Studies on Biological Activity of Wood Extractives (X VIII) -Isolation and Antioxidant Activity of Chemical Constituents from Maackia amurensis- (수목추출물의 생리활성에 관한 연구(X VIII) -다릅나무(Maackia amurensis) 수피의 추출성분의 분리 및 항산화 활성-)

  • The dried barks of Maackia amurensis were ground, extracted with 95% EtOH, concentrated, and fractionated with a series of light petroleum ether, dichloromethane, ethyl acetate and water on a separatory funnel. Each fraction was concentrated, then a portion of dichloromethane and ethyl acetate soluble was chromatographed on a Sephadex LH-20 and silica gel 60 column using a various solvent system as eluents. The isolated compounds were identified by cellulose TLC, $^1H-$, $^{13}C-NMR$, COSY, NOESY, HMQC, HMBC, FAB and EI-MS. The structures were determined as: 7-O-$\beta$-D-glucopyranosyl-4'-methoxyisoflavone, 7-O-$\beta$-glucopyranosyl(1'''->6'')-$\beta$-D-glucopyranosyl-4'-methoxyisoflavone, 7-O-$\beta$-D-glucopyranosyl(1''''->6''')-$\beta$-D-glucopyranosyl(1'''->6'')-$\beta$-D-glucopyransoyl-4'-methoxyisoflavone, 7-O-$\beta$-D-glucopyranosyl-4', 6-dimethoxyisoflavone. The Free radical scavenging activity using DPPH of the isolated compounds were similar with that of BHT but lower than of $\alpha$-tocopherol.

Analysis of soil moisture response due to Eco-hydrological change (생태수문 변화에 따른 토양수분의 영향 분석)

  • The main objective of this study is to estimate of the vegetation response induced by climate change to soil moisture. We investigated a relationship between vegetation activity and climate variables using Moderate Resolution Imaging Spectroradiometer (MODIS)-retrieved Normalized Difference Vegetation Index (NDVI) and soil moisture. NDVI which extracted from MODIS 13 Vegetation Indices Product was considered as an useful parameter to figure out a relationship with two types of soil moisture, which were observed at Rural Development Administration sites and estimated from Advanced Microwave Scanning Radiometer E (AMSR-E) satellite imagery. The correlation of MODIS-NDVI and ground measured soil moisture were observed, became much stronger when compared to soil moisture values with time lag (5days, 10days, 15days). The correlation patterns between NDVI and soil moisture with different time lag were related to soil texture. The results from this study will be useful to understand the role of vegetation in water balance control in various scales from regional to global climate change.

Effect of Content of Potato Tuber Component and Potato Variety on the Bioethanol Production (감자의 성분함량과 품종이 바이오에탄올 생산에 미치는 영향)

  • The contents of starch, moisture, crude protein, crude fat, crude fiber, and ash of different varieties of potatoes were analyzed. The average starch contents of Go-woon, Ha-ryoung, Dae-seo, Jo-won, Ga-won potatoes were $17.9{\pm}0.2$, $18.0{\pm}1.7$, $17.7{\pm}0.5$, $14.8{\pm}0.4$, and $16.2{\pm}1.0%$, respectively. The ground powder of each starchy substrate was suspended in distilled water, and then liquefied, saccharified, and fermented by Saccharomyces cerevisiae ATCC26603 at $30^{\circ}C$ for 4 days. By statistical analysis, the effectiveness of the contents of the different components of the potato tubers on the ethanol production were examined. The results showed that the starch content positively affected the ethanol production. while moisture content affected negatively the ethanol production. Ethanol production from the 5 different varieties of potato tubers harvested on different time were examined and the results indicated that both of potato variety and the harvesting-time significantly affected the ethanol production. Among the several varieties of potato, Ha-ryoung produced the highest yield of ethanol as much as $94.3{\pm}1.9$ L/ton or $3111{\pm}62.7$ L/ha.

