Pathogenicity and localization of the tobacco mosaic virus 4.8 kDa protein(oral)

  • In addition to the five well-characterized genes of Tobacco mosaic virus (TMV), this virus contains a sixth open reading frame (ORF6) that encodes a 4.8 kDa protein. TMV ORF6 overlaps the ORFs encoding the 30 kDa movement protein and the adjacent 17.5 kDa capsid protein. Although the 4.8 kDa protein could not be detected in vivo, alteration of the AUG codons of this ORF resulted in a mutant virus that attenuated the virulence of the mutated TMV in Nicotiana benthamiana, but not N. tabacum (tobacco). These sequence changes did not affect either the replication or movement of the mutated TMV. Expression of TMV ORF6 from the virus expression vector Potato virus X (PVX) intensified the virulence of this virus in N. benthmiana, but not tobacco, while expression of TMV ORF6 from the virus expression vector Tobacco rattle virus enhanced the pathogenicity observed in both N. benthamima and tobacco. Thus, the TMV ORF6 is a host- and virus-specific. virulence factor. However, two separate assays indicated that the TMV 4.8 kDa protein was not a suppression of RNA silencing. A fusion protein formed between the TMV 4.8 kDa protein and the green fluorescent protein was expressed from the PVX vector and localized to plasmodesmata. Possible roles of the 4.8 kDa protein in pathogenicity will be discussed

Construction of Environmental Friendly Special-Purpose Ship for the Removal of Blue-green Algae (친환경적 녹조 제거용 특수선박 건조)

  • This study note wished to introduce special-purpose ship for algae removal that is developed by core technology of our country. The ship is consisted of main frame and adjuvant that can attach and detach as cross (+) shape of a character. The characteristics of ship are super light weight and low draft. That is consisted of four devices as suction, collection, filtration and recovering units. Among these, filtration used screen filter (mesh size 30 ${\mu}m$). Also, can separate and remove water and algae by compression air participle notion. Percentage of moisture content of concentrated algal particle was 85%. Water parted with algae finally is exhausted to water area. Removal efficiency that compare by chlorophyll-$\alpha$ concentration was about 57% (inflow: 83.2 ${\mu}g\;L^{-1}$, outflow: 35.8 $[\mu}g\;L^{-1}$) without physical and chemical pretreatment. Forward, need to achieve effect test in various conditions (algal biomass, flow etc.) for efficiency and technological elevation of exclusion device. We wished to contribute in presuppression system construction of massive algal development that manage blue-green algae occurrence area effectively, and prevents spread as lower part of reservoir.

Trend and Development Direction of Restoration Technology Utilization in Ecosystem Conservation Charge Project - Focusing on Implementation Cases from 2014 to 2020 - (생태계보전부담금 반환사업의 복원기술 활용 경향과 방향 - 2014년부터 2020년까지 시행 사례를 중심으로 -)

  • The Ecosystem Conservation Levy (formerly known as the Ecosystem Conservation Cooperation Fund) system has been in place for 20 years, and it can be said that it has now entered the settlement stage. Based on an analysis of publicly available project implementation data from 2014 to 2020, we found that: 1) As the number of return projects increases, the targets of restoration technologies are also strengthening, and it is necessary to frame a series of processes from application, creation, and monitoring for some detailed projects to improve the effectiveness and efficiency of utilizing the levy. 2) Most of the implemented projects are applied as micro-ecosystem creation, but there are many cases where the contents of the project can be seen as other project categories. This shows that the purpose of the return project needs to be approached more clearly and suggests that institutional complementation is needed from the project application stage. 3) The detailed technologies applied tend to be gradually expanding, but most of them are technologies that are not differentiated from general development projects. It is urgent to secure a more technical identity, such as a range and list of utilized technologies suitable for the characteristics and purposes of return projects. 4) It is necessary to establish a relevant evaluation system or framework to utilize the monitoring results of restoration projects. 5) There have been few cases of application of single restoration technologies since the beginning, but the content and scope of the complexity tend to expand in recent years. Even if the objectives are not comprehensive and diverse, it can be seen that many parts of the projects are oriented toward convergence, so it is necessary to conduct separate research on this. 6) As for the direction of improvement of the return project, it is possible to consider expanding the restoration and conservation focus to partially accommodate the complexity of the natural environment and human ecology. It seems that the expansion of restoration technologies that consider the role and function of humans in the natural environment should be explored.

