Gallic Acid Enhancement of Gold Nanoparticle Anticancer Activity in Cervical Cancer Cells

  • Cervical cancer (CxCa) is the most common cancer in women and a prominent cause of cancer mortality worldwide. The primary cause of CxCa is human papillomavirus (HPV). Radiation therapy and chemotherapy have been used as standard treatments, but they have undesirable side effects for patients. It was reported that gallic acid has antioxidant, antimicrobial, and anticancer activities. Gold nanoparticles are currently being used in medicine as biosensors and drug delivery agents. This study aimed to develop a drug delivery agent using gold nanoparticles conjugated with gallic acid. The study was performed in uninfected (C33A) cervical cancer cells, cervical cancer cells infected with HPV type 16 (CaSki) or 18 (HeLa), and normal Vero kidney cells. The results showed that GA inhibited the proliferation of cancer cells by inducing apoptosis. To enhance the efficacy of this anticancer activity, 15-nm spherical gold nanoparticles (GNPs) were used to deliver GA to cancer cells. The GNPs-GA complex had a reduced ability compared to unmodified GA to inhibit the growth of CxCa cells. It was interesting that high-concentration ($150{\mu}M$) GNPs-GA was not toxic to normal cells, whereas GA alone was cytotoxic. In conclusion, GNPs-GA could inhibit CxCa cell proliferation less efficiently than GA, but it was not cytotoxic to normal cells. Thus, gold nanoparticles have the potential to be used as phytochemical delivery agents for alternative cancer treatment to reduce the side effects of radiotherapy and chemotherapy.

석암리 9호분 출토 금제띠고리의 제작 방법 고찰 (A Study of Metalworking Techniques Seen in the Gold Buckle from Seogam-ri Tomb No. 9)

  • 석암리 9호분 출토 금제띠고리(국보 제189호)는 한반도에서 출토된 가장 이른 시기의 누금세공품으로 금 순도 약 22.8K의 금판에 타출기법으로 일곱 마리의 용이 표현되어 있다. 그리고 약 23.8K의 금선과 금알갱이로 용의 몸통 및 윤곽선, 테두리 등이 장식되어 있다. 이 금제띠고리 곡선면의 일부에는 순도 23.8K의 금판이 덧대어져 있는데, 제작 당시 혹은 그 이후에 발생된 결함을 보완하기 위한 것으로 추정되었다. 금제띠고리에 장식된 용의 코를 표현하는데 사용된 금선은 순도가 23.3K로 약간 낮았다. 이는 동그랗게 감아 말아 만든 용의 코 모양이 잘 유지될 수 있도록 순도가 약간 낮은 금선을 사용하여 경도를 조금 높인 것으로 추정된다. 금알갱이는 크기에 따라 대, 중, 소로 구분되었으며 구리 확산법을 이용하여 접합한 것으로 추정되었다. 용의 이마 및 몸통을 장식하기 위하여 청색 터키석을 감입하였고, 일곱 마리 용의 눈 부분에는 적색 안료인 진사/주(HgS)가 감입되었음을 확인하였다.

백제 관모에 사용된 직물 연구 (A Study of the Fabrics Used for the Official Hats in Baekje Dynasty)

  • The kind and the use of the fabrics for crown manufacture in the Baekje period has been studied by characterizing the imprinted fabrics on the crowns and the diadem ornaments from the old tomb. The contact region with the skin inside of the crown and the region between the bark of white birch and the gilt bronze openwork plates contained fabrics. The fabrics used in the gilt-bronze crown were all plain weave silk except that of Yongwonri tomb where loosely woven thin tabby was used. There have been 4-types of iron framed diadem of the Baekje, which comprise the inverted triangle-shaped diadem only with iron frame, the diadem with gold plate ornament in the iron frame, the diadem decorated with mica plate and gold plate, and the diadem with silver ornament in the iron frame. The fabrics used in the triangle shaped iron frame diadem include plain weave silk, irregular plain woven silk, thin tabby, complex silk gauze, twill weave on plain ground, and warp-faced compound weave. The iron frames were wrapped with the fabrics from one layer up to three layers, and the iron diadem was covered with one later of loosely woven textile such as irregular plain woven silk, thin tabby, and complex silk gauze. But in case of decorating the iron diadem with gold Plate ornaments, multiple layers of fabric were used to sustain the weight of the ornaments. The fabrics in the iron diadem frame were sewed with running stitch, overedge stitch or hemming stitch, diagonal hemming stitch, half back stitch), and overcast stitch.

