finally, polygonlevel stand volume map was developed with spatial uncertainty map. Average stand volume was estimated at 85.7 $m^3$/ha in the study area, and at 95% significance interval it was ranged from 79.7 $m^3$/ha to 91.8 $m^3$/ha.

Environmental Factors on the Development of Root Rot on Ginseng Caused by Cylindrocarpon destructans (인삼 뿌리썩음병 발병에 미치는 환경 요인)

  • The fungus Cylindrocarpon destructans is the cause of root rot in many ginseng production areas in Korea. A total of 57 isolates of C. destructans were recovered from diseased roots in a survey of ginseng-growing fields from 2011-2012. Among these isolates, 37% were classified as highly virulent (causing lesions on unwounded mature roots) and 61% were weakly virulent(causing lesions only on previously wounded roots). Radial growth of highly and weakly virulent isolates on potato dextrose agar was highest at $20^{\circ}C$ and there was no growth at $35^{\circ}C$. Mycelial mass production was significantly (P = 0.05) lower at pH 7.0 compared with pH 5.0. To study the effects of pH (5.0 and 7.0) and wounding on disease development, ginseng roots were grown hydroponically in nutrient solution. Lesions were significantly larger (P < 0.01) at pH 5.0 compared with pH 7.0 and wounding enhanced disease by a highly virulent isolate at both pHs. In artificially infested soil, 2-yearold ginseng roots were most susceptible to Cylindrocarpon root rot among all root ages tested (1 to 4 years) when evaluated using a combined scale of disease incidence and severity. Root rot severity was significantly (P<0.05) enhanced by increasing the inoculum density from $3.5{\times}10^2cfu/g$ of soil to $2.0{\times}10^3cfu/g$ of soil.

Antibacterial Effect of Bacteria Isolated from the Korean Traditional Foods against Pathogenic Bacteria (한국전통식품으로부터 분리 된 세균의 항균활성 효과)

  • Aquaculture continues to be an ever-growing sector. However, high-density farming increases disease outbreaks due to deteriorating water quality and internal stress. To prevent disease, the most common method chemotherapy is using antibiotic administration. In this study, probiotic bacteria were isolated from Korean traditional foods, such a Gochu pickle and cutlassfish salted seafood. Various bacteria were isolated, and their 16S rDNA sequences were analyzed. The antimicrobial activities of four isolates from Gochu pickle and seven isolates from cutlassfish salted seafood were assayed, in addition to the antibacterial activity of culture pellet and supernatant. The antibacterial activity of the pellet was higher than that of the supernatant. Isolate JKM-2 showed the highest antibacterial activity against Streptococcus iniae (43 mm), S. parauberis (40 mm), S. mutans (35 mm), and Vibrio vuinificus (26.5 mm). The sequences of the isolated strains were compared with those of Bacillus subtilis (97.71%), B. tequilensis (97.71%), Brevibacterium halotolerans (97.71%), B. subtilis (97.63%), B. subtilis (97.63%), B. mojavensis (97.54%), B. vallismortis (97.46%), B. nanillea (97.45%), B. methylotrophicus (97.37%), and B. ssiamensis (97.37%). Future through analysis and new strains confirmed the bacterial cell material investigation of JKM-3, and to ensure sufficient stability, it is desired to verify the utility value as a substitute material for antibiotics by application to the form of the industry.

  • This study is to investigate the effect of split application and adapted fertilization application cultivation in stable medicinal herbs production of Acorus graminens S. And it accomplished one's purpose to GAP guide manual established standard method of cultivation technology, contribute to the enlargement of farm's income. Method of fertilization application were conducted under five condition compose to standard application(N-P-K-Compost: fermented mixed oil cake fertilizer applied at $4-2-6-300kg.10a^{-1}$, twofold application(N-P-K-Compost applied at $8-4-12-600kg.10a^{-1}$) cultivation. Compost (fermented mixed oil cake fertilizer) and fused phosphate applied at 100% of basal fertilizer in April 14. The split application level of standard application and twofold application(N-K) applied at 100%, 50%, 40% of basal fertilizer in April 14, 50% and 40% of top dressing were three times application (N-K) in June 1, July 15, August 15, 25% and 20% respectively. Transplanting with Naju varieties in May 12 by growing pot seedling for 40 days in 2015. Planting density were spaced 30 cm apart in rows 30 cm apart with black vinyl mulching on open cultivation. Plot design randomized block 3 repetition. Therefore, growth characteristics by 50% and 40% of top dressing were three times application (N-K) compared to standard application and twofold application (N-K) applied at 100% basal fertilizer, aerial part as a result were so many such amount of growth as number of leaf per plant, etc., Length of diameter and main root of subterranean part growth increment rising highest. Dried roots yields treated standard fertilizer 100% application($250kg.10a^{-1}$) compared to 100% from twofold application (N-K) applied at 50% basal fertilizer, 50% and 40% of top dressing were three times application increased by 16%, 20%. respectively.