Germ Line Transformation of the Silkworm, Bombyx mori L. with a piggyBac Vector Harboring the Human Lactoferrin Gene (락토페린 유전자도입 piggyBac 벡터에 의한 누에 형질전환)

  • Lactoferrin, an ion-binding 80-kDa glycoprotein, has been suggested to have many biologic activities, such as facilitating ion absorption and having antimicrobial and anti-inflammatory effects. Several of these activities are likely to only be facilitated by human lactoferrin because they depend on the binding of human lactoferrin to specific receptor. To produce recombinant human lactoferrin to animal foods using transgenic silkworm, Bombyx mori L, we have cloned and sequenced the cDNA encoding for a human lactoferrin (HLf) from the mRNA in mammary tumor line (GI-101). As a result, the 2.5-kb fragment of HLf gene was cloned with pGEM-T vector and then this fragment was sequenced. In the nucleotide sequence analysis, single open reading frame of the 2,136-bp encoding for a polypeptide of 712 amino acid residues was detected. On the other hand, we constructed a recombinant plasmid(pPT-HLf), containing human lactoferrin gene for germ line transformation of the silkworm using a piggyBac transposon-derived vector. A nonautonomous helper plasmid encodes the piggyBac transposase. Approximately 6.7% of individuals in the G0 silkworms expressed green fluorescent protein (GFP). PCR analyses of GFP-positive silkworms (G0 and G1) revealed that independent insertions occurred frequently. Furthermore, Western blot analysis showed that the recombinant HLf expressed in hemolymph has the same molecular weight (80 kDa) as a native protein. On the basis of these experiments, expression of HLf in next generation of transgenic silkworm is now in process.

Cloning and Characterization of an Endoglucanase Gene from Actinomyces sp. Korean Native Goat 40

  • A gene from Actinomyces sp. Korean native goat (KNG) 40 that encodes an endo-${\beta}$-1,4-glucanase, EG1, was cloned and expressed in Escherichia coli (E. coli) $DH5{\alpha}$. Recombinant plasmid DNA from a positive clone with a 3.2 kb insert hydrolyzing carboxyl methyl-cellulose (CMC) was designated as pDS3. The entire nucleotide sequence was determined, and an open-reading frame (ORF) was deduced. The ORF encodes a polypeptide of 684 amino acids. The recombinant EG1 produced in E. coli $DH5{\alpha}$ harboring pDS3 was purified in one step using affinity chromatography on crystalline cellulose and characterized. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis/zymogram analysis of the purified enzyme revealed two protein bands of 57.1 and 54.1 kDa. The amino terminal sequences of these two bands matched those of the deduced ones, starting from residue 166 and 208, respectively. Putative signal sequences, a Shine.Dalgarno-type ribosomal binding site, and promoter sequences related to the consensus sequences were deduced. EG1 has a typical tripartite structure of cellulase, a catalytic domain, a serine-rich linker region, and a cellulose-binding domain. The optimal temperature for the activity of the purified enzyme was $55^{\circ}C$, but it retained over 90% of maximum activity in a broad temperature range ($40^{\circ}C$ to $60^{\circ}C$). The optimal pH for the enzyme activity was 6.0. Kinetic parameters, $K_m$ and $V_{max}$ of rEG1 were 0.39% CMC and 143 U/mg, respectively.

Optimum Delivery Frequency for Reducing Construction Logistics Cost under a Carbon Taxation (탄소세 부과시 건설 물류비용 최소화를 위한 운송빈도 최적화)

  • The term 'green' has become an important way of survival for the construction industry in 21th century in accordance with the emergence of the environmental crisis due to the climatic change. Especially the policy of carbon taxation, planed to be introduced from 2012, is expected to be a considerable burden to the construction industry which has abundant carbon emission during the resource transportation due to the complexity of resources and local distribution of the construction sites. In this regard, this study shows an optimizing strategy for delivery frequency, which downsizes the net distribution costs based on the assumption that, despite of its other advantagements, the frequent small lot mode of JIT delivery would take negative effects due to the increase of costs of transportation and carbon emission once the carbon taxation policy carried out. To simulate the efficiency of the management strategies, the System Dynamics modeling has been used. The results show that the frequent small lot transportation strategy is now always efficient method to these changes, and that the frequency of transportation should be re-determinated according to the extent of the imposition of carbon tax. This study provides the conceptual frame for an efficient management of transportation system of the construction industry, showing necessity of change of the resource transportation systems through analysing JIT deliver system in accordance with the global changes in environmental economy.