고정화법을 달리하여 제조한 압전류적 항체 센서에 의한 Salmonella spp.의 신속 검출 (Rapid Detection of Salmonella spp. by Antibody-Immobilized Piezoelectric Crystal Biosensor)

  • Salmonella spp.의 신속한 검출을 위하여 엷은 박막형태의 수정결정을 사용하는 압전류적(piesoelectric) 항체센서 시스템을 개발하고 증류수, 완충용액, 식염용액 등의 여러 매질 중에서 보여주는 진동 특성을 검토하였다. Salmonella spp. 균 구조항원(Common structural antigen)에 대한 항체를 수정결정에 PEI pre-coating, BSA 가교화, 3-APTES silanizaition, protein A와 DTBP thiolation의 5가지 방법에 의해 고정화한 후 항체 센서의 안정성을 살펴보았다. Salmonella 균을 주입하였을 때 Salmonella 균과 수정 결정에 고정화한 항체와의 결함반응에 의해 수정결정의 질량증가와 이에 따른 진동수 감소가 나타났다. 고정화방법 중 protein A와 DTBP를 이용하여 고정화하는 방법이 센서반응을 가장 안정적이고 재현성 있게 나타내줌을 알 수 있었다. $7.45{\times}10^{7}\;CFU/ml$의 Salmonella 균을 반응 cell 내에 주입하였을 때 protein A를 이용한 고정화의 경우 80Hz, DTBP를 이용한 고정화의 경우 283 Hz의 진동수 감소가 나타났으며, 압전류적 항체센서를 이용할 경우 40분 이내에 Salmonella spp.의 검출이 가능하였다.

QDs를 이용한 키토산-골드와 키토산-실버 나노약물전달체 제조 (Preparation of Chitosan-Gold and Chitosan-Silver Nanodrug Carrier Using QDs)

  • 본 연구에서는 최근 많은 분야에서 응용되고 있는 형광물질인 양자점을 생명고분자인 키토산과 반응시켜 얻은 나노입자와 금속성 골드 나노입자, 그리고 실버 나노입자로 외부를 코팅하여 나노약물 전달체를 얻을 수 있었다. 키토산은 생체고분자로써 무독성이며 인체적합성 고분자이다. 양자점은 2~10 nm의 크기를 가지는 반도체성 나노입자이다. 양자점은 생명분자나 생명단백질의 비슷한 크기를 갖으며, 그 크기에 따라 알맞은 가시광선 영역의 빛을 발산할 수 있도록 조절 가능하므로, 세포 바이오 마킹, 약물전달체 등에 효과적으로 쓰일 수 있다. 따라서 키토산 나노입자 말단의 아민기와 양자점의 카르복실기가 아미드결합을 형성하여 반응하게 조절하였다. 양자점의 독성을 완화시키기 위해 코팅재료로 사용된 금속성 나노입자 중 골드나노입자는 약 5~10 nm의 크기를 가지고 있고, 인체에 무해하고 음전하를 띄어서 양전하를 띈 고분자와 쉽게 복합체를 형성할 수 있는 장점이 있다. 향균성으로 잘 알려진 실버나노입자는 약 5 nm의 크기를 가지고 있고, 은 나노입자로 코팅을 하면 미생물 감염을 미리 방지 할 수 있는 장점을 가지고 있다. 본 연구에서 만들어진 QDs-키토산-골드 & QDs-키토산-실버 나노쉘의 입자크기는 약 100 nm의 크기를 갖었으며, 목적하는 바 형광특성을 잘 보여주고 있었다. 이러한 입자들은 정전기적 상호작용에 의하여 각각 골드나노입자와 실버나노입자로 코팅되어 나노 약물전달체로 완성할 수 있었다.