  • The contents of starch, moisture, crude protein, crude fat, crude fiber, and ash of different varieties of various crops such as brown rice, barley, corn, sweet potato were analyzed. The average starch contents of brown rice, barley, corn, and sweet potato were $70.1{\pm}0.4\;{\sim}\;72.2{\pm}2.1$, $68.7{\pm}0.2\;{\sim}\;71.4{\pm}1.2$, $67.6{\pm}0.8\;{\sim}\;69.4{\pm}1.8$, and $21.7{\pm}0.9\;{\sim}\;28.3{\pm}0.5%$, respectively. The ground powder of each starchy substrate was suspended in distilled water, and then liquefied, saccharified, and fermented by dried active yeast at 32 for 4 days. By statistical analysis, the effectiveness of the contents of the different components such as starch, moisture, crude protein, crude fat, crude fiber, and ash of the crops on the ethanol production were examined. The results showed that the starch content positively affected the ethanol production in all the tested cereals and sweet potato. In brown rice, ash content affected negatively the ethanol production. In barley, protein content affected negatively the ethanol production, while fiber content affected positively the ethanol production. The sweet potato containing higher content of moisture produced less amount of ethanol.

  • This study was conducted to obtain basic information for soil improvement in flower crop cultivating greenhouse soil through survey on the chemical and physical properties of greenhouse soils. Total of 85 Flowcultivating farms were surveyed and analysis was done on the soil characteristics, amounts of chemical fertilizer and soil amendmentuse. The result are as follows: In soil properties of flower cultivating greenhousees, silt clay loam was 51%and 68% of the surveyed soils had good drainage condition. Ground water table was over 90-120cm which was optimum range for flower cultivation. Flower cultivating farms had problem with accumulation of fertility. Nitrate nitrogen was accumulated in Gypsophila paniculate farms and available phosphorus, and exchangeable postassium were significantly higher in greenhouse soils about 2 times than in open field soil. Application amount of chemical feltilizers in greenhouses were nitrate 211,phosphorus 135, and potassium 132kg/ha, respectively. Amount of organic matter used in greenhouse were high in order of cattle manure> compost> organic fertilizer> poultry manure> swine manure and their application amounts were69, 103, 32, 20, and 43 MT/ha, respectively.

Physicochemical Characteristics and Antioxidant Activity of Dutch Coffee Depending on Different Extraction Conditions and Storage (추출 및 저장 조건에 따른 더치커피의 이화학적 특성 및 항산화 활성)

  • This study was designed to evaluate the changes in the physicochemical properties and antioxidant activity of Dutch coffee (cold brew) under different conditions of extraction and storage. Dutch coffee was extracted from ground coffee soaked in water at 4 or $20^{\circ}C$ and stored for 8 weeks at 4 or $20^{\circ}C$. The storage temperature affected the decline in pH and increase in acidity compared to the extraction temperature. The total phenol content partly decreased during the storage period. As the extraction temperature increased, the ABTS [2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid)] and DPPH (2,2-diphenyl-1-picrylhydrazyl) radical-scavenging activities also increased; in fact, DPPH radical-scavenging activity showed a general increase. As the storage time prolonged, the caffeine content decreased, but the contents of caffeic acid and chlorogenic acid increased. The results for all kinds of samples indicated that the general bacterial count was <1 CFU/mL, which indicated that the coffee can be stored for and consumed within 8 weeks.

Effect of Alfalfa Extract, It's Concentration and Absorbents on Germination and Growth of Alfalfa (알팔파 추출물, 추출농도 및 흡착제 처리가 알팔파 발아와 생육에 미치는 영향)

  • Crude aqueous extracts from dried leaves, stems, roots, and flowers from both field grown and greenhouse grown alfalfa plants inhibited alfalfa seed germination and seedling growth. The degree of inhibition was greater in the field grown plant extracts. Flowers extract of field grown plant most inhibited alfalfa germination and seedling growth. In the concentration study, the highest concentration of extract (9.0%, w/v) significantly inhibited total alfalfa seed germination by 50% as compared to control. In partitioning study using pot hydroponic culture of plant biomass into leaves, stems, root, LAR products of LWR and SLA exhibited significant variation among four species. This result support that the inhibitory effect of autotoxic substances presenting in alfalfa tissue may be possible interference with the patitioning of biomass into leaf component relative to the total biomass produced by the alfalfa plant. Toxicity of extract was not reduced by adding activated charcoal, Dowex-50W, amberlite to the extract. Toxic substances existing in most plant tissues but mainly above ground foliage are water soluble and stable and may persist in old alfalfa fields. Thus, it is recommended to remove as much as possible of the above growth parts, especially vegetative stage, before one tries to re-establish alfalfa in former field of alfalfa.