Joint Design and Strength Evaluation of Composite Air Spoiler for Ship (선박용 복합재 에어 스포일러의 체결부 설계 및 강도 평가)

  • Air spoiler, which can reduce the drag during operation, can be considered as a possible means to reduce carbon dioxide emission and to increase fuel efficiency. In this study, a composite air spoiler was designed and tested by static and repeated loads. The Green Water Pressure of 0.1 MPa a ship experiences during operation was perpendicularly applied to the air spoiler. Air spoiler was manufactured with sandwich panel which has glass fabric face and balsa core. Multiple sandwich panels were assembled to steel frame by bolt joint. The joint was designed to have bearing failure and examined by static and fatigue tests. Tests showed that the designed joint has enough margin of safety to endure joint failure. The developed sandwich panel to air spoiler is planned to be applied to a large scale commercial ship.

A Research on the Creation of Floral Art Works as Public Installation : The Case of a Vertical Mini-Garden in Island Type Bus Stop (공공설치용 화예작품 창작에 관한 연구 -도심 중앙차로 버스정류장의 수직형 미니정원을 중심으로-)

  • This study is about the development and creation of the public installation to enhance the contact chances with plants for the city dwellers who are living in the environment of limited green space. The researcher tried to create a floral art work at an island type bus stop on the central bus only lane, where the passers-by can enjoy the contact with plants for a second on that crowded space. The researcher made the study of various related cased world-widely and existing island type bus stop on the central bus only lane. The researcher also made pre-creation study of applicable forms and elements, and developed the prototype of the installation on the base of that study. Therefore the researcher created 1 installable screen type floral art work offering abundant contact experience to the viewers which has multiple shelves in the vertical frame to fit and be substituted by the designated wall unit of the island type bus stop structure. Also the researcher developed the prototypes of 2 small interior pieces. The meaning of this study can be found in the contribution of increasing visual experiencing rate of green space and chances of experiencing plants to create positive psychological, emotional effects by developing floral art works to be installed at crowded urban public space.

  • In this paper, we proposed "Future Design and Success Engineering in Global Society by Self-Innovation" (an abbreviated name: FDSE). We started to make a small experiment between Korea Maritime University and Yamagata University at the current year. After inspecting the results, especially, successful and unsuccessful points in our lecture, we have to develop the strong points and amend the week points, through making the analysis of success-failure, which is his favorite management method. We have to level up the contents of our work in the future. The students for this lecture would learn from experiences of "the common knowledge (common field of discussion)", held common languages, common thought, common system. They filled up the blanks of "The Matrix Frame" which is mentioned in the previous report, and they expressed their own way of life (work, study). As a matter of course, their personal private affairs must not be made public. It is quite happy that the cultural, economical exchanges between Korea and Japan are now in full flourish and people do a lot of traveling each other. However, if this project will start on the educational global standard, we have a world-wide significance. Moreover, we can promote goodwill between Korea and Japan, and also we can contribute to a better international understanding and the peace of the world by students in both Universities. We hope that candidates for this lecture will have a rounded personality and be a life worthy of human being for realizing Self-Actualization in "Motivation and Personality" by Abraham Maslow.

Application of IUCN Category Regarding the Designation of Overlapping Protected Areas (중복지정된 보호지역을 고려하기 위한 IUCN 카테고리 적용)

  • The purpose of this study is to seek the application of IUCN categories of overlapping protected areas which is legally designated in South Korea. Different government departments in South Korea have managed and designated as protected areas. However, the protected areas due to different management agencies can be confused with restricting behaviors and supporting residents. The IUCN presents the reasonal standardization classifying the protected areas which could be applied all over the world. Six categories issued by the IUCN could be applied to deal with the problems of the overlapping protected areas. We suggested the application of the IUCN categories compared with legal frame in South Korea. Most areas are overlapped in designation, but the areas are important for ecology and landscape. Moreover, each protected areas in South Korea have zone districts. Comprehensively considered all these things, we made rationale matrix correlated with the IUCN categories and the zone districts of the protected areas in South Korea. For the result of this study, this matrix could be helped to the application of the IUCN categories in domestic protected areas. Although the protected areas has been recognized as regulatory regions, it is expected to expand and sustain the areas based on the matrix.