면역크로마토그라피 기법을 이용한 Salmonella 속균 신속 검출킷트 개발 (Development of the rapid detection kit for Salmonella spp. using immunochromatographic assay)

  • An immunochromatographic (IC) strip for the rapid detection of Salmonella spp. in the enriched sample was developed. Affinity purified Salmonella polyclonal antibody was conjugated with 40 nm colloidal gold particles which were prepared by citrate method in our laboratory. The antigen-antibody-gold complex was captured by Salmonella antibody attached to test line of nitrocellulose membrane during the capillary migration of sample. Specificity of the IC strip was calculated to be 100% (12/12) and sensitivity was 97.6% (41/42) in the test with pure cultured bacteria. Salmonella was artificially inoculated into raw pork macerated with enrichment broth. And then it was 10-fold diluted from $5.2{\times}10^{8}CFU/ml$ to 5.2 CFU/ml. The IC strip could detect $5.2{\times}10^{6}CFU/ml$ before enrichment. However, the lowest limit of detection was 5.2 CFU/ml after overnight incubation. The results indicated that the IC assay was a rapid, economical and simple method with high specificity and sensitivity for the detection of Salmonella spp. without using any equipment.

Evaluation of the cytotoxicity of gold nanoparticle-quercetin complex and its potential as a drug delivery vesicle

  • Recently, conjugates of medicinal herb-derived bioflavonoids, such as quercetin, and gold nanoparticles (GNPs) have gained attention as targeted drug delivery systems. In the present study, because quercetin is an important flavonoid with anti-cancer, anti-inflammatory, and anti-oxidant properties, GNP-quercetin complexes (GNPQs) were synthesized to investigate possible adverse effects such as cytotoxicity. We found that while quercetin was cytotoxic, GNPQs were not cytotoxic towards the RAW 264.7 and THP-1 cell lines. Therefore, GNPQs may serve as a potential drug delivery system for cancer treatment.

  • Small-film-type ion sensors are garnering considerable interest in the fields of wearable healthcare and home-based monitoring systems. The performance of these sensors primarily relies on electrode capacitance, often employing nanocomposite materials composed of nano- and sub-micrometer particles. Traditional techniques for enhancing capacitance involve the creation of nanoparticles on film electrodes, which require cost-intensive and complex chemical synthesis processes, followed by additional coating optimization. In this study, we introduce a simple one-step electrochemical method for fabricating gold nanoparticles on a carbon nanotube (Au NP-CNT) electrode surface through cyclic voltammetry deposition. Furthermore, we assess the improvement in capacitance by distinguishing between the electrical double-layer capacitance and diffusion-controlled capacitance, thereby clarifying the principles underpinning the material design. The Au NP-CNT electrode maintains its stability and sensitivity for up to 50 d, signifying its potential for advanced ion sensing. Additionally, integration with a mobile wireless data system highlights the versatility of the sensor for health applications.

Replica Correlation-Based Synchronization with Low Complexity and Frequency Offset Immunity

  • This paper investigates the multifarious nature of the long-term evolution (LTE) scheme and that of the modified LTE scheme for symbol timing synchronization (STS). This investigation allows us to propose a new replica correlation-based STS scheme to overcome the inherent weaknesses of the other two schemes. The proposed STS signal combines a gold sequence and a half sine wave in the time domain, whereas conventional STS signals specify either binary sequences or complex sequences in the time domain or in the frequency domain. In the proposed scheme, a sufficient correlation property is realized by the gold sequence, and robustness against the frequency offset (FO) is achieved through the sine wave. Compared to the existing LTE-related schemes, the proposed scheme can better achieve immunity to FO and reduction in detector complexity, as well as a low peak-to-average power ratio and a low detection error rate. Performance evaluations through analysis and simulation are provided in the paper to demonstrate these attributes.