Geometric Analysis of Fracture System and Suggestion of a Modified RMR on Volcanic Rocks in the Vicinity of Ilgwang Fault (일광단층 인근 화산암 암반사면의 단열계 기하 분석 및 암반 분류 수정안 제시)

  • The properties of fracture system on road-cut slopes along the Busan-Ulsan express way under construction are investigated and analyzed. Fracture spacing distributions show log-normal form with extension fractures and negative exponential form with shear fractures. Straight line segments in log-log plots of cumulative fracture length indicate a power-law scaling with exponents of -1.13 in site 1, -1.01 in site 2 and -1.52 in site 3. It is likely that the stability and strength of rock mass are the lowest in site 1 as judged from the analyses of spacing, density and inter-section of fractures in three sites. In contrast, the highest efficiency of the fracture network for conducting fluid flow is seen in site 3 where the largest cluster occupies 73% through the window map. Based on the field survey data, this study modified weighting values of the RMR system using a multiple regression analysis method. The analysis result suggests a modified weighting values of the RMR parameters as follows; 18 for the intact strength of rock; 61 for RQD; 2 for spacing of discontinuities; 2 for the condition of discontinuities; and 17 for ground water.

Risk Assessment of the Road Cut Slopes in Gyeoungnam based on Multiple Regression Analysis (다중회귀분석을 통한 경남 지방도로 절취사면의 안정성평가)

  • The purpose of this study is to capture the essentials in survey and evaluation scheme which are able to assess the hazard of a rock slope systematically. Statistical analysis are performed on slope instability parameters related to failure of the rock slope. As the slope instability parameters, twelve survey items are considered such as tension crack, surface deformation, deformation of retaining structures, volume of existing failures, angles between strike of discontinuity and strike of cut slope face, angles between dip of discontinuity and dip of cut slope face, discontinuity condition, cut slope angle, rainfall or ground water level, excavation condition, drainage condition, reinforcement. A total of 233 road cut slopes located in Gyeongnam were considered. The stability of the road cut slopes were evaluated by estimating the slope instability index(SII) and corresponding stability rank. 126 rock slopes were selected to analyze statistical relation between SII and slope instability parameters. The multiple regression analysis was applied to derive statistical models which are able to predict the SII and corresponding slope stability rank. Also, its applicability was explored to predict the slope failures using the variables of slope instability parameters. The results obtained in this study clearly show that the methodology given in this paper have strong capabilities to evaluate the failures of the road cut slope effectively.

Wetland Management Plan on Distributional Characteristics of Vegetation in Hwaeom Wetland (화엄늪 식생의 분포특성에 따른 습지 관리방안)

  • This study was carried out to understand the spatial distribution and conservation, restoration plan for vegetation in the Hwaeom wetland protected area. Total 29 phytosociological releves consisted of 117 plant species (unidentified species contained) was made during field studies conducted at 2008 and 2013 applying Braun-Branquet method. The collected vegetations were arranged 8 plant communities and thus divided hydric types (Drosera rotundifolia-Eleocharis congesta community, Carex forficula-Molinia japonica community) and xeric types (Miscanthus sinensis community, Rhododendron yedoense var. poukhanense community, Tripterygium regelii community, Carex gifuensis-Quercus mongolica community) under moisture conditions. The hydric types growing in hydric conditions distributed along hollowed out ground or the water channel that have a key qualification to protect. In the wetland protected area, the hydric vegetation types were mostly covered about 3.8% and the xeric types dominating by Miscanthus sinensis were most wildly distributed about 51.5%. Various woody plant species invaded the place. In 2008, invasion plants observed with total 14 species - 8 tree species (227 individual), 6 shrub species (51 individual) and alders (Aluns japonica) found in 2013. We suggested finding out that the sustainable conservation and management in Hwaeom wetland should be setting up plan focused on activity and variability such as continuous monitoring (changing vegetation types, spatial distribution, invasion plants) and zonation management (core zone, buffer zone, transition zone).
